A MINIATURIZED SYSTEM FOR RAPID AND QUANTITATIVE DETERMINATION OF A COCAINE METABOLITE BY A HOMOGENEOUS ENZYME IMMUNOASSAY

Abstract:Conventionally, drugs of abuse tests are performed on bulky and costly analytical instruments such as a spectrophotometer or chemistry analyzer, requiring large volumes of sample and reagents, normally more than 200 µL. This article presents a simple, low-cost, sensitive, and miniaturized light absorption detection device to conduct a homogenous immunoassay assay for detecting drugs of abuse. The concentrations of the target drug in samples are determined quantitatively by measuring the relative light intensity transmission of the reaction mixture. In this method, the reaction mixture is contained in a micro-well in a (Polydimethylsiloxane)-glass chip and exposed to UV light. The relative light intensity transmission is measured by a photo-detector. The desired temperature for incubation is provided by a thin-film heater under the chip. The automatic control of the heater, light source, data acquisition, and processing are achieved by a self-coded program. To verify the feasibility and performance of this device, a commercial homogenous assay kit for a cocaine metabolite was used. The effects of multiple parameters on the system's sensitivity were studied. The results show that the detection can be completed within 5 minutes with a ten-time smaller volume (∼22µL) of the reaction mixture (sample plus the reagents) than that of the conventional method. A detection limit of 50 ng/mL was achieved, comparable to that by a standard lab method with the same assay kit. This portable detection device is promising for on-site testing of drugs of abuse.
