Abstract:A new method based on the subtraction of images is proposed to enhance the resolution and contrast in CW-STED microscopy. It is realized by the subtraction of a confocal image from a STED image. Detailed theoretical analyses and a series of simulation tests are performed. The validity of this subtraction method in practice is demonstrated by experiments on both fluorescent nanoparticles and microtubule networks in which a better resolution and contrast than the STED image is achieved. Subtractive parameters are discussed in detail and a way of choosing the optimal value is proposed. Compared with other resolution enhancement concepts, this method is much simpler and free from using high intensity lasers.
Axial Resolution Enhancement for Light Sheet Fluorescence Microscopy Via Using the Subtraction Method

Abstract:Light sheet fluorescence microscopy is an imaging method in which the sample is illuminated from the side by a wide and relatively thin light sheet. This method minimizes the out-of-focus excitation, provides a good balance between spatial resolution and temporal resolution, and mitigates photobleaching and photodamage. Unfortunately, the axial resolution of light sheet fluorescence microscopy is much lower than lateral resolution, which limits the achievement of isotropic imaging. We propose to combine the subtraction method with light sheet fluorescence microscopy to improve the axial resolution. The experimental results demonstrate that the proposed method can produce higher axial resolution than that of conventional light sheet fluorescence microscopy. Abstract:Applying image subtraction strategy together with time-gated detection, we can reveal finer details in a fixed cell sample using moderate light intensities. The subtractive imaging is obtained from the subtraction of a suitably weighted short lifetime intensity-image from a long lifetime one, while the weight coefficient can be selected using the analysis of an optical transfer function. Compared with other subtractive methods, ours is regarded as an optimized variation of gated stimulated emission depletion microscopy and can be implemented in a simpler manner, providing lateral super-resolution, and it is theoretically possible to further expand its application scope. Abstract:Light sheet fluorescence microscopy (LSFM) with fluorescence emission difference (FED) obtains axial resolution-enhanced images by the subtraction of signals excited by different light sheets, Gaussian light sheet and negative light sheet. Negative values are inevitable in FED LSFM with conventional subtraction coefficient, due to mismatches in the outer contours of the raw signals excited by different light sheets, causing image distortion. In this paper, we propose a method based on tangent-function subtraction coefficient to reduce image distortion. This tangent-function subtraction coefficient is introduced to minimize image distortion without lowering the axial resolution. The value of the variable subtraction coefficient is obtained from the intensity value of raw signals. We performed experiments using fluorescent beads with a diameter of 1 micron and compared the subtraction results with constant and tangent-function subtraction coefficient. The experimental results demonstrated that the proposed tangent-function subtraction coefficient has an advantage over the conventional constant coefficient.

Abstract:A method based on the subtraction of images taken under different pinhole sizes is provided to improve the lateral resolution and SNR in confocal microscopy. Theoretical analyses have been performed and a notable enhancement in lateral resolution and SNR is shown by experimental results. Subtraction parameter and pinhole size have been discussed in detail. Compared with other resolution enhancement concepts, this method is much simpler and cheaper without using strong lasers. And it well suits for real time applications. It can be easily applied to a general confocal microscope and suitable for both fluorescence and non-fluorescence sample.

Abstract:We propose a novel method to improve the lateral resolution of a general confocal laser scanning microscopy (CLSM) by 10–35%, which can be readily realized by manipulating the shape of the excitation focal spot to a doughnut one. As the contrast of image is reversed synchronously, the raw data is further handled by the deconvolution algorithm to convert the doughnut point spread function (PSF) to a peak-centered distribution, so that a more intuitive vision will be given. Depending on the strategies like saturation excitation or doughnut size shrinkage, the resolution can be further optimized. This setup is valid for diverse systems and may find use in a variety of applications.

Abstract:There is always a tradeoff between the resolution and the signal-to-noise ratio (SNR) in confocal microscopy. In particular, the pinhole size is very important for maintaining a balance between them. In this paper, we propose a method for improving the lateral resolution and image SNR in confocal microscopy without making any changes to the hardware. By using the fluorescence emission difference (FED) approach, we divide the images acquired by different pinhole sizes into one image acquired by the central pinhole and several images acquired by ring-shaped pinholes. Then, they are added together with the deconvolution method. Simulation and experimental results for fluorescent particles and cells show that our method can achieve a far better resolution than a large pinhole and a higher SNR than a small pinhole. Moreover, our method can improve the performance of classic confocal laser scanning microscopy (CLSM) to a certain extent, especially CLSM with a continuously variable pinhole.

Abstract:Fluorescence emission difference microscopy (FED) obtains resolution-enhanced images by subtracting acquired solid and doughnut confocal images. Because of the mismatch of the outer contours of the two subtraction components, negative values are inevitable in the conventional FED method, giving rise to deformations. In this study, by using a saturation effect, we obtain imaging results with a profile-extended solid and center-shrunken doughnut point spread function. Owing to the nonlinear effect, two better-matched saturated images not only eliminate the deformations, but also enhance the resolving ability and signal to noise ratio compared to conventional FED. Simulations based on the saturated model of rhodamine 6G, as well as experiments on biological samples, are presented to verify the capability of the proposed concept, while experimental results show the unprecedented resolving ability of the saturated FED method.

Abstract:Fluorescence emission difference microscopy(FED) has been demonstrated to be capable of breaking the diffraction limit through the difference of two different confocal images. Here we propose and build a novel dual-color FED system that can provide fluorescence imaging with sub-diffraction resolution. The enhancement in the lateral resolution is verified theoretically and experimentally. Compared to standard STED system, FED enjoys the advantages of lower illumination power and higher flexibility. In addition, the dual-color configuration enables the presented system to be suitable for biological investigations.

Abstract:We propose a method for eliminating the deformations in fluorescence emission difference microscopy (FED). Due to excessive subtraction, negative values are inevitable in the original FED method, giving rise to deformations. We propose modulating the beam to generate an extended solid focal spot and a hollow focal spot. Negative image values can be avoided by using these two types of excitation spots in FED imaging. Hence, deformations are eliminated, and the signal-to-noise ratio is improved. In deformation-free imaging, the resolution is higher than that of confocal imaging by 32%. Compared to standard FED imaging with the same level of deformations, our method provides superior resolution. (C) 2014 Optical Society of America

Abstract:In this paper, we propose and demonstrate an improved fluorescence emission difference microscopy (FED) method, exploiting programmable phase modulation for image enhancement. The main novelty of the proposed approach lies in the matched size and intensity of two excitation spots. The proposed method improves the FED performance on image quality via artefact elimination. We demonstrate the feasibility of this method through theoretical studies and experimental tests. The experimental results of nanobeads and cells validate the practical performance of this method, which can enable reliable observations at superresolution in biomedical studies. LAY DESCRIPTION: In this paper, we propose a method to improve the imaging quality of regular fluorescence emission difference (FED) microscopy. In regular FED imaging, a solid and a doughnut excitation beam are successively used to acquire two images which are then subtracted with each other to improve the resolution of confocal microscopy. The doughnut beam can be generated by modulating the excitation beam with a vortex phase mask. Note that both of the excitation beam and the vortex phase mask must have the same handed direction in regular FED microscopy. However, some negative values may be produced and some information may be lost due to the subtraction process in regular FED imaging, which is mainly caused by the mismatched size and intensity of these two excitation spots. To address this issue, we propose conjugated FED (cFED) microscopy which additionally uses a conjugated vortex phase mask to modulate the solid beam to extend its focal spot size to be matched with the doughnut spot, which means the handed direction of the solid beam and the vortex phase mask is different. Besides, in order not to damage the resolution, the doughnut beam needs to be saturated to some degree. The experiment results show that, at the same resolution level, the negative values and the information loss in cFED image are all less than that of regular FED image.

Abstract:We propose a method theoretically to break the diffraction limit and to improve the resolution in all three dimensions for fluorescence emission difference microscopy. We produce two kinds of hollow focal spot by phase modulation. By incoherent superposition, these two kinds of focal spot yield a 3D hollow focal spot. The optimal proportion of these two kinds of spot is given in the paper. By employing 3D hollow focal spot, super-resolution image can be yielded by means of fluorescence emission difference microscopy, with resolution enhanced both laterally and axially. According to computation result, size of point spread function of three-dimensional super-resolution imaging is reduced by about 40% in all three spatial directions with respect to confocal imaging.

Abstract:Point spread function (PSF) engineering-based methods to enhance resolution and contrast of optical microscopes have experienced great achievements in the last decades. These techniques include: stimulated emission depletion (STED), time-gated STED (g-STED), ground-state depletion microscopy (GSD), difference confocal microscopy, fluorescence emission difference microscopy (FED), switching laser mode (SLAM), virtual adaptable aperture system (VAAS), etc. Each affords unique strengths in resolution, contrast, speed and expenses. We explored how PSF engineering generally could be used to break the diffraction limitation, and concluded that the common target of PSF engineering-based methods is to get a sharper PSF. According to their common or distinctive principles to reshape the PSF, we divided all these methods into three categories, nonlinear PSF engineering, linear PSF engineering, and linear-based nonlinear PSF engineering and expounded these methods in classification. Nonlinear effect and linear subtraction is the core techniques described in this paper from the perspective of PSF reconstruction. By comparison, we emphasized each method’s strengths, weaknesses and biologic applications. In the end, we promote an expectation of prospective developing trend for PSF engineering.

Abstract:We propose a single molecule localization method which takes advantage of stochastic photobleaching to improve the resolution of confocal fluorescence microscopy. By detecting the stochastic intensity loss of fluorophores, each fluorophore in the field can be localized. When all locations are known, a sub-diffraction image can be retrieved through single molecule localization algorithms. A confocal scheme is used to record the bleaching process of the sample. Each fluorophore can be localized from the recorded streaming followed by image subtraction. Compared with other single molecule localization concepts such as stochastic optical reconstruction microscopy (STORM) and photoactivated localization microscopy (PALM), this method does not require a laser cycling equipment and the pixel size is no longer limited by the size of CCD. This technique works well with common fluorescent dyes and does not require the use of engineered photoactivatable proteins or photoswitchable synthetic dye pairs.

Abstract:We report on the design and implementation of an integrated dual-color stimulated emission depletion (STED) microscopy and fluorescence emission difference (FED) microscopy. This highly integrated system combines the respective advantages of STED and FED microscopies, enabling acquisition of super-resolution STED images with specific fluorophores and high laser power, or resolution-enhanced FED imaging for ordinary fluorescent dyes and other materials with moderate demand for laser power. The functional integration enables researchers to select the more appropriate microscopy according to practical scenario and requirements, giving full play to their respective strengths and satisfying the imaging demands maximally. The system's resolution is characterized by dual-color imaging of fluorescent particles. Its practical applicability is verified through imaging various kinds of samples, including several fixed cells stained with different organic dyes and a given organic material-organometallic halide perovskite.

Abstract:We propose a novel imaging method that enables the enhancement of three-dimensional resolution of confocal microscopy significantly and achieve experimentally a new fluorescence emission difference method for the first time, based on the parallel detection with a detector array. Following the principles of photon reassignment in image scanning microscopy, images captured by the detector array were arranged. And by selecting appropriate reassign patterns, the imaging result with enhanced resolution can be achieved with the method of fluorescence emission difference. Two specific methods are proposed in this paper, showing that the difference between an image scanning microscopy image and a confocal image will achieve an improvement of transverse resolution by approximately 43% compared with that in confocal microscopy, and the axial resolution can also be enhanced by at least 22% experimentally and 35% theoretically. Moreover, the methods presented in this paper can improve the lateral resolution by around 10% than fluorescence emission difference and 15% than Airyscan. The mechanism of our methods is verified by numerical simulations and experimental results, and it has significant potential in biomedical applications.

Abstract:Fluorescence emission difference microscopy (FED) has been proved to be an effective method to break the diffraction barrier which had restricted the spatial resolution of optical microscopy in the far-field for a long time. In this paper, we theoretically demonstrate that the attainable resolution of FED can be further enhanced by using beam modulation. By applying beam shape modulating, polarization state modulating, phase encoding and beam blocking to the illumination beams in the process of FED imaging, we optimize the size of the corresponding PSFs and thus improve the resolving ability of FED. The simulation tests demonstrate that in the optimal case after beam modulation, the resolution of FED can be enhanced by a factor of 26% compared with a conventional one.
