Abstract:The sustained release of heparin in sufficient amounts and over long time is a challenge to drive the development of functional materials. In this paper SBA-15 mesoporous silica is selected in the search for a favorable morphology and optimized surface state for the sustained release of heparin. In situ carbonization of the template in the as-synthesized sample enables SBA-15 to possess narrowed channels with rougher surfaces, while modification with (aminopropyl)triethoxysilane (APTES) through a one-pot synthesis offers SBA-15 with positive charges to attract heparin through electro-static interactions. The structure of modified SBA-15 samples is assessed with XRD (powder X-ray diffraction analysis), nitrogen adsorption-desorption and electron microscopy techniques, and their performance is evaluated in adsorption and release of heparin. These modifications improve the heparin adsorption in SBA-15 and thus promote its sustained release, prolonging the release-equilibrium time up to 60 days. Among them, the SBA-15 sample modified with APTES can trap three times as much heparin as the parent SBA-15, and the release ratio is elevated to 80% (that of SBA-15 is 38%), realizing the best performance of controlling heparin release to date.
Multiple functionalization of SBA-15 mesoporous silica in one-pot: Fabricating an aluminum-containing plugged composite for sustained heparin release

Abstract:To fabricate an efficient releaser of heparin, which is an extensively used anticoagulant, aluminum containing plugged mesoporous silica SBA-15 is prepared using a one-pot synthesis process. A suitable aluminum additive in the starting mixture enables plugs to be formed inside hexagonal channels and Al species to be incorporated into the mesostructure, evidence for which is provided by powder X-ray diffraction analysis (XRD), transmission electron microscopy (TEM), N-2 sorption measurements and Al-27 MAS NMR tests. Incorporation of aluminum in SBA-15 reduces the negative charge (Zeta-potential) and tailors the surface roughness of the channel through forming plugs, and the former significantly improves heparin adsorption while the latter retards heparin release. As a result, these modified composites adsorb 2-4 times more heparin than parent SBA-15 and release 60-130% more of the drug over several weeks; the sample possessing both an Al-component and plug structure can sustain release of heparin for 6 weeks, demonstrating the increased efficiency of multiply functionalized SBA-15 in the controllable release of heparin and offering a valuable clue for design of novel drug releasers.
Porous Polymers as Universal Reversal Agents for Heparin Anticoagulants through an Inclusion–Sequestration Mechanism

Abstract:An inclusion–sequestration strategy is developed by making use of multicationic supramolecular organic frameworks and porous organic polymers as porous antidotes to neutralize heparins. In vitro and in vivo experiments and biocompatibility evaluations confirm that the porous polymers can realize rapid, safe, and universal reversal of heparin‐based anticoagulants, superior to that of clinically used protamine. Heparins are widely used anticoagulants for surgical procedures and extracorporeal therapies. However, all of them have bleeding risks. Protamine sulfate, the only clinically approved antidote for unfractionated heparin (UFH), has adverse effects. Moreover, protamine can only partially neutralize low‐molecular‐weight heparins (LMWHs) and is not effective for fondaparinux. Here, an inclusion–sequestration strategy for efficient neutralization of heparin anticoagulants by cationic porous supramolecular organic frameworks (SOFs) and porous organic polymers (POPs) is reported. Isothermal titration calorimetric and fluorescence experiments show strong binding affinities of these porous polymers toward heparins, whereas dynamic light scattering and zeta potential analysis confirm that the heparin sequences are adsorbed into the interior of the porous hosts. Activated partial thromboplastin time, anti‐FXa, and thromboelastography assays indicate that their neutralization efficacies are higher than or as high as that of protamine for UFH and generally superior to protamine for LMWHs and fondaparinux, which is further confirmed by tail‐transection model in mice and ex vivo aPTT or anti‐FXa analysis in rats. Acute toxicity evaluations reveal that one of the SOFs displays outstanding biocompatibility. This work suggests that porous polymers can supply safe and rapid reversal of clinically used heparins, as protamine surrogates, providing an improved approach for their neutralization.

Abstract:We report on a novel, efficient, and one-step approach to prepare single-layer reduced graphene oxide (RGO) suspensions and films using hydroxylamine hydrochloride. The effective chemical reduction of GO was evidenced by a significant increase in the C/O ratio and five orders of magnitude decrease in the GO resistance. Field-effect transistor gas sensors were fabricated using as-produced RGO sheets and the resulting sensor exhibited a fast response and a high sensitivity to low-concentration target gases at room temperature.

Abstract:Commercial heparin production, a widely applied anticoagulant, requires a series of time-consuming and costly separation units to achieve isolation and purification and the challenging removal of macromolecular impurities, such as proteins. We report a novel separation strategy for the first time that can capture heparin while simultaneously recovering proteins through an efficient one-step separation. Novel adsorptive membranes were prepared by grafting polymer brushes of (3-acrylamidopropyl) trimethylammonium chloride (APTMAC) onto the surface of poly(vinylidene fluoride) (PVDF) membrane via atom transfer radical polymerization (ATRP) by controlling the polymerization time. The prepared membrane exhibited excellent hydrophilicity with water contact angle of 26.57° and adsorption capacity for heparin (598.6 μg/cm 2 ). XPS, FTIR measure and density functional theory (DFT) simulation demonstrated that the adsorptive membrane could effectively adsorb heparin on the surface via the electrostatic interaction between -N(CH 3 ) 3 + groups of the grafted APTMAC on the membranes and the reactive functional groups (OSO 3 − , NHSO 3 − , COO − ) of heparin. Membrane filtration results showed that the adsorptive membranes created one-step separations that could selectively capture heparin via electrostatic interaction while rejecting and concentrating bovine serum albumin (BSA) because of size sieving and hydrated layer repulsion. Besides, the purity of crude heparin increased from 60 % to 93 % through using adsorptive membrane filtration. Moreover, the heparin adsorption capacity remained at 82.3 % of its original capacity after six cycles. This study provides a novel strategy for efficient, economic and selective multi-functional separations for heparin and its similar biomedical medicines.

Abstract:In this paper, epigallocatechin gallate (EGCG) was used as a green reductant both for the fabrication of soluble reduced graphene oxide (rGO) and the synthesis of Au nanoparticles/rGO nanocomposite. Fourier transform infrared (FTIR) spectra confirmed the efficient removal of the oxygen-containing groups in graphene oxide (GO) through the reduction act of EGCG. Au nanoparticles (AuNPs) were anchored onto the rGO sheets by heating the mixed solution of rGO and chloroauric acid at 65 degrees C using EGCG as reductant. Transmission electron microscopy (TEM), atomic force microscope (AFM) and X-ray photoelectron spectroscopy (XPS) were employed to characterize the resulting nanocomposite. Due to the chelating effect of polyhydroxy EGCG, AuNPs with diameters of similar to 20-50 nm were stably decorated onto both sides of the rGO sheets. Because this reduction method avoids the use of toxic reagents, AuNPs/rGO nanocomposite would be eco-friendly, and it might be useful not only for electronic devices but also for biocompatible materials in the future applications.

Abstract:The ability to sieve ions through nanopores with high throughput has significant importance in seawater desalination and other separation applications. In this study, a plasma etching process has been demonstrated to be an efficient way to produce high-density nanopores on graphene membranes with tunable size in the sub-nanometer range. Besides the pore size, the nanopore density is also controllable through adjusting the exposure time of the sample to argon or oxygen plasma. The plasma-treated graphene membranes can selectively transport protons, Na+ and Cl- ions. Density function theory calculations uncover that the sp3 and vacancy-type defects construct different energy barriers for different ions, which allow the defected graphene membrane to selectively transport ions. Our study indicates that oxygen plasma etching can be used as a very convenient and efficient method for fabricating a monolayer filtration graphene membrane with tunable sub-nanometer pores.

Abstract:Here, a new hierarchical porous carbon was first prepared from sodium lignosulphonate (SLS), an industrial by-product, via a confined carbonization and alkali activation methods using graphene oxide (GO) as template, and was used for ciprofloxacin (CIP) removal. The addition of GO nanosheets remarkably affected the morphology and pore structure. GLHPC-1, obtained at GO/SLS weight ratio of 1:100, possess a largest specific surface area of 3223m2g−1 and pore volume of 2.275cm3g−1, and thus exhibited an ultrahigh adsorption of 980.4mgg−1 at 318K and fast adsorption rate, as well as a good recycle ability in practice.

Abstract:Mesoporous silica MCM-41 and SBA-15 were chosen to study the adsorption and release of bulky biomolecule heparin, in order to develop new heparin controlled delivery system and expand the application of mesoporous materials in life science. To explore how the structure of support such as pore size and surface state affects the accommodation and release of heparin, we used decane as swelling agent to enlarge pores of MCM-41, introduced amino groups for improving the biocompatibility of support, and controllably retained templates in the as-synthesized sample. The influence of modification on the structure of samples was investigated by XRD and N(2) adsorption-desorption, whereas their performance of adsorbing and releasing heparin was assessed with that of toluidine blue method. Both enlarged pore and organic modification significantly promoted the adsorption and prolonged the release of heparin in MCM-41, and the release was characterized with a three-stage release model. The mechanism of heparin release from mesoporous material was studied by fitting the release profiles to the theoretical equation. As expected, some mesoporous composites could release heparin in the long term with tuned dosage.

Abstract:Motivated by adhesive proteins in mussels, strategies using dopamine to modified surface have become particularly attractive. In the present work, we developed a novel and convenient method to modify the biodegradable Fe plates with heparin. Iron was first treated by a facile one-step pH-induced polymerization of dopamine, and then a high density heparin was successfully grafted onto the surface via coupling with polydopamine (PDA) active layer. Heparin immobilization contributed much longer blood clotting coagulation time than the pure Fe sample, and hence reduced the risk of thrombosis. Cell viability tests suggested that the heparin modified Fe plates were more favorable to the proliferation of ECV304 cells. In summary, the heparin modified Fe plates with good anti-thrombus properties and inhibiting the proliferation of VSMC cells provide great prospects for biodegradable iron.
Development of Substrate-Independent Heparin Coating to Mitigate Surface-Induced Thrombogenesis: Efficacy and Mechanism

Abstract:Heparin coatings are widely applied on blood-contact materials to reduce the use of anticoagulants during blood treatment. However, the previous heparin coatings formed via covalent binding or electrostatic bonding commonly require complex surface premodification, and the blood coagulation pathway was significantly inhibited to potentially increase the bleeding risk. This contradicts the intended purpose and deviates from the anticoagulation mechanism of the heparin coatings. Herein, we present a facile and substrate-independent coating, achieved through the co-deposition of dopamine/chitosan followed by electrostatic interaction between heparin and the immobilized chitosan, which could be prepared within 1 hour. This coating prolonged the plasma re-calcification time (PRT) to over 60 minutes, effectively preventing surface-induced thrombosis. Favorable hemocompatibility was reflected in a hemolysis ratio of less than 2%, low levels of platelet adhesion and activation, and low levels of fibrinogen adhesion. We also systematically elucidate the anticoagulant mechanism of the coating, demonstrating why the coating can prevent thrombogenesis without the bleeding risk. Our work not only offers a promising and readily available heparin coating for blood-contact materials, but more importantly, the mechanism exploration supports the practical feasibility of heparin coating in various applications.

Heparin-engineered mesoporous iron metal-organic framework nanoparticles: toward stealth drug nanocarriers

Abstract:The specific modification of the outer surface of the promising porous metal-organic framework nanocarriers (nanoMOFs) preserving their characteristic porosity is still a major challenge. Here a simple, fast, and biofriendly method for the external functionalization of the benchmarked mesoporous iron(III) trimesate nanoparticles MIL-100(Fe) with heparin, a biopolymer associated with longer-blood circulation times is reported. First, the coated nanoparticles showed intact crystalline structure and porosity with improved colloidal stability under simulated physiological conditions, preserving in addition its encapsulation and controlled release capacities. The effect of the heparin coating on the nanoMOF interactions with the biological environment is evaluated through cell uptake, cytotoxicity, oxidative stress, cytokine production, complement activation, and protein adsorption analysis. These results confirmed that the heparin coating endowed the nanoMOFs with improved biological properties, such as reduced cell recognition, lack of complement activation, and reactive oxygen species production. Overall, the ability to coat the surface of the nanoMOFs using a simple and straight-forward approach could be taken as a way to enhance the versatility and, thus, the potential of porous MOF nanoparticles in biomedicine.
Preparation of porous graphene oxide by chemically intercalating a rigid molecule for enhanced removal of typical pharmaceuticals

Abstract:Porous graphene oxide (GO) adsorbents were successfully prepared by connecting GO sheets with tetrafluoroterephthalonitrile (TFT) or decafluorobiphenyl (DFB) through a nucleophilic aromatic substitution reaction. Textural characterization indicated that the enlarged surface area and pore size of the as-synthesized GO-based adsorbents were favorable for the diffusion and adsorption of the typical pharmaceuticals. The GO reacted with 20 mmol/L DFB (GO-DFB20) exhibited the highest removal for six pharmaceuticals among the prepared adsorbents, and can be separated easily. The adsorption capacities of GO-DFB20 for carbamazepine (CBZ), sulfamethoxazole (SMZ), sulfadiazine (SDZ), ibuprofen (IBP), paracetamol (PCT) and phenacetin (PNT) were 340.5, 428.3, 214.7, 224.3, 350.6 and 316.1 μmol/g, respectively. The adsorption kinetics of PCT on the GO-DFB20 was faster than SMZ. According to the Langmuir fitting, the maximum adsorption capacities of GO-DFB20 for SMZ and PCT were 749.6 and 663.9 μmol/g, respectively. The spent GO-DFB20 was successfully regenerated by methanol with little loss of adsorption capacity in five successive adsorption cycles. This study shows that the porous GO adsorbent has a promising application for the removal of typical pharmaceuticals from water or wastewater.
A Facile Method to Prepare Reduced Graphene Oxide with a Large Pore Volume

Abstract:Large size reduced graphene oxide (RGO) tends to restack or form irreversible agglomerates during the conventional drying. In this work, a facile and efficient drying method, involving freezing–thawing–drying approach, is developed to prepare porous RGO. The RGO exhibits a large pore volume (0.414cm3/g) and specific surface area (294.9m2/g). The RGO can be easily ground using mortar and pestle and be dispersed in solvent by ultrasonication. The RGO shows porous nanostructure and poorly-stacked layered structure. Compared with the RGO dried by conventional method, the RGO dried by this modified process possesses similar structural defect density. The ice crystal formed by liquid nitrogen is expected to prevent the restacking of graphene nanosheets and be beneficial to obtain porous structure.
Green Synthesis of Porous Cocoon-Like rGO for Enhanced Microwave Absorbing Performances.

Abstract:A novel porous cocoon-like reduced graphene oxide (rGO) with high porosity and low density was fabricated by a simple and green reduction reaction using ascorbic acid as a reductant in combination with a freeze-drying process without annealing. The bulk density of porous cocoon-like rGO is only 28.49 mg/cm3 and the porosity reaches 94.57%. The reaction times have an important influence on the formation of porous cocoon-like rGO and the reduction degree of rGO. The porous cocoon-like rGO exhibits an excellent microwave absorbing property with a low mass filling ratio of 7 wt%, its minimum reflection loss is -29.05 dB at 15.96 GHz with the sample thickness of 2.0 mm, the effective absorption bandwidth (EAB, RL < -10 dB) is 5.27 GHz. The microwave absorbing property of porous cocoon-like rGO is much better than that of GO and other porous rGO. The in-depth analyses of the reduction degree, porosity and microwave absorbing performance illustrate that the microwave absorbing performance of rGO is significantly related to the reduction degree and porosity. In addition, the synthetic route for porous cocoon-like rGO is simple, low energy consumption and environmentally friendly. Our work demonstrates that the porous cocoon-like rGO is a promising light weight microwave absorber with high-performances.

Abstract:Hypothesis: Graphene-based microparticles materials are broadly utilized in all sorts of fields owing to their outstanding properties. Despite great progress, the present graphene microparticles still face challenges in the aspects of size uniformity, motion flexibility, and tailorable surface chemistry, which limit their application in some specific fields, such as versatile adsorption. Hence, the development of novel graphene microparticles with the aforementioned characteristics is urgently required. Experiments: We presented a simple microfluidic electrospray strategy to generate magnetic Janus reduced graphene oxide/carbon (rGO/C) composite microspheres with a variety of unique features. Specifically, the microfluidic electrospray method endowed the obtaiend microspheres with sufficient size uniformity as well as magnetic responsive motion ability. Additionally, magnetic-mediated surface assembly of phase transition lysozyme (PTL) nanofilm on the microspheres rendered the deposited area hydrophilic while non-deposited area hydrophobic. Findings: Such magnetic Janus rGO/C composite microspheres with regionalized wettability characteristics not only showed prominent performance in adsorbing organic liquids with high adsorption capacity and remarkable reusability but also displayed satisfying biocompatibility for the efficient uptake of bilirubin. More encouragingly, the microspheres could serve as adsorbents in a simulative hemoperfusion setup, which further demonstrated the clinical application potential of the magnetic Janus rGO/C microspheres. Thus, we anticipate that the obtained magnetic Janus rGO/C composite microspheres could show multifunctional properties toward water treatment and blood molecule cleaning.
