Abstract:Research on the design of heparin-analogue hydrogels is of tremendous importance and fuelled by diverse emerging biomedical applications, such as cancer inhibition, treatments of genetic diseases, growth factor carriers, and scaffolds for regeneration medicine, due to their specific biological and biocompatible properties. In this study, by taking inspiration from recent advancements of graphene nanomaterials and heparin-analogue polymers, we designed a kind of highly swellable, elastic, hemo- and cyto-compatible graphene oxide (GO) hybridized heparin-analogue hydrogels for potential drug and protein delivery. The fabricated GO/heparin-analogue hydrogels (GHHs) exhibited an inner-interpenetrated porous structure and robust mechanical properties compared to the GO absent heparin-analogue hydrogel (HH). Notably, the GHHs showed excellent results for in vitro biocompatibility, such as red blood cell compatibility, anti-platelet adhesion and activation, low inflammation potential, high endothelial cell compatibility. Furthermore, after adding GO, the hydrogels showed improved loading and persistent release abilities of doxorubicin hydrochloride (DOX); the GHHs also demonstrated their potential for efficient protein loading and long-term releasing. Due to the integration of elastic mechanical properties, hemo-and cyto-compatibility, as well as drug and protein delivery abilities, the GO hybridized heparin-analogue hydrogels open up a new potential protocol for implantable drug and protein delivery therapies, and bioactive scaffolds for tissue regeneration.

Abstract:Studies on the design of heparin and heparin-mimicking polymer based hydrogels are of tremendous interest and are fuelled by diverse emerging biomedical applications, such as antithrombogenic materials, growth factor carriers, and scaffolds for tissue engineering and regeneration medicine. In this study, inspired by the recent developments of heparin-based hydrogels, graphene oxide (GO) based heparin-mimicking hydrogels with hemocompatibility and versatile properties were prepared via free radical copolymerization, and poly(ethylene glycol) methyl ether methacrylate (PEGMA) and 2-hydroxyethyl methacrylate (HEMA) hydrogels were used as the control samples. The GO based heparin-mimicking polymeric hydrogels exhibited interconnected structures with thin pore walls and high porosity. Because of the increased ionization and electrostatic repulsion of sodium styrene sulfonate (SSNa) segments, the swelling ratios of the SSNa added hydrogels were dramatically increased; after incorporating flexible GO nanosheets, as the 3D skeleton of the hydrogels, the swelling ability was further increased. In addition, the GO based heparin-mimicking hydrogels showed superior red blood cell compatibility, anti-platelet adhesion ability and anticoagulant ability. Furthermore, drug release data indicated that the GO based heparin-mimicking hydrogels had high drug loading ability and prolonged drug releasing ability; the antibacterial tests showed coincident results with large inhibition zones and long effective periods. Due to the integration of blood compatibility, drug loading and releasing abilities, as well as an excellent ability for the removal of toxic molecules, the GO based heparin-mimicking hydrogels can be used for versatile biomedical applications.

Abstract:Hydrogels with injectability, self-healing ability and adhesiveness have great potential for hemostasis and full-thickness skin wound repair, which are usually fabricated by multistep chemical synthesis and the use of organic solvents and catalyst. Herein, we report an injectable and self-healing hydrogel facilely prepared through one-pot heating of chitosan and graphene oxide mixture solution, without any pollutant and waste generated. The dynamic reversible breakage and recombination of noncovalent bonds between chitosan and graphene oxide endows the hydrogel injectability and self-healing ability. In addition, the mechanical and rheological properties of the hydrogels can be controlled by varying the dosage of graphene oxide. Meanwhile, hydrogels exhibited good adhesiveness and hemocompatibility. Finally, in vivo experiments in a rat liver bleeding model and full-thickness skin defect model verified the outstanding hemostatic and wound healing capability of the hydrogels, indicating the promising future for use as wound dressing.

Abstract:Elastomeric, robust, and biocompatible hydrogels are rare, while the need for these types of biomaterials in biomedical-related uses remains high. Here, a new family of genetically engineered silk-elastin copolymers (SELPs) with encoded enzymatic crosslinking sites is developed for a new generation of stimuli-responsive yet robust hydrogels. Input into the designs is guided by simulation, and realized via genetic engineering strategies. The avoidance of gamma irradiation or chemical crosslinking during gel fabrication, in lieu of an enzymatic process, expands the versatility of these new gels for the incorporation of labile proteins and cells. In the present study, the new SELP hydrogels offers sequence dependent, reversible stimuli-responsive features. Their stiffness covers almost the full range of the elasticity of soft tissues. Further, physical modification of the silk domains provided a secondary control point to fine-tune mechanical stiffness while preserving stimuli-responsive features, with implications for a variety of biomedical materials and device needs.

Abstract:In the present study, novel heparin-mimetic polyurethane hydrogels were prepared by introducing chemical crosslinked sulfated konjac glucomannan (SKGM). Scanning electron microscopy (SEM) results indicated that the introduction of SKGM and the increase of the molecular weight of diol segments could enlarge the pore sizes of the hydrogels. The swelling behavior corresponded with the SEM results, and the hydrogels could absorb more water after the modification. The modification also led to an improvement in the mechanical property. Meanwhile, the SKGM and the modified polyurethane hydrogels showed excellent hemocompatibility. The thromboplastin time of SKGM could reach up to 182.9s. Gentamycin sulfate (GS) was used as a model drug to be loaded into the hydrogels, and the loading amount was increased ca. 50% after the introduction of SKGM, thus resulting in high bactericidal efficiency. The results indicated that the introduction of SKGM and the alternation in the diol's molecular weight bestowed polyurethane hydrogels with promising properties of integrated blood-compatibility, mechanical properties and drug loading-releasing behavior. Therefore, the heparin-mimetic multifunctional polyurethane hydrogels have great potential to be used in biomedical applications.

Abstract:In this work, we designed a robust and heparin-mimetic hydrogel thin film coating via combined layer-by-layer (LbL) self-assembly and mussel-inspired post-cross-linking. Dopamine-grafted heparin-like/-mimetic polymers (DA-g-HepLP) with abundant carboxylic and sulfonic groups were synthesized by the conjugation of adhesive molecule, DA, which exhibited substrate-independent adhesive affinity to various solid surfaces because of the formation of irreversible covalent bonds. The hydrogel thin film coated substrates were prepared by a three-step reaction: First, the substrates were coated with DA-g-HepLP to generate negatively charged surfaces. Then, multilayers were obtained via LbL coating of chitosan and the DA-g-HepLP. Finally, the noncovalent multilayers were oxidatively cross-linked by NaIO4. Surface ATR-FTIR and XPS spectra confirmed the successful fabrication of the hydrogel thin film coatings onto membrane substrates; SEM images revealed that the substrate-independent coatings owned 3D porous morphology. The soaking tests in highly alkaline, acid, and concentrated salt solutions indicated that the cross-linked hydrogel thin film coatings owned high chemical resistance. In comparison, the soaking tests in physiological solution indicated that the cross-linked hydrogel coatings owned excellent long-term stability. The live/dead cell staining and morphology observations of the adhered cells revealed that the heparin-mimetic hydrogel thin film coated substrates had low cell toxicity and high promotion ability for cell proliferation. Furthermore, systematic in vitro investigations of protein adsorption, platelet adhesion, blood clotting, and blood-related complement activation confirmed that the hydrogel film coated substrates showed excellent hemocompatibility. Both the results of inhibition zone and bactericidal activity indicated that the gentamycin sulfate loaded hydrogel thin films had significant inhibition capability toward both Escherichia coli and Staphylococcus aureus bacteria. Combined the above advantages, it is believed that the designed heparin-mimetic hydrogel thin films may show high potential for applications in various biological and clinical fields, such as long-term hemocompatible and drug-loading materials for implants.
Super-Anticoagulant Heparin-Mimicking Hydrogel Thin Film Attached Substrate Surfaces to Improve Hemocompatibility.

Abstract:In this study, heparin-mimicking hydrogel thin films are covalently attached onto poly(ether sulfone) membrane surfaces to improve anticoagulant property. The hydrogel films display honeycomb-like porous structure with well controlled thickness and show long-term stability. After immobilizing the hydrogel films, the membranes show excellent anticoagulant property confirmed by the activated partial thromboplastin time values exceeding 600 s. Meanwhile, the thrombin time values increase from 20 to 61 s as the sodium allysulfonate proportions increase from 0 to 80 mol%. In vitro investigations of protein adsorption and blood-related complement activation also confirm that the membranes exhibit super-anticoagulant property. Furthermore, gentamycin sulfate is loaded into the hydrogel films, and the released drug shows significant inhibition toward E. coli bacteria. It is believed that the surface attached heparin-mimicking hydrogel thin films may show high potential for the applications in various biological fields, such as blood contacting materials and drug loading materials.

Abstract:A new class of hydrogels distinguished by electro-conductivity and high hemo-compatibility, which resemble biological tissues, are promising candidates for a plethora of biomedical applications. This work describes the design of highly elastic and biocompatible hydrogels using polypyrrole (PPy) as the conductivity media. The bio-inspired design of heparin-mimicking sodium alginate (HMSA) combined with tough alginate/polyacrylamide (PAM) is selected to form chemical and physical cross-linked binary hydrogel networks, while the incorporation of interfacial polymerized PPy endows the hydrogels with electro-conductivity. The resulting hydrogels exhibit a mesh-like hierarchical structure with excellent mechanical strength; moreover, the hydrogels show greatly enhanced electro-conductivity (up to 0.63 S m-1). In addition, due to the presence of the unique heparin-mimicking structure, the hydrogels possess a desirable anticoagulant ability and biocompatibility demonstrated via antithrombotic evaluations together with cell culture observations. The facile synthesis of HMSA/PAM/PPy hydrogels and their robust physical, chemical and biological performance make them attractive components for the future generation of biosensors and bioelectrodes.
Highly Tough, Stretchable, and Enzymatically Degradable Hydrogels Modulated by Bioinspired Hydrophobic β‑Sheet Peptides

Abstract:Peptide-based supramolecular hydrogels have attracted great attention due to their good biocompatibility and biodegradability and have become promising candidates for biomedical applications. The bottom-up self-assembly endows the peptides with a highly ordered secondary structure, which has proven to be an effective strategy to improve the mechanical properties of hydrogels through strong physical interactions and energy dissipation. Inspired by the excellent mechanical properties of spider-silk, which can be attributed to the rich β-sheet crystal formation by the hydrophobic peptide fragment, a hydrophobic peptide (HP) that can form a β-sheet assembly was designed and introduced into a poly(vinyl alcohol) (PVA) scaffold to improve mechanical properties of hydrogels by the cooperative intermolecular physical interactions. Compared with hydrogels without peptide grafting (P-HP0), the strong β-sheet self-assembly domain endows the hybrid hydrogels (P-HP20, P-HP29, and P-HP37) with high strength and toughness. The fracture tensile strength increased from 0.3 to 2.1 MPa (7 times), the toughness increased from 0.4 to 21.6 MJ m–3 (54 times), and the compressive strength increased from 0.33 to 10.43 MPa (31 times) at 75% strain. Moreover, the hybrid hydrogels are enzymatically degradable due to the dominant contribution of the β-sheet assembly for network cross-linking. Combining the good biocompatibility and sustained drug release of the constructed hydrogels, this hydrophobic β-sheet peptide represents a promising candidate for the rational design of hydrogels for biomedical applications.

Abstract:The application of functional self-healing and mechanically robust hydrogels in bioengineering, drug delivery, soft robotics, etc., is continuously growing. However, fabricating hydrogels that simultaneously possess good mechanical and self-healing properties remains a challenge. Developing robust hydrogel formulations for the encapsulation and release of hydrophobic substances is a major challenge especially in some pharmaceutical treatments where the many of drugs show incompatibility with the hydrophilic hydrogel matrices. Schiff base hydrogels have been developed using a benzaldehyde multifunctional amphiphilic polyacrylamide crosslinker in conjunction with glycol chitosan. The polymeric crosslinker was synthesized by a two-step reaction using aqueous Cu-RDRP to give an ABA telechelic copolymer of N,N-dimethyl acrylamide (DMAc) and N-hydroxyethyl acrylamide (HEAm) from a bifunctional PEG. The polymer was then modified by post functionalization leading to a multifunctional benzaldehyde crosslinker that was shown to be capable of self-assembly into aggregates in aqueous media serving as a possible candidate for the entrapment of hydrophobic substances. Aqueous solutions of the crosslinker spontaneously formed hydrogels when mixed with glycol chitosan due to the in situ formation of imine bonds. Hydrogels were characterized while additional comparisons were made with a commonly used bifunctional PEG crosslinker. The effect of introducing partially reduced graphene oxide (GO) nanosheets was also examined and led to enhancements in both mechanical properties (2.0 fold increase in modulus and 1.4 fold increase in strain) and self-healing efficiencies (>99% from 60% by rheology) relative to the pristine polymer hydrogels.

Abstract:An interpenetrating network (IPN) strategy has been widely facilitated to construct strong and tough hydrogels, but most of the efforts have been focused on organic/organic networks. Herein, aqueous dispersible 2,2'-(ethylenedioxy)-diethanethiol (EDDET) cross-linked graphene oxide (E-cGO) skeleton was in situ incorporated into a PVA matrix, resulting in novel inorganic/organic IPN hydrogels with super mechanical and chondrocyte cell-adhesion properties. The unique interpenetrating structure and hydrogen bonding were demonstrated to play critical roles in enhancing the compressive property of the IPN hydrogels, in comparison to the GO and thermally reduced graphene oxide (T-rGO) filled hydrogels. It is critical that the E-cGO/PVA hydrogels have been demonstrated as being biocompatible, which make the E-cGO/PVA hydrogels promising candidate biomaterials for load-bearing biotissue substitution.

Abstract:The application fields of hydrogels are often severely limited by their weak mechanical performance. It is therefore highly demanded to develop an effective strategy to fabricate mechanically strong hydrogels. Herein, a kind of bioinspired ternary hydrogel consisting of graphene oxide (GO) nanosheets, xonotlite nanowires, and polyacrylamide (PAM) is constructed under the synergy of hydrogen bonding-induced GO/xonotlite network and the penetrated PAM chain network. Benefiting from the effective energy dissipation mechanism caused by double-network structural design and the strong hydrogen bonding interaction between two nanobuilding blocks, the gel exhibits a high toughness of 22 MJ m(-3) at an elongation of 2750%. Even notched with 1/4 size, it still holds a large extensibility of 2180% its initial length. These high-performance hydrogels could be of great interest in the fields of tissue engineering and biomedical areas.
Biologically Derived Soft Conducting Hydrogels Using Heparin-Doped Polymer Networks

Abstract:The emergence of flexible and stretchable electronic components expands the range of applications of electronic devices. Flexible devices are ideally suited for electronic biointerfaces because of mechanically permissive structures that conform to curvilinear structures found in native tissue. Most electronic materials used in these applications exhibit elastic moduli on the order of 0.1-1 MPa. However, many electronically excitable tissues exhibit elasticities in the range of 1-10 kPa, several orders of magnitude smaller than existing components used in flexible devices. This work describes the use of biologically derived heparins as scaffold materials for fabricating networks with hybrid electronic/ionic conductivity and ultracompliant mechanical properties. Photo-cross-linkable heparin methacrylate hydrogels serve as templates to control the microstructure and doping of in situ polymerized polyaniline structures. Macroscopic heparin-doped polyaniline hydrogel dual networks exhibit impedances as low as Z = 4.17 Omega at 1 kHz and storage moduli of G' = 900 +/- 100 Pa. The conductivity of heparin/polyaniline networks depends on the oxidation state and microstructure of secondary polyaniline networks. Furthermore, heparin/polyaniline networks support the attachment, proliferation, and differentiation of mu rifle myoblasts without any surface treatments. Taken together, these results suggest that heparin/polyaniline hydrogel networks exhibit suitable physical properties as an electronically active biointerface material that can match the mechanical properties of soft tissues composed of excitable cells.

Abstract:Large-scale fabrication of heparinized hydrogel could be accomplished by free-radical crosslinking polymerization (FRCP), whilst its bioactivity is often partially lost. Heparinized hydrogel based implantable medical devices, therefore, frequently encounter the compelling problems of thrombosis and microbial infection. In this study, a facile FRCP approach was proposed to fabricate heparinized hydrogels by preforming polydopamineheparin (PDA-HEP) complex, which bypassed the steps of heparin functionalization and effectively preserved the function of heparin in the meantime. Compared with heparin mixed hydrogels, heparin in PDA-HEP complex composite hydrogels augmented the antimicrobial activity of PDA by enhancing the combined bactericidal action of cations and reactive oxygen species (ROS), while still maintaining its anticoagulant function. Since its integration of extremely low hemolysis rate, remarkable anticoagulation property and efficient bactericidal performance, we believe that this universal method of preparing heparinized hydrogels based on PDA-HEP complex will speed up the versatile application of hydrogel implants in clinical medicine.
General and Biomimetic Approach to Biopolymer-Functionalized Graphene Oxide Nanosheet Through Adhesive Dopamine.

Abstract:Graphene oxide (GO), reduced graphene oxide (rGO), and their derivatives are investigated for various biomedical applications explosively. However, the defective biocompatibility was also recognized, which restricted their potential applications as biomaterials. In this study, a facile biomimetic approach for preparation of biopolymer adhered GO (rGO) with controllable 2D morphology and excellent biocompatibility was proposed. Mussel-inspired adhesive molecule dopamine (DA) was grafted onto heparin backbone to obtain DA grafted heparin (DA-g-Hep) by carbodiimide chemistry method; then, DA-g-Hep was used to prepare heparin-adhered GO (Hep-a-GO) and heparin-adhered rGO (Hep-a-rGO). The obtained heparin-adhered GO (rGO) showed controllable 2D morphology, ultrastable property in aqueous solution, and high drug and dye loading capacity. Furthermore, the biocompatibility of the heparin-adhered GO (rGO) was investigated using human blood cells and human umbilical vein endothelial cells, which indicated that the as-prepared heparin-adhered GO (rGO) exhibited ultralow hemolysis ratio (lower than 1.2%) and high cell viability. Moreover, the highly anticoagulant bioactivity indicated that the adhered heparin could maintain its biological activity after immobilization onto the surface of GO (rGO). The excellent biocompatibility and high bioactivity of the heparin-adhered GO (rGO) might confer its great potentials for various biomedical applications.

Abstract:Novel heparin-conjugated PLGA-PEG-PLGA hydrogels were prepared via Michael-type addition between thiolated heparin and PLGA-PEG-PLGA diacrylate. The thiolated heparin (HP-SH) was conjugated with thiolacid dihydrazide followed by reduction. The structure and the thiol determination of obtained HP-SH were characterized by 1 H NMR and the Ellman method. Anticoagulant activity and pK a of the HP-SH were determined by aPTT test and UV absorbance measurement which were 79.3% and 10.5, respectively. The PLGA-PEG-PLGA diacrylate was synthesized by bulk ring-opening polymerization of D,L-lactide (DLLA) and glycolide (GA) with PEG and stannous 2-ethylhexanoate, followed by the acrylation of the terminal groups. HP—SH was then conjugated to PLGA-PEG-PLGA diacrylate by Michael addition. Phase diagrams of the hydrogels were obtained by vial tilting; the release of heparin from the hydrogels exhibited temperature dependent sol—gel transition behavior. These in situ-forming heparin-conjugated hydrogels are novel as injectable and tissue-compatible scaffold formation, thermo-sensitivity, and growth factor binding.

Abstract:The importance of hydrogels in tissue engineering cannot be overemphasized due to their resemblance to the native extracellular matrix. However, natural hydrogels with satisfactory biocompatibility exhibit poor mechanical behavior, which hampers their application in stress-bearing soft tissue engineering. Here, we describe the fabrication of a double methacrylated gelatin bioink covalently linked to graphene oxide (GO) via a zero-length crosslinker, digitally light-processed (DLP) printable into 3D complex structures with high fidelity. The resultant natural hydrogel (GelGOMA) exhibits a conductivity of 15.0 S m-1as a result of the delocalization of theπ-orbital from the covalently linked GO. Furthermore, the hydrogel shows a compressive strength of 1.6 MPa, and a 2.0 mm thick GelGOMA can withstand a 1.0 kg ms-1momentum. The printability and mechanical strengths of GelGOMAs were demonstrated by printing a fish heart with a functional fluid pumping mechanism and tricuspid valves. Its biocompatibility, electroconductivity, and physiological relevance enhanced the proliferation and differentiation of myoblasts and neuroblasts and the contraction of human-induced pluripotent stem cell-derived cardiomyocytes. GelGOMA demonstrates the potential for the tissue engineering of functional hearts and wearable electronic devices.
