Abstract:The human tissues most sensitive to electrical activity such as neural and muscle tissues are relatively soft, and yet traditional conductive materials used to interface with them are typically stiffer by many orders of magnitude. Overcoming this mismatch, by creating both very soft and electroactive materials, is a major challenge in bioelectronics and biomaterials science. One strategy is to imbue soft materials, such as hydrogels, with electroactive properties by adding small amounts of highly conductive nanomaterials. However, electroactive hydrogels reported to date have required relatively large volume fractions (>1%) of added nanomaterial, have shown only modest electroactivity, and have not been processable via additive manufacturing to create 3D architectures. Here, we describe the development and characterization of improved biocompatible photo-cross-linkable soft hybrid electroactive hydrogels based on gelatin methacryloyol (GelMA) and large area graphene oxide (GO) flakes, which resolve each of these three limitations. The addition of very small amounts (less than a 0.07% volume fraction) of GO to a 5% w/v GelMA hydrogel resulted in a dramatic (∼35-fold) decrease in the impedance at 1 Hz compared with GelMA alone. The GelMA/GO coated indium tin oxide (ITO) electrode also showed a considerable reduction in the impedance at 1 kHz (down to 170 Ω compared with 340 Ω for the GelMA-coated ITO), while charge injection capacity increased more than 6-fold. We attribute this enhanced electroactivity to the increased electroactive surface area contributed by the GO. Despite this dramatic change in electroactivity, the GelMA/GO composite hydrogels' mechanical properties were only moderately affected. Mechanical properties increased by ∼2-fold, and therefore, the hydrogels' desired softness of <4 kPa was retained. Also, we demonstrate how light attenuation through the gel can be used to create a stiffness gradient with the exposed surface of the gel having an elastic modulus of <1.5 kPa. GO addition also enhanced the rheological properties of the GelMA composites, thus facilitating 3D extrusion printing. GelMA/GO enhanced filament formation as well as improved printability and the shape fidelity/integrity of 3D printed structures compared with GelMA alone. Additionally, the GelMA/GO 3D printed structures presented a higher electroactive behavior than nonprinted samples containing the same GelMA/GO amount, which can be attributed to the higher electroactive surface area of 3D printed structures. These findings provide new rational choices of electroactive hydrogel (EAH) compositions with broad potential applications in bioelectronics, tissue engineering, and drug delivery.

Dual-Crosslinked Oxidized Pectin/N-Succinyl Chitosan Hydrogel Containing Graphene Oxide Nanosheets for Tissue Engineering Application

Abstract:Abstract Biopolymer-based hydrogels are commonly used in clinical applications. In the present study, N- succinyl chitosan (NSC), oxidized pectin (OP), and graphene oxide (GO) were used to develop a new dual-crosslinked hydrogel system. The dynamic OP/NSC/GO hydrogel showed quick gelation and great injectability due to the cooperation of hydrogen interaction between the GO nanosheets and the NSC and OP macromolecules and Schiff-based crosslinking by amino and aldehyde functional groups of polysaccharide derivatives. The performance of the above-mentioned hydrogel was improved when the GOs were embedded. When the GO content was 6 (mg/ml), the hydrogel showed the best overall performance, with a 10-minute healing time, a quick gelation time (~ 13s), acceptable swelling ability, suitable conductivity, great hemocompatibility, and strong biological compatibility. These results showed that the composite hydrogel could be used as a promising conductive injectable self-healing hydrogel for tissue engineering applications.
A New Shear-thinning Nanocomposite Hydrogel from GelMA- GO for Soft Tissue Engineering

Abstract:The use of bio ink materials for fabricating scaffolds is the best option to replace aggressive surgery. This study aims to formulate an injectable hydrogel with suitable structural, mechanical, and rheological properties for tissue engineering applications. In this respect, gelatin methacrylate (GelMA) nanocomposites with different amounts of the modified graphene oxide nanoplates (0.25, 0.5, 0.75, and 1 wt.%) were fabricated. The effect of the nanoplates on the structural, degradability, mechanical, rheological, and biological properties of the nanocomposites were evaluated. The results revealed that the functionalization of graphene oxide with dopamine (GOPD) resulted in the formation of a layer of polydopamine on the surface of the nanoplates, which developed the interaction of graphene oxide with GelMA chains. Adding GOPD to GelMA from 0 to 1 wt.% decreased the water absorption (0.3 times) and its weight loss percentage (0.2 times). The addition of 0.5 wt.% of GOPD to GelMA improved the compressive strength (7.5 times) and compressive modulus (7 times) compared to the GelMA hydrogel. Also, a similar amount of GOPD developed the thin shear properties of GelMA by three logarithmic units. Besides, the addition of the modified graphene oxide nanoplates to GelMA enhanced cell viability (93.26%) after 72 hours of cell seeding. The proper behavior of the GelMA hydrogel with 0.5% GOPD , and its stability during the 3D printing process was confirmed. Accordingly, the injectable GelMA hydrogel containing dopamine-modified graphene oxide could be a proper candidate for tissue engineering expenditures.

Abstract:Gelatin and chitosan (CS) are widely used natural biomaterials for tissue engineering scaffolds, but the poor mechanical properties of pure gelatin or CS hydrogels become a big obstacle that limits their use as scaffolds, especially in load-bearing tissues. This study provided a novel mechanism of forming interpenetrating network (IPN) of gelatin methacryloyl (GelMA) and CS hydrogels by covalent bonds and hydrophobic interactions through photocrosslinking and basification, respectively. By characterization of the compressive and tensile moduli, ultimate tensile stress and strain, it was found that semi-IPN and IPN structure can greatly enhance the mechanical properties of GelMA-CS hydrogels compared to the single network CS or GelMA. Moreover, the increase of either GelMA or CS concentration can strengthen the hydrogel network. Then, the swelling, enzymatic degradation, and morphology of GelMA-CS hydrogels were also systematically investigated. The excellent biocompatibility of GelMA-CS hydrogels was demonstrated by large spreading area of bone mesenchymal stem cells on hydrogel surfaces when CS concentration was <2% (w/v). According to this study, the multiple requirements of properties can be fulfilled by carefully selecting the GelMA and CS compositions for IPN hydrogels.

Abstract:For conductive hydrogels applied in biosensors, wearable devices and so forth, multifunctionality is an inevitable trend of development to meet various practical requirements and enhance human experience. Herein, inspired by nanocomposite, double-network (DN) and mussel chemistry, a new Graphene oxide@Dopamine/Alginate/Poly (acrylic acid-co-acrylamide) [GO@DA/Alginate/P(AAc-co-AAm)] hydrogel was fabricated through one-pot in-situ radical copolymerization. GO@DA nanofillers, prepared via GO confined DA polymerization, imparted the hydrogel with remarkable adhesiveness. Alginate/P(AAc-co-AAm) DN matrix, physically and chemically crosslinked by Fe3+ and N,N'-Methylenebisacrylamide, made hydrogels ultrastretchable, self-healing and biocompatible. With contents of DA and alginate accurately regulated, the tensile strength, elongation, adhesion strength and conductivity of the optimal hydrogel could reach 320.2 kPa, 1198 %, 36.9 kPa and 3.24 +/- 0.12 S/m, respectively. What's more notable was that the synergistic integration of repeatable adhesiveness, strain sensitivity, use stability, self-healing ability and biocompatibility provided such hydrogels with tremendous possibility of practical application for strain sensors.

Abstract:Gelatin methacrylate (GelMA)-based hydrogels are gaining a great deal of attention as potentially implantable materials in tissue engineering applications because of their biofunctionality and mechanical tenability. Since different natural tissues respond differently to mechanical stresses, an ideal implanted material would closely match the mechanical properties of the target tissue. In this regard, applications employing GelMA hydrogels are currently limited by the low mechanical strength and biocompatibility of GelMA. Therefore, this review focuses on modifications made to GelMA hydrogels to make them more suitable for tissue engineering applications. A large number of reports detail rational synthetic processes for GelMA or describe the incorporation of various biomaterials into GelMA hydrogels to tune their various properties, e.g., physical strength, chemical properties, conductivity, and porosity, and to promote cell loading and accelerate tissue repair. A novel strategy for repairing tissue injuries, based on the transplantation of cell-loaded GelMA scaffolds, is examined and its advantages and challenges are summarized. GelMA-cell combinations play a critical and pioneering role in this process and could potentially accelerate the development of clinically relevant applications.

Abstract:Conductive hydrogels have garnered considerable attention as novel materials for biomedical devices and tissue engineering because they exhibit electroactivity and tissue-like softness. Various composites and manufacturing techniques have been developed in this regard. However, conductive hydrogels often exhibit poor mechanical properties (e.g., low toughness), which impedes their biomedical application. In this study, we fabricated double network (DN) hydrogels composed of sodium alginate (SA), polyacrylamide (PAAm), and graphene oxide (GO) to promote elasticity, toughness, and mechanical strength. Subsequently, we reduced the composite hydrogels to improve electrical conductivity by converting GO to more conductive reduced graphene oxide (rGO). The electrical, electrochemical, and mechanical properties of the produced hydrogels, r(GO/SA/PAAm), were characterized. Particularly, crosslinking density and reducing time were varied to obtain optimal conditions. The produced r(GO/SA/PAAm) demonstrated excellent electrical conductivity, mechanical strength, and toughness compared to homopolymers and unreduced DN hydrogels. We demonstrated that conductive DN hydrogels are suitable strain sensors. Electrically conductive and mechanically strong hydrogels will be beneficial in various biomedical applications, such as tissue engineering scaffolds and bioelectrodes.

Development of 3D printable graphene oxide based bio-ink for cell support and tissue engineering

Abstract:Tissue engineered constructs can serve as in vitro models for research and replacement of diseased or damaged tissue. As an emerging technology, 3D bioprinting enables tissue engineering through the ability to arrange biomaterials and cells in pre-ordered structures. Hydrogels, such as alginate (Alg), can be formulated as inks for 3D bioprinting. However, Alg has limited cell affinity and lacks the functional groups needed to promote cell growth. In contrast, graphene oxide (GO) can support numerous cell types and has been purported for use in regeneration of bone, neural and cardiac tissues. Here, GO was incorporated with 2% (w/w) Alg and 3% (w/w) gelatin (Gel) to improve 3D printability for extrusion-based 3D bioprinting at room temperature (RT; 25°C) and provide a 3D cellular support platform. GO was more uniformly distributed in the ink with our developed method over a wide concentration range (0.05%-0.5%, w/w) compared to previously reported GO containing bioink. Cell support was confirmed using adipose tissue derived stem cells (ADSCs) either seeded onto 3D printed GO scaffolds or encapsulated within the GO containing ink before direct 3D printing. Added GO was shown to improve cell-affinity of bioinert biomaterials by providing more bioactive moieties on the scaffold surface. 3D cell-laden or cell-seeded constructs showed improved cell viability compared to pristine (without GO) bio-ink-based scaffolds. Our findings support the application of GO for novel bio-ink formulation, with the potential to incorporate other natural and synthetic materials such as chitosan and cellulose for advanced in situ biosensing, drug-loading and release, and with the potential for electrical stimulation of cells to further augment cell function.

Cytotoxicity assays indicated robust cell survival rates of 90%, confirming the biocompatibility of the developed materials. Cryoprinting effectively preserved scaffold integrity and functionality. The CAM assay validated the biocompatibility and angiogenic potential, demonstrating substantial vascularization upon scaffold implantation onto chick embryo CAM. Conclusion: Integrating graphene into GelMA hydrogels, coupled with low-temperature 3D printing, represents a potent strategy for enhancing scaffold fabrication. Abstract:Cardiac failure can be a life-threatening condition that, if left untreated, can have significant consequences. Functional hydrogel has emerged as a promising platform for cardiac tissue engineering. In the proposed study, gelatin methacrylate (GelMA) and alginate, as a primary matrix to maintain cell viability and proliferation, and polypyrrole and carboxyl-graphene, to improve mechanical and electrical properties, are thoroughly evaluated. Initially, a polymer blend of GelMA/Alginate (1:1) was prepared, and then the addition of 2–5 wt% of polypyrrole was evaluated. Next, the effect of incorporating graphene-carboxyl nanosheets (1, 2, and 3 wt%) within the optimized scaffold with 2 wt% polypyrrole was thoroughly studied. The electrical conductivity of the hydrogels was significantly enhanced from 0.0615 ± 0.007 S/cm in GelMA/alginate to 0.124 ± 0.04 S/cm with the addition of 5 wt% polypyrrole. Also, 3 wt% carboxyl graphene improved the electrical conductivity to 0.27 ± 0.09 S/cm. The compressive strength of carboxyl-graphene-containing hydrogel was in the range of 175 to 520 kPa, and tensile strength was 61 and 133 kPa. The simplicity and low-cost fabrication, tunable mechanical properties, optimal electrical conductivity, blood compatibility, and non-cytotoxicity of GelMA/alginate/polypyrrole/graphene biocomposite hydrogel is a promising construct for cardiac tissue engineering.

Abstract:The mechanical performances of hydrogels are greatly influenced by the functionality of cross-linkers and their covalent and non-covalent interactions with the polymer chains. Conventional chemical cross-linkers fail to meet the demand of large toughness and high extensibility for their immediate applications as artificial tissues like ligaments, blood vessels, and cardiac muscles in human or animal bodies. Herein, we synthesized a new graphene oxide-based two-dimensional (2D) cross-linker (GOBC) and exploited the functionality of the cross-linker for the enhancement of toughness and stretchability of a poly(acrylic acid) (PAA) hydrogel. The 2D nanosheets of GO were modified in such a way that they could provide multifunctional sites for both physical and chemical bonding with the polymer chains. Carboxylic acid groups at the surfaces of the GO sheets were coupled with the acrylate functional groups for covalent cross-linking, while the other oxygen-containing functional groups are responsible for physical cross-linking with polymers. The GOBC had been successfully incorporated into the PAA hydrogel and the mechanical properties of the GOBC cross-linked PAA hydrogel (PAA-GOBC) were investigated at various compositions of cross-linker. Seven times enhancement in both toughness and elongation at break has been achieved without compromising on the modulus for the as-synthesized PAA-GOBC compared to the conventional N,N'-methylenebis(acrylamide) (MBA) cross-linked PAA hydrogel. This facile and efficient way of GO modification is expected to lead the development of a high-performance nanocomposite for cutting-edge applications in biomedical engineering.

Abstract:The ideal bone repair materials possess a series of properties, such as injectability, good mechanical properties and bone inducibility. In the present study, gelatin methacryloyl (GelMA) and graphene oxide (GO) were selected to prepare conductive hydrogel by changing the concentration of GelMA and GO during the cross-link process. The effects of different contents of GelMA and GO to the hydrogel performance were investigated. The results showed that the mechanical properties of the hydrogel kept 16.37 ± 1.89 KPa after adding 0.1% GO, while the conductivity was improved to 1.36 ± 0.09 μS/cm. The porosity of hydrogel before and after mineralization could reach more than 90%. The mechanical properties of mineralized hydrogel was improved significantly, could reach 26.38 ± 2.29 KPa. Cell experiments indicated that the mineralized hydrogel with electrical stimulation obviously improve the alkaline phosphatase activity of the cells. GelMA/GO conductive hydrogel could be a promising candidate for bone repair and bone tissue engineering.

Abstract:In this study we consider a mathematical model of a sexual population that lives in a changing environment. We find that a low rate of environmental change can produce a very large increase in genetic variability. This may help to explain the high levels of heritability observed in many natural populations. We also study asexuality and find that a modest rate of environmental change can be very damaging to an asexual population, while leaving a sexual population virtually unscathed. Furthermore, in a changing environment, the advantages of sexuality over asexuality can be much greater than suggested by most previous studies. Our analysis applies in the case of very large populations, where stochastic forces may be neglected.

3D Printing and Additive Manufacturing

Abstract:Bioprinting has emerged as a powerful manufacturing platform for tissue engineering, enabling the fabrication of 3D living structures by assembling living cells, biological molecules, and biomaterials into these structures. Among various biomaterials, hydrogels have been increasingly used in developing bioinks suitable for 3D bioprinting for diverse human body tissues and organs. In particular, hydrogel blends combining gelatin and gelatin methacryloyl (GelMA; "GG hydrogels") receive significant attention for 3D bioprinting owing to their many advantages, such as excellent biocompatibility, biodegradability, intrinsic bioactive groups, and polymer networks that combine the thermoresponsive gelation feature of gelatin and chemically crosslinkable attribute of GelMA. However, GG hydrogels have poor electroactive properties, which hinder their applications in neural tissue engineering where electrical conductivity is required. To overcome this problem, in this study, a small amount of highly electroactive graphene oxide (GO) was added in GG hydrogels to generate electroactive hydrogels for 3D bioprinting in neural tissue engineering. The incorporation of GO nanoparticles slightly improved mechanical properties and significantly increased electrical conductivity of GG hydrogels. All GO/GG composite hydrogels exhibited shear thinning behavior and sufficient viscosity and hence could be 3D printed into 3D porous scaffolds with good shape fidelity. Furthermore, bioinks combining rat bone marrow-derived mesenchymal stem cells (rBMSCs) with GO/GG composite hydrogels could be 3D bioprinted into GO/GG constructs with high cell viability. GO nanoparticles in the constructs provided ultraviolet (UV) shading effect and facilitated cell survival during UV exposure after bioprinting. The GO/GG composite hydrogels appear promising for 3D bioprinting applications in repairing damaged neural tissues.

Abstract:A novel gelatin‐based hydrogel (GelDT), modified with dithiolane groups, is engineered to improve tissue engineering. This multi‐functional biomaterial offers long‐term stability, adjustable mechanical properties, and responsive drug release. Its versatility includes support for 3D bioprinting and strong tissue adhesion via disulfide bonds with native tissue thiols, making it a powerful tool for tissue repair and regeneration. Biomaterials that integrate multiple functionalities, mimic the extracellular matrix (ECM) microenvironment to support cellular growth, and adhere robustly to damaged tissues are highly needed to advance tissue engineering. Protein‐based biomaterials are promising due to their inherent biocompatibility, biomimicry, biodegradation, and cell‐supportive properties. Herein, by leveraging the unique ability of dithiolanes to generate on‐demand in situ thiols, a new class of dithiolane‐modified, protein‐based biomaterial that combines unique, seemingly opposing functions for tissue engineering is developed. Dithiolane‐modified gelatin, a model protein used herein, enabled photoinitiator‐free photo‐crosslinking to form multi‐functional gelatin‐dithiolane (GelDT) hydrogels, which displayed exceptional long‐term stability in cell culture media (>28 days) to support the growth of both surface‐seeded and encapsulated cells. GelDT hydrogels allowed pre‐gelation tuning of biomechanical properties and biodegradation via introducing physical crosslinks, and post‐gelation tuning of matrix stress‐relaxation rate, via responding to exogenous thiols, independently of other parameters. Furthermore, GelDT enabled covalent immobilization of bio‐active molecules, glutathione‐responsive drug release, supported efficient 3D bioprinting due to its shear‐thinning ability, and demonstrated robust tissue adhesion in various contexts (bare skin, ex‐vivo, in‐vivo) due to covalent disulfide coupling with endogenous tissue thiols. Together, this study presents a novel multi‐responsive and multi‐functional protein‐based biomaterial, anticipated to advance tissue engineering and regeneration.

Abstract:Currently, a major challenge in material engineering is to develop a cell-safe biomaterial with significant utility in processing technology such as 3D bioprinting. The main goal of this work was to optimize the composition of a new graphene oxide (GO)-based bioink containing additional extracellular matrix (ECM) with unique properties that may find application in 3D bioprinting of biomimetic scaffolds. The experimental work evaluated functional properties such as viscosity and complex modulus, printability, mechanical strength, elasticity, degradation and absorbability, as well as biological properties such as cytotoxicity and cell response after exposure to a biomaterial. The findings demonstrated that the inclusion of GO had no substantial impact on the rheological properties and printability, but it did enhance the mechanical properties. This enhancement is crucial for the advancement of 3D scaffolds that are resilient to deformation and promote their utilization in tissue engineering investigations. Furthermore, GO-based hydrogels exhibited much greater swelling, absorbability and degradation compared to non-GO-based bioink. Additionally, these biomaterials showed lower cytotoxicity. Due to its properties, it is recommended to use bioink containing GO for bioprinting functional tissue models with the vascular system, e.g., for testing drugs or hard tissue models.

Abstract:In this research, carboxymethyl cellulose (CMC)/gelatin (Gel)/graphene oxide (GO)-based scaffolds were produced by using extrusion-based 3D printing for cardiac tissue regeneration. Rheological studies were conducted to evaluate the printability of CMC/Gel/GO inks, which revealed that CMC increased viscosity and enhanced printability. The 3D-printed cardiac patches were crosslinked with N-(3-dimethylaminopropyl)-n'-ethylcarbodiimide hydrochloride (EDC)/ N-hydroxysuccinimide (NHS) (100:20 mM, 50:10 mM, 25:5 mM) and then characterized by mechanical analysis, electrical conductivity testing, contact angle measurements and degradation studies. Subsequently, cell culture studies were conducted to evaluate the viability of H9C2 cardiomyoblast cells by using the Alamar Blue assay and fluorescence imaging. A high concentration of EDC/NHS (100:20 mM) led to the stability of the patches; however, it drastically reduced the flexibility of the scaffolds. Conversely, a concentration of 25:5 mM resulted in flexible but unstable scaffolds in PBS solution. The suitable EDC/NHS concentration was found to be 50:10 mM, as it produced flexible, stable, and stiff cardiac scaffolds with high ultimate tensile strength (UTS). Mechanical characterization revealed that % the strain at break of C15/G7.5/GO1 exhibited a remarkable increase of 61.03% compared to C15/G7.5 samples. The improvement of flexibility was attributed to the hydrogen bonding between CMC, Gel and GO. The electrical conductivity of 3D printed CMC/Gel/GO cardiac patches was 7.0×10-3 S/cm, demonstrating suitability for mimicking the desired electrical conductivity of human myocardium. The incorporation of 1 wt% of GO and addition of CMC concentration from 7.5 wt % to 15 wt % significantly enhanced relative % cell viability. Overall, although this research is at its infancy, CMC/Gel/GO cardiac patches have potential to improve the physiological function of cardiac tissue.

Abstract:Decellularized extracellular matrix (dECM) has emerged as an exceptional biomaterial that effectively recapitulates the native tissue microenvironment for enhanced regenerative potential. Although various dECM bioinks derived from different tissues have shown promising results, challenges persist in achieving high-resolution printing of flexible tissue constructs because of the inherent limitations of dECM's weak mechanical properties and poor printability. Attempts to enhance mechanical rigidity through chemical modifications, photoinitiators, and nanomaterial reinforcement have often compromised the bioactivity of dECM and mismatched the desired mechanical properties of target tissues. In response, this study proposes a novel method involving a tissue-specific rheological modifier, gelatinized dECM. This modifier autonomously enhances bioink modulus pre-printing, ensuring immediate and precise shape formation upon extrusion. The hybrid bioink with GeldECM undergoes a triple crosslinking system—physical entanglement for pre-printing, visible light photocrosslinking during printing for increased efficiency, and thermal crosslinking post-printing during tissue culture. A meticulous gelatinization process preserves the dECM protein components, and optimal hybrid ratios modify the mechanical properties, tailoring them to specific tissues. The application of this sequential multiple crosslinking designs successfully yielded soft yet resilient tissue constructs capable of withstanding vigorous agitation with high shape fidelity. This innovative method, founded on mechanical modulation by GeldECM, holds promise for the fabrication of flexible tissues with high resilience.

Abstract:Flexible and soft bioelectronics used on skin tissue have attracted attention for the monitoring of human health. In addition to typical metal-based rigid electronics, soft polymeric materials, particularly conductive hydrogels, have been actively developed to fabricate biocompatible electrical circuits with a mechanical modulus similar to biological tissues. Although such conductive hydrogels can be wearable or implantable in vivo without any tissue damage, there are still challenges to directly writing complex circuits on the skin due to its low tissue adhesion and heterogeneous mechanical properties. Herein, we report cellulose-based conductive hydrogel inks exhibiting strong tissue adhesion and injectability for further on-skin direct printing. The hydrogels consisting of carboxymethyl cellulose, tannic acid, and metal ions (e.g., HAuCl4) were crosslinked via multiple hydrogen bonds between the cellulose backbone and tannic acid and metal-phenol coordinate network. Owing to this reversible non-covalent crosslinking, the hydrogels showed self-healing properties and reversible conductivity under cyclic strain from 0 to 400%, as well as printability on the skin tissue. In particular, the on-skin electronic circuit printed using the hydrogel ink maintained a continuous electrical flow under skin deformation, such as bending and twisting, and at high relative humidity of 90%. These printable and conductive hydrogels are promising for implementing structurally complicated bioelectronics and wearable textiles.

Abstract:The conductive and hemostatic cryogel capable of mimicking skeletal muscle's conductivity and extracellular matrix component presents huge potential for in situ skeletal muscle tissue engineering. Herein, elastic and conductive nanocomposite cryogels were synthesized using amide bond crosslinked gelatin and poly(tannic acid) reduced graphene oxide () via cryopolymerization. The cryogels possessed excellent elasticity, blood-trigged shape memory and 3D macroporous structure. The incorporation of enhanced the mechanical property and conductivity of the cryogel, and endowed the cryogels with anti-oxidation, and without disturbing the elasticity and macroporous structure. Besides, the improved the fast degradation of gelatin cryogel to prolong its service life. The cell proliferation and myogenic differentiation of C2C12 myoblasts during 3D culture were significantly improved by the conductive cryogel scaffold. Especially, the cryogel with 4 mg/mL presented the best cell proliferation and myogenic differentiation, and significantly improved the repair efficiency of volumetric muscle loss. Furthermore, it had better hemostatic efficiency than those of commercial gelatin sponge and gelatin cryogel. This study provides a new strategy to develop highly elastic, hemostatic and conductive scaffolds, which integrates hemostasis and tissue repair to promote skeletal muscle or other electrical signal responsive soft tissue repair via in situ tissue engineering approach.
