Abstract:In order to monitor the process of cellular metabolism and electrophysiology,we developed a novel biochip,on which interdigited electrodes(IDE),microelectrode arrays(MEA) and light-addressable potentiometric sensor(LAPS) were integrated.It can be used in real time detection of cell-electrode impedance,extracellular field potential and cellular ionic metabolism.In the design,the three parts of functions were integrated in a multi-channel way.The size of electrodes,inter-electrode distance and surface properties of electrodes were optimized to reduce the electric disturbance between electrodes and in the mean time to improve performance.In the fabrication,the SiO2 layer was used as the insulation layer of MEA and IDE as well as the protective layer of LAPS,by which three types of sensors were integrated onto a single silicon wafer compatibly.The testing result showed that this integrated cellular chip could provide more than a single parameter with similar performance of separately fabricated chips.Above all,it would be a potential platform to study the electrophysiology and metabolism of cells,and further the pharmacological assays and bionic research.

Abstract:Bio-cellular chips have been important tools to detect extracellular physiology in post-gene era,but most of these tools are confronted with the embarrassments of mono-function,unstable parameters,low S/N ratio and difficulty of repetition.However,the integrated chip can transform the electrical,chemical and motile signals of cells into detectable ones by the integration and network of different sensor units with on-line analysis.The research on a novel integrated cellular physiologic sensor is reported in this article,which includes Micro-electrodes Array Sensors(MEAS),Interdigitated Array(IDA) for impedimetric analysis,and Light-addressable Potentiometric sensors(LAPS),etc.to record the extracellular metabolism chemical and biological substances,action potential and livable characteristics of living cells.The cell-sensor interfacial model of each unit was first introduced,the characteristic curves were then measured,and the fundamental electro-physiological cellular analysis was finally tested and analyzed.The average noise level of MEA is about 80 μV after measuring solution was added,the chip is biocompatible for cell culture and suitable for AP measurement;cultured kidney cells on the surface of IDA chip will cause an cellular impedance change of 12%～16% when exposed to 5-Fluororacil;and the sensitivity of H+-LAPS is 50.65 mV/pH.When kidney cells were cultured on LAPS and exposed to high concentration of 5-Fluororacil,the normal metabolism of cells will be affected,which was similar with the IDA results.This research will be interesting and useful for multi-functionalized cellular detecting platform and facilitate a lot to the lab-on-chip research in near future.
An integrated micro-chip for cell array positioning, in-situ impedance measurement and electroperation

Abstract:This paper reports a biocompatible microchip integrating function of dielectrophoresis (DEP)-based cell array positioning with in-situ real-time impedance measurement and electroporation (EP) of living cells. The DEP-based cell manipulation [1], impedance measurement [2], and cell electroporation [3] are commonly-used label-free techniques for studying of living cells, but they are generally applied separately and rare to be integrated in one chip due to complex structure and crosstalk among them. In this paper we present a novel microchip, in which the cells are manipulated and trapped onto the electrode array (4×8). The impedance spectroscopy of individual cell trapped on electrode array can be measured in situ and the trapped cells can be also electroporated with a high transfection rate (about 90%).

Abstract:These days, multiplex assay with diverse functions on a single chip has become more and more imperative for biological cell research. Multipoint and multistep manipulation for single cells on a chip plays a significant role for cell characterization, immunoassays, and rare cell isolation, etc. In this article, a novel dielectrophoresis (DEP)-based manipulation method is proposed to flexibly move and position cells on a chip via applying various electrical signals onto microelectrodes. By modulating phases and amplitudes of alternating current (ac) signals applied onto the microelectrodes, single cells can be controllably moved from one position to another along diverse directions on a chip. Quantitative analysis is conducted for position and direction controls via simulation, which are validated through experiments. With this flexible manipulation method, single-cell biophysical parameters can be estimated in situ by moving the cell, and as an example, single HeLa and MCF-7 cells are measured. This method allows an efficient and flexible transportation of single cells in lab-on-a-chip systems and provides a fundamental platform for multioriented and multipoint manipulation. The chip is easy to scale up by means of array design for more multifunctional cell assays.

Abstract:Bioanalysis at a single-cell level has yielded unparalleled insight into the heterogeneity of complex biological samples. Combined with Lab-on-a-Chip concepts, various simultaneous and high-frequency techniques and microfluidic platforms have led to the development of high-throughput platforms for single-cell analysis. Dielectrophoresis (DEP), an electrical approach based on the dielectric property of target cells, makes it possible to efficiently manipulate individual cells without labeling. This review focusses on the engineering designs of recent advanced microfluidic designs that utilize DEP techniques for multiple single-cell analyses. On-chip DEP is primarily effectuated by the induced dipole of dielectric particles, (i.e., cells) in a non-uniform electric field. In addition to simply capturing and releasing particles, DEP can also aid in more complex manipulations, such as rotation and moving along arbitrary predefined routes for numerous applications. Correspondingly, DEP electrodes can be designed with different patterns to achieve different geometric boundaries of the electric fields. Since many single-cell analyses require isolation and compartmentalization of individual cells, specific microstructures can also be incorporated into DEP devices. This article discusses common electrical and physical designs of single-cell DEP microfluidic devices as well as different categories of electrodes and microstructures. In addition, an up-to-date summary of achievements and challenges in current designs, together with prospects for future design direction, is provided.

Abstract:This paper reports a novel microarray chip for in-situ , real-time and selective electroporation on individual cells integrated with cell positioning and impedance monitoring. An array of quadrupole-electrode units (termed positioning electrodes) and pairs of planar center electrodes located at the centers of each quadrupole-electrode unit were fabricated on the chip. The positioning electrodes are used to trap and position living cells onto the center electrodes based on negative dielectrophoresis (nDEP). The center electrodes are used for in-situ cell electroporation and also used to measure cell impedance for monitoring cellular dynamics in real time. Controllably selective electroporation and electrical measurement on the cells in array are realized. We present an evidence of selective electroporation through use of fluorescent dyes. Subsequently we use in-situ and real-time impedance measurement to monitor the process, which demonstrates the dynamic behavior of the cell electroporation. Finally, we show the use of this device to perform successful transfection onto individual HeLa cells with vector DNA encoding a green fluorescent.

Abstract:Cells are the fundamental unit of life. Owing to cellular heterogeneity, traditional biochemical assays which analyze cells in bulk often overlook the rich information available when single cells are studied. Accurate and reliable determination of the chemical composition in individual cells would greatly improve the probability of discriminating infected cells from healthy ones and provide a solid foundation on study and development in various fields including biochemistry, medicine, and pathology clinic. Microfluidic chip system for single-cell analysis is now attracting broad interests. The ability of integrating the whole process on one microfluidic chip, including single cell injection, lysis, separation and detection of cellular constituents by chip-based CE, offers a new technical platform for single cell analysis. This article provides a review on recent advances in determining intracellular contents in individual cells using microfluidic devices. Focus areas include on-chip single cell manipulation by means of electroosmotic flow (EOF), pressure-driven flow combined with EOF and optical tweezer (OT) to perform sequentially single cell injection, lysis, separation and detection of cellular constituents by chip-based CE with highly sensitive detection. The approaches on on-chip derivatization are also addressed.
Controllably Moving Individual Living Cell in an Array by Modulating Signal Phase Difference Based on Dielectrophoresis.

Abstract:This paper reports a novel dielectrophoresis (DEP) based method for manipulating individual living cells by modulating phase difference of electrical signals applied on DEP electrodes. A novel microchip with an array structure is also proposed, consisting of a plurality of quadrupole-electrode units patterned into array on a glass substrate with a pair of center electrodes locating at the center of each quadrupole-electrode unit. Living cells can be trapped and positioned at the center of each quadrupole-electrode unit by using negative DEP (nDEP) manipulation and form an array. The trapped cells in the array can be controllably moved from one position to another and even from one of quadrupole-electrode units to adjacent unit by changing the phase difference of the signals applied on the two pairs of opposite electrodes in each quadrupole-electrode unit. The microchip allows an efficient and flexible manipulation of individual living cells that can be applied to study single cells. The experiments are performed to verify that different types of cells (MCF-7 cell and HeLa cell) can be effectively distinguished between each other using the method without label and fluorometric measurements. An identification of individual living cell from dead cells is also well demonstrated.

Abstract:The physical manipulation of biological cells is a key point in the development of miniaturized systems for point-of-care analyses. Dielectrophoresis (DEP) has been reported by several laboratories as a promising method in biomedical research for label-free cell manipulation without physical contact, by exploiting the dielectric properties of cells suspended in a microfluidic sample, under the action of high-gradient electric fields. In view of a more extended use of DEP phenomena in lab-on-chip devices for point-of-care settings, we have developed a portable instrument, integrating on the same device the microfluidic biochip for cell manipulation and all the laboratory functions (i.e., DEP electric signal generation, microscopic observation of the biological sample under test and image acquisition) that are normally obtained by combining different nonportable standard laboratory instruments. The nonuniform electric field for cell manipulation on the biochip is generated by microelectrodes, patterned on the silicon substrate of microfluidic channels, using standard microfabrication techniques. Numerical modeling was performed to simulate the electric field distribution, quantify the DEP force, and optimize the geometry of the microelectrodes. The developed instrument includes an electronic board, which allows the control of the electric signal applied to electrodes necessary for DEP, and a miniaturized optical microscope system that allows visual inspection and eventually cell counting, as well as image and video recording. The system also includes the control software. The portable and integrated platform described in this work therefore represents a complete and innovative solution of applied research, suitable for many biological applications.

Abstract:Chips-based platforms intended for single-cell manipulation are considered powerful tools to analyze intercellular interactions and cellular functions. Although the conventional cell co-culture models could investigate cell communication to some extent, the role of a single cell requires further analysis. In this study, a precise intercellular interaction model was built using a microelectrode array [microelectrode array (MEA)]-based and dielectrophoresis-driven single-cell manipulation chip. The integrated platform enabled precise manipulation of single cells, which were either trapped on or transferred between electrodes. Each electrode was controlled independently to record the corresponding cellular electrophysiology. Multiple parameters were explored to investigate their effects on cell manipulation including the diameter and depth of microwells, the geometry of cells, and the voltage amplitude of the control signal. Under the optimized microenvironment, the chip was further evaluated using 293T and neural cells to investigate the influence of electric field on cells. An examination of the inappropriate use of electric fields on cells revealed the occurrence of oncosis. In the end of the study, electrophysiology of single neurons and network of neurons, both differentiated from human induced pluripotent stem cells (iPSC), was recorded and compared to demonstrate the functionality of the chip. The obtained preliminary results extended the nature growing model to the controllable level, satisfying the expectation of introducing more elaborated intercellular interaction models.

Abstract:The design and fabrication of a self-digitization dielectrophoretic (SD-DEP) chip with simple components for single-cell manipulation and downstream nucleic acid analysis is presented. The device employed the traditional DEP and insulator DEP to create the local electric field that is tailored to approximately the size of single cells, enabling highly efficient single-cell capture. The multistep procedures of cell manipulation, compartmentalization, lysis, and analysis were performed in the integrated microdevice, consuming minimal reagents, minimizing contamination, decreasing lysate dilution, and increasing assay sensitivity. The platform developed here could be a promising and powerful tool in single-cell research for precise medicine.

Abstract:Biochemical analysis is crucial in understanding mechanism of life activities and giving biological insights into life process. With inherent merits in flexible design, microscale of operation, good incorporation with other techniques for manipulation and detection, microfluidic chip has introduced new paradigms in biochemical analysis field. Considering the explosive development of microfluidics over the past decades, in this review, we summarized recent advances in biochemical analysis on microfluidic platforms. Highlight was put on the integrated technologies such as optical, electrical, acoustic and magnetic techniques. Focus also was given on relative applications in biomimetics, drug screening, biomolecular detection, single cell and stem cell analysis.
A Biocompatible Microchip and Methodology for Efficiently Trapping and Positioning Living Cells into Array Based on Negative Dielectrophoresis

Abstract:We present a microchip and trapping methodology based on negative dielectrophoresis (nDEP), whereby living cells were manipulated and positioned into an array with high trapping efficiency while maintaining good viability. The main factors that ensured good viability of cells were investigated including temperature of medium, extra transmembrane potential on cells, and electrolysis effect in DEP-based trapping. Optimum DEP conditions for the microchip were determined by considering both biocompatibility and trapping efficiency. Experiments demonstrated that under a voltage of 3.6–4 Vpp and at a frequency of 100 kHz, HeLa cells could be trapped and positioned into an array in less than 10 s while maintaining good viability. The normal adherence morphology and fluorescence of the cells, dyed with propidium iodide and Calcein-AM, were observed and verified the biocompatibility of the microchip and trapping methodology.

Abstract:Magnetic beads manipulation in microfluidic chips is a promising research field for biological application, especially in the detection of biological targets. In this review, we intend to present a thorough and in-depth overview of recent magnetic beads manipulation in microfluidic chips and its biological application. First, we introduce the mechanism of magnetic manipulation in microfluidic chip, including force analysis, particle properties, and surface modification. Then, we compare some existing methods of magnetic manipulation in microfluidic chip and list their biological application. Besides, the suggestions and outlook for future developments in the magnetic manipulation system are also discussed and summarized.

Abstract:The real-time screening of biomolecules and single cells in biochips is extremely important for disease prediction and diagnosis, cellular analysis, and life science research. Barcode biochip technology, which is integrated with microfluidics, typically comprises barcode array, sample loading, and reaction unit array chips. Here, we present a review of microfluidics barcode biochip analytical approaches for the high-throughput screening of biomolecules and single cells, including protein biomarkers, microRNA (miRNA), circulating tumor DNA (ctDNA), single-cell secreted proteins, single-cell exosomes, and cell interactions. We begin with an overview of current high-throughput detection and analysis approaches. Following this, we outline recent improvements in microfluidic devices for biomolecule and single-cell detection, highlighting the benefits and limitations of these devices. This paper focuses on the research and development of microfluidic barcode biochips, covering their self-assembly substrate materials and their specific applications with biomolecules and single cells. Looking forward, we explore the prospects and challenges of this technology, with the aim of contributing toward the use of microfluidic barcode detection biochips in medical diagnostics and therapies, and their large-scale commercialization.

Abstract:We demonstrate a method of concentrating and patterning of biological cells on a chip, exploiting the confluence of electric and thermal fields, without necessitating the use of any external heating or illuminating sources. The technique simply employs two parallel plate electrodes and an insulating layer over the bottom electrode, with a drilled insulating layer for inducing localized variations in the thermal field. A strong induced electric field, in the process, penetrates through the narrow hole and generates highly nonuniform heating, which in turn, results in gradients in electrical properties and induces mobile charges to impose directional fluid flow. The toroidal vortices, induced by secondary electrokinetic forces originating out of temperature-dependent electrical property variations, transport the suspended cells toward a hot-spot site of the chip, for rapid concentrating and patterning into different shaped clusters based on predesigned conditions, without exceeding safe temperature limits that do not result in damage of thermally labile biological samples. We characterize the efficacy of the cell trapping process for two different biological entities, namely, Escherichia coli bacteria and yeast cells. These results have importance toward developing biomedical microdevices for drug discovery, antibiotic resistance assessment, and medical diagnostics.

Abstract:In basic cell biology research and drug discovery, it is important to rapidly introduce genes, proteins or drug compounds into cells without permanent damage. Here, we report a three dimensional SU-8 micro-well structure sandwiched with an indium tin oxide (ITO) electrode-covered slide from the top and an individually addressable array of microelectrodes on the bottom to allow parallel delivery of exogenous molecules into various cells in a spatially specific manner. A positive dielectrophoretic force was selectively applied by energizing appropriate electrodes to capture the dispersed cells at the bottom electrode, while the micro-wells were designed to confine cells in situ when the positive dielectrophoretic force is removed. The combination of spatial positive dielectrophoresis (pDEP) and micro-wells made it possible to construct cell microarrays with specific patterns. Once the cells become attached to the electrodes, different plasmids can be introduced sequentially for selective electroporation. The present cell arraying-assisted electroporation chip integrates a pDEP-assisted cell positioning function with selective electroporation to provide a simple and efficient method for gene transfer. This platform is ideal for high throughput screening of compounds in parallel and thus holds promise for applications in cellular and molecular research.

Abstract:Presented is the design, fabrication, and evaluation of 72-channel microcircuit electrode arrays (biochips) built to interface neurons and computers. Contributions include a detailed mathematical model of the electrode/electrolyte interface and subsequent electrode design optimization (in terms of maximum signal-to-noise ratio). Electrode impedance measurements were obtained with the electrodes submerged in electrolytic neural solution. Large sample results indicate a 7% discrepancy between theoretical and measured impedances. In addition to the engineering efforts, biological cell culturing techniques were developed to enable the growth of synaptically interconnected, electrically excitable cells on the biochip. Both dissociated invertebrate neurons and clonal mammalian cell systems were employed. Together these contributions enabled microvolt signals to be recorded from individual neurons grown and synaptically interconnected on the biochip surface. Also recorded were neuronal responses to on-chip stimulation. Consequently, the biochip technology presented represents a step towards a long-term, noninvasive, multisite electrical stimulation and recording capability necessary for extracting the mechanisms governing the way living neural networks connect, integrate, and self-organize.

Abstract:This paper reports a method for label‐free single‐cell biophysical analysis of multiple cells trapped in suspension by electrokinetic forces. Tri‐dimensional pillar electrodes arranged along the width of a microfluidic chamber define actuators for single cell trapping and selective release by electrokinetic force. Moreover, a rotation can be induced on the cell in combination with a negative DEP force to retain the cell against the flow. The measurement of the rotation speed of the cell as a function of the electric field frequency define an electrorotation spectrum that allows to study the dielectric properties of the cell. The system presented here shows for the first time the simultaneous electrorotation analysis of multiple single cells in separate micro cages that can be selectively addressed to trap and/or release the cells. Chips with 39 micro‐actuators of different interelectrode distance were fabricated to study cells with different sizes. The extracted dielectric properties of HeLa, HEK 293, and human immortalized T lymphocytes cells were found in agreements with previous findings. Moreover, the membrane capacitance of M17 neuroblastoma cells was investigated and found to fall in in the range of 7.49 ± 0.39 mF/m2.This article is protected by copyright. All rights reserved
