Abstract:The hollow fiber membranes (HFMs) were widely used as tissue engineering scaffold The efficacy of the HFMs were affected by the porosity of the HFMs. In this study, the HFMs were prepared with dry-jet wet spinning method. In order to improve the performance of the HFMs, different spinning I parameters were applied to systematically investigating the effects of the fabrication conditions on the porosity of the HFMs. A 1-D diffusion model was set to numerically study the volume fraction distribution along the wall thickness. The porosity of the HFMs show a significant decrease in the porosity as the dope concentration increases. The morphology analyses showed that a higher dope concentration leads to a denser porous structure which result in a lower porosity. The results were discussed based on the phase inversion process and the I-D model was built to numerically analyze the polymer volume fraction distribution in the HFMs cross section. This study can help to better understand the effects of the different fabrication conditions on the morphology of the HFMs.

Abstract:Recently the semi-permeable hollow fiber membrane (HFM) is finding promising applications in promoting axonal Outgrowth for nerve repair and regeneration. It is of interest to model the phase inversion-based HFM fabrication process and control the fabricated HFM geometry. The effect of gravity and surface tension which is frequently ignored in general fiber spinning should be carefully addressed in HFM fabrication modeling. Both the volume Of fluid (VOF) method and the scale analysis have been applied to appreciate the effect of gravity and surface tension on the HFM geometry profile. The VOF method-based Simulation results reveal that both the gravity and/or Surface tension significantly reduce the predicted radii/diameters, while the scale analysis reveals that the gravity or Surface tension affects the HFM fabrication process dynamics. Both the approaches warrant the need of including the gravity and surface tension in HFM fabrication process modeling.

Abstract:Alginate microfibres were fabricated by a simple microfluidic spinning device consisting of a coaxial flow. The inner profile and spinnability of polymer were analysed by rheology study, including the analysis of viscosity, storage modulus and loss modulus. The effect of spinning parameters on the morphological structure of fibres was studied by SEM, while the crystal structure and chemical group were characterized by FTIR and XRD, respectively. Furthermore, the width and depth of grooves on the fibres was investigated by AFM image analysis and the formation mechanism of grooves was finally analysed. It was illustrated that the fibre diameter increased with an increase in the core flow rate, whereas on the contrary of sheath flow rate. Fibre diameter exhibited an increasing tendency as the concentration of alginate solution increased, and the minimum spinning concentration of alginate solution was 1% with the finest diameter being around 25 µm. Importantly, the grooved structure was obtained by adjusting the concentration of solutions and flow rates, the depth of groove increased from 278.37 ± 2.23 µm to 727.52 ± 3.52 µm as the concentration varied from 1 to 2%. Alginate fibres, with topological structure, are candidates for wound dressing or the engineering tissue scaffolds.

Abstract:Material extrusion, a filament-based three-dimensional (3D) bioprinting technique, is commonly adopted to fabricate many complex constructs for its high efficiency, compatibility with a variety of biomaterials, and easy realization. During extrusion bioprinting, the morphology (including the shape and size) of extruded filaments is of great interest since the filaments are the basic building blocks for printed 3D structures and the filament morphology determines the printing resolution, surface quality, and part mechanical strength. The objective of this study is to computationally analyze the printing performance of the jammed gelatin microgel-based composite ink during extrusion in terms of the filament cross-sectional morphology and the influence of ink yield-stress fluid property on the structural printability. As seen from the rheological measurements, the jammed gelatin microgel composite ink is a viscoplastic fluid with the shear-thinning property, and its yield-stress fluid property enables it for self-supported printing applications. The ink printing process has been computationally modeled by using a fitted Herschel–Bulkley model to simulate the behavior of the jammed gelatin microgel composite ink for the first time, resulting in good modeling performance. In particular, the filament cross-sectional morphology under different printing conditions has been satisfactorily modeled. It is found that the cross-sectional shape turns flat rectangular under a small normalized gap distance (less than 0.6). Furthermore, the achievable maximum length (without collapse) of the jammed gelatin microgel-based composite ink, deposited both between two supporting substrates and over a supporting substrate, has been satisfactorily estimated.
The polymer melt swelling behavior in micro-extrusion process for preparation of continuous carbon fiber reinforced PP prepreg filament

Abstract:The micro-extrusion process for polymer melt has been increasingly used for the production of high value-added prepreg filaments with applications in the fields of fused deposition modeling (FDM). In this study, the effect of characteristic scale, defined as the gap of die land in an annular micro-extrusion die, on the extrudate swelling behaviour of viscoelastic melt is analyzed through numerical simulations and micro-extrusion experiments. The results show that the swelling behaviour displays an obvious dependence on the characteristic scale. An increase in the characteristic scale reduces the swell ratio and retards the process to reach the equilibrium state. In contrast, a decrease in the characteristic scale results in a larger magnitude of change in velocity field and faster relaxation development of stress field. The location of the maximum velocity layer for the laminar flow gradually deviates from the geometric center of channel toward the wall of mandrel with the increase in the characteristic scale. It is imperative to consider the complicated swelling behavior and remarkable viscoelastic effect simultaneously in micro-extrusion process.
Porous Morphology and Mechanical Properties of Poly(lactide-Co-glycolide) Hollow Fiber Membranes Governed by Ternary-Phase Inversion

Abstract:Due to the controllable biodegradability and good biocompatibility, poly(lactide-co-glycolide acid) (PLGA) hollow fiber membranes (HFMs) have been successfully used for various biomedical applications, where the morphological and mechanical characteristics of HFMs are of crucial importance. This study focused on the effects of fabrication conditions on the morphology and mechanical performance of PLGA HFMs, which were manufactured by a phase inversion-based dry-jet wet spinning process. The cross-sectional morphology of HFMs was found to be significantly influenced by PLGA concentration of dope fluid. A 5-layered microstructure with macrovoids varied to a 6-layered microstructure with sponge-like microvoids when the PLGA concentration increased from 14% to 22%. Also, Young's modulus and compressive modulus of these PLGA HFMs were found to dramatically increase with PLGA concentration. To understand the underlying mechanism of morphological formation in PLGA HFMs, a physical mass transfer model was developed to describe the mutual diffusion process of PLGA/dimethyl sulfoxide (DMSO)/ water ternary system. This model was able to quantify the PLGA distribution within the HFM cross sections by considering the interaction terms of polymer/solvent/ nonsolvent in the governing equations, which was validated by the overall agreement between the experimental and numerical results. The present study should be helpful for understanding the formation mechanism of porous morphology during the phase inversion-based dry-jet wet spinning process, which may guide the optimization of PLGA HFMs for their applications as tissue engineered scaffolds.
A Systematic Thermal Analysis for Accurately Predicting the Extrusion Printability of Alginate-Gelatin-Based Hydrogel Bioinks

Abstract:Three-dimensional (3D) bioprinting has significant potential for addressing the global problem of organ shortages. Extrusion printing is a versatile 3D bioprinting technique, but its low accuracy currently limits the solution. This lack of precision is attributed largely to the complex thermal and dynamic properties of bioinks and makes it difficult to provide accurate estimations of the printed results. It is necessary to understand the relationship between printing temperature and materials' printability to address this issue. This paper proposes a quantitative thermal model incorporating a system's printing temperatures (syringe, ambient, and bioink) to facilitate accurate estimations of the printing outcomes. A physical model was established to reveal the relationship between temperature, pressure, and velocity in guiding the printing of sodium alginate-gelatin composite hydrogel (a popular bioink) to optimize its extrusion-based printability. The model considered the phenomenon of bioink die swells after extrusion. A series of extrusion experiments confirmed that the proposed model offers enhanced printing outcome estimations compared with conventional models. Two types of nozzles (32- and 23-gauge) were used to print several sets of lines with a linewidth step of 50 mm by regulating the extrudate's temperature, pressure, and velocity separately. The study confirmed the potential for establishing a reasonable, accurate open-loop linewidth control based on the proposed optimization method to expand the application of extrusion-based bioprinting further.

Abstract:Numerical simulation of the high-speed fiber spinning process was carried out using Polyflow software, based on the models proposed by Doufas et at. Calculation predicts not only the variation of parameters along the spinline, but also the radially distribution of flow field variables, Such as temperature, crystallinity and molecular structure. Considering combined effects on the process Such as flow-induced crystallization, viscoelasticity, filament cooling, air drag, inertia, surface tension and gravity, the Simulated material flow behaviors are consistent with that observed for semi-crystalline polymer under various spinning conditions. The structure change of polymer coils in the necking region described by the evolution of conformation tensor is also investigated. Moreover, three-dimensional (3D) simulation approach was also used to predict the fiber shape given the spinneret geometry, combined with the die design capability. It is helpful to design die geometry for generating fibers with complex cross-sections.

Abstract:Ideal tissue engineering scaffolds need interconnected pores and high porosity to enable cell survival, migration, proliferation, and differentiation. However, obtaining a high-resolution structure is difficult with traditional one-temperature control fused deposition modeling (FDM). In this study, we propose a dual-temperature control method to improve printability. A numerical model is developed in which the viscosity is a function of temperature and shear rate to study the influence of two different temperature control modes. Quantitative tests are used to assess filament formation and shape fidelity, including one-dimensional filament printing, deposition at corners, fusion, and collapse. By using dual-temperature control, the width of the deposited poly(ε-caprolactone) filament is reduced to 50 μm. The comparative results of both the experimental method and numerical simulation suggest that the dual-temperature control FDM can manufacture spatially arranged constructs and presents a promising application in tissue engineering.

Abstract:Alginate fibers were prepared by extruding a concentrated sodium alginate solution into calcium chloride coagulation bath. The rheological behavior of different concentrations of sodium alginate solutions were investigated to evaluate the spinnability. The effect of dope concentration on the morphologies and mechanical properties of resulting alginate fibers were examined. The results showed that sodium alginate solutions were pseudoplastic fluid and had good spinnability with concentration ranging in 1.5%-3.5%. Moreover, the morphology of obtained fibers became smoother with increasing sodium alginate concentrations. When the sodium alginate concentration was 3.5%, the tensile strength and elongation at break of alginate fibers reach to 13.6±1.2 cN/tex and 8±0.5%, respectively.

Abstract:Microfluidic wet spinning has gained increasing interest in recent years as an alternative to conventional wet spinning by offering higher control in fiber morphology and a gateway for the development of multi-material fibers. Conventionally, microfluidic chips used to create such fibers are fabricated by soft lithography, a method that requires both time and investment in necessary cleanroom facilities. Recently, additive manufacturing techniques were investigated for rapid and cost-efficient prototyping. However, these microfluidic devices are not yet matching the resolutions and tolerances offered by soft lithography. Herein, we report a facile and rapid method using selected arrays of hypodermic needles as templates within a silicone elastomer matrix. The produced microfluidic spinnerets display co-axially aligned circular channels. By simulation and flow experiments, we prove that these devices can maintain laminar flow conditions and achieve precise 3D hydrodynamic focusing. The devices were tested with a commercial polyurethane formulation to demonstrate that fibers with desired morphologies can be produced by varying the degree of hydrodynamic focusing. Thanks to the adaptability of this concept to different microfluidic spinneret designs—as well as to its transparency, ease of fabrication, and cost-efficient procedure—this device sets the ground for transferring microfluidic wet spinning towards industrial textile settings.

Numerical modeling of hydrogel scaffold anisotropy during extrusion-based 3D printing for tissue engineering

Abstract:Extrusion-based 3D printing is a recent and widely used tissue engineering tool that provides precise 3D control of bioinks to create organ-size biomaterial-based objects composed of hierarchically organized cellularized scaffolds. The internal organization of scaffold constituents should mimic the structural anisotropy of targeted tissue to stimulate proliferative cellular behavior during 3D cell culture. Both the choice of polymers constituting the bioink and the topological properties during the extrusion process greatly influence the structural anisotropy and cellular response of tissue engineering constructs. The bioink used in our study was a hydrogel made of three constituents: fibrinogen, alginate and gelatin. These components provide biocompatibility, printability and conservation of 3D shape after printing. The topological properties in flowing polymers are dictated by macromolecule conformation i.e. their orientation and degree of stretch. In this study, we used the micro-macro approach to describe the orientation state of hydrogel macromolecules during the extrusion process, offering a two-scale description of fluid behavior. The goal of our study was to use the Fokker-Planck equation, which describes the evolution of probability distribution function over time, to represent the real state of the constituent population in the representative elementary volume within a hydrogel during extrusion-based 3D printing. Our data suggest that for a tubular nozzle syringe, constituent orientation is driven by a high shear rate, which overcomes the fluid rheological behavior. Also, the interaction coefficient, which represents the microscopic interaction between fluid particles overcomes hydrogel behavior for constituent orientation in the prediction model.

3D Printing Process for Hydro Gel with the Three-dimensional Micro Tubes to Mimic Vascular Network

Abstract:A coaxial nozzle printing system is built to investigate the influence of ferric chloride solution to the mechanical strength and shape structure of sodium alginate hollow tube. The optimization process of 3D printing is further determined. The concentration of 2%- 4% sodium alginate and the concentration of 4% of calcium chloride is used to print hollow tubes through the coaxial probe. The concentration of 0.25% of atomizing ferric chloride solution is used to reinforced to the calcium alginate hollow tubes. Mechanics experiment prove that the Young's modulus of the reinforced hollow tube (0.4677 MPa±0.279 MPa(2% sodium alginate), 2.0153 MPa±0.221 MPa(3% sodium alginate),11.684 MPa±0.332 MPa(4% sodium alginate)) are 5.4 times(2%),14.2 times(3%) and 43.3 times(4%) as the sodium alginate hollow tubes. The reinforced hollow tubes are used to print complex vascular networks such as lattice structure and spiral structure. The vascular networks are encapsulated with hydro gel solution to get the biological scaffold. Perfusion experiment and cell toxicity test show that the scaffolds had good biocompatibility and it is feasible to be effectively transported cells culture medium. Therefore, printed scaffolds can meet the demand of manufacturing large soft tissues in tissue engineering.
Study on filament deflection in lattice porous structures fabricated through direct ink writing

Abstract:Direct ink writing (DIW) technology supersedes traditional mold‐based forming methods, significantly enhancing the fabrication of personalized and customized products with complex structures. This technology particularly excels in achieving precise control over the porosity of porous constructs. This study employs inorganic Al2O3 as raw material, sodium hexametaphosphate as dispersing agent, and inorganic SiO2 micropowder as binding medium to fabricate lattice porous structures. One challenge encountered is the viscoelastic behavior of the extruded filament. When spanning the unsupported segments of the lower layer, the upper extruded filaments are susceptible to deflection or collapse, adversely affecting the porosity and dimensional fidelity of the final specimens. Experimental results revealed that a larger span and smaller modulus will cause the extruded filament to be more prone to deformation at the midpoint. The introduction of 2 wt% polyethylene glycol as a plasticizer mitigates this issue, ensuring nondeflection of the extruded filaments at a span of 6 mm. The deflection model for the extruded filament about span and modulus identifies the minimum modulus necessary to prevent or minimize deflection under given spans, which closely approximates our experimental findings, offering a valuable framework for guiding the production of high‐precision, porosity‐controlled porous structures.

Abstract:In response to an investigation on the paths of changes in the crystallization and radial differences during the forming process of nascent fibers, in this study, we conducted numerical simulation and analyzed the changes in crystallization mechanical parameters and tensile properties through a fluid dynamics two-phase model. The model was based on the melt-spinning method focusing on melt spinning, the environment of POLYFLOW, and the method of joint simulation, coupled with Nakamura crystallization kinetics, including the development of process collaborative parameters, stretch-induced crystallization, viscoelasticity, filament cooling, gravity term, inertia, and air resistance. Finally, for nylon 6 BHS and CN9987 resin spinning, the model successfully predicted the distribution changes in temperature, velocity, strain rate tensor, birefringence, and stress tensor along the axial and radial fibers and obtained the variation pattern of fibers' crystallinity along the entire spinning process under different stretching rates. Furthermore, we also explored the effects of spinning conditions, including inlet flow rate, winding speeds, and the extrusion temperature, on the fibers' crystallization process and obtained the influence rules of different spinning conditions on fiber crystallization. Knowing the paths of changes in mechanical performance can provide important guidance and optimization strategies for the future industrial preparation of high-performance fibers.

Abstract:Geometry design of spinneret is highly associated with nanofiber production in needleless electrospinning technologies. Current free-liquid-surface spinnerets have disadvantages, including large air/solution contact surface area, uneven electric field and fiber diameter distribution, and poor stability of polymer solution, which prevents further industrial progress. This study presented a strategy from design to simulation and experiment to develop a novel annular slit needleless electrospinning system. The effects of spinneret geometries on electric field distribution and strength were discussed by the finite element method. Numerous experiments were carried out to verify the feasibility of annular slit electrospinning. Main process parameters including solution concentration, applied voltage, and airflow velocity were studied by the response surface method. This novel needleless electrospinning apparatus shows great potential for producing nanofibers on an industrial scale.

Novel microfluidic strategy for the production of sodium alginate fibers with regular inclusions at very high throughput

Abstract:Scalable technologies for the production of bio-compatible complex microfibers with controllable size and composition at competitively high throughput are urgently needed in order to meet the growing demand for such microstructures in pharmaceutical or biomedical applications. Here, we introduce a new in-air microfluidic strategy with throughput of > 1400 ml/h (corresponding to > 17000 m fiber per h). The microfibers of uniform diameter have regular inclusions, which can potentially be used for encapsulating cells into a protecting and nutrient environment, or for finely tuning the release of various actives at individualized doses. With the help of a newly developed prototype, we test seven different liquid combinations and obtain seven types of fibers, whose "dry" diameter range between 73um and 141um. The principle of our approach is to solidify the complex liquid structures generated by the controlled collisions of a drop stream with a continuous liquid jet, in air, via ionic crosslinking. After the stream of water-based droplets, which constitute the inclusions, collides in-air with the alginate-based jet (jet 1), the generated "drops-in-jet" compound is brought in contact with a second jet (jet 2) containing divalent strontium or calcium cations to initiate the solidification. Finally, the fibers are collected via a horizontal spinning plate, let to dry, i.e. to fully equilibrate under controlled conditions, and characterized by their elongation at break and Young's modulus.
Wet Electrospun Alginate/gelatin Hydrogel Nanofibers for 3D Cell Culture

Abstract:Convergence of biological and biofabrication approaches is necessary to progress new biomaterials promoting three-dimensional (3D) cell growth and maturation towards tissue regeneration and integration. Here, we have developed a novel approach to fabricate 3D macroporous, alginate/gelatin hydrogel nanofibers (Alg/GelF-MA), which provide superior cell adhesion, motility, proliferation and maturation. The electrospinning process greatly depends on the ionic strength and viscoelastic behavior of the solution. The polyelectrolyte nature of alginate favors intramolecular bundles over intermolecular entanglement, which hinders its electrospinnability. Electrospinning of alginate was achieved by the aid of a supporting polymer, polyethylene oxide and a surfactant, Pluronic (R) F127. Furthermore, the Ca2+-mediated coagulation process of alginate was realized in situ during wet electrospinning, where the rapid physical crosslink-ability of alginate was applied in conjunction with the jet entrance into the wet electrospinning collector, a coagulation bath. Consequently, the rapid formation of Ca2+-alginate complex stabilized the nanofiber morphology. The low surface tension of the non-solvent ethanol used in the bath prevented fibers from dense packing, thus allowing the generation of 3D macroporous structure favoring cell motility. The subsequent UV-mediated chemical crosslinking further stabilized the gelatin content in the Alg/GelF-MA hydrogel nanofibers. It is demonstrated that the Alg/GelF-MA nanofibers with low cytotoxicity (below 10%) supported an over 8-fold proliferation of mesenchymal stem cells over 5 weeks and supported the maturation of human iPSC-derived ventricular cardiomyocytes, which significantly outperform the cell encapsulated bulk GelF-MA hydrogel. Abstract:Fibrous structures with anisotropic fillers as composites have found increasing interest in the field of biofabrication since they can mimic the extracellular matrix of anisotropic tissues such as skeletal muscle or nerve tissue. In the present work, the inclusion of anisotropic fillers in hydrogel-based filaments with an interpenetrating polymeric network (IPN) was evaluated and the dynamics of such fillers in the composite flow were analyzed using computational simulations. In the experimental part, microfabricated rods (200 and 400 μm length, 50 μm width) were used as anisotropic fillers in extrusion of composite filaments using two techniques of wet spinning and 3D printing. Hydrogels such as oxidized alginate (ADA) and methacrylated gelatin (GelMA) were used as matrices. In the computational simulation, a combination of computational fluid dynamics and coarse-grained molecular dynamics was used to study the dynamics of rod-like fillers in the flow field of a syringe. It showed that, during the extrusion process, microrods are far from being well aligned. Instead, many of them tumble on their way through the needle leading to a random orientation in the fiber which was confirmed experimentally.
