Abstract:A mild and efficient method for the synthesis of 2,5-disubstituted chalcogenophenes is reported. This method involves two steps of reactions via one-pot approach by employing commercially available and cost-effective alkynes and element chalcogen. Further, by using LiNH2 as the promoter and DMSO/EtOH as the solvent, Cu-catalyzed Glaser coupling-cycloaddition processes are dramatically enhanced that a diverse range of chalcogenophenes can be obtained in good to excellent yields under mild conditions. Controlled experiments show that trisulfur radical anion (S3 & sdot;-) is the key intermediate and DMSO/EtOH serves as the hydrogen source, while alcohol may facilitate the transition state. The mild and highly efficient constructions of 2,5-disubstitued thiophenes, selenophenes and tellurophenes are achieved by using a one-pot, two-step protocol that combine Cu-catalyzed Glaser coupling with LiNH2-mediated cycloaddition. And the DMSO/EtOH mixture solvent is found to short the reaction time and improve the reaction yield by promoting the production of S3 & sdot;- and stabilizing the transition state.image

Abstract:A one-pot two-step reaction of 4-aminocoumarin with arylglyoxal monohydrates and p-toluenesulfonates has been developed, which offers 3-alkoxy-substituted chromeno[4,3-b]pyrrol-4(1H)-ones in moderate to good yields. At first, arylglyoxal monohydrates and 4-aminocoumarin were converted into 3-hydroxychromeno[4,3-b]pyrrol-4(1H)-one intermediates in AcOH under reflux. Then the title compounds in an in situ reaction of the intermediates with p-toluenesulfonates catalyzed by DBU in refluxing toluene were obtained.

Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A mild, efficient, and environmentally benign one-pot synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones by a three-component domino reaction of 4-aminocoumarins, arylglyoxal monohydrates, and 1,3-dicarbonyl compounds in refluxing ethanol without the use of catalyst is reported. Abstract:The reaction of o-aminoacetophenones and arylboronic acids catalyzed by copper(II) salts in the presence of pyridine under an O2 atmosphere provides a general and efficient one-pot preparation of biologically interesting acridones. This relay reaction comprises an intermolecular Suzuki cross-coupling, intramolecular oxidative C(sp3)-H amination, and C(sp2)-H activation with simultaneous rearrangement of the generated isatin intermediates. This strategy tolerates both electron-donating and -withdrawing functionalities to afford various acridones in good to excellent yields.

Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:In organic chemistry, dimerization reactions represent a synthetically powerful strategy for the rapid construction of molecular complexity. This study introduces a copper-catalyzed radical dimerization of 1-alkynylnaphthols, enabling chemo- and diastereoselective synthesis of bridged tetrahydro-benzo[ no ]tetraphenone products. The approach boasts the use of an economical catalyst and a green oxidant under mild conditions. Different heteroaryls, such as indole and thiophene, are well-tolerated in the reaction. Control experiments provided valuable insights, revealing a sequence involving the formation of a C1–radical intermediate, intermolecular (3+2) cyclization, and an unusual intramolecular C4–C8′ coupling.

Abstract:We herein disclose a highly efficient one-pot synthetic strategy for dihydrobenzo[a]fluorenes via cascade rhodium(III)-catalyzed ortho-C-H activation/annulation of thiobenzamides with aryl ethynyl ketones and subsequently copper(II)-promoted intramolecular C-H/C-H cross-coupling reactions. Mechanistic investigations suggest that Cu(II) plays two crucial roles by serving as a sulfide scavenger to regenerate the Rh(III) catalyst and promoting the intramolecular C-H/C-H cross-coupling reaction. This protocol greatly streamlines accesses to a variety of appealing tetracyclic benzo[a]fluorene skeletons, which may have potential biological activity and medicinal properties.

Abstract:A series of non-annulated products: trifluoromethylpyrazolone-tethered trisubstituted methane derivatives were synthesized in high yields in acetonitrile solvent catalyzed by NH4OAc via one-pot, three-component reactions of 2-hydroxy-1,4-naphthoquinone, aromatic aldehydes and 1-aryl-3-trifluoromethyl-5-pyrazolone. Treatment of trifluoromethylpyrazolone-tethered trisubstituted methane derivatives with the dehydrating agent SOCl2/Pyridine in acetonitrile afforded the corresponding annulated fused polyheterocyclic products trifluoromethylated benzo[6,7]chromeno[2,3-c]pyrazole-5,10-dione derivatives. Furthermore, an efficient, straightforward protocol has been developed for the synthesis of trifluoromethylated benzo[6,7]chromeno[2,3-c]pyrazole-5,10-dione derivatives at medium yields via one-pot, two-step, three-component reactions. Meanwhile, the structures of typical products were further confirmed by XRD-analysis.

Abstract:A highly regioselective synthesis of 2,3-disubstituted chromen-4-one derivatives is accomplished from readily available internal alkynes and 2-methoxybenzoyl chlorides. The reaction proceeds via a domino intermolecular Friedel-Crafts acylation/intramolecular vinyl carbocation trapping (or oxa-Michael addition)/demethylation reaction sequence. This Lewis acid promoted method features relatively mild reaction conditions to synthesize a variety of 2,3-disubstituted chromen-4-one derivatives in one pot with up to 93% yield. The chromen-2-one (coumarin) product was obtained when 2,6-dimethoxybenzoyl chloride was used as a starting material via an electrophilic aromatic substitution/rearrangement process.
