Abstract:We report a metal-, acid-, and base-free 2(bromomethyl)naphthalene (2-BMN)-promoted organocatalytic Cloke-Wilson rearrangement of chain doubly activated cyclopropanes for the construction of 2,3-dihydrofurans via a carbocation-initiated tandem intramolecular ring-opening/recyclization process. The strategy is especially suitable for the construction of furan units in complex molecules, providing a solution to the problem of heavy-metal residues in dihydrofuran-containing drugs synthesized by traditional metal-based protocols. Thus, it is of potential interest in synthetic and medicinal chemistry.

Abstract:Although various methods for the preparation of dibenzofurans are available, an efficient, general synthesis of dibenzofurans from benzofurans remains an unsolved problem. Herein, we provide the first benzannulation of readily prepared 2‐nitrobenzofurans with alkylidene malononitriles. This methodology enables facile assembly of a wide variety of highly functionalized dibenzofurans in moderate to excellent yields under metal‐free conditions. Gram‐scale synthesis and further elaborations of the product were succesfully performed, demonstrating synthetic utility of this method.

Abstract:A transition-metal-free Claisen rearrangement/cyclization reaction was developed for the synthesis of naphthofuran derivatives from bromonaphthyl 3-phenylallyl ethers. The nature of the base employed in this reaction plays an important role in determining the ratio for the formation of naphthofuran and naphthol products. By using K2CO3 as base and DMF as solvent, we have synthesized a variety of functionalized naphthofurans in good to high yields (49-92%) and with satisfactory selectivities.

Abstract:A variety of 2-aroyl(acyl, or carboxyl)-3-vinyl benzo[b]furans have been prepared via C–C bond formationin good to excellent yields by Pd/C-catalyzed cyclization/isomerization of propargylic compounds. Thereaction proceeded with easily accessible starting materials and inexpensive catalyst under mild condi-tions. Furthermore, a number of benzo[b]furan derivatives including an allene functional group have beenprepared too.

Abstract:A new copper(I)-catalyzed cycloetherification followed by an acid-catalyzed allylic nucleophilic substitution have been developed for the one-pot synthesis of 2-substituted benzofurans. This one-pot reaction proceeds efficiently under extremely mild conditions with simple and inexpensive catalysts, providing diversely substituted benzofurans in good to excellent yields.

Abstract:Benzofuran is an important backbone for molecules that make up several pharmaceuticals, herbicides/pesticides, and organo-electronics. An environmentally benign dimethyl(methylthio)sulfonium tetrafluoroborate salt was used as an electrophile to induce cyclization of o-alkynyl anisoles to form 2,3-disubstituted benzofurans. The cyclization is performed at ambient reaction conditions, only takes 12 hours to get excellent yields, and shows a high tolerance for various substituted alkynes. Also, a trimethyl group obtained after the cyclization reactions allows for a cascade cyclization, and an alkyne is used in the reaction to create a thieno[3,2-b]benzofuran core structure.

Abstract:A novel method for the synthesis of benzofuran analogues via condensation of 2-hydroxybenzaldehyde or 2-hydroxyphenyl ketone with methyl 4-bromocrotonate was reported. Moderate to good yield was obtained with cesium carbonate as base in DMF at 70 degrees C. The new-generated double bond was (E)-configuration determined by X-ray diffraction. All benzofuran derivatives were determined by H-1 NMR, C-13 NMR and HRMS techniques.

Abstract:The expeditious construction of tetrasubstituted furans via Cu/base-promoted cascade reaction of terminal alkynes with 1,2-diketones is described. This reaction proceeds smoothly involving the formation of multiple chemical bonds with tolerating a wide range of functional groups. The mechanism underlying these transformations has been thoroughly investigated, suggesting that 1,3-diyne serves as a key intermediate generated in situ through oxidative coupling facilitated by the Cu catalyst.

Abstract:Herein, the de novo synthesis of atropisomeric benzofurans through the Cu/spirocyclic-pyrrolidine-oxazoline (SPDO) complex catalyze asymmetric formal [3 + 2] cycloaddition between quinones and ynamides is explored, producing a range of structurally diverse axially chiral benzofurans in satisfactory yields with good to excellent enantioselectivities. Furthermore, density functional theory calculations were applied to explain the possible reaction mechanism.

Abstract:Objective To simplify the synthesis of 3-aryl-2,2-disubtituted dihydrobenzofurans.Methods It involves a selective cross McMurry reaction coupling of a 2-hydroxyphenyl-aryl-methanone with an aldehyde as a pivotal step and a sequential acid-promoted cyclization of the resulting ortho-vinylphenols.Results The structure of 3-aryl-2,2-disubtituted dihydrobenzofurans was confirmed by IR,1H-NMR,13C-NMR and HRMS(EI).An array of 3-aryl-2,2-disubtituted dihydrobenzofurans were conveniently and efficiently obtained with two-step total yields of 89%~98%.Conclusion The advantages of this synthetic approach are as follows: the reagents are readily available,the conditions are mild and the operations are simple,and the preparations are suitable for scale-up.

Abstract:A simple and convenient synthetic approach to dihydrofurans has been developed from the cyclization of 2-propynyl-1,3-dicarbonyl compounds catalyzed by potassium tert-butoxide, copper(II) trifluoromethanesulfonate and triphenylphosphine using methanol/dichloromethane as the solvent under very mild conditions. Moreover, dihydrofurans could be easily transformed into the corresponding furans in the presence of trifluoroacetic acid.

Abstract:The 2,3‐dihydrobenzofuran ring systems are a privileged class of oxygen heterocycles, and their synthesis continues to attract considerable effort. Herein, a visible‐light‐driven photoredox‐catalyzed radical multicomponent cyclization of 2‐vinyl phenols, N ‐alkoxypyridinium salts, and sulfur ylides is described. Key to success of the reaction involves the use of both N ‐alkoxypyridinium salts and sulfur ylides as radical precursors. This redox‐neutral protocol features good functionality tolerance, easily available starting materials, simple operation, and mild reaction conditions, enabling modular synthesis of various 2,3‐disubstituted dihydrobenzofurans. Mechanistic studies have also elucidated some of the aspects associated with the key radical intermediates.

Abstract:A palladium(0)-catalyzed cascade reaction for the efficient synthesis of 2,3-disubstituted benzofuran derivatives 3 containing a 3-trisubstituted alkene functional group in moderate yields from alkynyl-substituted benzynes 1 and aryl halides 2 has been developed. This method provides an efficient and alternative approach to benzofurans which are very useful heterocyclic compounds with biological and pharmacological potentials. A plausible mechanism involving intramolecular ene reaction, intermolecular insertion, and β-H elimination is proposed.
