Abstract:Recently, we have developed highly regio- and stereoselective carbometalation of 2-alkynols and 2,3-allenols. The organometallacyclic intermediates may be trapped with I(2) to afford 3-iodoalk-2-en-1-ols. These 3-iodoalk-2-en-1-ols may readily undergo the Sonogashira coupling with terminal propargyl alcohols to form 4-alkyn-2-ene-1,6-diols. Subsequent cycloisomerization in DMA or CH(2)Cl(2) with Au(PPh(3))Cl and AgOTf as the catalyst would afford polysubstituted 2-(1-alkenyl)furans; with PdCl(2) as the catalyst and the reaction in DMA in the presence of allylic bromides, the same substrates afforded polysubstituted 2-(1,4-alkadienyl)furans. In both types of catalyzed cyclization reactions, the elimination of H(2)O promoted the aromatization to form the furan ring. Different alkyl or aryl groups could be introduced into different positions of furans due to the substituent-loading capability of 3-iodoalkenols and diversity of the terminal propargyl alcohols and allylic bromides.

Abstract:A facile preparation of 3-aminofuran derivatives via multicomponent reactions of thiazole carbenes, aldehydes, and dimethyl acetylenedicarboxylate (DMAD) is reported. In this process, the thiazole carbenes, generated in situ from thiazolium salts, reacted with aldehydes and DMAD at -78 to 0 degrees C in CH2Cl2 to afford the substituted furans in moderate to good yields. Eight substituted thiazolium salts were employed as carbene precursors in the reaction. Besides aryl aldehydes, alpha beta-unsaturated aldehydes, aliphatic aldehydes, and arenedial were also investigated and found to be applicable to this reaction.

Abstract:Although various methods for the preparation of dibenzofurans are available, an efficient, general synthesis of dibenzofurans from benzofurans remains an unsolved problem. Herein, we provide the first benzannulation of readily prepared 2‐nitrobenzofurans with alkylidene malononitriles. This methodology enables facile assembly of a wide variety of highly functionalized dibenzofurans in moderate to excellent yields under metal‐free conditions. Gram‐scale synthesis and further elaborations of the product were succesfully performed, demonstrating synthetic utility of this method.

Abstract:The expeditious construction of tetrasubstituted furans via Cu/base-promoted cascade reaction of terminal alkynes with 1,2-diketones is described. This reaction proceeds smoothly involving the formation of multiple chemical bonds with tolerating a wide range of functional groups. The mechanism underlying these transformations has been thoroughly investigated, suggesting that 1,3-diyne serves as a key intermediate generated in situ through oxidative coupling facilitated by the Cu catalyst.

Abstract:We report a metal-, acid-, and base-free 2(bromomethyl)naphthalene (2-BMN)-promoted organocatalytic Cloke-Wilson rearrangement of chain doubly activated cyclopropanes for the construction of 2,3-dihydrofurans via a carbocation-initiated tandem intramolecular ring-opening/recyclization process. The strategy is especially suitable for the construction of furan units in complex molecules, providing a solution to the problem of heavy-metal residues in dihydrofuran-containing drugs synthesized by traditional metal-based protocols. Thus, it is of potential interest in synthetic and medicinal chemistry.

Abstract:A method for the synthesis of tri-substituted furans has been developed based on Cu(I)-catalyzed reaction of terminal alkynes with β-keto α-diazoesters. This method for the synthesis of 2,3,5-trisubstituted furans is operationally simple and applicable to wide substrate scope. Moreover, this synthesis employs cheap Cu(MeCN)4PF6 as the catalyst and no additional ligand is needed. Similar reaction has also been applied to ethyl (E)-2-diazo-3-(methoxyimino)butanoate for the synthesis of 2,3,5-trisubstituted N-methoxypyrroles with limited success.

Abstract:In the presence of boron trifluoride, a variety of alkynyl sulfides and alkynyl sulfoxides undergo tandem cross-coupling/[3,3]-sulfonium rearrangement followed by 5-exo-dig heterocyclization. The strategy provides concise access to novel tetrasubstituted furans in good to high yields with 100% atom-economy efficiency. Further derivatization of the resultant furans was feasible by utilizing the incorporated alkylthio groups.

Abstract:The in situ formed furan-fused cyclobutenes via Cu(I)-catalyzed cycloisomerization of readily available allenyl ketones bearing a cyclopropyl moiety are a highly reactive and powerful species, which undergo annulative fragmentation with terminal ynones to afford a wide variety of functional furans in moderate to high yields. Abstract:We have developed a facile and effective synthesis of 3-iodofurans from 3-monosubstituted 1,2-allenyl perfluoroalkyl ketones or 2-hydroxy-4-iodo-2,5-dihydrofurans from 3,3-disubstituted 1,2-allenyl perfluoroalkyl ketones. The perfluoroalkyl substituent and the amount of water in the solvent are important for the success of this electrophilic cyclization.

Abstract:A highly efficient Au(I)-catalyzed cascade reaction between bispropargylic alcohols and pyridine-N-oxides has been realized. The reaction process involved a gold(I)-catalyzed sequential oxidation/Pinacol rearrangement/oxacyclization. Moreover, 1,2-aryl, 1,2-alkyl, or 1,2-vinyl migration was favored over 1,2-alkynyl in the crucial gold(I)-catalyzed Pinacol rearrangement step. A range of 2,4,5-trisubstituted 3(2H)-furanones were synthesized in high yields (up to 88%) with excellent regioselectivities (up to >19:1) under mild reaction conditions.

Abstract:Polysubstituted furans were prepared in moderate to good yields from various sulfur ylides and alkyl acetylenic carboxylates. The direct reactions of dimethylsulfonium acylmethylides with dialkyl acetylenedicarboxylates afforded dialkyl furan-3,4-dicarboxylates through a tandem sequence of Michael addition, intramolecular nucleophilic addition, 4 pi ring opening, intramolecular Michael addition, and elimination. The method was extended to synthesize furan-3-carboxylate, -2,4-dicarboxylates, and -2,3,4-tricarboxylates as well. The current method provides a direct and simple strategy in the synthesis of structurally diverse polysubstituted furans with mono to tricarboxylate groups from safe and readily available dimethylsulfonium acylmethylides and different alkyl acetylenic carboxylates.

Abstract:A cyclodimerization of readily accessible acetylenic ketones facilitated by phosphane-borane complex under base is achieved. This methodology allows one pot synthesis of phosphorus-involved tetrasubstituted furans via the construction of a C−P bond and a furan ring within a single procedure. Abstract:Ag(I)-catalyzed synthesis of diazo-trisubstituted furans starting from diazo-cumulated allenyl ketones has been investigated. The Rh2(OAc)4-catalyzed reaction of the diazo 2,3,5-trisubstituted furans provided penta-substituted aromatics via cascade intermolecular cyclopropanation/rearrangement/isomerization. The cyclopropanation on the furan ring/rearrangement of cyclopropane moiety has been reported. A reasonable mechanism is proposed.
