Abstract:Although various methods for the preparation of dibenzofurans are available, an efficient, general synthesis of dibenzofurans from benzofurans remains an unsolved problem. Herein, we provide the first benzannulation of readily prepared 2‐nitrobenzofurans with alkylidene malononitriles. This methodology enables facile assembly of a wide variety of highly functionalized dibenzofurans in moderate to excellent yields under metal‐free conditions. Gram‐scale synthesis and further elaborations of the product were succesfully performed, demonstrating synthetic utility of this method.

Abstract:We report a metal-, acid-, and base-free 2(bromomethyl)naphthalene (2-BMN)-promoted organocatalytic Cloke-Wilson rearrangement of chain doubly activated cyclopropanes for the construction of 2,3-dihydrofurans via a carbocation-initiated tandem intramolecular ring-opening/recyclization process. The strategy is especially suitable for the construction of furan units in complex molecules, providing a solution to the problem of heavy-metal residues in dihydrofuran-containing drugs synthesized by traditional metal-based protocols. Thus, it is of potential interest in synthetic and medicinal chemistry.

Abstract:Objective To simplify the synthesis of 3-aryl-2,2-disubtituted dihydrobenzofurans.Methods It involves a selective cross McMurry reaction coupling of a 2-hydroxyphenyl-aryl-methanone with an aldehyde as a pivotal step and a sequential acid-promoted cyclization of the resulting ortho-vinylphenols.Results The structure of 3-aryl-2,2-disubtituted dihydrobenzofurans was confirmed by IR,1H-NMR,13C-NMR and HRMS(EI).An array of 3-aryl-2,2-disubtituted dihydrobenzofurans were conveniently and efficiently obtained with two-step total yields of 89%~98%.Conclusion The advantages of this synthetic approach are as follows: the reagents are readily available,the conditions are mild and the operations are simple,and the preparations are suitable for scale-up.

Abstract:Abstract A concise method for the synthesis furan and benzene fused 2‐tetralones via photo‐induced rearrangement of 2′‐(furan‐2‐yl)‐[1,1′‐biphenyl]‐4‐ols was developed. Under argon atmosphere and room temperature, 6,7‐dihydrophenanthro[9,10‐ b ]furan‐5(4 H )‐ones were obtained by irradiating 2′‐(furan‐2‐yl)‐[1,1′‐biphenyl]‐4‐ols with a 254 nm UV lamp (64 W) in dichloromethane (DCM) for 1 hour. The reaction mechanism includes 6π‐electrocyclization, thermal suprafacial [1,5]‐H shift, double thermal suprafacial [1,3]‐H shift and keto‐enol isomerization.

Abstract:A highly regio-and stereoselective Pd-catalyzed cascade reaction of allenoate adducts has been developed. Various allenoate adducts of phosphine-catalyzed addition of allenoates are found to be efficient substrates for the preparation of 2,3-dihydrobenzofuran. It is the first example of allenoate adducts used as ideal and practical precursors of hard-to-get functionalized 1,3-butadienes for heterocycle synthesis.

Abstract:A novel method for the synthesis of benzofuran analogues via condensation of 2-hydroxybenzaldehyde or 2-hydroxyphenyl ketone with methyl 4-bromocrotonate was reported. Moderate to good yield was obtained with cesium carbonate as base in DMF at 70 degrees C. The new-generated double bond was (E)-configuration determined by X-ray diffraction. All benzofuran derivatives were determined by H-1 NMR, C-13 NMR and HRMS techniques.

Abstract:The 2,3‐dihydrobenzofuran ring systems are a privileged class of oxygen heterocycles, and their synthesis continues to attract considerable effort. Herein, a visible‐light‐driven photoredox‐catalyzed radical multicomponent cyclization of 2‐vinyl phenols, N ‐alkoxypyridinium salts, and sulfur ylides is described. Key to success of the reaction involves the use of both N ‐alkoxypyridinium salts and sulfur ylides as radical precursors. This redox‐neutral protocol features good functionality tolerance, easily available starting materials, simple operation, and mild reaction conditions, enabling modular synthesis of various 2,3‐disubstituted dihydrobenzofurans. Mechanistic studies have also elucidated some of the aspects associated with the key radical intermediates.

Abstract:A general and efficient synthesis of indeno[1,2]furans via a Pd-catalyzed bicyclization reaction between 2-alkynyliodobenzenes and propargylic alcohols is described. The procedure furnishes indeno[1,2]furans with moderate to excellent yields (51%-78%) and a broad substrate scope. The cascade process combines the formation of one C-O bond and two C-C bonds in a single step.

Abstract:A metal-free synthetically useful catalytic approach for the preparation of 2-acyl benzofurans and indoles from easily accessible o-hydroxyphenyl and o-aminophenyl propargylic alcohols in the presence of pyridine N-oxide is presented. This protocol comprises the acid promoted formation of carbocation and subsequent nucleophilic trap by pyridine N-oxide, which leads to umpolung of the allenyl ether moiety. The following intramolecular attack at the umpolung allenyl carbon furnishes the final benzofurans and indoles via a nucleophile-intercepted Meyer–Schuster rearrangement.

Abstract:A novel solvent-free, TfOH-promoted decyanative cyclization approach for the synthesis of 2,1-benzisoxazoles has been developed. The reactions are complete instantly at room temperature and result in the formation of the desired 2,1-benzisoxazoles in a 34-97% isolated yield.

Abstract:Herein, a hydroxyl assisted, photoredox/cobalt co‐catalyzed semi‐hydrogenation and tandem cyclization of o‐alkynylphenols is developed towards direct assembly of 2,3‐dihydrobenzofurans. Moderate to good yields were obtained for a range of sterically and electronically diverse 2‐propynolphenols under mild conditions. Mechanistic studies demonstrated the inevitable role of the alcoholic hydroxyl group with (Z)‐alkene as the real intermediate. Finally, a key low‐valent cobalt catalyzed intramolecular hydroetherification of alkene is proposed.

Abstract:A palladium-catalyzed domino cyclization/cycloisomerization reaction of alkyne-tethered carbamoyl chlorides with (E)-β-chloroenones is reported. This reaction proceeds via a syn-carbopalladation of the alkyne, followed by a vinyl-PdII-catalyzed cycloisomerization of the (E)-β-chloroenone cascade, which provides an efficient method to synthesize furan-linked methylene oxindoles. The reaction features stereodefined vinyl-PdII species, high to excellent 5-exo/6-endo selectivity, excellent Z/E selectivity, and the sequential formation of three bonds and bis-heterocycles. The strategy for the synthesis of furan-containing benzofurans has also been demonstrated.

Abstract:A convenient synthesis of monosubstituted 3-alkynylfurans by Suzuki coupling reaction of 3-furanylboronic acid with substituted acetylene bromides is described. The internal alkyne products were attainable in 50-89% yields from substrates free of relatively acidic protons. Our protocol is expected to find applications in the synthesis of structurally related natural products.

Abstract:A convenient and efficient method was developed for the synthesis of naphtho[2,1-b]furans via 4-dimethylaminopyridine(DMAP)-catalyzed cascade reaction of 2-hydroxy-1-naphthaldehydes and α-halogenated ketones in moderate to good yields in the presence of Na2CO3 at 80 ºC for 6 h. The mechanism for this process was briefly discussed with a tentative catalytic cycle proposed. Moreover, this method features organocatalysts and high step-economy, which make it practical and attractive.
