Abstract:Three microbial methods for determinating antibiotic in milk were compared, and the critical parameters of three methods for detecting penicillin G, the residue limits, detection time, cost price and difficulty to operate were studied. At the same time, the study pointed out some disadvantages of TTC method on residue limits, and the tube diffusion method is introduced as a good supplement for the two methods of TTC and AOAC paper disk to determinate β-lactams in milk, inexpensive, quick, simple and convenient, and the low limits of detection for the operation.

Abstract:Two microbial standard methods to detect β-lactam residues in milk were compared in this paper.The detection limits of two methods for six kinds of β-lactams were determined.The results showed that the TTC method could not detect the MRL antibiotic residual concentration regulated by EU,while the AOAC paper disk method could.The AOAC method was suggested to be a supplement of TTC method to determine residual β-lactams in milk because of its specificity,sensitivity,convenience and economy.The detection limit of AOAC method for the six β-lactams also measured up the MRLS regulation.

Abstract:Less than 1% of microorganisms taxa can be isolated and cultured from soil microbial community by traditional culturing approaches,which had great limitation for studying soil microbial diversity.Phospholipids fatty acid profile analysis,BIOLOG micro-plate analysis and DNA fingerprinting technique,can be employed to describe and characterize soil microorganisms which cannot be cultured currently,and get more perfect information of soil microbial diversity.The application principle,research status,advantage and disadvantage of the three methods were reviewed.Different methods should be combined in order to integrate the advantages of different methods,and ensure the efficiency and scientificity for studying microbial diversity of farmland soil.

Abstract:Six kinds ofβ-lactams antibiotics fiequently existing in milk were detected by the method of Bacillus stearothemophilus tube diffusion.It was found that the method was semi-quantitative,which was sensitive to detectingβ-lactams antibiotic in milk.The determination limits to penicillin G,ampicillin and amoxicillin were all 6μg/kg.The detection limits to cefalexin,oxacillin,cephapirin were 150μg/kg,15μg/kg and 30μg/kg,re- spectively.It was proved that the method was quick and convenient compared to the method of B.sterothermophilus Disc Assay(BSDA)and could be used to detectβ-lactams in milk

Abstract:A broad-spectrum microbiological inhibition method has been developed for rapidly screening different kinds of antibiotics such as beta-lactam, aminoglycosides, tetracyclines, sulfonamides, macrolides, lincosamides and quinolones in milk, chicken egg and honey by using an easy sample preparation. The microbiological system in microtiter plates consists of an agar medium, a mixture of nutrients, test bacteria (Geobacillus stearothermophilus var C953), bromocresol purple, and other supplements such as trimethoprim, chloramphenicol, streptomycin and enrofloxacin which helps to improve the detection capability of the microbiological system toward the chosen antibiotics. It was observed that the limit of detection of the kit used in present study for all kinds of antibiotics in milk were lower than or close to maximum residue limits determined by EU or CODEX. For chicken egg and honey, the detection capability of the kit was similar to that determined in milk. Moreover, it was revealed that the kit in present study was more sensitive to aminoglycosides, macrolides and quinolones in various matrixes than internationally available commercial kits. The false-positive and false-negative rates for both were 0%. The coefficient of variations among various factors was all less than 4%. Additionally, the quality guarantee period of the kit was more than 6 months at 4 degrees C. A good correlation between the kit results and the LC-MS/MS results for milk was also observed, which revealed that the kit was reliable to screen antibiotics residues in incurred samples.

Abstract:Adding exogenous β-lactamase to the raw milk is banned in Chinese laws and regulations. Presented method promulgated by Chinese government is time-consuming and inappropriate for the test of the raw milk receiving. Thus it is imperative to develop a method for rapid detection ofβ-lactamase in the raw milk. In this study, we used the capacity of theβ-lactamase to decompose the penicillin G and attempted to establish a rapid detection method for detecting β-lactamas in raw milk. The results showed that the detection limit of method was 4 IU/mL. The repetitive veriifcation showed that the result 100%ift among 10 groups of parallel samples. The anti-interference performance of negative samples was good. The average positive detection rate of raw milk of ten regions was 4.46%with the rapid detection method, then the positive samples were quantitative validated and the veriifcation results were all positive, so the rapid method had high detection accuracy and good universality, it was suitable for the raw milk purchasing in dairy companies.

Abstract:In this paper, a chemiluminescence (CL) micro-flow system combined with on-line solid phase extraction (SPE) is presented for determination of β-lactam antibiotics (penicillin, cefradine, cefadroxil, cefalexin) in milk. It is based on the enhancement effect of β-lactam antibiotics on the luminol-K3Fe(CN)6 CL system. The micro-flow system was fabricated from two polymethyl methacrylate (PMMA) plates (50mm×40mm×5mm) with the microchannels of 200μm wide and 150μm deep. C18-modified silica gel was packed into the microchannel (length: 10mm; width: 1mm; depth: 500μm) to serve as SPE device. Extraction and preconcentration of the analytes were carried out using on-line SPE micro-flow system and the selectivity of CL detection was improved. The detection limits were 0.5μgmL−1 of penicillin, 0.04μgmL−1 of cefradine, 0.08μgmL−1 of cefadroxil and 0.1μgmL−1 of cefalexin. The proposed method was also applied to analyze the β-lactam antibiotics in milk. Experimental results were in good agreement with those obtained by high performance liquid chromatography (HPLC) method with UV detection.

Abstract:In this study, we developed a microbiological inhibition method for the rapid screening of antibiotics in milk with Geobacillus stearothermophilus ATCC12980 as an indicator bacterium and an easy sample pretreatment. We observed that the limits of detection of the kit for 34 common antibiotic residues in milk, including β-lactams (13), aminoglycosides (6), tetracyclines (4), sulfonamides (6), macrolides (4), lincosamides (1), were lower than or close to the maximum residue limits formulated by the European Union and China. Moreover, the false-positive rate was 1% and the false-negative rates were less than 5%. The ruggedness of the method (the reproducibility of detection capability of different batches of medium) met requirements at determined levels and residual limits. The shelf life of the kit was more than 6 mo at 4°C. Additionally, we observed good correlations between the kit results and ultra-high-performance liquid chromatography-tandem mass spectrometry results for incurred milk (samples taken from animals treated with antibiotics according to the pre-slaughter medication data), which indicated that the kit was reliable for screening antibiotics in incurred samples. In conclusion, the kit has a broad application potential with high sensitivity, specificity, and reproducibility, stability, and reliability, combined with simple operation, low cost, and high-throughput capacity.

Abstract:In this paper,effects of Land-Eynon's method and colorimetric method were compared,and then we used HPLC(high performance liquid chromatography) to detect lactose.These two methods were consistent,and the accuracy of Land-Eynon's method was better than that of colorimetric method.We found both two methods couldn't detect lactose accurately in low lactose milk while HPLC method could,this is most probably because the two methods can't distinguish lactose from other reducing sugars.

Abstract:The TTC method and SNAP kit method were used to detect the antibiotic residue in milk,including 9 typical antibiotics of β-Lactam,Tetracycline,Macrolide,Aminoglycoside,Chloramphenicol,Chlorpromazine and Fluoroquinolone antibiotics.According to test results,both of the two methods had some certain defects.There's some risk if each of them were used alone in the quality control of raw milk.While on the basis of general survey of the antibiotics used in dairy cow feeding,SNAP method can be used to control the β-Lactam residue in raw milk.

Abstract:Amoxicillin (AMOX), clavulanic acid (CLAV) and prednisolone (PSL) are widely used in combination for the treatment of mastitis in lactating dairy cows. However, no method has been reported to detect these three chemicals in milk in a single assay. In the present work, a reliable and sensitive UPLC-MS/MS method was developed and validated for simultaneous determination of AMOX, CLAV and PSL in cow's milk. The analytes were determined by a positive and negative ionization electrospray mass spectrometer via multiple reaction monitoring. The linear ranges of AMOX, CLAV and PSL were from 2 to 1000 ng/mL, 20-1000 ng/mL and 1-1000 ng/mL, respectively, with the correlation coefficients greater than 0.999. The limits of quantification (LOQs) were 2 ng/mL (AMOX), 20 ng/mL (CLAV) and 1 ng/mL (PSL). Recoveries of the analytes of interest in milk samples were in the ranges of 84.2-101.4%. The intra-day and inter-day precisions ranged from 1.8% to 11.9%. This method was successfully applied to investigate the pharmacokinetics of AMOX, CLAV and PSL in milk from healthy and mastitic cows. The elimination times of AMOX and PSL in mastitic cows were longer than that in healthy cows, but the elimination times of CLAV did not show significant difference. (C) 2015 Elsevier B.V. Abstract:Penicillin-Micrococcus luteus-TSA medium plates were used to establish a method for the rapid isolation of β-lactamase-producing bacteria in raw milk.This method revealed a more convenient and simpler observation than conventional methods,and could provide highly specific isolation.16S rRNA gene sequence analysis allowed the assignment of the β-lactamase-producing bacterial isolates obtained in this study.As a result,Escherichia coli was the dominant β-lactamase-producing strain in raw milk.

Abstract:This present study was designed to develop a novel microbiological inhibition-based method for the rapid screening and identification of antibiotic residues in milk, chicken egg and honey. Geobacillus stearothermophilus C953 was used as test bacterium in the detection system of this study. The optimization of nutrients and other supplements were performed to promote the growth of test bacterium and thus shorten the detection time. Furthermore, the synergetic agents were added to improve the sensitivity of test bacterium to more antibiotics. Additionally, confirmatory solutions such as beta-lactamase, p-aminobenzoic acid, MgSO4 and cysteine were added to classify and identify different kinds of antibiotics. We observed that the LOD of this detection system was at or close to maximum residue limits established by EU for beta-lactams, aminoglycosides, tetracyclines, sulfonamides, macrolides and quinolones in milk. The LOD of different kinds of antibiotics in chicken egg was less than or similar to the MRL and the LOD of Premi (R) test (except sulfonamides). For honey, there are no MRL, the LOD was less than or similar to the recommended concentration and the LOD of Premi (R) test. Noteworthy, the detection system also can identify these six kinds of antibiotics in milk, chicken egg and honey, and there were satisfactory results of specificity experiments and confirmation experiments by LC-MS/MS. Accordingly, the present study provides a reliable preliminary characterization of antibiotic residues in animal foods and improves the detection efficiency for the following chemical confirmation experiments by HPLC, LC-MS/MS, immunological and receptor-based tests.

Abstract:In order to compare MTT method and traditional disc agar diffusion test,nine kinds of antibiotics-Amikacin,rimethoprim and sulphame-thoxazole,Fluperacid,Gentamycin,Cefotaxime,Cefazolin sodium,Ampicillin and Streptomycin were used.The result showed that bacteria in milk samples were tolerance to streptomycin,erythromycin,trimethoprim and sulphame-thoxazole,were moderate sensitivity to Gentamycin and Amikacin,were sensitivity to Ampicillin,Cefotaxime and Cefazolin sodium besides Fluperacid.These two methods had same result.Therefore it can be concluded that MTT test may be used to make antibiotic susceptibility test to detect cow mastitis.

Abstract:Objective An ultra pressure liquid chromatography(UPLC) tandem mass spectrometric(MS/MS) me- thod has been developed for the simultaneous determination of two β-lactam antibiotics(cefotaxime and cefoperazone) and two β-lactamase inhibitors(tazobactam and sulbactam) in bovine milk and infant formula. Methods The analysts were ex- tracted from milk sample with acetonitrile, defatted with liquid-liquid extraction by hexane, and further purified by Oasis HLB solid phase extraction cartridges. The extracted analysts were separated by UPLC BEH C18 column(50 mm × 2.1 mm, 1.7 μm), and determined by UPLC-MS/MS under negative ionization mode. Results The method showed a good linearity over the range of 1 100 μg/L, with the linear correlation coefficient r0.997. The mean recoveries for four analysts in bovine milk ranged from 85.5% to 95.5%. The limit of detection was 0.1 0.2 μg/L. Conclusion This method is rapid, sensitive and re- peatable, and it could be performed for the determination of β-lactam antibiotics and β-lactamase inhibitors in milk-marix.

Abstract:Based on the fact that Bacillus stearothermophilus can degrade lactose to produce acid,Bacillus stearothermophilus was used as indicator strain to detect antibiotics residues in the raw milk.The composition of mixed indicators was modified and lactase was added to milk sample to enhance detection rate.Other detection parameters were also studied.The optimal parameters for detection were as follows:the viable cells of Bacillus stearothermophilus 5×106 in 0.1 mL solution,addition of 30 g·L-1 lactase solution 0.1 mL,addition volume of indicator 0.1 mL,detection volume of milk 0.1 mL,fermentation temperature 64 ℃ and detection time 2.5 h.Compared with national standard method (TTC method),this new method has lower detection limits for five antibiotics,and is easy to judge result by nude eyes.

Abstract:In order to understand the antibiotic residues in milk,the antibiotics should not be detected according to the calling standard of no social effects of pollution of food issued by the Ministry of Agriculture in 2001.The 32 samples of milks was randomly selected and detected by TTC methods.The results indicated that the positive number of antibacterial residues was 5,the rate of positive 15.63%.The sulfadimidine was not detected by ELISA methods, the number positive of tetracycline residues was 3,the positive rate was 9.38%,the positive number of streptomycin was 7,the positive rate of was 21.88%.The sterilized milk of different pock had antibiotic residues excepted for fresh milk.The antibiotic was not detected in entrance sterilized milk powder.The 10 positive antiobiotics was detected in 32 sterilized milk samples,and the 11 positive antiobiotics was detected in 20 milk powder samples,the rates were 31.25% and 55%.The test results showed that the positive rate of antibiotic residues in milk powder was significantly higher than that in sterilized milk,and that in ordinary packaged milk was higher than that in elegant packaged milk,No antibiotic was detected in the import sterilized milk.The test results showed that residues of antibiotics in sterilized milk from market is higher.

Abstract:The detecting method of tube diffusion was confirmed with PCA medium as testing medium, Bacillus stearothermophilus c953 as indicative bacteria and bromocresol purple as indicator. In this study, six kinds of β-lactam antibiotics were detected by the two methods of tube diffusion and 2,3,5-triphenyl tetrazolium chloride test. On the foundation of efficient parameters and actual experiment, it is indicated that the method of tube diffusion is economic, quick, convenient and stable,and the limit of detection measures up. So it is a good method for detecting β-lactam antibiotic residues in raw milk in China.
