Abstract:Polylactic acid is one of most important and promising biodegradable and bioabsorbable materials.It has been widely applied as biodegradable plastics and fibers as well as biomaterials,and therefore,is of tremendous market value.Polylactic acid has polyolefins-comparative mechanical and processing properties but unsatisfactory heat and impact resistance.Its thermal,mechanical and gas barrier properties need to be enhanced to satisfy various applications.Preparation of nanocomposites of polylactic acid with inorganic nanoparticles is an effective approach to remarkably improve the properties.The progress in the preparation,structure and properties of polylactic acid-based nanocomposites as well as the relationships among them are reviewed,and the prospect of this field is discussed.
Cytocompatibility Evaluation of a Novel Series of PEG-Functionalized Lactide-Caprolactone Copolymer Biomaterials for Cardiovascular Applications

Abstract:Although the use of bioresorbable materials in stent production is thought to improve long-term safety compared to their durable counterparts, a recent FDA report on the 2-year follow-up of the first FDA-approved bioresorbable vascular stent showed an increased occurrence of major adverse cardiac events and thrombosis in comparison to the metallic control. In order to overcome the issues of first generation bioresorbable polymers, a series of polyethylene glycol-functionalized poly-L-lactide-co-ε-caprolactone copolymers with varying lactide-to-caprolactone content is developed using a novel one-step PEG-functionalization and copolymerization strategy. This approach represents a new facile way toward surface enhancement for cellular interaction, which is shown by screening these materials regarding their cyto- and hemocompatibility in terms of cytotoxicity, hemolysis, platelet adhesion, leucocyte activation and endothelial cell adhesion. By varying the lactide-to-caprolactone polymer composition, it is possible to gradually affect endothelial and platelet adhesion which allows fine-tuning of the biological response based on polymer chemistry. All polymers developed were non-cytotoxic, had acceptable leucocyte activation levels and presented non-hemolytic (<2% hemolysis rate) behavior except for PLCL-PEG 55:45 which presented hemolysis rate of 2.5% ± 0.5. Water contact angles were reduced in the polymers containing PEG functionalization (PLLA-PEG: 69.8° ± 2.3, PCL-PEG: 61.2° ± 7.5) versus those without (PLLA: 79.5° ± 3.2, PCL: 76.4° ± 10.2) while the materials PCL-PEG550, PLCL-PEG550 90:10 and PLCL-PEG550 70:30 demonstrated best endothelial cell adhesion. PLLA-PEG550 and PLCL-PEG550 70:30 presented as best candidates for cardiovascular implant use from a cytocompatibility perspective across the spectrum of testing completed. Altogether, these polymers are excellent innovative materials suited for an application in stent manufacture due to the ease in translation of this one-step synthesis strategy to device production and their excellent in vitro cyto- and hemocompatibility.
[Preliminary Evaluation of the Biological Properties of Poly-Lactide-co-glycolic Acid (PLGA) Knitted Mesh].

Abstract:This is a work aimed to investigate the biodegradability, biocompatibility and mechanical property of the poly-lactide-co-glycolic acid (PLGA) knitted mesh preliminarily and to further explore its applications in tissue engineering and regenerative medicine. The biological property of PLGA mesh was investigated comprehensively with the degradation experiment in vitro, the acute cytotoxicity assay, the intradermal irritation test and the subcutaneous implantation test in vivo utilized. The degradation experiment in vitro demonstrated that the pH value of the removed solution fluctuated between 6.68 and 7.33. The elastic modulus of the PLGA mesh increased at first and then decreased afterwards. The acute toxicity test and the intradermal irritation test indicated that the PLGA mesh was with innocuity safety. The PLGA mesh accelerated degradation and was replaced gradually by the neotissue. The results of immunohistochemical staining demonstrated that the number of ED-1+ cells increased at first and then decreased afterwards. The PLGA mesh with excellent mechanical properties, good biocompatibility and favorable degradation ratio has the potential to be employed as a "skeleton" to reinforce the mechanical property of collagen-based dermal substitutes in tissue engineering.

Abstract:The biocompatibility of biomaterials is essentially for its application. The aim of current study was to evaluate the biocompatibility of poly(lactic-co-glycolic acid) (PLGA)/gelatin/nanohydroxyapatite (n-HA) (PGH) nanofibers systemically to provide further rationales for the application of the composite electrospun fibers as a favorable platform for bone tissue engineering. The PGH composite scaffold with diameter ranging from nano- to micrometers was fabricated by using electrospinning technique. Subsequently, we utilized confocal laser scanning microscopy (CLSM) and MTT assay to evaluate its cyto-compatibility in vitro. Besides, real-time quantitative polymerase chain reaction (qPCR) analysis and alizarin red staining (ARS) were performed to assess the osteoinductive activity. To further test in vivo, we implanted either PLGA or PGH composite scaffold in a rat subcutaneous model. The results demonstrated that PGH scaffold could better support osteoblasts adhesion, spreading, and proliferation and show better cyto-compatibility than pure PLGA scaffold. Besides, qPCR analysis and ARS showed that PGH composite scaffold exhibited higher osteoinductive activity owing to higher phenotypic expression of typical osteogenic genes and calcium deposition. The histology evaluation indicated that the incorporation of Gelatin/nanohydroxyapatite (GH) biomimetics could significantly reduce local inflammation. Our data indicated that PGH composite electrospun nanofibers possessed excellent cyto-compatibility, good osteogenic activity, as well as good performance of host tissue response, which could be versatile biocompatible scaffolds for bone tissue engineering.

Abstract:BACKGROUND:Polylactic acid and its copolymers have been widely used in biomedical fields because of their good biocompatibility and biodegradability, which have become a hotspot in the research of biomaterials. OBJECTIVE:To review the biocompatibility of fully degradable polylactic acid and its copolymers. METHODS:A computer-based online retrieval of PubMed and CNKI was performed to search relevant papers published from 2006 to 2016, with the key words of "polylactide, polylactic acid, copolymer, biodegradability, biocompatibility, animal" in English and Chinese, respectively. RESULTS AND CONCLUSION:Polylactic acid and its copolymers as polyester compounds are approved by the US Food and Drug Administration for use in the biomedical field because of their good biocompatibility mainly as drug delivery carriers and temporary implants. Moreover, their side effects in clinical application have attracted attentions. Polylactide copolymers can cause some adverse reactions when used in drug delivery systems, orthopedic and skin care, and other clinical medical fields. These copolymers are deemed to have no impact on the central nervous system, eyes, cardiovessels and other tissues and organs. They also have no virtually genotoxic and carcinogenic effects. Currently, the polylactide copolymer implant mainly results in local reactions in the surrounding tissues, and no systemic reactions have been found.
[Mechanical Properties and Biocompatibility of Nanometer Calcium Carbonate/poly (L-Lactide) Composites].

Abstract:The purpose of this study is to synthesize nanometer calcium carbonate/poly(L-lactide) composites and evaluate its biocompatibility, and mechanical properties. The nanometer calcium carbonate/poly(L-lactide) composites was prepared by the method of solution mixing and cell culture. Its properties were analyzed using, mechanical tests, MTT assay, transmission electron microscope and scanning electron microscope. It was found that the new kind copolymer had good biocompatibility, nanometer calcium carbonate could enhance the strength of the pure poly (L-lactide) matrix. The nanometer calcium carbonate/poly (L-lactide) composites is a promising bioabsorbable material. It is worth further medical research.

Abstract:OBJECTIVE:The super-high-molecular-weight poly-DL-lactic acid (PDLLA), with the molecular weight of 900 kD, is a newly emerging biomaterial and potentially used in the therapy of bone fracture because of its excellent mechanical property. However the biocompatibility of this material has not been reported so far, therefore this experiment was designed to examine whether the super-high-molecular-weight PDLLA was harmful to creatures, when it was implanted in the body of animals for a long period.METHODS:The material was prepared in small cuboids, with the size of 1.0 mm x 1.5 mm x 2.0 mm, and these blocks were implanted into the masseteric space of SD rats and, the activity of the SD-rats was monitored continuously. The animals were sacrificed in the 3rd, 6th, 9th, 12th months after the operation and, the specimens were taken out from the animals. The examination included anatomical, pathological and haematological methods. The data were analyzed with SPSS 8.0.RESULTS:The wound healed well after the operation. Super-high-molecular-weight PDLLA degraded 6 months after the implantation. In the 3rd month after the operation, a thin fiber membrane around the materials was formed. In the 6th month, the membrane was much thinner than that in the 3rd month and completely disappeared in the 9th month. The pathological examination showed that slightly inflammatory reaction appeared in the tissue around these blocks in the 3rd month, but the inflammatory reactions were gradually remitted in the following 6th, 9th and 12th months. Further, the haematological examination did not show any abnormity during the 12-month observation period.CONCLUSION:The super-high-molecular-weight PDLLA can be degrade when it is implanted into the body of creatures, which proves its good biocompatibility.
Molecular biocompatibility evaluation of poly(D,L-lactic acid)-modified biomaterials based on long serial analysis of gene expression

Abstract:In this work, long serial analysis of gene expression (LongSAGE) technology was used to investigate the molecular mechanism of the interaction between cells and poly(D,L-lactic acid)-modified biomaterials. After mouse osteoblast-like MC3T3-E1 cells were cultured on poly(D,L-lactic acid) (PDLLA) and a novel maleic anhydride-modified poly(D,L-lactic acid) (MPLA) films, the morphology, proliferation activity and alkaline phosphatase (ALP) activity of MC3T3-E1 cells were assessed by laser confocal microscope, cell counting assay and ALP assay, and the gene expression profiles of the cells were detected and compared at the transcript levels, respectively. 202 tags were found differentially expressed (p<0.05, and fold change >2) between PDLLA and MPLA LongSAGE libraries. Gene ontology functional analysis of the differentially expressed genes indicates that surface modification of MPLA biomaterial has an extensive influence on cells by regulating expression of genes related to cell proliferation, cell cycle, cytoskeleton organization, ossification, bone remodeling, metabolism, and eventually induces osteoblast proliferation and differentiation. The approach presented here provides a new insight in the molecular biocompatibility evaluation of biomaterials, contributing to the development of biomaterials in tissue engineering field.

Abstract:OBJECTIVE:To examine the biocompatibility of polylactic/glycolic acid (PLGA) and Schwann cells.METHOD:Schwann cells were harvested from rat brachial and sciatic nerves. Schwann cells were cultured with PLGA, observed by phase-contrast microscopy and electron microscopy.RESULT:Schwann cells could attach and proliferate on the surface of the PLGA.CONCLUSION:The PLGA has good cellular biocompatibility. It can be used as biomaterial for tissue engineering.

Abstract:In this study, poly(l-lactide-co-β-malic acid) (PLMA) copolymer was synthesized via ring-opening polymerization of l-lactide and β-benzyloxycarbonyl-β-propiolactone, followed by removal of benzyl protective groups through hydrogenolysis. Novel biomorphic PLMA scaffolds with a honeycomb-like structure were prepared using cane as template. We investigated the chemical structure and physicochemical properties of PLMA copolymer and the morphology and biocompatibility of biomorphic PLMA scaffolds. The results confirmed the chemical structure of PLMA copolymer. The amorphous copolymer had a glass transition temperature of 49°C. The PLMA scaffolds well retained the morphology of cane, and had a porosity of 85.5% and a compressive modulus of 766.2±36.1kPa. in vitro biocompatibility evaluation with MC3T3-E1 cells demonstrated the potential of the biomorphic PLMA scaffolds for bone tissue engineering.

Abstract:Chitin short fiber reinforced polycaprolactone composite was prepared by melting blending method. The cytotoxicity and biocompatibility of pure polycaprolactone and of chitin short fiber reinforced polycaprolactone composite were investigated in order to provide useful scientific basis for clinical application. The biocompatibility of pure polycaprolactone and that of chitin short fiber reinforced polycaprolactone composite were evaluated by a series of tests, including cytotoxicity test in vitro, acute systemic toxicity test, hemolysis test, pyrogen test and sensitivity test. The results showed that the cytotoxicity scores of the two materials were grade 0 and the growth and proliferation of the cultured cells were not significantly inhibited by the two materials. There were no potential allergic materials in the composites and the maceration extract showed no hemolytic reaction, no acute systemic toxicity and no pyrogen reaction. We conclude that the composites have fine biocompatibility and are safe for clinical use in the reconstruction treatment for chest wall defect.

Abstract:OBJECTIVE: When preliminary tests have confirmed a nano-hydroxyapatite (Nano-HA) content of 20% of the polylactic acid (PLA) composite material of Nano-HA interface fixation material for biomechanical requirements, there is a need for further observation of its biocompatibility and clinical applications, to provide reference data.METHODS: Preparation of Nano-HA content of 20% PLA composite Nano-HA bone substitute material and extract. The establishment of the negative control group (containing 10% fetal bovine serum in DMEM complete medium), experimental group (extract), the positive control group (mass concentration of 0.64% phenol), and a co-culture of rabbit bone marrow mesenchymal stem cells (rBMSC) and materials extraction liquid. Observation of the morphological changes in rBMSC in culture at time points of 3, 5, and 7 days, the use of the MTT assay, and determination of the relative growth in the above set of rBMSC in cell culture at 3, 5, and 7 days, to judge the material's cytotoxicity.RESULTS: With time, the absorbance value of the three groups of cells were significantly increased (P < 0.01). The relative growth of the rBMSCs in experimental group in the first 3, 5, and 7 days was 95.3%, 96.8% and 97.6%; the cytotoxicity was according to the national standards I; the difference was not significant (P > 0.05) between the the experimental group and the negative control group; there was a significant difference between the positive control group and the other 2 groups (P < 0.05). Cells in the experimental group were seen having normal morphology, and spindle-shaped adherent growth.CONCLUSION: PLA composite artificial bone materials and Nano-HA show good cell compatibility, and the values for cytotoxicity, with reference to GB/T16886.5.2003 (China) standards, are in the safe range.

Abstract:PLLA-TMC-GA terpolymer was prepared by ring-opening polymerization of l-lactide, 1, 3-trimethylene carbonate (TMC), and glycolide (GA). The biocompatibility of terpolymer was evaluated in comparison with PLLA and PLLA-TMC with the aim of assessing their potential in the development of bioresorbable cardiovascular stents. Various aspects of in vitro biocompatibility were considered, including MTT assay, hemolytic test, dynamic clotting time, platelet adhesion, platelet activation, protein adsorption, plasma recalcification time and release of cytokines. The results revealed that the terpolymer presents good cytocompatibility and hemocompatibility. Moreover, no significant increase in the release of cytokines was detected. It is thus concluded that these polymers, in particular PLLA-TMC-GA terpolymer present good biocompatibility for cardiovascular applications.

Abstract:Abstract Poly(lactic acid) (PLA) and poly( ε -caprolactone) (PCL) are two important aliphatic esters known for their biodegradability and bioresorbability properties; the former is stiffer and brittle while the smaller modulus of the latter allows a suitable elongation. The new biomaterials being developed from the blend of these two polymers (PLA and PCL) is opportune due to the reducing interfacial tension between their immiscible phases. In a previous study, PLA/PCL immiscible blend when compatibilized with poly( ε -caprolactone- b -tetrahydrofuran) resulted in enhanced ductility and toughness no cytotoxic effect in vitro tests. There is little published data on the effect of poly( ε -caprolactone- b -tetrahydrofuran) on PLA and PCL biocompatibility and biodegradability in vivo tests. This study focuses on evaluating the behavioral response and polymer-tissue interaction of compatibilized PLA/PCL blend compared to neat PLA implanted via intraperitoneal (IP) and subcutaneous (SC) in male Wistar rats, distributed in four experimental groups: neat PLA, PLA/PCL blend, sham, and control at 2-, 8- and 24-weeks post-implantation (WPI). An open-field test was performed to appraise emotionality and spontaneous locomotor activity. Histopathological investigation using hematoxylin-eosin (H&E) and picrosirius-hematoxylin (PSH) was used to assess polymer-tissue interaction. Modifications in PLA and the PLA/PCL blend’s surface morphology were determined by scanning electron microscopy (SEM). PLA group defecated more often than PLA/PCL rats 2 and 8 WPI. Conjunctive capsule development around implants, cell adhesion, angiogenesis, and giant cells of a foreign body to the biomaterial was observed in light microscopy. Both groups displayed a fibrous reaction along with collagen deposition around the biomaterials. In the SEM, the images showed a higher degradation rate for the PLA/PCL blend in both implantation routes. The polymers implanted via IP exhibited a higher degradation rate compared to SC. These findings emphasize the biocompatibility of the PLA/PCL blend compatibilized with poly( ε -caprolactone- b -tetrahydrofuran), making this biopolymer an acceptable alternative in a variety of biomedical applications.

Abstract:Over the past few decades, with the development of science and technology, the field of biomedicine has rapidly developed, especially with respect to biomedical materials. Low toxicity and good biocompatibility have always been key targets in the development and application of biomedical materials. As a degradable and environmentally friendly polymer, polylactic acid, also known as polylactide, is favored by researchers and has been used as a commercial material in various studies. Lactic acid, as a synthetic raw material of polylactic acid, can only be obtained by sugar fermentation. Good biocompatibility and biodegradability have led it to be approved by the U.S. Food and Drug Administration (FDA) as a biomedical material. Polylactic acid has good physical properties, and its modification can optimize its properties to a certain extent. Polylactic acid blocks and blends play significant roles in drug delivery, implants, and tissue engineering to great effect. This article describes the synthesis of polylactic acid (PLA) and its raw materials, physical properties, degradation, modification, and applications in the field of biomedicine. It aims to contribute to the important knowledge and development of PLA in biomedical applications.

Abstract:In this study, poly(l-lactic acid) (PLLA) fibers were prepared by the dry–wet-spinning method, while chitosan (CHS) fibers were prepared via the wet-spinning method. The two fibers were blend spun and then fabricated into PLLA/CHS fabrics. In vitro degradation experiments of the fabrics were carried out in a phosphate-buffered solution at 37°C with a pH of 7.4. Changes in molecular parameters (molecular weights and molecular weight distributions), phase structures (crystallinities), morphologies (fiber surface topologies) of the PLLA fibers, and their macroscopic properties (the fabric weight losses and mechanical strengths) were monitored with degradation times. These results were compared with control samples with no degradation. The hydrolysis mechanism of PLLA/CHS fabrics was analyzed. It was found that the degradation rate of dry–wet-spun PLLA fibers was higher than those of the melt-spun or dry-spun ones. Furthermore, the compatibility between PLLA/CHS fabrics and osteoblast under the in vitro degradation was investigated for the potential application of using the PLLA/CHS fabrics as supporting materials for chest walls and bones. Cell strain hFOB1.19 human SV40-transfected osteoblast and PLLA/CHS mixed fabrics were incubated. The cell morphology at early stages of cultivation was also studied. Excellent adhesion between osteoblast and PLLA/CHS fabrics was observed, indicating good biocompatibility of the fabrics with osteoblast.

Abstract:OBJECTIVETo evaluate the biocompatibility of the super high molecular weight poly D,L-lactic acid (SHMW-PDLLA) implant.METHODSThe SHMW-PDLLA plates were implanted into the SD-rats between the masseter and ramus of the mandible. The blood specimens were gained at 3, 6, 9, 12 months after the operation. The proteins, electrolyte, enzyme and other indices were tested by use of Beckman automatic biochemical analysis device. The soft tissue specimens around the SHMW-PDLLA plates were gained at 3, 6, 9, 12 months after the operation and the tissue reaction was observed with the pathological and haematological methods.RESULTSThere were not any abnormal findings in the blood after the SHMW-PDLLA plates implanted in the body of SD-rats. The implanted SHMW-PDLLA plates were degraded gradually in 6 to 12 months after the operation. There was not any abnormal tissue reaction found to the soft tissue around the SHMW-PDLLA plates by histological and pathological observations.CONCLUSIONSThe SHMW-PDLLA implant has a good biocompatibility to SD-rats.

Abstract:Biodegradable chitosan-graft-polylactide (PLA-CS) copolymers were prepared by the grafting of a poly(L-lactide) (PLLA) or poly(D-lactide) (PDLA) precursor to the backbone of chitosan using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC center dot HCl) and N-hydroxysuccinimide (NHS) as a coupling agent. The blood and cell compatibility of the graft copolymers were investigated in comparison to PLLA and PDLA homopolymers. The coagulation properties of PLA-CS were evaluated by hemolysis, plasma recalcification, dynamic blood clotting and protein absorption assays. PLA-CS copolymers present similar hemolysis ratio and plasma recalcification time as PLA, but slower dynamic blood clotting and lower protein absorption. The cell viability was assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), agar diffusion and lactate dehydrogenase (LDH) experiments. All the samples presented no effect on the viability to cells. Inflammatory cytokine analysis using sandwich ELISAs revealed that PLA-CS would not stimulate inflammatory activity.
Interfacial Compatibilization into PLA/Mg Composites for Improved In Vitro Bioactivity and Stem Cell Adhesion

Abstract:The present work highlights the crucial role of the interfacial compatibilization on the design of polylactic acid (PLA)/Magnesium (Mg) composites for bone regeneration applications. In this regard, an amphiphilic poly(ethylene oxide-b-L,L-lactide) diblock copolymer with predefined composition was synthesised and used as a new interface to provide physical interactions between the metallic filler and the biopolymer matrix. This strategy allowed (i) overcoming the PLA/Mg interfacial adhesion weakness and (ii) modulating the composite hydrophilicity, bioactivity and biological behaviour. First, a full study of the influence of the copolymer incorporation on the morphological, wettability, thermal, thermo-mechanical and mechanical properties of PLA/Mg was investigated. Subsequently, the bioactivity was assessed during an in vitro degradation in simulated body fluid (SBF). Finally, biological studies with stem cells were carried out. The results showed an increase of the interfacial adhesion by the formation of a new interphase between the hydrophobic PLA matrix and the hydrophilic Mg filler. This interface stabilization was confirmed by a decrease in the damping factor (tanδ) following the copolymer addition. The latter also proves the beneficial effect of the composite hydrophilicity by selective surface localization of the hydrophilic PEO leading to a significant increase in the protein adsorption. Furthermore, hydroxyapatite was formed in bulk after 8 weeks of immersion in the SBF, suggesting that the bioactivity will be noticeably improved by the addition of the diblock copolymer. This ceramic could react as a natural bonding junction between the designed implant and the fractured bone during osteoregeneration. On the other hand, a slight decrease of the composite mechanical performances was noted.
