Abstract:A novel composite of carbon fiber(CF) reinforced hydroxyapatite(HA)/ploylactide(PLA) degradable biomaterial was developed by solvent blending.The mechanical properties and degradation properties in vitro of the composite were researched.The CF/HA/PLA composites have excellent mechanical properties.As the hydroxyapatite mass fraction increases,there are respectively peak values with flexural strength and flexural modulus,including maximums of flexural strength of 430MPa and flexural modulus of 26GPa.After degradating in vitro for 3 months,the flexural strength and flexural modulus fall 30% and 36% respectively.The SEM photos indicate that the degradation begins from the interfaces of the composite,after degradating,there are gaps at the interfaces,and the water uptake increases to 5%,but the mass loss is only 1.6%.The pH values of the PBS drop less than 0.1,which can do some favors to the recoveries of the fracture.The results show that the mechanical properties of the composite can meet the requirements of internal fracture fixation.
[Preliminary Evaluation of the Biological Properties of Poly-Lactide-co-glycolic Acid (PLGA) Knitted Mesh].

Abstract:This is a work aimed to investigate the biodegradability, biocompatibility and mechanical property of the poly-lactide-co-glycolic acid (PLGA) knitted mesh preliminarily and to further explore its applications in tissue engineering and regenerative medicine. The biological property of PLGA mesh was investigated comprehensively with the degradation experiment in vitro, the acute cytotoxicity assay, the intradermal irritation test and the subcutaneous implantation test in vivo utilized. The degradation experiment in vitro demonstrated that the pH value of the removed solution fluctuated between 6.68 and 7.33. The elastic modulus of the PLGA mesh increased at first and then decreased afterwards. The acute toxicity test and the intradermal irritation test indicated that the PLGA mesh was with innocuity safety. The PLGA mesh accelerated degradation and was replaced gradually by the neotissue. The results of immunohistochemical staining demonstrated that the number of ED-1+ cells increased at first and then decreased afterwards. The PLGA mesh with excellent mechanical properties, good biocompatibility and favorable degradation ratio has the potential to be employed as a "skeleton" to reinforce the mechanical property of collagen-based dermal substitutes in tissue engineering.

Abstract:In this study, poly(l-lactic acid) (PLLA) fibers were prepared by the dry–wet-spinning method, while chitosan (CHS) fibers were prepared via the wet-spinning method. The two fibers were blend spun and then fabricated into PLLA/CHS fabrics. In vitro degradation experiments of the fabrics were carried out in a phosphate-buffered solution at 37°C with a pH of 7.4. Changes in molecular parameters (molecular weights and molecular weight distributions), phase structures (crystallinities), morphologies (fiber surface topologies) of the PLLA fibers, and their macroscopic properties (the fabric weight losses and mechanical strengths) were monitored with degradation times. These results were compared with control samples with no degradation. The hydrolysis mechanism of PLLA/CHS fabrics was analyzed. It was found that the degradation rate of dry–wet-spun PLLA fibers was higher than those of the melt-spun or dry-spun ones. Furthermore, the compatibility between PLLA/CHS fabrics and osteoblast under the in vitro degradation was investigated for the potential application of using the PLLA/CHS fabrics as supporting materials for chest walls and bones. Cell strain hFOB1.19 human SV40-transfected osteoblast and PLLA/CHS mixed fabrics were incubated. The cell morphology at early stages of cultivation was also studied. Excellent adhesion between osteoblast and PLLA/CHS fabrics was observed, indicating good biocompatibility of the fabrics with osteoblast.

Abstract:The present work focusses on developing a completely sustainable, ecological, biodegradable, and eco-friendly composite material for edible as well as health product packaging utilizing an existing production technology. Ground chitin and chitosan powder were blended with eco-friendly polylactic acid (PLA) pellets and transformed into filaments by an extrusion process; thereafter composite parts produced by 3D printing were subjected to mechanical testing, morphological examination, crystallographic examination, chemical composition analysis and thermal analysis. Composite specimens containing chitin and chitosan, respectively, were obtained with varying mass fraction. The 3D printed composite specimen containing respectively chitin and chitosan yielded reduced tensile and flexural properties; for instance, for Chitin/PLA composite with 0.5 wt% (chitin), the tensile strength and flexural strength were 6.9 MPa and 10.4 MPa respectively. The addition of chitin reinforcement to the eco-friendly composite led to increased density with a maximum value of 1.285 g/cm 3 for Chitin/PLA composite at 0.5 wt% of chitin reinforcement. However, both Chitosan/PLA and Chitin/PLA composite exhibited enhanced ductility compared to neat PLA. Additionally, the micrographs of the composites revealed the presence of voids and impurity-like particle reflecting decomposed chitin and chitosan. Insights into the chemical bonding and composition were gained by analysing the Fourier transform infrared spectra; analysis of the X-ray diffraction pattern revealed the crystal size and crystallinity index with values of 7.7 nm and 62%, respectively (e.g. Chitin/PLA composite at 0.5 wt%). Finally, the thermogravimetric analysis revealed that both composite materials exhibited a thermal stability of around 257 °C. Altogether, these findings support the use of the Chitosan/PLA and Chitin/PLA composites in applications such as food product packaging.

Polylactic acid (PLA) is a thermoplastic polyester that has received widespread attention for its environmentally friendly origin and excellent performance, and its potential to address the current problems of severe white pollution and scarcity of petroleum resources. This article focuses on the synthesis, modification, degradation and application of PLA. The main focus is on the modification of PLA with different environmentally friendly materials (natural organic materials, biodegradable polymers, inorganic minerals) in blends for defects such as the brittleness of PLA. In addition, the applications of PLA composites in the fields of construction, medical, packaging and oil and water separation are also introduced, which hopefully will provide some help to the promotion of PLA.

Abstract:Calcium Sulfate Dihydrate/beta-Tricalcium Phosphate/Polylactide Composite was prepared and characterized. CS and beta-TCP were first mixed through ball milling, and then added into PLA solution with a mass fraction of 20%. After drying, composite sheets were obtained. The sheets were cut into pieces with a size of around 2 mmx5 mmx5 mm, and then used to injection molding. The tensile strength test shows that the tensile strength decreases with increasing CS content at the fixed total mass fraction of Cs and beta-TCP. After soaking in the Tris solution, the composites' mass is slowly reduced due to degradation. With increasing CS content, the degradation rate decreases. SEM of composites indicates that both pits and deposits appear on the surface after soaking. The results of cell culture in vitro prove that the cell adhesion and proliferation on the composites surface exhibit no significant difference with that of PLA's. The mechanical properties, degradation and biocompatibility of the CS/beta-TCP/PLA composites could meet the requirements of degradable bone fixation materials.

Abstract:In this study, composite films and scaffolds of polyester poly(3-hydroxybutyrate) and polysaccharide chitosan obtained via a simple and reproducible blending method using acetic acid as a solvent were considered. The degradation process of the films was studied gravimetrically in a model biological medium in the presence of enzymes in vitro for 180 days. The kinetics of weight reduction depended on the amount of chitosan in the composition. The biocompatibility of the films was evaluated using the Alamar blue test and fluorescence microscopy. The materials were non-cytotoxic, and the addition of poly(3-hydroxybutyrate) to chitosan improved its matrix properties on mesenchymal stem cells. Then, the 3D composites were prepared by freeze-drying. Their structure (using SEM), rheological behavior, moisture absorption, and porosity were investigated. The addition of different amounts of chitosan allowed us to vary the chemical and biological properties of poly(3-hydroxybutyrate) materials and their degradation rate, which is extremely important in the development of biomedical poly(3-hydroxybutyrate) materials, especially implantable ones.

Abstract:In this study, biodegradable polylactic acid (PLA)/chitosan (Cs) composites were produced via melt compounding and compression molding techniques. Various chitosan loadings of 2.5, 5, 7.5 and 10 parts per hundred parts of polymer (php) were incorporated into PLA and its effects on thermal, water absorption kinetics, tensile and morphological characteristics were investigated systematically. Thermal analysis indicated that an increase in chitosan loading of up to 10 php enhanced the crystallinity percentage (χc) of neat PLA to an extent of 51%, yet reduced the thermal stability of the resulting biocomposites. The kinetic study results revealed that water absorption of PLA/Cs biocomposites approached the Fickian diffusion behavior. The maximum water uptake (Msat) increased with chitosan addition, which can be attributed to stronger water–filler interaction. This was correlated to higher diffusion (D), solubility (S) and permeability (P) coefficients, which suggested the acceleration in diffusion rate and better water permeation through the biocomposites. In addition, the tensile results of dry samples showed enhancement in tensile strength and tensile modulus by 2% and 14%, respectively, relative to neat PLA through the incorporation of 2.5 php of chitosan loading. However, the water-immersed biocomposites demonstrated deterioration in all tensile properties (tensile strength, tensile modulus, and elongation-at-break values) which signified hydrolytic polymer degradation. This was confirmed by the FESEM micrographs of the fractured surfaces which exhibited filler pulled-out phenomenon and cavity formation after 50 days of water immersion.

Homogeneous Chitosan/Poly(L-Lactide) Composite Scaffolds Prepared by Emulsion Freeze-Drying

Abstract:The combination between chitosan (CS)-based hydrophilic extracellular matrix polysaccharide and polylactide (PLA)-based hydrophobic biodegradable aliphatic polyester is a challenge in the biomaterials field. This study investigated the formation of homogeneous chitosan/poly(L-lactide) (CS/PLLA) porous composite scaffold using a novel emulsion freeze-drying technique. An oil-in-water (O/W) emulsification system was used in the presence of surfactant Tween-80, in which CS solution was used as the water phase and PLLA solution was used as the oil phase. The composite scaffolds showed well interconnected pore structures and homogenous distribution of CS and PLLA when the PLLA volume fraction was not higher than 50%. Once the PLLA content increased to 75%, SEM micrographs demonstrated that the two components present phase separation region. FT-IR analysis revealed that there are strong hydrogen bond interactions between CS and PLLA components. The porosity of the CS/PLLA composites was in the range of 85-90% and showed a slight decrease with increasing PLLA dose. The mechanical properties of the composites lay between that of the pure CS and the PLLA scaffold. The compressive strength increased from 0.17 to 0.21 MPa, while the compressive modulus increased from 2.37 to 3.38 MPa as the PLLA contents increased from 25 to 75%. In vitro cytotoxicity was evaluated by MTT assay. The results indicated that MC3T3-E1 cell viability and proliferation in the CS/PLLA scaffold were comparable to that in the CS scaffold, and much higher than that in the PLLA scaffold. The successful hydrophilic polysaccharide and hydrophobic polyester system offers a new delivery method of growth factors and a novel scaffold design for tissue engineering.

Abstract:Polylactic acid (PLA) is considered a mature alternative to synthetic plastics made from petroleum by-products, possessing the advantages of good mechanical strength. However, it also has some disadvantages such as brittleness and low toughness. In order to overcome and improve some of these unfavorable properties, PLA/PBAT composites were prepared by blending PLA with Poly (butylene adipate-co-terephthalate) (PBAT), and adding 4,4′-methylene diphenyl diisocyanate (MDI) and chitosan nanoparticles (ChNPs) as compatibilizers to investigate the effects of different compatibilizers on the properties of the composites. The main observations are as follows: FT-IR indicated that MDI did not add new groups, while the addition of ChNPs added a substantial amount of hydroxyl and methylene groups. The addition of both MDI and ChNPs did not have any effect on the crystalline shape of the composites, but could potentially reduce their crystallinity, increase the melt peak temperature, wet the boundary of the PLA and PBAT phases, decrease the size of the dispersed phases, reduce the number of dispersed phases, and improve interfacial compatibility. The incorporation of MDI increased the tensile strength from 13.02 MPa to 19.24 MPa, whereas the addition of ChNPs substantially enhanced the elongation at the break from 3.84% to 19.24%. Furthermore, the inclusion of MDI conferred enhanced moisture resistance, whereas the addition of ChNPs seemed to weaken the resistance to moisture.

Abstract:Hydroxyapatite (HA)/polycaprolactone (PCL)-chitosan (CS) composites were prepared by melt-blending. For the composites, the amount of HA was varied from 0% to 30% by weight. The morphology, structure and component of the composites were evaluated using environmental scanning electron microscope, X-ray diffraction and Fourier transform infrared spectroscope. The tensile properties were evaluated by tensile test. The bioactivity and degradation property were investigated after immersing in simulated body fluid (SBF) and physiological saline, respectively. The results show that the addition of HA to PCL-CS matrix tends to suppress the crystallization of PCL but improves the hydrophilicity. Adding HA to the composites decreases the tensile strength and elongation at break but increases the tensile modulus. After immersing in SBF for 14 days, the surface of HA/PCL-CS composites are covered by a coating of carbonated hydroxyapatite with low crystallinity, indicating the excellent bioactivity of the composites. Soaking in the physiological saline for 28 days, the molecular weight of PCL decreases while the mass loss of the composites and pH of physiological saline increase to 5.86% and 9.54, respectively, implying a good degradation property of the composites.
Chitosan microspheres/nano-hydroxyapatite/collagen/polylactide composite

Abstract:Chitosan microspheres (CMs) loaded with bovine serum albumin (BSA) were prepared by an emulsion-ionic crosslinking method in the presence of tripolyphosphate (TPP). Porous CMs/nano-hydroxyapatite/collagen/polylactide (CMs/nHAC/PLA) composite was developed by thermally induced phase separation. The properties of the CMs and composite were investigated by SEM, laser particle size analyzer, mercury porosimeter and universal testing machine. The results indicate that the CMs are spherical in shape, with a regular surface. The diameters of the CMs are in the range of 20-50μm. With the increase of initial BSA dosage, the loading capacity is also increased from 0.78% to 2.74%, whereas the encapsulation efficiency is decreased from 86.9% to 78.4%. Controlling the CMs dosage to be not higher than 30% on a PLA weight basis, the CMs can distribute perfectly in the whole CMs/nHAC/PLA composite. The total porosity of the porous composite is over 83.1%. The pore diameter is about 100-200μm, and the compressive strength is about 1-2 MPa. This porous composite is a promising material for non-loaded bone implants and tissue engineering scaffolds.
Evaluation and Enhancement of Polylactic Acid Biodegradability in Soil by Blending with Chitosan

Abstract:This study highlights the soil burial degradation of polylactic acid/chitosan (PLA/Cs) biocomposites prepared by the melt compounding technique. The effect of various Cs loadings (2.5, 5, 7.5, 10 parts per hundred parts of polymer (php)) and soil burial periods (0, 2, 6, 12 months) on visual observation, weight loss, changes in functional groups, as well as tensile, thermal, and morphological properties were analyzed. The PLA/Cs biocomposites became brittle and showed more fragmentation with increasing Cs content and buried time. The result correlates with a remarkable increase in weight loss percentage of about ~ 192%, with Cs addition from 2.5 to 10 php at the end of soil degradation. Besides, a decrement in peak intensity at 1751 cm−1 and 1087 − 1027 cm−1 after 12 months signifies the breakdown of PLA ester bonds due to the hydrolytic degradation. This correlates to a significant drop of 60% and 55% in tensile strength and elongation at break, respectively, in the 2.5 php sample, whilst further Cs addition resulted in the broken of the biocomposites at the end of the soil degradation. Yet, no significant difference was observed in the tensile modulus. A consistent stiffness in the biocomposite suggests the degradation occurs in the amorphous region and leaves the crystalline part. This is proven by the 70% increment in crystallinity degree in all samples after 12 months of soil burial. Moreover, surface morphology showed numerous and extended crack formations. It proposes a notable deterioration effect of the biocomposite due to biodegradation. The hydrophilicity of Cs enhances water-polymer interaction, thereby accelerating the biodegradation of polymer components. Therefore, Cs could be a good candidate for facilitating PLA biodegradation in the natural soil environment.

Abstract:Polylactic acid (PLA) has garnered global attention as a potential biomaterial thanks to its biodegradability and renewability. However, the drawbacks of its high flammability impede its broader application. In this work, two different types of all bio-based charring agents (named as CFU, CVA) were prepared in a green and environmentally friendly ethanol solvent by using chitosan derived from corn starch and sugarcane, as well as bio-based vanillin and furfural, as raw materials. The CFU (or CVA) in conjunction with ammonium polyphosphate (APP) were incorporated into PLA to prepare flame-retardant and high-performance composite materials. The addition of 3.75 wt% CFU (or CFA) and 11.25 wt% APP to PLA resulted in the attainment of the UL-94 V-0 rating, a nearly 28% of limiting oxygen index, as well as a decrease of ∼35% and ∼90% in total heat and smoke releases, respectively. Besides, the addition of mixed flame retardants significantly reduced the viscosity of PLA by an order of magnitude, which was greatly beneficial for processing and production. The mechanical performance testing results demonstrated an increase in the elongation at break and impact strength of PLA composites containing CFU (or CVA) and APP. Moreover, the heat resistance and crystallinity of the PLA composites were also improved.

Abstract:Bacterial cellulose (BC) has become a universal biomaterial owing to its intrinsic properties. BC fibers are composed of microfibers with a diameter of 3–4 nm into fiber bundles with a thickness of 40–60 nm, and interweave with each other to form a well-developed ultra-fine network structure. Polylactic acid (PLA) has good mechanical properties and excellent biocompatibility and biodegradability. Therefore, PLA has been widely applied in tissue engineering. Addressed herein is a novel type of PLA/BC (PLA/BC) composite scaffold prepared by 3D printing (3D), 3D modeling of the required porous membrane material support established in the computer, and decomposition of the model into 5 layer 20 μM sheets. The range of PLA loadings assessed in this work was 1.0 wt.%, 1.5 wt.%, and 2.0 wt.%, and its physicochemical properties and biological properties were characterized and evaluated. Tensile strength of PLA/BC composite scaffolds increased to 66.49 MPa compared to that of a pure BC film (25.61 MPa). Hydrophilicity was tunable with the amount of added PLA. In this paper, the effects of 3D round hole and stripe surface topology on cell growth behavior were characterized. Schwann cells (SCs) adhered to the surface of the 3D composite membrane successfully, and their proliferation rate on the surface of the regular circular pore and stripe structure was better than that of the smooth surface. Erythrocyte fixation and platelet adhesion experiments showed that the 3D composite scaffold had excellent blood compatibility. Further degradation studies showed that loose structures appeared after 1 week, and structural defects began after 3 weeks. The in vitro degradation results showed that the degradation rate of the BC membrane in simulated body fluid after 6 weeks was 14.38%, while the degradation rate of the PLA/BC composite scaffold was 18.75%.

Abstract:Amphiphilic N,N-dilauryl chitosan/PLLA (NCS/PLLA) composite films with three different weight ratios were prepared by solution approach using CHCl3 as co-solvent. The miscibility of NCS/LLA composite systems is studied by FTIR, DSC, XRD and SEM techniques. It mainly arises from hydrogen-bonding interactions between the hydroxyl groups of NCS and the carbonyl groups of PLLA. The cell adhesion, cell viability and SEM experimental results indicate that NCS is benefit to improve the cell affinity of PLLA.

In this paper, a composite scaffold composed of polylactic acid (PLA) film and 3D printing scaffold was fabricated which could provide structural and mechanical support for tissue application. The effects of PLA film/3D printing composite scaffold on scaffold morphology, average diameter, pore diameter, mechanical properties, themal conductivity, electrical conductivity, and surface hydrophilicity were characterized. PLA film/3D printing composite scaffold had larger thickness, better mechanical and thermal conductivity properties in comparison with pure PLA film and 3D printed scaffold. These results suggested that composite scaffold with relatively good overall performance could be a potential candidate for tissue engineering application.</p>

Novel Biodegradable Films and Scaffolds of Chitosan Blended with Poly(3-Hydroxybutyrate).

Abstract:In order to develop a novel biomaterial, films of chitosan blended with poly(3-hydroxybutyrate) (PHB) were prepared by an emulsion blending technique and their properties were characterized. Scanning electron microscopy (SEM) showed that PHB microspheres were formed and were entrapped in chitosan matrices, which made the film surface rough. With increasing PHB content, the roughness of the film surface increased, while the swelling capability of the films decreased. In a wet state, the blended films exhibited a lower elastic modulus, a higher elongation-at-break and a higher tensile strength compared with chitosan films. Cell-culture experiments revealed that the blended films had better cytocompatibility than chitosan films. To explore the potential application of the blended material in tissue engineering, the porous blended scaffolds were fabricated and their pore morphology was observed by SEM. The results revealed that not only pore structure but also pore wall morphology of the blended scaffolds could be controlled by selecting the parameters of the fabrication process. These advantageous properties indicate that the blended chitosan/PHB material is promising for tissue engineering applications.

Abstract:The feasibility of perlite particles used in poly(ε-caprolactone) (PCL)/poly(lactic acid) (PLA) composite films by melt blending is explored to improve their mechanical property and analyze their antibacterial effect. The effect of perlite content on the mechanical, thermal, hydrophilic, and antibacterial properties of composite films is investigated. Results show that incorporation of 10 wt% perlite in PCL/PLA film improves the tensile strength and hydrophilicity by 1.2 times and 25 %, respectively. After perlite addition, the melting crystallization and glass transition temperature of PCL/PLA film are improved. The presence of perlite also confers antibacterial benefits to the composite film. PLA-based materials are used in the fields of medical materials and food packaging, and their ability to degrade in seawater has been a long-standing goal. In this study, the addition of PCL and perlite not only increases various properties and antibacterial effects, but the blending of inorganic materials and organic materials can destroy the link strength of polymer chain segments of organic materials and help them degrade in seawater. The prepared composite film features broad prospects for the development and application of various fields, such as food packaging and medical materials, reduce white pollution in the ocean.

Abstract:Composite materials prepared from polylactic acid have been used more and more widely in many fields. However, PLA has high brittleness, which largely restricts its application. Blending PLA with polycaprolactone (PCL) helps improve the disadvantages of pure PLA. In this study, different molecular weights of PCL were used and blended with PLA in different proportions to form a composite film by melt blending. The mechanical properties, thermal properties, and water wettability of Composite films are evaluated to examine the influences of the relative molecular weight of PCL and PCL/PLA ratio. When the relative molecular weight of PCL is 50,000, Composite films have high stress, strain, hydrophilicity, thermal properties, and UV-resisting property, but their thermal properties are not affected. At a specific relative molecular weight of PCL and PCL/PLA ratio of 30/70, the composite films reach their maximum stress and exhibit high strain, good hydrophilicity, and UV transmittance lower than 10%. Moreover, the PCL/PLA ratio has a marginal effect on the thermal properties of the composite films. This research has certain significance for promoting the application of PCL/PLA composite film in many fields.
