Abstract:A novel composite of carbon fiber(CF) reinforced hydroxyapatite(HA)/ploylactide(PLA) degradable biomaterial was developed by solvent blending.The mechanical properties and degradation properties in vitro of the composite were researched.The CF/HA/PLA composites have excellent mechanical properties.As the hydroxyapatite mass fraction increases,there are respectively peak values with flexural strength and flexural modulus,including maximums of flexural strength of 430MPa and flexural modulus of 26GPa.After degradating in vitro for 3 months,the flexural strength and flexural modulus fall 30% and 36% respectively.The SEM photos indicate that the degradation begins from the interfaces of the composite,after degradating,there are gaps at the interfaces,and the water uptake increases to 5%,but the mass loss is only 1.6%.The pH values of the PBS drop less than 0.1,which can do some favors to the recoveries of the fracture.The results show that the mechanical properties of the composite can meet the requirements of internal fracture fixation.
Characterization and Cytocompatibility of Premineralized Silk Fibroin/Chitosan Composite Scaffolds

Abstract:Silk fibroin/chitosan scaffolds were prepared through lyophilization, then hydroxyapatite was deposited by premineralization to obtain porous hydroxyapatite (HA)-silk fibroin/chitosan composite scaffolds.Composite scaffolds were characterized by Fourier transform infrared analysis (FT-IR), X-ray diffraction (XRD), scanning electronic microscopy (SEM), thermogravimetry analysis (TGA), compressive modulus and porosity, as well as the proliferation of MG-63 cells on composite scaffolds was detected by WST-1 method. The results reveal that the structure of deposited HA is a weak crystallinity, which deposits on the wall of pore. With the increase of premineralization cycles, the amount of HA and compressive modulus of premineralized composite scaffolds are significantly increased, the porosity keeps about 90%. WST-1 assay shows that premineralized composite scaffolds deposited with HA dramatically enhance the proliferation of cells, and possess a great cytocompatibility.
Preparation and mechanical properties of carbon fiber reinforced hydroxyapatite/polylactide biocomposites

Abstract:A novel biocomposite of carbon fiber (CF) reinforced hydroxyapatite (HA)/polylactide (PLA) was prepared by hot pressing a prepreg which consisting of PLA, HA and CF. The prepreg was manufactured by solvent impregnation process. Polymer resin PLA dissolved with chloroform was mixed with HA. After reinforcement CF bundle was impregnated in the mixture, the solvent was dried completely and subsequently hot-pressed uniaxially under a pressure of 40 MPa at 170 degrees C for 20 min. A study was carried out to investigate change in mechanical properties of CF/HA/PLA composites before and after degradation in vitro. The composites have excellent mechanical properties. A peak showed in flexural strength, flexural modulus and shear strength aspects, reaching up 430 MPa, 22 GPa, 212 MPa, respectively, as the HA content increased. Degraded in vitro for 3 months, the flexural strength and flexural modulus of the CF/HA/PLA fell 13.2% and 5.4%, respectively, while the shear strength of the CF/HA/PLA composites remains at the 190 MPa level. The SEM photos showed that there were gaps between the PLA matrix and CF after degradation. Water uptake increased to 5%, but the mass loss rate was only 1.6%. The pH values of the PBS dropped less than 0.1. That's because the alkaline of HA neutralize the acid degrades from PLA, which can prevent the body from the acidity harm.

Abstract:Nano-fibrous hydroxyapatite/chitosan (HA/CS) composites with HA contents of 070 wt % were prepared by in situ hybridization with a preprepared chitosan film as semipermeable membrane to slow down the hybridization process. The phase composition, microstructure, and morphology of the composites were characterized by means of Fourier transform infrared spectroscopy, X-ray diffraction, and high-resolution transmission electron microscopy. It was found that the in situ prepared inorganic phase was hydroxyapatite nano-fibers that were uniformly dispersed in the chitosan matrix. The average diameter of the fibers were about 3 nm, while the length of the fibers increases from 20 to 60 nm when the hydroxyapatite content increased from 10 to 70 wt %. The compressive strength and Young's modulus of these nano-fibrous HA/CS composites increased with the increasing HA content and reached the highest values of 170 and 1.7 GPa, respectively, at the HA content of 5070 wt %, which were much higher than the values of samples prepared by coprecipitation. Abstract:Chitosan(CS)/hydroxyapatite(HA) nanocomposite with high strength were synthesized via in-situ compositing at room temperature. The key of in-situ compositing is that a pre-precipitated CS membrane was used to obstruct the CS/HA precursor from NaOH aqueous solution. The CS membrane controlled the process of CS precipitation and transformation of HA from the precursor simultaneously and tardily. XRD, TEM and SEM were performed to investigate CS/HA nanocomposite. Results shown that the in-situ formed calcium phosphate in CS was exactly HA, and that nano-size HA granules were dispersed in CS matrix uniformly, and that the structure of CS/HA composite prepared by in-situ compositing is layered structure. The bending strength and modulus of CS/HA (10015, wt/wt) were 86MPa and 3.4GPa, which is much higher than that of cancellous bone, 22MPa and 1.2MPa, respectively.
Preparation and Characterization of Bionic Bone Structure Chitosan/hydroxyapatite Scaffold for Bone Tissue Engineering.

Abstract:Three-dimensional oriented chitosan (CS)/hydroxyapatite (HA) scaffolds were prepared via in situ precipitation method in this research. Scanning electron microscopy (SEM) images indicated that the scaffolds with acicular nano-HA had the spoke-like, multilayer and porous structure. The SEM of osteoblasts which were polygonal or spindle-shaped on the composite scaffolds after seven-day cell culture showed that the cells grew, adhered, and spread well. The results of X-ray powder diffractometer and Fourier transform infrared spectrometer showed that the mineral particles deposited in the scaffold had phase structure similar to natural bone and confirmed that particles were exactly HA. In vitro biocompatibility evaluation indicated the composite scaffolds showed a higher degree of proliferation of MC3T3-E1 cell compared with the pure CS scaffolds and the CS/HA10 scaffold was the highest one. The CS/HA scaffold also had a higher ratio of adhesion and alkaline phosphate activity value of osteoblasts compared with the pure CS scaffold, and the ratio increased with the increase of HA content. The ALP activity value of composite scaffolds was at least six times of the pure CS scaffolds. The results suggested that the composite scaffolds possessed good biocompatibility. The compressive strength of CS/HA15 increased by 33.07% compared with the pure CS scaffold. This novel porous scaffold with three-dimensional oriented structure might have a potential application in bone tissue engineering.

Abstract:Magnetite/hydroxyapatite/chitosan rods have been prepared via in situ compositing method. The mechanical properties were characterized and evaluated for suitability as a bone repair materials. Scanning electron microscopy (SEM) was carried out to investigate the microstructure of CS/HA/Fe3O4 composite. Effect of inorganic content on the mechanical properties of the CS/HA/Fe3O4 composite was also investigated. It was observed that the mechanical properties of the composite in the dry condition improved as the percentage of CS content increased. Mechanical properties of CS/HA/Fe3O4 composite rod are much better than that prepared via traditional method. Bending strength, bending modulus of CS/HA/Fe3O4 composite are 118.5 MPa, 2.3 GPa, respectively. All of these material properties are twice or three times stronger compared with that of nature bone of human being.

Abstract:Chitosan (CS) bicomponent nanofibers with an average diameter controlled from 100 to 50 nm were successfully prepared by electrospinning of CS and poly (vinyl alcohol) (PVA) blend solution. Finer fibers and more efficient fiber formations were observed with increased PVA contents. On this contribution, a uniform and ultrafine nanofibrous CS bicomponent mats filled with hydroxyapatite (HA) nanoparticles were successfully electrospun in a well devised condition. An increase in the contents of HA nanoparticles caused the conductivity of the blend solution to increase from 1.06 mS/cm (0 wt % HA) to 2.27 mS/cm (0.5 wt % HA), 2.35 mS/cm (1.0 wt % HA), respectively, and the average diameter of the composite fibers to decrease from 59 +/- 10 nm(0 wt % HA) to 49 +/- 10 nm (0.5 wt % HA), 46 +/- 10 nm (1.0 wt % HA), respectively. SEM images showed that some particles had filled in the nanofibers whereas the others had dispersed on the surface of fibers, and EDXA results indicated that both the nanoparticles filled in the nanofibers and those adhered to the fibers were HA particles. Effect of chitosan as a dispersant on collagen-hydroxyapatite composite matrices

Abstract:Purpose: Hydroxyapatite (HA) powder is difficult to disperse evenly in collagen (Col) gelatum, which can affect the properties of the final Col-HA composite. Here, we used chitosan (CS) as a dispersant in a bone matrix constructed with Col and Ca(5)(PO(4))(3)OH (HA), which are the main components of natural bone. Solid-liquid phase separation was used to shape a three-dimensional porous structure to support cell growth. Methods: The two bone matrices (Col-CS-HA and Col-HA) were investigated by transmission electron microscopy, scanning electron microscopy, and electron spectroscopy for chemical analysis for structural characteristics and physicochemical properties. Their ability to repair bone defects was evaluated in vivo. Results: HA particles were dispersed in Col gelatum evenly by mixing with CS gelatum. The surface morphology and three-dimensional structure of the matrix became more regular, which improved cell growth. After CS modification, the percentage of C-N groups increased to 6.40% and C-OH groups increased from 1.05% to 6.92%. The total amount of nonpolar groups (C-C groups and C-H groups) decreased from 82.48% to 74.05%. The vitrification and denaturation temperatures increased from 40.59 degrees C and 106.36 degrees C, respectively, to 42.23 degrees C and 111.04 degrees C, respectively. Conclusion: CS modified the surface chemistry to create a favorable environment for osteoblast adhesion and proliferation in vivo, which would accelerate the process of bone regeneration. The Col-CS-HA matrix possessed superior biological and mechanical properties for bone defect repair.
In vitro study of hydroxyapatite/polycaprolactone (HA/PCL) nanocomposite synthesized by an in situ sol-gel process

Abstract:Hydroxyapatite (HA) is the most substantial mineral constituent of a bone which has been extensively used in medicine as implantable materials, owing to its good biocompatibility, bioactivity high osteoconductive, and/or osteoinductive properties. Nevertheless, its mechanical property is not utmost appropriate for a bone substitution. Therefore, a composite consist of HA and a biodegradable polymer is usually prepared to generate an apt bone scaffold. In the present work polycaprolactone (PCL), a newly remarkable biocompatible and biodegradable polymer, was employed as a matrix and hydroxyapatite nanoparticles were used as a reinforcement element of the composite. HA/PCL nanocomposites were synthesized by a new in situ sol-gel process using calcium hydroxide and phosphoric acid precursors in the presence of Tetrahydrofuran (THF) as a solvent. Chemical and physical characteristics of the nanocomposite were studied by X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM) and Fourier transform infrared (FTIR) analyses. The results indicated that pure HA nanoparticles were well-incorporated and homogenously dispersed in the PCL matrix. It was found that the mechanical property of PCL was improved by addition of 20wt.% HA nanoparticles. Furthermore, the biological property of nanocomposites was investigated under in vitro condition. For this purpose, HA/PCL scaffolds were prepared through a salt leaching process and immersed in a saturated simulated body fluid (SBF) after 3 and 7days. It was found that a uniform layer of biomimetic HA could be deposited on the surface of HA/PCL scaffolds. Therefore, the prepared HA/PCL scaffolds showed good potential for bone tissue engineering and could be used for many clinical applications in orthopedic and maxillofacial surgery.
Preparation and Properties of Calcium Sulfate Dihydrate/beta-Tricalcium Phosphate/Polylactide Composite

Abstract:Calcium Sulfate Dihydrate/beta-Tricalcium Phosphate/Polylactide Composite was prepared and characterized. CS and beta-TCP were first mixed through ball milling, and then added into PLA solution with a mass fraction of 20%. After drying, composite sheets were obtained. The sheets were cut into pieces with a size of around 2 mmx5 mmx5 mm, and then used to injection molding. The tensile strength test shows that the tensile strength decreases with increasing CS content at the fixed total mass fraction of Cs and beta-TCP. After soaking in the Tris solution, the composites' mass is slowly reduced due to degradation. With increasing CS content, the degradation rate decreases. SEM of composites indicates that both pits and deposits appear on the surface after soaking. The results of cell culture in vitro prove that the cell adhesion and proliferation on the composites surface exhibit no significant difference with that of PLA's. The mechanical properties, degradation and biocompatibility of the CS/beta-TCP/PLA composites could meet the requirements of degradable bone fixation materials.

Abstract:Nanofibrous biocomposite scaffolds of chitosan (CS), PVA, and hydroxyapatite (HA) were prepared by electrospinning. The scaffolds were characterized by FTIR, SEM, TEM, and XRD techniques. Tensile testing was used for the characterization of mechanical properties. Mouse fibroblasts (L929) attachment and proliferation on the nano-fibrous scaffold were investigated by MTT assay and SEM observation. FTIR, TEM, and XRD results showed the presence of nanoHA in the scaffolds. The scaffolds have porous nanofibrous morphology with random fibers in the range of 100-700 nm diameters. The CS/PVA (90/10) fibrous matrix (Without HA) showed a tensile strength of 3.1 +/- 0.2 MPa and a tensile modulus 10 I MPa with a strain at failure of 21.1 +/- 0.6%. Increase the content of HA up to 2% increased the ultimate tensile strength and tensile modulus, but further increase HA LIP to 5-10% caused the decrease of tensile strength and tensile modulus. The attachment and growth of mouse fibroblast was on the Surface of nanofibrous structure, and cells' morphology characteristics and viability were unaffected. A combination of nanofibrous CS/PVA and HA that mimics the nanoscale features of the extra cellular matrix Could be promising for application as scaffolds for tissue regeneration, especially in low or nonload bearing areas. [A review of recent researches on correlation between ATP and acupuncture efficacies].

Abstract:It has been documented that adenosine triphosphate (ATP) is a multifunctional nucleoside triphosphate used in cells, including chemical energy transportation, extra- and intracellular signaling, cell structure maintaining, DNA and RNA synthesis, etc. In the present paper, the authors reviewed studies on the involvement of ATP in different efficacies of acupuncture intervention from the following four aspects. 1) ATP release in the stimulated acupoint area is one of the key factors for producing acupuncture analgesia; 2) Acupuncture induced suppression of ATP activity in the central nervous system results in pain relief; 3) ATP application on the human body surface may strengthen the sensation propagation along the meridian; 4) Favorable regulation of acupuncture intervention on the abnormal functional activities of some viscera often accompanies with an increase of ATP content and ATPase activity in the related internal organs. It has been proposed that ATP, Ca2+ and reactive oxygen species (ROS) are closely related each other in the life activities of the organism. Hence, a reasonable regulation on ATP levels in the related organs of the body may be a new approach for raising clinical therapeutic effects of acupuncture therapy.
Thermal behavior and water absorption kinetics of polylactic acid/chitosan biocomposites

Abstract:In this study, biodegradable polylactic acid (PLA)/chitosan (Cs) composites were produced via melt compounding and compression molding techniques. Various chitosan loadings of 2.5, 5, 7.5 and 10 parts per hundred parts of polymer (php) were incorporated into PLA and its effects on thermal, water absorption kinetics, tensile and morphological characteristics were investigated systematically. Thermal analysis indicated that an increase in chitosan loading of up to 10 php enhanced the crystallinity percentage (χc) of neat PLA to an extent of 51%, yet reduced the thermal stability of the resulting biocomposites. The kinetic study results revealed that water absorption of PLA/Cs biocomposites approached the Fickian diffusion behavior. The maximum water uptake (Msat) increased with chitosan addition, which can be attributed to stronger water–filler interaction. This was correlated to higher diffusion (D), solubility (S) and permeability (P) coefficients, which suggested the acceleration in diffusion rate and better water permeation through the biocomposites. In addition, the tensile results of dry samples showed enhancement in tensile strength and tensile modulus by 2% and 14%, respectively, relative to neat PLA through the incorporation of 2.5 php of chitosan loading. However, the water-immersed biocomposites demonstrated deterioration in all tensile properties (tensile strength, tensile modulus, and elongation-at-break values) which signified hydrolytic polymer degradation. This was confirmed by the FESEM micrographs of the fractured surfaces which exhibited filler pulled-out phenomenon and cavity formation after 50 days of water immersion.

Characterization, Thermal and Biological Properties of PCL/PLA/PEG/N-HA Composites

Abstract:Initially, the use of Additive Manufacturing (AM) technology is mainly for the production of prototypes of a new invention and design. However, the AM technology had extended to the tooling industry, medical scaffolding, and direct part production. This is due to the extensive research and development carried out around the globe regardless of the material, equipment, and technology of AM. In this study, characterization and properties of PCL/PLA/PEG/Nano-HA polymer composites were investigated. Polycaprolactone (PCL), polylactic acid (PLA), polyethylene glycol (PEG), and nano-hydroxyapatite (n-HA) composites were prepared by melting and compounding the mixture using a Brabender machine. The composition of the materials was determined by previous research that it is the optimum recipe, with the ratio of PCL:PLA is 7:3 while the composition of PEG and nano-HA is 5% per hundred resin each. The temperature of the Brabender machine was set at 160°C, and the speed of the mixer was set at 30 rpm. After the mixing, the composites were characterized by using Fourier Transform Infrared Spectroscopy (FTIR) to determine the spectrum and quality of the composite mixed. Based on the spectrum, it can be verified that the characteristic of n-HA powder had been incorporated well into the PCL/PLA/PEG polymer blend. The thermal properties of the composites were investigated by using Thermogravimetry Analyzer (TGA). The results showed that the addition of n-HA lowered the initial degradation temperature and peak degradation temperature. Apparently, the addition of n- HA did not increase the degradation temperature of the composites. Furthermore, Simulated Body Fluid (SBF) test was carried out to access the bioactivity properties of the composites after the addition of n-HA. The results had proved that the addition of n-HA had triggered the growth of apatite layer on the surface of the samples treated with SBF. The growth of the apatite layer was verified by the X-ray Diffraction pattern, and the results proved the initial assumptions.

Abstract:Organic and inorganic composite material is becoming a solution on making the mechanical and degradation properties of biomaterial more suited. Porous calcium polyphosphate was immersed into different concentrations of carboxymethyl chitosan before immersing 10% alginate dialdegyde. After freeze-drying, the scaffolds were performed in physiologic saline. At stated day, the weightloss, Ca2+ concentration, pH value and morphology were measured. The biocompatibility of the composite was demonstrated by extract and direct contact tests. As the results showed, the degradation rates of composites were faster, and the compressive strength became bigger because of the cross-linked network formed by Carboxymethyl chitosan (CMC) and alginate dialdehyde (ADA). The pH value of composite was higher than that of calcium polyphosphate (CPP) due to the organic part of composite’s pH was in slight alkaline. From the SEM, the cross-linked network structure could be observed clearly. Because the glycosaminoglycans-like chains in CMC molecules, which are typically presented in extracellular matrix (ECM), extractions of composite material gave the cells good adhesion and growth condition. All the results testified the composite scaffold was a good candidate for bone repair.

Abstract:Hydroxyapatite (HA) is widely used in bone repair and regeneration because of its composition and structure similar to human bone. However, the artificially synthesized HA has poor mechanical properties, so it is often necessary to add a second phase to the powder as a supporting phase to enhance its mechanical properties. Strontium (Sr) has the properties of promoting the growth of bone cells, which can increase bone formation in the body and reduce bone self-absorption. Nevertheless, Sr-doped HA composites have been rarely reported in the literature in recent years. In this paper, Sr-doped HA was employed to be complexed with chitosan (CS) for the first time to prepare a composite coating that is dense and uniform, with good adhesion and ductility, in order to expand its clinical applications. Therefore, a series of HA samples doped with different Sr mole fraction (0 mol% (pure HA), 0.5 mol%, 1 mol%, 2 mol%, 4 mol%) were prepared by chemical precipitation method. Then the HA/CS composite coating doped with Sr was prepared on the stainless steel substrate by spin-coating technology. The results showed that Sr2+ ions had replaced part of Ca2+ ions and have been successfully doped into the HA lattice. The synthesized HA particles showed a shortrod-like morphology, and the substitution of Sr ions promoted the growth of Sr-doped HA crystals and increased its grain size. In addition, the synthesized Sr-doped HA/CS composite coating is dense and uniform. In vitro cell culture experiments showed that all composite coatings have good biocompatibility. The HA/CS coating with 4 mol% Sr ion doping is more conducive to cell proliferation, adhesion and osteogenic differentiation.

Abstract:Familial chylomicronemia is a rare genetic disorder attributable to the absence of lipoprotein lipase activity or the absence of apo-CII, i.e., the cofactor for the same enzyme. Plasma lipoproteins were analyzed by zonal ultracentrifugation under rate flotation conditions in four patients with lipoprotein lipase deficiency and two patients with apo-CII deficiency. Lipoproteins of density less than 1.006 gm/ml, and particularly lipoproteins with Sf greater than 100, were present in very high concentrations. Low levels of density greater than 1.006 gm/ml lipoproteins were observed. This fraction was composed of some different and discrete lipoprotein populations: intermediate-density lipoproteins (in three of six patients, density = 1.006 to 1.019 gm/ml); low-density lipoprotein LDL2 (in all patients, density = 1.019 to 1.045 gm/ml); low-density lipoprotein LDL3 (in all patients, density = 1.045 to 1.063 gm/ml); high-density lipoprotein HDL2 (in four of six patients); and high-density lipoproteins HDL3 (in all patients). LDL3 was never observed in normal participants by means of zonal ultracentrifugation; this subclass of low-density lipoproteins seems to correspond to LDL particles of very low Sf (2 to 5) previously identified by analytical ultracentrifugation in patients with severe hypertriglyceridemia. LDL3 was isolated by means of zonal ultracentrifugation as a single and discrete peak in all patients. Lipoproteins of density greater than 1.006 gm/ml were rich in triglycerides and poor in cholesterol in comparison with normal lipoproteins. The heterogeneity of low-density lipoproteins (particularly the appearance of LDL3), low levels of total high-density lipoproteins, and lower HDL3 flotation rate than normal are typical aspects of serum lipoproteins in these patients. No significant differences in the lipoprotein profiles of the patients with lipoprotein lipase deficiency in comparison with patients with apo-CII deficiency were found. In both groups of patients, the plasma lipoproteins profile and the altered lipoprotein composition could be related to the impaired catabolism of triglyceride-rich lipoproteins caused by the absence of lipoprotein lipase activity.

Abstract:In this study, orthogonal experiments were designed to explore the optimal process parameters for preparing polycaprolactone(PCL) scaffolds by the near-field direct-writing melt electro-spinning(NFDWMES) technology. Based on the optimal process parameters, the PCL scaffolds with different thicknesses, gaps and structures were manufactured and the corresponding hydrophilicities were characterized. The PCL scaffolds were modified by chitosan (CS) and hyaluronic acid(HA) to improve biocompatibility and hydrophilicity. Both Fourier transform infrared spectroscopy(FTIR) analysis and antibacterial experimental results show that the chitosan and hyaluronic acid adhere to the surface of PCL scaffolds, suggesting that the modification plays a positive role in biocompatibility and antibacterial effect. The PCL scaffolds were then employed as a carrier to culture cells. The morphology and distribution of the cells observed by a fluorescence microscope demonstrate that the modified PCL scaffolds have good biocompatibility, and the porous structure of the scaffolds is conducive to adhesion and deep growth of cells.

Abstract:Hydroxyapatite/polycaprolactone (HA/PCL) composites have been extensively explored in laser powder bed fusion (L-PBF) for bone tissue engineering. However, conventional mechanical mixing methods for preparing composite powders often yield inhomogeneous compositions and suboptimal flowability. In this study, HA/PCL powders were prepared and optimized for L-PBF using the modified emulsion solvent evaporation method. The morphology, flowability and thermal and rheological properties of the powders were systematically investigated, along with the mechanical and biological properties of the fabricated specimens. The HA/PCL powders exhibited spherical morphologies with a homogeneous distribution of HA within the particles. The addition of small amounts of HA (5 wt% and 10 wt%) enhanced the processability and increased the maximum values of the elastic modulus and yield strength of the specimens from 129.8 MPa to 166.2 MPa and 20.2 MPa to 25.1 MPa, respectively, while also improving their biocompatibility. However, excessive addition resulted in compromised sinterability, thereby affecting both mechanical and biological properties.
