Amperometric Ethanol Biosensor Based on Layer-by-Layer Deposition of Alcohol Oxidase on the Platinum Electrode

Abstract:A novel amperometric ethanol biosensor was developed based on layer-by-layer (LbL) technique to immobilize alcohol oxidase (AOx) on the Platinum (Pt) electrode surface. First, Pt, glassy carbon (GC), and gold (Au) electrodes were used as base electrodes for constructing ethanol biosensors, and the responses of these biosensors were compared. Second, The LbL films were formed by an alternate adsorption of poly(diallydimethylammonium chloride) (PDDA) and AOx on the surfaces of the electrodes. The results showed that Pt electrode coated with the five-bilayer (PDDA/AOx), film was the best one among the ethanol biosensors prepared. Third, the pH values and applied potential were studied. The performance of the optimized biosensor showed sensitive determination of ethanol with a linear range from 2.0 x 10(-4) M to 4.0 x 10(-1) M, with a remarkable sensitivity of 38 mu A mM(-1) cm(-2), a detection limit of 3.0 x 10(-6) M estimated at a signal-to-noise ratio of 3, and a response time of less than 10 s. The ethanol sensors could be used to determine ethanol in serum without any significant interference originated from electroactive compounds. The results showed that LbL deposition was a useful technique for AOx immobilization in ethanol biosensors fabrication.
