Chitosan-Silica Hybrid Matrix For Immobilized Metal Chelated Affinity Chromatography

Abstract:Chitosan-silica hybrid beads (CSHB) bearing with metal ion chelating group was prepared by sol-gel method and micro-emulsion technique. The condition for the preparation of CSHB, as well as its structure and morphology were investigated. The experimental results show that rigid beads with narrow size distribution could be obtained, when the component in miero-emulsion system was controlled in the following conditions: 100 mL chitosan (2%, m/V), 100 mL span emulsifier, 250 mL cyclohexane, 13. 3 mL Tetraethoxysilane (TEOS), 1.33 mL 3-Glyeldoxypropyltrimrthoxysilane (GPTMS) and 0.802 g Iminodiacetic acid (IDA). FTIR verified that the prepared beads were multi-component hybrid materials. The data of DSC-TG show that phase transition peak of the beads shift to higher temperature with the increasing of GPTMS content, which indicate that GPTMS plays an important role in enhancing the interaction of different component. The CSHB could adsorb metal chelating protein through coordinating force after the beads was loaded with heavy metal ions such as CU2+ and Ni2+. Bovine serum albumin (BSA) could be reversibly adsorbed on CU2+-CSHB column and eluted by imidazole with recovery rate of 76.6%; meanwhile only 4.7% BSA was adsorbed on CSHB column, indicating the low nonspecific adsorption of BSA on the column. Ni2+-CSHB column also show some purification effect for the catalase (CAT). The enzyme purification fold could reach 2.43 after one step of process. This novel hybrid beads could be potentially use in the purification of his-tagged protein in bioengineering process.
