Abstract:In order to monitor the process of cellular metabolism and electrophysiology,we developed a novel biochip,on which interdigited electrodes(IDE),microelectrode arrays(MEA) and light-addressable potentiometric sensor(LAPS) were integrated.It can be used in real time detection of cell-electrode impedance,extracellular field potential and cellular ionic metabolism.In the design,the three parts of functions were integrated in a multi-channel way.The size of electrodes,inter-electrode distance and surface properties of electrodes were optimized to reduce the electric disturbance between electrodes and in the mean time to improve performance.In the fabrication,the SiO2 layer was used as the insulation layer of MEA and IDE as well as the protective layer of LAPS,by which three types of sensors were integrated onto a single silicon wafer compatibly.The testing result showed that this integrated cellular chip could provide more than a single parameter with similar performance of separately fabricated chips.Above all,it would be a potential platform to study the electrophysiology and metabolism of cells,and further the pharmacological assays and bionic research.

Abstract:Bio-cellular chips have been important tools to detect extracellular physiology in post-gene era,but most of these tools are confronted with the embarrassments of mono-function,unstable parameters,low S/N ratio and difficulty of repetition.However,the integrated chip can transform the electrical,chemical and motile signals of cells into detectable ones by the integration and network of different sensor units with on-line analysis.The research on a novel integrated cellular physiologic sensor is reported in this article,which includes Micro-electrodes Array Sensors(MEAS),Interdigitated Array(IDA) for impedimetric analysis,and Light-addressable Potentiometric sensors(LAPS),etc.to record the extracellular metabolism chemical and biological substances,action potential and livable characteristics of living cells.The cell-sensor interfacial model of each unit was first introduced,the characteristic curves were then measured,and the fundamental electro-physiological cellular analysis was finally tested and analyzed.The average noise level of MEA is about 80 μV after measuring solution was added,the chip is biocompatible for cell culture and suitable for AP measurement;cultured kidney cells on the surface of IDA chip will cause an cellular impedance change of 12%～16% when exposed to 5-Fluororacil;and the sensitivity of H+-LAPS is 50.65 mV/pH.When kidney cells were cultured on LAPS and exposed to high concentration of 5-Fluororacil,the normal metabolism of cells will be affected,which was similar with the IDA results.This research will be interesting and useful for multi-functionalized cellular detecting platform and facilitate a lot to the lab-on-chip research in near future.

Abstract:Microfluidic devices provide powerful platforms for single-cell transcriptome analysis for many advantages such as highly parallel processing and low consumption of reagents. We have elaborated an integrated system with individual controller for effective single-cell transcriptome analysis. Inside this integrated microfluidic device, the cells are individually manipulated and isolated in an array using micro sieve structures, then transferred into different nanoliter reaction chambers for parallel processing of single-cell transcriptome analysis. The result demonstrates that this platform has the potential for assessment of genetic stability in cancer diagnosis.

Abstract:The life science has entered the era of post-genomics.One of the most important tasks of present life science research is to annotate and verify human genomic data already obtained.Microarrays of DNA and protein and lab-on-chip all play crucial roles in uncovering the functions of the genomes and in analyzing the specific genomic relationships of the interacting networks.At the same time, microarray technologies can provide tremendous amount of data for biomedical researchers, and is becoming an ever-powerful tool for system biology.All these shall contribute to the eventual elucidation of the functional genomics.

Abstract:Biochemical analysis is crucial in understanding mechanism of life activities and giving biological insights into life process. With inherent merits in flexible design, microscale of operation, good incorporation with other techniques for manipulation and detection, microfluidic chip has introduced new paradigms in biochemical analysis field. Considering the explosive development of microfluidics over the past decades, in this review, we summarized recent advances in biochemical analysis on microfluidic platforms. Highlight was put on the integrated technologies such as optical, electrical, acoustic and magnetic techniques. Focus also was given on relative applications in biomimetics, drug screening, biomolecular detection, single cell and stem cell analysis.

On-line cell lysis and DNA extraction on a microfluidic biochip fabricated by microelectromechanical system technology

Abstract:Integrating cell lysis and DNA purification process into a micrototal analytical system (microTAS) is one critical step for the analysis of nucleic acids. On-chip cell lysis based on a chemical method is realized by sufficient blend of blood sample and the lyzing reagent. In this paper two mixing models, T-type mixing model and sandwich-type mixing model, are proposed and simulation of those models is conducted. Result of simulation shows that the sandwich-type mixing model with coiled channel performs best and this model is further used to construct the microfluidic biochip for on-line cell lysis and DNA extraction. The result of simulation is further verified by experiments. It asserts that more than 80% mixing of blood sample and lyzing reagent which guarantees that completed cell lysis can be achieved near the inlet location when the cell/buffer velocity ratio is less than 1:5. After cell lysis, DNA extraction by means of a solid-phase method is implemented by using porous silicon matrix which is integrated in the biochip. During continuous flow process in the microchip, rapid cell lysis and PCR-amplifiable genomic DNA purification can be achieved within 20 min. The potential of this microfluidic biochip is illustrated by pretreating a whole blood sample, which shows the possibility of integration of sample preparation, PCR, and separation on a single device to work as portable point-of-care medical diagnostic system.

Abstract:The human body can be viewed as an organism consisting of a variety of cellular and non-cellular materials interacting in a highly ordered manner. Its complex and hierarchical nature inspires the multi-level recapitulation of the human body in order to gain insights into the inner workings of life. While traditional cell culture models have led to new insights into the cellular microenvironment and biological control in vivo, deeper understanding of biological systems and human pathophysiology requires the development of novel model systems that allow for analysis of complex internal and external interactions within the cellular microenvironment in a more relevant organ context. Engineering organ-on-chip systems offers an unprecedented opportunity to unravel the complex and hierarchical nature of human organs. In this chapter, we first highlight the advances in microfluidic platforms that enable engineering of the cellular microenvironment and the transition from cells-on-chips to organs-on-chips. Then, we introduce the key features of the emerging organs-on-chips and their proof-of-concept applications in biomedical research. We also discuss the challenges and future outlooks of this state-of-the-art technology.

Abstract:The real-time screening of biomolecules and single cells in biochips is extremely important for disease prediction and diagnosis, cellular analysis, and life science research. Barcode biochip technology, which is integrated with microfluidics, typically comprises barcode array, sample loading, and reaction unit array chips. Here, we present a review of microfluidics barcode biochip analytical approaches for the high-throughput screening of biomolecules and single cells, including protein biomarkers, microRNA (miRNA), circulating tumor DNA (ctDNA), single-cell secreted proteins, single-cell exosomes, and cell interactions. We begin with an overview of current high-throughput detection and analysis approaches. Following this, we outline recent improvements in microfluidic devices for biomolecule and single-cell detection, highlighting the benefits and limitations of these devices. This paper focuses on the research and development of microfluidic barcode biochips, covering their self-assembly substrate materials and their specific applications with biomolecules and single cells. Looking forward, we explore the prospects and challenges of this technology, with the aim of contributing toward the use of microfluidic barcode detection biochips in medical diagnostics and therapies, and their large-scale commercialization.

Abstract:Microsystems create new opportunities for the spatial and temporal control of cell growth and stimuli by combining surfaces that mimic complex biochemistries and geometries of the extracellular matrix with microfluidic channels that regulate transport of fluids and soluble factors. Further integration with bioanalytic microsystems results in multifunctional platforms for basic biological insights into cells and tissues, as well as for cell-based sensors with biochemical, biomedical and environmental functions. Highly integrated microdevices show great promise for basic biomedical and pharmaceutical research, and robust and portable point-of-care devices could be used in clinical settings, in both the developed and the developing world.

Abstract:In vitro models are essential to a broad range of biomedical research, such as pathological studies, drug development, and personalized medicine. As a potentially transformative paradigm for 3D in vitro models, organ‐on‐a‐chip (OOC) technology has been extensively developed to recapitulate sophisticated architectures and dynamic microenvironments of human organs by applying the principles of life sciences and leveraging micro‐ and nanoscale engineering capabilities. A pivotal function of OOC devices is to support multifaceted and timely characterization of cultured cells and their microenvironments. However, in‐depth analysis of OOC models typically requires biomedical assay procedures that are labor‐intensive and interruptive. Herein, the latest advances toward intelligent OOC (iOOC) systems, where sensors integrated with OOC devices continuously report cellular and microenvironmental information for comprehensive in situ bioanalysis, are examined. It is proposed that the multimodal data in iOOC systems can support closed‐loop control of the in vitro models and offer holistic biomedical insights for diverse applications. Essential techniques for establishing iOOC systems are surveyed, encompassing in situ sensing, data processing, and dynamic modulation. Eventually, the future development of iOOC systems featuring cross‐disciplinary strategies is discussed.
