Abstract:Isozymes have been extensively employed for the past 30 years to investigate a wide variety of aspects of pure and applied entomology, including phylogeny and taxonomy, population genetics, insecticide resistance, development and infection by hymenopterous parasitoids and entomopathogenic fungi. Recently, entomologists have also exploited the potential of DNA techniques for such studies. These have proved particularly useful to investigate mating relationships in the Hymenoptera and clonal structure in aphids, both groups having generally lower levels of enzyme polymorphism than many other insects, and in insect phylogeny. Since DNA techniques reveal greater amounts of genetic variability compared with isozymes, they allow examination of phylogenetic relationships and population structure and dynamics hitherto impossible with enzyme markers. Some DNA techniques, unlike isozymes, also work with dried or ancient samples. Examination of DNA variability from dried insect specimens has potential for study of long-term temporal as well as spatial genetic changes.This paper briefly describes the main DNA techniques now in use and their application, particularly in relation to agricultural entomology.
