Abstract:The principle and characteristics of RAPD molecular marker were introduced. Its applications on germplasm genetic diversity analysis, phylogenetic relationship study and cultivars purity identification of turfgrass plant were reviewed. Its problems and prospect in the study of turfgrass plant were also explained.

Abstract:About the research of the plant genetics breeding, people always tended to use the genetic marker methods, which were easy to identify the results, to study the heredity and variation rules. The genetic markers that were involved should possess these characteristics, which were heritable, special, and easily identified expression form. And currently these genetic markers contained four types which were morphology marker, cell marker, biochemistry marker and molecular marker. Among them, molecular marker technique has been widely used in plant genetic map construction, system development analysis, germplasm resources appraisal, molecular marker-assisted breeding, virus identification, somatic hybrid identification, registration of new crops, and the protection of patent -related, and other aspects. This paper mainly introduces the generation, development, principle, and application of the Random Amplified Polymorphic DNA (RAPD).

Abstract:Random amplified polymorphic DNA(RAPD), a polymerase chain reaction based technique testing DNA polymorphisms, has been widely used in genome research. In the field of animal genetics and breeding, RAPD technique provides a powerful tool for detection of gene diversity of populations and analysis of genetic relationship. It also can be used for analysis of linkage between RAPD markers and quantitative trait loci (QTL) and Markers Assisted Selection (MAS). Theory and features of RAPD were also discussed in this paper. According to the authors, RAPD technique has good perspective.

Abstract:Bermudagrass (Cynodon ssp.) germplasm is genetically diverse and widely distributed in the world. The study was conducted to identify and assess the molecular variation and relationship among 24 cultivars developed in China, Australia and the USA. Sequence-Related Amplified Polymorphism (SRAP) was applied to cultivars identification in this study for the first time. Thirty of the 90 SRAP primer combinations generated a total of 274 clearly bands encompassing 249 (91%) polymorphic. Each bermudagrass cultivar has its unique binary code and can be distinguished from the others. Three distinct clusters were obtained by unweighted pair-group method with arithmetic averages based on the polymorphic markers. Coefficients of genetic distance among the genotypes ranged from 0.57 to 0.97. The results demonstrated that SRAP marker is a stable molecular marker technique for the identification of bermudagrass cultivars and their genetic relationships.

Abstract:In recent years, application of molecular markers such as AFLP, RAPD and SSR markers have speeded up the step of utilization in cucurbita genetics and breeding. The progress of genetic map construction, genetic diversities, relationship of species (varieties), marker-assisted selection, construction of DNA fingerprint library and identification of F1 hybrid purity by using molecular markers was reviewed in watermelon and melon. Random amplified polymorphic DNA (RAPD) is a high-efficiency technology for the polymorphism of genome DNA analysis; it has such advantages as quickness, simplicity and effectiveness. Application of RAPD is mainly summarized in this article. The problem existent in application and its prospect are also discussed.

Abstract:Sequence characterized amplified region(SCAR) is a new molecular single-site markers based on PCR.Compared with other markers,it has such merit as low price,high stability,site-specific and so on.It has been applied in molecular assisted selection,linkage analysis,precisely location of target gene,discriminate in cultivars and so on in recent years.After brief review on recent development of SCAR on plant,it was suggested that research should be enhanced on application of SCAR in grass for such purpose as molecular assisted selection,variety evaluation.

Abstract:The article discussed the advantages and disadvantages of using morphological markers, cytological markers, biochemical markers and molecular markers in the identification of crop cultivars, focusing on the application of moleuclar marker fingerprint in identifying crop cultivars. In addition to the discussion of the application of protein eletrophoresis fingerprint, which is produced by the electrophoresis of isozyme and depot protein (prolamin, albumin, glutelin, globulin, etc.), recent DNA fingerprint techniques of RFLP, RAPD, minisatelite DNA, microsatellite DNA, ISSR, AFLP, CAPS and SNPS, and their contribution to the identification of crop cultivars, new cultivars registration, the inspection of cultivar purity, the relationship among cultivars and the studies in classification are thoroughly discussed as well.
