Abstract:Regioselective acetylation of per-O-TMS carbohydrates was achieved preferentially at the 6 position or 1,6-position under mild conditions involving the eco-friendly solvent acetonitrile, at room temperature, in ambient atmosphere and in a shorter time. Good or moderate yields were obtained via 4-dimethylaminopyridine, without auxiliary equipment. A single α-O-Acetyl acetylation anomer was exclusively defined for the involved substrates. A 6-O-monoacetate derivative was applied and used as a stable glycosyl donor in the disaccharides construction. The methodology was successful for a range of substrates, which include the following: d-lactose, d-trehalose, d-galactose, methyl α-d-galactose, d-glucose, d-mannose, d-xylose, and l-fucose.

Abstract:A convenient method to perdeuterate aromatic compounds is reported. Commercially available nonaflic acid (C4F9SO3H) in combination with C6D6 as the deuterium source provided access to a series of highly labeled arenes. Some of them were then used as building blocks for the preparation of functionalized deuterated products. A wide range of synthetic procedures were investigated. They involved traditional methods such as halogenation or nucleophilic substitution, but also metal‐catalyzed cross coupling and C−X bond activation. The transformations comprised redox‐neutral, oxidative as well as reductive reactions, and mechanistically, ionic, radical and pericyclic processes were covered. Under many conditions, the overall degree of deuteration remained high; in C−X activation reactions, a more pronounced re‐protonation was observed.

Abstract:The acetyl ester plays an important role for protection of the hydroxyl groups in carbohydrates synthesis. In the present study, we described an efficient deprotection of acetyl group of pentacyclic triterpenoid by using methanolic ammonia in THE solution. Good selectivity for cleaving gal-C2-OAc group of 3 beta-hydroxy-olean-12-en-28-oic acid 28-N-2,3,4,6-tetra-O-acetyl-beta-D-galactopyranoside (3) was achieved in the presence of methanolic ammonia within 4 h at low temperature (-60 degrees C) in a yield of 56%. The reaction disclosed here provides a new method for the synthesis of C2 selective modified carbohydrates, which is more useful than conventional synthesis procedure that usually requires many steps including temporary regioselective protection and deprotection. When the reaction temperature was increased from -60 degrees C to room temperature, the cleavage of the other three acetyl groups of galactose in an order of C4-OAc > C3-OAc > C6-OAc was observed. Abstract:An efficient method for selective anomeric deacetylation of peracetylated mono-, di-, and trisaccharides has been developed by using 2 mol% Hf(OTf)4 as catalyst in acetonitrile. Employment of ultrasonic irradiation could significantly accelerate the reaction rate. Mechanistic study confirmed the hydrolysis nature of this reaction, and NMR experimental data suggested that multiple peracetylated saccharide molecules may ligate to Hf(IV) cation primarily via the anomeric acetate to promote its specific hydrolysis.

Abstract:First use of benzylamine as a mild and efficient reagent for removal of the anomeric acetyl group of 1,3,4,6-tetra-O-acetyl-2-deoxy-2-phthalimido-b-D-galactopyranose (2) is described. Treatment of 2 with an excess of benzylamine in anhydrous THF at room temperature for 24h gave 3,4,6-tetra-O-acetyl-2-deoxy-2-phthalimido-b-D-galactopyranose with the yield of more than 80%.

Abstract:A convenient methodology has been developed for the regioselective removal of primary trimethylsilyl group (TMS) of various per-O-TMS-protected carbohydrates by inexpensive ammonium acetate. After acetylation and trichloroacetimidation of 6-hydroxyl sugar 1b, other TMS groups of 1d and 1c were inert to ammonium acetate in the same conditions, and this approach was also successfully applied in TMS-protected sphingosine.

Abstract:The synthesis of 2, 4-diisopropyl-5,5-dimethyl-1,3-dioxane through the acetalization of isobutyraldehyde with 2, 2, 4-trimethyl-1,3-pentanediol (TMPD) catalyzed by the novel carbon based acid was first carried out. High conversion (≥98%) and specific selectivity were obtained using the novel carbon based acid, which kept high activity after it was reused 5 times. Moreover, the catalyst could be used to catalyze the acetalization and ketalization of different aldehydes and ketones with superior yield. The yield of several products was over 90%. The novel heterogeneous catalyst has the distinct advantages of high activity, strikingly simple workup procedure, non-pollution, and reusability, which will contribute to the success of the green process greatly.

Abstract:Herein, a selective protocol for the complete removal of acetyl protecting groups from carbohydrate derivatives based on copper nanoparticles supported on activated carbon is presented. The deacetylation procedure occurs under neutral conditions, is applicable to a wide range of substrates and tolerates a variety of functional groups (e.g. azido, allyl and silyl groups). Quantitative yields of the deacetylated product were obtained for all evaluated derivatives using catalyst amounts between 1 and 10 mol%. As a special highlight, the dual behavior of the nanocatalyst in both deacetylation and click reactions is reported.

Abstract:Treatment of methyl 5-acetamido-2,4,7,8,9-penta-O-acetyl-3,5-dideoxy-β-l-glycero-d-galacto-2-nonulopyranosidonate (1) with cyclic secondary amines in pyridine at room temperature for 24h afforded unusual products (2a–g). Related experiments were carried out to explain the formation of 4-amination and 2-O-deacetylation of peracetylated sialic acid derivatives (2a–g). Abstract:A simultaneous stereoselective 2-O-deacetylation and 4-amination reaction of peracetylated Neu5Ac 1 has been established with cyclic secondary amines, such as 1-N-Boc-piperazine. Four C2-symmetric and two asymmetric sialic acid dimers with (4→4)-piperazine derivatives linked were synthesized. They may serve as precursors of unnatural polysialic acids.

Abstract:AbstractA cobalt‐catalyzed deuteration of amidoacrylates using deuterated methanol afforded α,β‐dideuterio‐α‐amino esters in excellent enantiomeric ratios (mostly >95 : 5) and almost complete deuteration (99 %). The new protocol was used to prepare dideuterio‐α‐amino acid fragments in some drugs. Furthermore, the stereoselective deuteration was applied in a concise synthesis of dideuterio l‐DOPA.

Abstract:This study aimed to optimize the per‐O‐acetylation, deacetylation, and benzylidene protection of carbohydrates and investigate the scope of saccharides via alum‐catalysis. Initially, optimization was based on the previously reported PTA‐catalyzed acetylation of glucose, but alum‐catalysis required heating conditions. The study expanded to investigate the per‐O‐acetylation of different saccharides, including galactose and mannose, which produced stereoisomeric mixtures. The study proposed a plausible reaction mechanism for the isomerization observed in galactose and xylose after acetylation. The mechanism involved chelation of the active alum‐catalyst with the O‐6 position and β‐type O‐1 position of galactopyranose to form a transition state complex via ring opening. This study investigated the generality of the de‐O‐acetylation method on a variety of carbohydrates using per‐O‐acetyl saccharide substrates bearing different functional groups. The direct protection of benzylidene in addition to acetylation/de‐O‐acetylation with a focus on optimizing reaction conditions for the synthesis of carbohydrate derivatives. Overall, the study offers insights into optimizing de‐acetylation, per‐O‐acetylation, and benzylidene protection investigating the scope of saccharides via alum‐catalysis.

Abstract:A new method for cleaving methyl ethers off protected saccharides using Co2(CO)8/Et3SiH/CO system was developed. The method showed regioselectivity in different protected monosaccharides with various anomeric groups. The primary and equatorial secondary methyl ethers were preferentially removed. This method was successfully applied to the synthesis of the DEF trisaccharide segment of Idraparinux, which is highly methylated.
