Abstract:A convenient method to perdeuterate aromatic compounds is reported. Commercially available nonaflic acid (C4F9SO3H) in combination with C6D6 as the deuterium source provided access to a series of highly labeled arenes. Some of them were then used as building blocks for the preparation of functionalized deuterated products. A wide range of synthetic procedures were investigated. They involved traditional methods such as halogenation or nucleophilic substitution, but also metal‐catalyzed cross coupling and C−X bond activation. The transformations comprised redox‐neutral, oxidative as well as reductive reactions, and mechanistically, ionic, radical and pericyclic processes were covered. Under many conditions, the overall degree of deuteration remained high; in C−X activation reactions, a more pronounced re‐protonation was observed.

Abstract:A novel synthesis of α,α-difluoro-γ-butyrolactones was realized by the alkaline hydrolysis of the adducts of ethyl iododifluoroacetate and alkenes. Ethyl iododifluoroacetate reacted with alkenes in the presence of sodium dithionite to give the addition products in 62–78% yields. The products were hydrolyzed in 10% aqueous sodium carbonate solution to give α,α-difluoro-γ-butyrolactones in 93–98% yields.

Abstract:We report a catalytic late‐stage bromination strategy for the α‐C‐H functionalization of aryl acetic acid. The aryl acetic acid was transformed into enediolate in the presence of an (AcO)4B2O with the combination of DBU, and consequently underwent an electrophilic bromination reaction with pyridinium tribromide. A wide range of α‐bromo aryl acetic acids were isolated in yields of 19‐65%. In addition, the reaction could be performed on a gram scale, and several transformations of the products were carried out.

Abstract:Photoinduced acylmethyl radical procedures for the efficent and simple preparation of highly functionalized 1,5-dicarbonyl compounds from Morita-Baylis-Hillman acetates with a-bromo ketones are developed. The acylmethylation is carried out with rose bengal as an organophotocatalyst at room temperature under metal- and oxidant-free conditions. Particularly, the simple photoinduced bromoacylmethylation was exploited via a fac-Ir(ppy)3-catalyzed atom transfer radical addition (ATRA) strategy under visible light irradiation. This method are attractive and valuable due to its mild reaction conditions, broad substrate scope and good functional group tolerance.

Abstract:Functionalized ketene dithioacetals are important intermediates in organic synthesis.α-Functionalization reactions based on them provide potential precursors for the construction of usefully carbo-and heterocyclic compounds.In this paper,a versatileα-bromination of functionalized ketene dithioacetals was developed.In the presence of H2 O,the reactions of functionalized ketene dithioacetals(1) with CuBr 2 in THF at room temperature gave variousα-bromo ketene dithioacetals(2) in excellent yields with high regioselectivities.The mechanism of thisα-bromination is proposed to undergo an addition of bromine formed in situ to compound 1 and sequontial elimination of theα-proton.The simplicity of execution,mild conditions,inexpensive,easily usable brominating agent and high yields,make this synthetic method attractive for practical applications.
A New Method for the Synthesis of Α-Acetyl Ketene Dithioacetals

Abstract:The α-acetyl ketene dithioacetals 2, which bear various alkylthio groups, are a kind of important intermediates in organic synthesis. In this paper, dithioacetals 2 were prepared in very high yields (90%—100%) via the deacetylation reaction of the corresponding α,α-diacetyl ketene dithioacetals 1 in the presence of concentrated sulfuric acid. This reaction involves an %in-situ% electrophilic addition-deacetylation mechanism and shows the nucleophilicity of the α-carbon atom in α-oxo ketenedithioacetals. Meanwhile, when the reaction time was prolonged to 22—25 h, the β-keto thiolesters 3a and 3c were produced in good yields.

Abstract:A novel DABCO-catalyzed Ireland-Claisen [3,3]-rearrangement of allylic acrylates to give alpha-methylene-gamma,delta-unsaturated carboxylic acids in the presence of an excess of TMSCl and DBU in refluxing acetonitrile was developed. The protocol provides an easy entry to alpha-methylene-gamma,delta-unsaturated carboxylic acids from allylic alcohols in good yields.

Abstract:Mono and dichloro‐substituted organic molecules are shown to possess significant roles in various fields, especially in medicinal chemistry. One approach to synthesizing these compounds involves the selective dechlorination of easily prepared trichloromethyl compounds. Nevertheless, developing a practical approach that allows selective mono‐ and dihydrodechlorination is a challenging task. Herein, we introduced a method for selective mono‐ and dihydrodechlorination of trichloroacetamides and acetates which was promoted by two different Lewis base‐boryl radicals. Accordingly, 4‐dimethylaminopyridine (DMAP)‐borane enabled mono‐substitution of chlorine atom by hydrogen while dihydro‐substitution of chlorine atoms was promoted by N‐heterocyclic carbene (NHC)‐boryl radical. Using deuterated Lewis base‐borane as deuterium atom source, mono‐ and dideuterodechlorination were also successfully took place. This protocol features broad substrate scope and operates under mild reaction conditions.

Abstract:An efficient and chemoselective deprotection protocol for aryl silyl ethers using LiOAc as a bifunctional Lewis acid-Lewis base catalyst was described. Acetates, epoxides, and aliphatic silyl ethers were preserved, whereas aryl TBS and TBDPS ethers can be differentiated.

Abstract:Selective syn and anti diacetoxylations of alkenes have been achieved using a PhI(OAc)(2)/BF3 center dot OEt2 system in the presence and absence of water, respectively. A broad range of substrates including electron-deficient alkenes (such as alpha,beta-unsaturated esters) could be elaborated efficiently at room temperature with this methodology, furnishing the desired products in good to excellent yields and diastereoselectivity. In particular, a multigram-scale diastereoselective diacetoxylation of methyl cinnamate (5.00 g) was also accomplished in a few hours, maintaining the same efficiency as small-scale reaction. This novel methodology provides an alternative approach for the preparation of various 1,2-diols.

Abstract:Herein, the use of triphenylmethylium tetrakis(pentafluorophenyl)borate as an efficient catalyst for the intermolecular dehydrative coupling of p‐hydroxymandelic acid hydrate with substituted aromatic amines and (thio)phenols was developed, which allows for the preparation of aryl acetic acid derivatives in a convenient manner. Notably, this method offers several advantages, including being metal‐free, prefunctionalization free, high atom efficiency, highc regioselective, and utilizing easily accessible starting materials. Additionally, it exhibits a broad substrate scope, enabling the synthesis of a range of aryl acetic acid derivatives. Furthermore, this method can also be applied to the etherification of arylthiol derivatives, facilitating the preparation of various thiolated products. The readily available starting materials and broad substrate scope enhance the applicability and potential utility of this method in organic synthesis.

Abstract:A highly selective photocatalytic deuterodefluorination of fluorinated esters with D2O is presented. By tailoring the catalytic system, a variety of α,α-difluoroaryl acetates underwent selective defluorination/deuteration to afford the 2-fluoro-2-arylacetate-d and 2-arylacetate-d2 in good yields with excellent deuteration rate. Satisfactorily, this protocol was successfully applied to deuterate some drug molecules, such as the felbinacethyl-d2 and felbilnac-d2.

Abstract:(E)-4-bromo-3-methyl-2-butenoic acid was conveniently synthesized from 3-methyl-2-butenoic acid in 29% overall yield.In this procedure,the title compound was easily obtained by hydrolysis of (E)-trimethylsilyl 4-bromo-3-methyl-2-butenoate,which could be isolated from its mixture of Z-/E-isomer by distillation in vacuo.The isomeric mixture was prepared via the esterification of 3-methyl-2-butenoic acid with chlorotrimethylsilane,followed by bromination with NBS.

Abstract:AbstractA cobalt‐catalyzed deuteration of amidoacrylates using deuterated methanol afforded α,β‐dideuterio‐α‐amino esters in excellent enantiomeric ratios (mostly >95 : 5) and almost complete deuteration (99 %). The new protocol was used to prepare dideuterio‐α‐amino acid fragments in some drugs. Furthermore, the stereoselective deuteration was applied in a concise synthesis of dideuterio l‐DOPA.

Abstract:A facile one-pot synthesis of functionalized (E)-β -arylvinyl bromides bearing various 4-alkoxycarbonyl and 4-aryloxycarbonyl groups was achieved by debrominative decarboxylation of anti-2,3- dibromo-3-(4-carboxyphenyl)propanoic acid in the presence of AgOAc and subsequent esterification in the presence of dicyclohexyl carbodiimide and dimethylaminopyridine. This method gives (E)-β - arylvinyl bromides in high stereoselectivities and high yields, and tolerates aldehyde, ester, and nitro functional groups.

Abstract:The primary amino group has been seldom utilized as a transformable functionality in organic synthesis. Reported herein is a deaminative halogenation of primary amines using <i>N</i>-anomeric amide as the nitrogen-deletion reagent. Both aliphatic and aromatic amines are competent substrates for direct halogenations. The mildness and robustness of the protocol are evidenced by the successful reactions of several complex- and functional group-enriched bioactive compounds or drugs. Elaboration of the resulting products provides interesting analogues of drug molecules.
