Determination of Trace Amino Acid Neurotransmitters in the Plasma of Rats with Capillary Electrophoresis Coupled Diode Laser-induced Fluorescence Detector

Abstract:Trace amino acid neurotransmitters, glycine and glutamic, were deriviatized with cyanine dyes-5 (Cy5) and separated and determined by capillary electrophoresis with diode laser-induced fluorescence detector. Compared with other amino-labeled fluorescence reagent, such as fluorescein isothiocyanate (FITC) and tetramethyl rhodamine isothiocyanate (TRITC) , Cy5 showed the higher reaction selectivity and shorter reaction time for primary amion-group. The effect of some experimental conditions, Cy5 concentration, buffer concentration, pH and reaction time, on the derivatization and separation was investigated in detail. The fluorescence intensity of the derivative has a linear relationship with amino acid concentration between 7.5 x 10 ~(-8) ~ 3.5 x 10~(-6) mol/L and the correlation coefficient was 0.999 for both. The minimum concentrations of detection limit of glycine and glutamic acid were 3.3 X 10~(-9) mol/L and 1.0 x 10~(-9) mol/L, respectively. Finally, the concentration of glycine and glutamic acid in the plasma of rats was determined. The values of glycine and glutamic acid were (62 ± 14) x 10~(-6) mol/L and (19 ±2) x 10~(-6) mol/L, respectively.
