Abstract:Two new sesquiterpenes were isolated from the aerial parts of Inula japonica. Their structures were elucidated as 1beta-hydroxy-8beta-acetoxycostic acid methyl ester and 1beta-hydroxy-8beta-acetoxyisocostic acid methyl ester by spectral methods.

Abstract:Three new sesquiterpenes, 1beta-hydroxy-8beta-acetoxycostic acid methyl ester, 1beta-hydroxy-8beta-acetoxyisocostic acid methyl ester and 1beta-hydroxy-4alpha,11alpha-eudesma-5-en-12,8beta-olide, along with fourteen known compounds were isolated from the aerial parts of Inula japonica. The structures of these new compounds were elucidated by spectroscopic methods (IR, EIMS, HRMS, 1D and 2D NMR). 1,6alpha-dihydroxy-4alphaH-1,10-secoeudesma-5(10),11(13)-dien-12,8beta-olide exhibited appreciable cytotoxic activity against cultured SMMC-7721 (human hepatoma cell) and HO-8910 (human ovarian carcinoma cell) with IC 50 values of 52.22 and 21.32 microg/mL, while 5alphaH-eudesma-4(15),11(13)-dien-12,8beta-olide showed remarkable cytotoxic activity against cultured SMMC-7721 and HO-8910 with IC 50 values of 6.21 and 5.28 microg/mL, respectively.

Abstract:Fourteen sesquiterpenoids with an eudesmane C-atom skeleton, including four new ones, (1β,5α,7β,8β,11β)-5-hydroperoxy-1-hydroxyeudesm-4(15)-eno-12,8-lactone (1), (1β,5α,7β,8β)-8-(acetyloxy)-5-hydroperoxy-1-hydroxycostic acid methyl ester (12), and a mixture of (1β,3β,4β,7β,8β)-1,3-dihydroxyeudesma-5,11(13)-dieno-12,8-lactone (7) and (1β,3β,4β,7β,8β,11β)-1,3-dihydroxyeudesm-5-eno-12,8-lactone (8), were isolated from the aerial parts of Inula japonica (Asteraceae). Their structures were determined by extensive spectroscopic methods, and those of 7 and 12 confirmed by means of single-crystal X-ray diffraction analysis.

Abstract:Five new dimeric sesquiterpene lactones, neojaponicone A (1) and japonicones M-P (2-5) were isolated from the aerial part of Inula japonica. Neojaponicone A (1) has an unprecedented carbon skeleton with an unusual valerolactone system formed between a guaianolide and an eudesmane. The structure and absolute configuration of 1 were established by extensive analysis of spectroscopic data and computational approaches.

Abstract:Sesquiterpene dimers are mainly found in the Asteraceae family. However, conflicting reports on the structures of these compounds can be found in the literature. Herein, we describe ten sesquiterpene dimers isolated from the flowers of Inula japonica, including configurational revisions of japonicone H (1-1), japonicone D (2-1), inulanolide A (4-1), japonicone X (5-1), and inulanolide F (5-2) to compounds 1, 2, 4, and 5, respectively. Five new related metabolites (3 and 6-9) are also described. Application of GIAO NMR/DP4+ analyses and ECD/OR calculations enabled us to revise the absolute configurations of an additional 13 sesquiterpene dimers isolated from plants of the genus Inula. Compounds 1, 2, 4, and 6 exhibited inhibition of nitric oxide production in lipopolysaccharide activated RAW264.7 macrophages with IC50 values of 4.07-10.00 μM.

Abstract:Two new insecticidal sesquiterpene esters with a -dihydroagarofuran sesquiterpene skeleton, ejaponine A (1) and ejaponine B (2), and two known compounds, 3 and 4, were isolated from the EtOAc extract of the root bark of Euonymus japonicus Thunb. by bioassay-guided fractionation. Their chemical structures were elucidated mainly by analysis of MS and NMR spectral data. The LD50 values of ejaponine A (1), ejaponine B (2), 3 and 4 showed against Mythimna separata were 89.2, 98.6, 181.4 and 109.2 mu g g-1, respectively.

Abstract:Eight new (1-8) and two known (9 and 10) sesquiterpenic acids featuring α-methylene-γ-carboxyl units were isolated from the whole plants of Inula wissmanniana, along with two new germacranolides (11 and 12). Their structures were elucidated based on detailed spectroscopic analysis, including HRESIMS, 1D and 2D NMR, and X-ray crystallography. Notably, the skeleton of 1 was firstly discovered from nature, while that of 2 was discovered for the second time. All the compounds were evaluated for their inhibition against LPS-induced nitric oxide (NO) production in RAW264.7 macrophages. Compound 11 exhibited the strongest activity with the IC50 value of 1.04 μM.

Abstract:Two new sesquiterpene lactone dimers, neojaponicone B (1) and inulanolide E (2) along with five known sesquiterpene lactone dimers (3–7, resp.) were isolated from the aerial parts of Inula japonica Thunb. The chemical structures of 1 and 2 were elucidated by detailed spectroscopic analysis. The relative configuration of 2 was confirmed by biomimetic transformation from the known sesquiterpene lactone dimer inulanolide A (3). The cytotoxicities of the isolated sesquiterpene lactone dimers were evaluated against 6T-CEM and Jurkat cell lines. All compounds showed potent cytotoxicities with IC50 value of 2.2–5.9μm.

Abstract:OBJECTIVE:To investigate the sesquiterpene lactones of the aerial parts of Inula helianthus-aquatica.METHOD:Compounds were isolated and purified by silica gel, Sephadex LH-20 and preparative HPLC. On the basis of physicochemical properties and spectroscopic data, their structures were identified.RESULT:Seven sesquiterpene lactones and four other compounds were obtained and identified as 2-desoxy-4-epi-pulchellin (1), 6-acetoxy-4-hydroxy-1, 10H-pseudoguaia-11 (13)-en-12,8-olide (2), 4-acetoxy-6-hydroxy-1, 10H-pseudoguaia-11(13)-en-12,8-olide (3), 8-epi-inuviscolide (4), 2,3,11,13-tetrahydroaromaticin (5), 11,13-dihydro-ergolide (6), 4-epipulchellin-2-O-acetate (7), 7-epiloliolide (8), loliolide (9), beta-sitosterol (10) and daucosterol (11).CONCLUSION:All the compounds were isolated from this plant for the first time.
