Abstract:Two new labdane diterpenes isolated from the rhizomes of Hedychium forrestii were determined by spectroscopic evidence to be labda-8(17), 11, 13-trien-7 beta-hydroxyl-15(16)-olide (1, hedyforrestin B) and labda-8(17), 11, 13-trien-7beta, 16-dihydroxyl-16(15)-olide (2, hedyforrestin C).

Abstract:An abietane diterpenoid, triptobenzene Y and six sesquiterpene polyol esters, wilforsinines C–H, together with 14 known compounds, have been isolated from the roots of Tripterygium wilfordii. The structures of the compounds were elucidated on the basis of spectroscopic analyses. The quinone reductase (QR) induction assay indicated that two compounds showed moderate QR-inducing activities at concentrations of 25μM and 50μM, respectively.

Abstract:Two new 18-norschiartane-type schinortriterpenoids, namely wuwezidilactones Q (1) and R (2) were isolated from the stems of Schisandra lancifolia. Their structures were determined on the basis of extensive spectroscopic analysis. The absolute configurations of the new compounds were further determined by ROESY and an empirical comparison of their experimental ECD spectra with literature.

Abstract:Forrestiacids C (1) and D (2), a pair of C-25 epimeric triterpene–diterpene adducts were isolated from the needles and twigs of the vulnerable conifer Pseudotsuga forrestii. This unprecedented class of compounds might be generated via an intermolecular Michael addition reaction of a rearranged 6/6/5/5-fused spiro-lanostene with an abietene. Their structures were established by spectroscopic data and X-ray crystallography. The adducts showed inhibitory activities against the ATP-citrate lyase (ACL) and acetyl-CoA carboxylase 1 (ACC1), two rate-limiting enzymes in the de novo lipogenesis pathway.

Abstract:A systematic phytochemical investigation on Abies forrestii afforded two new and 20 known compounds. Abieseconordines A and B (1 and 2) are the first two examples of norditerpenes with a novel 18-nor-5,10?:?9,10-disecoabietane skeleton. Their structures were established mainly by analysis of 1D- and 2D-NMR spectroscopic data. In addition, electronic circular-dichroism calculations and molecular-orbital analysis were utilized to confirm the absolute configuration of 1. Both compounds exhibited a potent effect in a bioassay inhibiting LPS-stimulated NO production in RAW264.7 macrophages.

Abstract:Two new ent-labdane diterpenoids, hypoestesins A-B (1–2) and five new labdane diterpenoids, hypopurolides H-L (3–7), were isolated from the aerial parts of Hypoestes purpurea. All of the structures were fully determined based on extensive analysis of 1H, 13C, 2D NMR, and HRESIMS data. The absolute configurations of 1–3 was established through comparing the experimental and calculated ECD curves and the structure of 5 was confirmed by single crystal X-ray diffraction experiment. Compounds 5–7 were unusual C23 labdane diterpenoids having a γ-acetonyl-α, β-unsaturated γ-lactone unit and each assigned as C-15 epimeric mixture. Furthermore, cytotoxic and anti-inflammatory activities of 3–7 were evaluated. The results showed that 3 had remarkable cytotoxic activity against HL-60, A549, SMMC-7721, MDA-MB-231, and SW480 cancer cell lines with IC50 values ranging from 2.35 to 17.06 μM. Compound 4 showed moderate cytotoxic activity against HL-60 and SMMC-7721 cancer cell lines with IC50 values of 15.12 ± 0.53 and 12.92 ± 0.60 μM, respectively. Furthermore, compound 4 was also found to exhibit inhibitory activity against NO production in RAW 264.7 macrophages with IC50 values of 23.56 ± 0.99 μM, compared to the positive control L-NMMA with an IC50 value of 41.11 ± 1.34 μM.

Abstract:Four new lanostane triterpenoids, namely (3 ss)-3-hydroxy-24-methylenelanost-8-ene-7,11-dione (1), (3 ss)-3-hydroxylanosta-8,24-diene-7,11-dione (2), (3 ss,7 alpha)-3,7-dihydroxylanosta-8,24-dien-11-one (3), and (3 ss,11 ss)-3,11-dihydroxylanosta-8,24-dien-7-one (4) were isolated from Euphorbia humifusa, together with 2 known compounds. The structures of these new compounds were elucidated on the basis of extensive spectroscopic analysis and comparison with the related known compounds.

Abstract:Two new lignans, named as 6-hydroxyhinokinin and 6-hydroxyhinokinin-6- O-β-D-glucopyranoside, together with five known compounds: elemol, oplodiol, catechin, β-sitosterol and daucosterol were isolated from Kadsura heteroclita. The structures of these compounds were characterized by extensive 1D and 2D NMR spectral experiments, chemical analysis and comparison with literature data. The absolute structures of the novel lignans were elucidated as 8 S, 8′ S, based on their optical rotations.

TJ403-LS1, which was originally isolated from roots of the medicinally valuable plantAnoectochilus roxburghii. The structures of compounds1and2were established based on widespread spectroscopic methods, mainly including 1D & 2D NMR and HRESIMS analyses, and the absolute configuration of1was further confirmed by electronic circular dichroism (ECD) calculation. All new compounds were tested for the in vitro cytotoxicity against five human cancer cell lines.

Abstract:12,15-Epoxy-5alpha H,9beta H-labda-8(17),13-dien-19-oic acid (1) and 14-methoxy-15,16-dinor-5alpha H,9alpha H-labda-13(E),8(17)-dien-12-one (2), two new labdane-type diterpenes, and 1alpha-hydroxy-8,12-epoxyeudesma-4,7,11-triene-6,9-dione (3), one new eudesmane-type sesquiterpene, were isolated from Chloranthus henryi Hemsl, along with two known compounds, 8beta H-eudesma-4(14),7(11)-dien-12,8-olide (4) and 8beta-hydroxyeudesma-4(14),7(11)-dien-12,8-olide (5). Their structures were established by a combination of 1D- and 2D-NMR spectroscopic techniques. The new compounds 2, 3 and the known compound 4 exhibited antitumor activities against Hela and K562 human tumor cell lines.

Abstract:Two new bislabdane-type diterpenoids lanceolatin (1), lancelatol (2) and three new diterpenoids 15-nor-14-oxo-8(17),12-labdanien-14,18-dionic acid (3), 8(17),12,14-labdantrien-18-oic acid (4) and 8(17),12,14-labdantrien-18-ol (5), together with 7 known diterpenoids were isolated from the roots of Cunninghamia lanceolata. Their structures were determined by spectroscopic and chemical methods. All of these compounds were isolated from the species for the first time. One of the isolated compounds showed a significant inhibition effect on mouse hind-paw edema induced by carrageenan.

Their chemical structures have been suggested as ent- 11α-acetoxy-13β,19-dihydroxykaur-16-en-15-one (1)and ent-11α,19-diacetoxy -7β,13β-dihydroxykaur -16-en-15-one (2) respectively on the basis of biogenesis, detailed spectroscopic analysis (COSY) and chemical conversions.

Abstract:Two new triterpenoids, namely 24-methylene-5,24-dien-19(10→9)- abeo -8 α ,9 β ,10 α -eupha-3 β -ol ( 1 ) and 24-methyl-5,23-dien-19(10→9)- abeo -8 α ,9 β ,10 α -eupha-3 β -ol ( 2 ) were isolated from the stems of Euphorbia royleana , together with three known analogs. The structures of the new compounds were elucidated by extensive 1 H NMR, 13 C NMR, HSQC, HMBC, 1 H- 1 H COSY, ROESY and HR-MS spectroscopic analyses.

Abstract:AbstractForrestiacids A (1) and B (2) are a novel class of [4+2] type pentaterpenoids derived from a rearranged lanostane moiety (dienophile) and an abietane unit (diene). These unprecedented molecules were isolated using guidance by molecular ion networking (MoIN) from Pseudotsuga forrestii, an endangered member of the Asian Douglas Fir Family. The intermolecular hetero‐Diels–Alder adducts feature an unusual bicyclo[2.2.2]octene ring system. Their structures were elucidated by spectroscopic analysis, GIAO NMR calculations and DP4+ probability analyses, electronic circular dichroism calculations, and X‐ray diffraction analysis. This unique addition to the pentaterpene family represents the largest and the most complex molecule successfully assigned using computational approaches to predict accurately chemical shift values. Compounds 1 and 2 exhibited potent inhibitory activities (IC50s <5 μM) of ATP‐citrate lyase (ACL), a new drug target for the treatment of glycolipid metabolic disorders including hyperlipidemia. Validating this activity 1 effectively attenuated the de novo lipogenesis in HepG2 cells. These findings provide a new chemical class for developing potential therapeutic agents for ACL‐related diseases with strong links to traditional medicines.
