Abstract:Two new diterpenoids, forskolin 6 and H were isolated from the chloroform extract of the roots of Coleus forskohlii, and based oil spectroscopic data, their structures were identified as 1alpha-hydroxy-6beta,7beta-diacetoxy-8,13-epoxylabd-14-ene-11-one (1), and 1alpha,6beta-diacetoxy-8,13-epoxy labd-14-ene-11-one (2), respectively.

Abstract:OBJECTIVE:To study the chemical constituents in the aerial parts of Coleus forskohlii.METHODS:The compounds were isolated by various column chromatographic methods, and their structures were identified by spectroscopic methods.RESULTS:Twelve compounds were isolated and identified as chamaecydin (1), 6 alpha-hydroxydemethylcryptojaponol (2), alpha-cedrene (3), oleanolic acid (4), forskolin G (5), forskolin J (6), 1,6-diacetyl-9-deoxyforskolin (7), forskolin A (8), forskolin H (9), 6-acetyl-1-deoxyforskolin (10), betulinic acid (11), beta-sitosterol (12).CONCLUSION:Compounds 1 - 3 are isolated from Coleus genus for the first time, and compound 4 is isolated from C. forskohlii for the first time.

Abstract:Two new stilbenoids, stemobenoids A (1) and B (2), together with three known compounds were obtained from the roots of Stemona tuberosa. The structures of the new compounds were established by extensive spectroscopic analysis, including HRMS, 1D and 2D NMR data. Compounds 1 and 2 displayed potent quinone reductase inducing activity in Hepa 1c1c7 cells.

Abstract:Two new C(20)-oxygenated ent-kaurene diterpenoids, taibaihenryiins A (1) and B (2), along with five known compounds, shikokianin (3), longikaurin D, 2 alpha -hydroxyursolic acid, longikaurin F, and lasiodin, were isolated from the EtOH extract of the leaves and tender branches of Isodon henryi (HEMSL.) KUDO. The structures of the new compounds were determined as 11 alpha -acetoxy-7,20-epoxy-1 alpha ,6 beta ,7 beta -trihydroxy-ent-kaur-16-en-15-one (1) and 7,20-epoxy-3 beta ,6 beta ,7 beta ,11 alpha -tetrahydroxy-ent-kaur-16-en-15-one (2) on the basis of detailed, spectroscopic analyses.

Abstract:Eleven new diterpenoids, adenanthins B-L (1-11), together with five known analogues, calcicolin B (12), adenanthin (13), weisiensin A (14), nervosanin (15), and forrestin C (16), were isolated from the aerial parts of Isodon adenantha. The structures of 1-11 were elucidated on the basis of spectral methods, as well as the X-ray crystallographic analysis of 11. Compounds 2, 5, and 14 showed significant inhibitory activities against K562 cells, with IC(50) values less than 4.0 microg/mL, respectively.

Abstract:A phytochemical investigation of the leaves of Callicarpa macrophylla led to the isolation of five new diterpenoids (1-5), macrophypenes A-E and nine known analogues (6-14). The structures of 1-5 were established on the basis of extensive analysis of NMR spectroscopic data, X-ay diffraction data, and experimental and calculated electronic circular dichroism spectra. Compound 1 is a spiroditerpenoid with a novel skeleton, and compound 5 is a rare ent-abietane diterpenoid possessing a peroxide bridge. Compounds 1, 5-7, and 11-14 stimulate nerve growth factor mediated neurite outgrowth from PC12 cells.

Abstract:Forskolin is a complex labdane plant diterpenoid, which has been used in the treatment of a variety of diseases based on its activity as an activator of adenosine monophosphate(cAMP) cyclase. Natural forskolin exists only in the cork layer of the root of Coleus forskohlii. Due to the complexity of the extraction and chemical synthesis processes, the yield and purity of forskolin cannot meet commercial requirements. In recent years, with the rapid development of synthetic biology and the analysis and interpretation of many diterpene biosynthetic pathways, a new approach has been provided for the green production of forskolin. In this paper, the structure, activity, biosynthetic pathway and the heterologous biosynthesis of forskolin were reviewed. The problems and solutions in the heterologous biosynthesis of forskolin were also discussed and summarized, which will provide references for the construction of high-yielding forskolin engineering strains.
A New Dinor-cis-Labdane Diterpene and Flavonoids with Antimycobacterium Activity from Colebrookea oppositifolia

Abstract:The new 14,15-dinor-cis-labdane diterpene, named (+)-14,15-dinor-9α-hydroxy-cis-labd-11(E)-en-13-one (1), was isolated from the acetone extract of the aerial parts of Colebrookea oppositifolia, along with the known compounds alnustin (2), mosloflavone (3), flindulatin (4), 5,6,7-trimethoxy baicalein (5), tanetin (6), scutellarein 4′-methyl ether (7), apigenin (8), caffeic acid (9), anisofolin A (10), apigetrin (11), and forsythoside A (12). Structures of the new and known compounds were established by detailed analysis of 1D and 2D nuclear magnetic resonance studies. The isolated compounds 1–12 were evaluated for their antimycobacterium activity against Mycobacterium tuberculosis H37Ra and Mycobacterium bovis in both dormant and active phases. Compounds 1, 7, and 8 exhibited inhibitory activity against M. tuberculosis with IC50 values in the range of 8.1–55.0 µM (MIC 14.4–119.7 µM) in the active phase and 7.4–43.5 µM (MIC 11.5–123.3 µM) in the dormant phase. Similarly 1, 7, and 8 exhibited inhibitory activity against M. bovis with IC50 values in the range of 4.1–98.5 µM (MIC 13.7–161.0 µM) in the active phase and 4.1–111.1 µM (MIC 13.0–166.4 µM) in the dormant phase.

Abstract:Two new triterpenoids, (3 beta)-olean-12-ene-3,23-diol (1) and (6 alpha)-6-hydroxyolean-12-en-3-one (2) were isolated from the MeOH extract of the stalks of Celastrus hypoleucus (OLIV.) WARB., together with the seven known compounds (3p,22a)-3,22-dihydroxyolean-12-en-29-oic acid, beta-amyrin and beta-amyrin palmitate, wilforlide A, wilforilide B, palmitic acid, and beta-sitosterol. Their structures were elucidated on the basis of spectroscopic data and, in the case of 2, by X-ray crystallography. Compound 1 showed moderate cytotoxicity against human cervical squamous carcinoma (Hela) cells.

Abstract:Two new ent-kaurane-derived diterpene derivatives, phyllostacins A (1) and B (2), were isolated from the aerial parts of Isodon phyllostachys, together with two known compounds, irroratin A (3) and serrin B (4). Both 1 and 2 were found to be present as diastereoisomers. In the case of 1, the corresponding diastereoisomeric diacetates 5 and 6 were prepared and separated. The structures of the new compounds were elucidated by extensive 1D- and 2D-NMR spectroscopic methods, in combination with MS experiments. In (D-5)pyridine solution, the two epimers of 1 are present in equal amounts, but in CDCl3 or CD3OD, the (S)-epimer predominates in the mixture of hemiacetals.

Abstract:Nine new diterpenoids, albopilosins B-J (1-9), together with six known analogues, albopilosin A (10), macrocalyxin C (11), rabdokunmin C (12), excisanin (13), amethystonoic acid (14), and coetsanoic acid (15), were isolated from the aerial parts of Isodon albopilosus. The structures of 1-9 were established using spectroscopic methods including extensive 1D and 2D NMR analysis. The diterpenoids isolated were evaluated for their inhibitory activities against HepG2 (hepatoma) cells. Compounds 7 and 13 were the most active, with both having IC(50) values of <15 microM.

Abstract:Two new bislabdane-type diterpenoids lanceolatin (1), lancelatol (2) and three new diterpenoids 15-nor-14-oxo-8(17),12-labdanien-14,18-dionic acid (3), 8(17),12,14-labdantrien-18-oic acid (4) and 8(17),12,14-labdantrien-18-ol (5), together with 7 known diterpenoids were isolated from the roots of Cunninghamia lanceolata. Their structures were determined by spectroscopic and chemical methods. All of these compounds were isolated from the species for the first time. One of the isolated compounds showed a significant inhibition effect on mouse hind-paw edema induced by carrageenan.

Abstract:Eight previously undescribed diterpenoids, crolaevinoids A-H, including two halimanes, four clerodanes, and two laevinanes, along with six known analogues were isolated from the twigs of Croton laevigatus. The structures of the previously undescribed were elucidated by spectroscopic analysis, and their absolute configurations were determined by combination of a single crystal X-ray diffraction and CD analysis (exciton chirality and Rh-2(OCOCF3)(4)-induced methods). Crolaevinoids A and B represent the first halimane diterpenoids with a unique lactone bridge between C-12 and C-17. All compounds were evaluated for their inhibitory effects on the nitric oxide (NO) production induced by lipopolysaccharide (LPS) in RAW264.7 macrophage cells. Furocrotinsulolide A and 3,4,15,16-diepoxy-cleroda-13(16),14-diene-12,17-olide exhibited pronounced inhibition of NO production with IC50 values of 10.4 +/- 0.8 and 6.0 +/- 1.0 mu M, respectively, being more potent than the positive control, quercetin (IC50 = 13.1 +/- 1.9 mu M). Abstract:This study is to investigate the chemical constituents from the whole plant Corydalis edulis. The chemical constituents were separated and purified by macroporous resin D101, silica gel, Sephadex LH-20, ODS, and semi-preparative HPLC. Their structures were determined by physicochemical properties and spectroscopic data. Four compounds were isolated from the dichloromethane and water extracts of the whole plant C. edulis, and identified as 6'-β-D-xylosylicariside B2(1),(3S,5R,6S,7E)-5,6-epoxy-3-hydroxy-7-megastigmen-9-one(2), loliolide(3), and 5,5'-dimethoxybiphenyl-2,2'-diol(4), respectively. Compound 1 is a new compound, of which the absolute configuration was established by electronic circular dichroism(ECD) calculations. Compound 4 is obtained from the plants of Papaveraceae family for the first time. Compounds 2 and 3 are firstly isolated from the Corydalis genus.

Abstract:Forskolin is mainly found in the root of a plant called Coleus forskohlii (Willd.) Briq., which has been used in the traditional medicine of Indian Ayurvedic and Southeast Asia since ancient times. Forskolin is responsible for the pharmacological activity of this species. Forskolin is a labdane diterpenoid with a wide biological effect. Several studies suggested a positive role of forskolin on heart complications, respiratory disorders, high blood pressure, obesity, and asthma. There are numerous clinical and pre-clinical studies representing the effect of forskolin on the above-mentioned disorders but more clinical studies need to be performed to support its efficacy.
