Abstract:beta-Phenylseleno-alpha-tolylsulfonyl-substituted alkenes were synthesized via the three-component conjugate -nucleophilic addition of acetylenic sulfones, phenylselenomagnesium bromide, and carbonyl compounds, such as aldehydes, aliphatic ketones, or alpha,beta-unsaturated enals or enones. The reaction is highly regio- and stereoselective with moderate to good yields. Functionalized allylic alcohols were obtained in the case of aldehydes and aliphatic ketones. In the case of alpha,beta-unsaturated enones, functionalized allylic alcohols or functionalized gamma,delta-unsaturated ketones were obtained, depending on the structures of the ketones.

Abstract:[GRAPHICS] A mixture of phenylselenomagnesium bromide, an acetylenic sulfone, and an aldehyde in THF/CH2Cl2 afforded Michael-aldol tandem adduct, i.e., (Z)-beta-phenylseleno-alpha-(p-tolylsulfonyl)allylic alcohol, in good yield with high stereoselectivity. The stereoselectivity greatly depended on solvent.

Abstract:Phenylseleno- and p-tolylsulfonyl substituted 1,3-dienes were conveniently prepared with high stereoselectivity by the elimination reaction of phenylseleno- and p-tolylsulfonyl substituted allyl alcohols in the presence of BF3·Et2O in acetic anhydride. The products were characterized by 1H NMR, MS, IR and elemental analysis. The single crystal structure of 2a was determined by X-ray diffraction analysis.

Abstract:An enantioselective sulfenylation/semipinacol rearrangement of 1,1-disubstituted and trisubstituted allylic alcohols was accomplished with a chiral Lewis base and a chiral Brønsted acid as cocatalysts, generating various β-arylthio ketones bearing an all-carbon quaternary center in moderate to excellent yields and excellent enantioselectivities. These chiral arylthio ketone products are common intermediates with many applications, for example, in the design of new chiral catalysts/ligands and the total synthesis of natural products. Computational studies (DFT calculations) were carried out to explain the enantioselectivity and the role of the chiral Brønsted acid. Additionally, the synthetic utility of this method was exemplified by an enantioselective total synthesis of (-)-herbertene and a one-pot synthesis of a chiral sulfoxide and sulfone.

Abstract:A homo-Michael addition reaction of lithium selenolates with 1-(1-alkynyl)cyclopropyl ketones and the subsequent reaction with electrophiles such as PhSeBr, NFSI and NCS is reported. Based on the nature of electrophiles, this reaction may afford highly substituted 1,2-allenyl ketones or furans (E(+) = PhSe(+)) and 2-alkynyl ketones (E(+) = F(+), Cl(+), active halides) as the final products, respectively.

Abstract:Sulfonyl hydrazones have been identified as an excellent sulfonyl anion surrogate in the base and Pd-0-catalyzed three-component tandem reaction with aryl iodides and allenes for the synthesis of functionalized allylic sulfones. By forming a stabilized six-membered palladacycle intermediate, the reaction led to the desired higher substituted allylic sulfones with excellent Z selectivities and high yields.

Abstract:A novel three-component stereoselective synthesis of (E)-4-alkene 1,3-diketones from lithium selenolates, 1-(1-alkynyl)cyclopropyl ketones, and aldehydes is reported. This reaction afforded the products in moderate to good yields with the formation of a new C-Se single bond, a new C-C double bond, and a new C-O double bond.

Abstract:The hydrochalcogenation of allenyl sulfones with sodium organyl chalcogenolates can proceed smoothly to give beta-phenylchalcogenyl allyl sulfones in high yields with exclusive regioselectivity. For the asymmetric allenyl sulfone, the reaction has both high regio- and stereoselectivity with the predominace of E-isomer. Some applications of beta-phenyltelluro and beta-phenylselanyl allyl sulfone in organic synthesis are also investigated.

Abstract:A one‐pot Pd‐catalyzed tandem process to prepare (2‐arylallyl) aryl sulfones has been developed. This strategy is based on the modular assembly of a boronic acid, a sodium sulfinate and 2‐bromoallyl acetate. The reaction is completely regioselective towards the terminal alkene, yielding (2‐arylallyl) aryl (or alkyl) sulfones with yields ranging from 56 to 93%. Control experiments together with DFT calculations allowed to propose a plausible reaction mechanism of the tandem reaction. The usefulness of this methodology has been demonstrated with the formal synthesis of the marketed drug Apremilast and of several natural products by asymmetric hydrogenation. Using the commercially available UbaPHOX iridium complex, chiral β‐methyl sulfones with up to 98% ee were obtained

Abstract:Multicomponent reactions that involve interception of onium ylides through Aldol, Mannich, and Michael addition with corresponding bench-stable acceptors have demonstrated broad applications in synthetic chemistry. However, because of the high reactivity and transient survival of these in situ generated intermediates, the substitution-type interception process, especially the asymmetric catalytic version, remains hitherto unknown. Herein, a three-component asymmetric allylation of α-diazo carbonyl compounds with alcohols and allyl carbonates is disclosed by employing a ternary cooperative catalysis of achiral Pd-complex, Rh2(OAc)4, and chiral phosphoric acid CPA. This method represents the first example of three-component asymmetric allylic alkylation through an SN1-type trapping process, which involves a convergent assembly of two active intermediates, Pd-allyl species, and enol derived from onium ylides, providing an expeditious access to chiral α,α-disubstituted ketones in good to high yields with high to excellent enantioselectivity. Combined experimental and computational studies have shed light on the mechanism of this novel three-component reaction, including the critical role of Xantphos ligand and the origin of enantioselectivity.

Abstract:Here, we demonstrated a transition metal-mediated/monophosphorus ligand system for the selective synthesis of ketones or chiral allylic alcohols in high yields/enantiomeric excess from the 1,2-addition of arylboronic acids to α,β-unsaturated aldehydes. Notably, isomerization of the chiral allylic alcohols to ketones was suppressed by the Ru-catalyzed/monophosphorus ligand system. The asymmetric catalytic system provides an alternative and efficient method of preparing chiral allylic alcohols.

Abstract:A general method is reported for the stereo selective preparation of highly functionalized allylic thioethers. This protocol is based on a Pd-catalyzed thiolation of modular vinyl cyclic carbonate substrates and features high (Z) selectivity, good yields, minimal waste, ample product scope, and operational simplicity. A one-pot strategy was used for the stereoselective formation of pharma-relevant allylic sulfones derived from their in situ prepared thioether precursors.

Abstract:The present Letter demonstrates a photoswitched stereodivergent synthesis of allylic sulfones from sodium sulfinates, triphenylvinylphosphonium chloride, and (hetero)aromatic aldehydes in a single step. Mechanistically, cis-allylic sulfones, generated from the unstabilized ylide intermediates and aldehydes in situ, could be finally converted to trans-allylic sulfones via photochemical isomerization in the presence of a catalytic amount of bis(2-thienyl) ketone.
