Abstract:Carbon-fluorine bond activation of the trifluoromethyl group represents an important approach to fluorine-containing molecules. While selective defluorinative functionalization reactions of CF3 -containing substrates have been achieved by invoking difluorocarbocation, difluorocarboradical, or difluoroorganometallic species as the key intermediates, the transformations via fluorocarbanion mechanism only achieved limited success. Furthermore, the enantioselective defluorinative transformation of the CF3 group remained a formidable challenge. Here we report a defluorinative functionalization reaction of 4-trifluoromethylpyridines involving difluoro(pyrid-4-yl)methyl anion as the key intermediate, which was developed based upon our previous studies on the N-boryl pyridyl anion chemistry. In particular, asymmetric defluoroallylation of 4-trifluoromethylpyridines and -pyrimidines could be achieved by using Ir-catalysis to forge a difluoroalkyl-substituted chiral center.
Highly efficient heterogeneous procedure for the synthesis of fructone fragrancy

Abstract:A novel heterogeneous strong acid catalyst was synthesized through the copolymerization of p -toluenesulfonic acid and paraformaldehyde and utilized for the synthesis of fructone. The results showed that the catalyst was very efficient for the reaction with the yield over 95%. The advantages of extremely high density of acidity, high thermal and chemical stability, low cost for the simple synthetic procedure, and reusability made the catalyst one of the best choices for the reaction.

Abstract:We developed an amide-based Cinchona alkaloids catalyzed enantioselective fluorination of 4-substituted pyrazolones, the products with a quaternary chiral fluorine-attached carbon centre were afforded in high yields (up to 99%) with excellent enantioselectivities (up to 98% ee). Our method is highlighted by low catalyst loading (0.05-0.5 mol %), short reaction times (30 min) and broad substrate scopes (35 examples). Furthermore, we carried out this asymmetric fluorination under sustainable conditions in 10 cylces, the optically pure product (>99.5% ee) could be easily obtained by simple extraction and recrystallization on a multigram scale. Controlled experiments and DFT studies showed that the multiple hydrogen bonding interactions and the steric hindrance at ortho-position in benzene ring of catalyst were the main reasons for high enantioselectivity and reactivity. With a simple operational protocol and low catalyst loading, this transformation provides a practical method for preparation of chiral fluorine-containing compounds.

Abstract:A rapid synthesis of β-arylethenesulfonyl fluorides is realized using ligand-accelerated Pd(II)-catalyzed nondirected C–H alkenylation of simple arenes. The newly developed electron-deficient 2-pyridone ligand is crucial for this transformation with arenes as limiting reagent. This protocol features a broad substrate scope and good functional group tolerance. Late-stage functionalization of various pharmaceuticals and their further click manipulations demonstrate this procedure to be highly valuable.

Abstract:Fluoranthenes have attracted tremendous attention due to their unique optoelectronic properties and ex-tensive applications.Although several synthetic methodologies have been developed for the preparation of fluoranthene derivatives,it is still unfavorable to functionalize the fluoranthene framework at different positions due to the relatively low selectivity and reactivity.Herein,a catalyst-free intramolecular[4+2]annulation between thiophenes and alkynes is developed towards the synthesis of fluoranthenes.Alto-gether 20 examples have been demonstrated using this method.Various functional groups can be pre-cisely introduced into the fluoranthene skeleton at different positions by simply tuning the substituents on the thiophenes and alkynes.The conjugation of the fluoranthene can be facilely extended through different directions.Furthermore,the feasibility of this[4+2]annulation reaction is also investigated by density functional theory calculations.Therefore,this protocol provides not only a synthetic methodol-ogy towards fluoranthenes with substituents functionalized at different positions,but also an effective pathway to construct large polycyclic aromatic hydrocarbons containing fluoranthene moieties.

Abstract:Fluorination using chiral catalytic methods could result in a direct access to asymmetric fluorine chemistry. However, challenges in catalytic asymmetric fluorinations, especially the longstanding stereochemical challenges existed in BF3·Et2O-based fluorinations, have not yet been addressed. Here we report the catalytic asymmetric nucleophilic fluorination using BF3·Et2O as the fluorine reagent in the presence of chiral iodine catalyst. Various chiral fluorinated oxazine products were obtained with good to excellent enantioselectivities (up to >99% ee) and diastereoselectivities (up to >20:1 dr). Control experiments (the desired fluoro-oxazines could not be obtained when Py·HF or Et3N·3HF were employed as the fluorine source) indicated that BF3·Et2O acted not only as a fluorine reagent but also as the activating reagent for activation of iodosylbenzene.

Abstract:Polyfluoroaryl ethers represent an important framework of biologically active molecules and materials. Owing to the strong bond dissociation energy of C‐F bond, selectivity and other issues, transition metal‐catalyzed synthesis of polyfluoroaryl ethers from perfluoroarenes via the activation of C‐F bond is challenging and underdeveloped, as compared to the well‐documented C‐O bond formation starting from aryl iodides, aryl bromides or aryl chlorides. Herein, an unprecedented Pd‐catalyzed defluorinative etherification for the synthesis of polyfluoroaryl ether skeletons using hydrobenzoxazoles as phenol surrogate, has been reported. The substrate scope for this protocol is broad, with respect to hydrobenzoxazoles and perfluoroarenes, under mild reaction conditions. More importantly, challenging alkenyl and alkynyl substituted polyfluoroarenes could be successfully used as the coupling component for Pd‐catalyzed etherification reaction. Density functional theory (DFT) calculations were employed to investigate the reaction mechanism, which suggested that oxidative addition between polyfluorobenzene and Pd(0) constituted the ratedetermining step.

Abstract:A wide range of organic molecules can participate in hydrogen atom transfer (HAT) processes as both HAT-donors and photo-HAT-acceptors simultaneously. Therefore, it opens the possibilities to exclude catalysts for C-H functionalization in such cases. This is the underlying idea of the presented catalyst-free C-H bond fluorination approach. We demonstrated that broad range of aryl alkyl ketones can be efficiently fluorinated with Selectfluor-based reagents at benzylic position under UV-A irradiation without any added catalyst. The selectivity of mono- and difluorination can be controlled by con-trolling the excess of fluorinating reagent. Additionally, we propose an analogue of Selectfluor reagent - F-TEDA-TFSI with much greater solubility in acetonitrile which makes our protocol solvent-economical. By the same manner benzaldehydes can be transformed to corresponding benzoyl fluorides with almost quantitative yields. The protocol was successfully applied in late-stage fluorination of complex molecules Tonalide and Tolperisone. Kinetic measurements demonstrated zero-order kinetics which indicates that light flux is the limiting factor. A tentative mechanism was proposed based on the selectivity of fluorination of primary, secondary, and tertiary benzylic C-H bonds.

Abstract:The defluoroborylation of fluoroarenes by chromium-catalyzed cleavage of unactivated C–F bonds is described. The reaction uses HBpin as the boron source, low-cost and commercially available chromium salt as the precatalyst, and terpyridine as crucial ligand, providing a protocol with atom-efficient benefits and a wide range of applicable substrates for the functionalization of aryl C–F bonds. Preliminary mechanistic studies indicate an unprecedant Cr-catalyzed magnesation of unactivated C–F bond occurred. The generated arylmagnesium intermediates then participated in the subsequent borylation reaction. The application of the stratey in the preparation of valuable derivatives is demonstrated by the late-stage functionalization of boronate ester groups.

Abstract:Aryl fluorosulfates of varying complexities have been used in amination reactions in water using a new Pd oxidative addition complex (OAC) developed specifically to match the needs of the fine chemicals industry, not only in terms of functional group tolerance but also reflecting time considerations associated with these important C–N couplings. Also especially noteworthy is that they replace both PFAS-related triflates and nonaflates, which are today out of favor due to recent government regulations. The new complex based on the BippyPhos ligand is used at low loadings and under aqueous micellar conditions. Moreover, it is easily prepared and stable to long-term storage. DFT calculations on the OAC precatalyst compare well with the X-ray structure of the crystals with -complexation to the aromatic system of the ligand and also confirm the NMR data showing a mixture of conformers in solution that differ from the X-ray structure in rotation of the phenyl and t-butyl ligand substituents. An extensive variety of coupling partners, including pharmaceutically relevant APIs, readily participate under mild and environmentally responsible reaction conditions

Abstract:A cooperative Pd/Cu-catalyzed dual C-H bond activation of electron-deficient polyfluoroarenes with aryl halides is developed. This protocol provides a unique and strategically distinct access to a variety of polyfluoroarene-substituted benzofuran derivatives. Abstract:A decarboxylative/dehydrofluorinative formal [3 + 2] cycloaddition aromatization of isoquinolinium N-ylides with difluoroenoxysilanes has been developed. This methodology provides a facile and straightforward synthetic pathway to afford highly functionalized fluorinated pyrrolo[2,1-a]isoquinolines in good to excellent yields under mild conditions. Moreover, gram-scale and synthetic derivatization experiments for the late-stage functionalization of drug molecules have also been demonstrated.

Abstract:<p>A method for the synthesis of polysubstituted 4-difluoromethyl and perfluoroalkyl pyrazole derivatives by the Lewis acid- and base-<em>co</em>-mediated reaction of perfluoroacetyl diazoester with ketones is reported. A series of structurally diverse 4-difluoromethylated and perfluoroalkylated pyrazoles were obtained in moderate to good yields. The reactions proceeded smoothly under mild conditions with wide functional group tolerance.</p>

Abstract:A [3 + 2]/[2 + 1] cycloaddition reaction of gem‑difluorocyclopropenes is presented, offering a mild and efficient approach to accessing tri- and tetra-substituted 4-fluoropyridines in moderate to good yields with excellent regioselectivity. Multiple synthetic applications, including process-scale reactions, modification of bioactive molecules, derivatization reactions and synthesis of the analogue of the PKM2 modulator, are subsequently described.

The diazo functionality (RR'C[double bond, length as m-dash]N) is successfully converted to RR'CF, RR'CHF, RR'CFBr or RR'CFNR'' groups by employing different fluorination reagents.
