Abstract:The transition-metal-catalyzed α-arylation of carbonyl compounds is a widely practiced method for C-C bond formation. Several enantioselective versions of this process have been reported, but intermolecular, enantioselective coupling reactions of aryl electrophiles with α-fluoro carbonyl compounds have yet to be disclosed. We report enantioselective coupling of aryl and heteroaryl bromides and triflates with α-fluoroindanones catalyzed by palladium complexes of a BINOL-derived monophosphine and Segphos, respectively. The enolates were generated directly from α-fluoroindanones in the presence of potassium phosphate base during the reactions. We also report that reactions of α-fluorotetralones occur in high yields and enantioselectivities when conducted with enolates generated by elimination of trifluoroacetate from trifluoromethyl β-diketone hydrates. These reactions were catalyzed by palladium complexes of the commercially available bisphosphine Difluorphos. Thus, the formation of enantioenriched α-aryl-α-fluoroketones can be readily achieved by C-C bond formation when the appropriate palladium catalyst and α-fluoro enolate precursor were used.

Abstract:Pentaphenylferrocenyl di-tert-butylphosphine has been prepared in high yield from a two-step synthetic procedure, and the scope of various cross-coupling processes catalyzed by complexes bearing this ligand has been investigated. This ligand creates a remarkably general palladium catalyst for aryl halide amination and for Suzuki coupling. Turnovers of roughly 1000 were observed for aminations with unactivated aryl bromides or chlorides. In addition, complexes of this ligand catalyzed the formation of selected aryl ethers under mild conditions. The reactions encompassed electron-rich and electron-poor aryl bromides and chlorides. In the presence of catalysts containing this ligand, these aryl halides coupled with acyclic or cyclic secondary alkyl- and arylamines, with primary alkyl- and arylamines, and with aryl- and primary alkylboronic acids. These last couplings provide the first general procedure for reaction of terminal alkylboronic acids with aryl halides without toxic or expensive bases. The ligand not only generates highly active palladium catalysts, but it is air stable in solution and in the solid state. Palladium(0) complexes of this ligand are also air stable as a solid and react only slowly with oxygen in solution.

Abstract:Fluorine-containing compounds have shown unparalleled impacts in the realm of functional molecules, and the ability to prepare novel structures has been crucial in unlocking new properties for pharmaceutical and materials science. Herein, we report a copper-catalyzed, photoinduced defluorinative C‒O coupling between trifluoromethylarenes and alcohols. This method allows for direct access to a wide selection of difluorobenzylether (ArCF2OR) molecules, including a com-pound displaying liquid crystal behavior. Through slight modification of the protocol, we were able to generate ArCF2I products, another class of synthetically useful fluorine-bearing molecules. Comprehensive mechanistic investigations first suggested ArCF2I as a reservoir to steadily supply the key ArCF2• radical species. Furthermore, experimental evi-dence supported a mechanism consisting of two collaborative cycles: C‒F activation operated by a homoleptic Cu(I) co-ordinated by two bisphosphine ligands as the photocatalyst and C‒O coupling promoted by a Cu(I) ligated by a single bisphosphine ligand. The critical roles of the two salt additives, lithium iodide and zinc acetate, in orchestrating the two cycles were also elucidated. This dual-role copper catalyst demonstrates the power of base metal photoredox catalysis in achieving both substrate activation and chemical bond formation via a single catalytic system.

Abstract:Direct CAr-F arylation is effective and sustainable for synthesis of polyfluorobiaryls with different degrees of fluorination, which are important motifs in medical and material chemistry. However, with no aid of transition metals, the engagement of CAr-F bond activation has proved difficult. Herein, an unprecedented transition-metal-free strategy is reported for site-selective CAr-F arylation of polyfluoroarenes with simple (het)arenes. By merging N,N-bis(2,6-diisopropylphenyl)perylene-3,4,9,10-bis(dicarboximide)-catalyzed electrophotocatalytic reduction and anodic nitroxyl radical oxidation in an electrophotocatalytic cell, various polyfluoroaromatics (2F-6F and 8F), especially inactive partially fluorinated aromatics, undergo sacrificial-reagents-free C-F bond arylation with high regioselectivity, and the yields are comparable to those for reported transition-metal catalysis. This atom- and step-economic protocol features a paired electrocatalysis with organic mediators in both cathodic and anodic processes. The broad substrate scope and good functional-group compatibility highlight the merits of this operationally simple strategy. Moreover, the easy gram-scale synthesis and late-stage functionalization collectively advocate for the practical value, which would promote the vigorous development of fluorine chemistry.

Abstract:An imino-N-heterocyclic carbene palladium(II) complex with a bulky substituted group on the imino nitrogen was used to catalyze the direct arylation of electron-deficient fluoroarenes with aryl halides. A series of electron-poor substrates and aryl bromides could be coupled in good to excellent yields with satisfactory position selectivity (20 examples, up to 93%). These arylations could proceed at a relatively low temperature (80 °C, 20 examples, up to 95%) with mono-N-protected amino acid assistance. Some of them even gained higher yields than those at high temperature (110 °C). Otherwise, some aryl iodides can forge cross-coupling products in yields of nearly 30% under the optimized conditions. The rate profiles for arylation of electron-poor arenes were measured in the presence of the imino-N-heterocyclic carbene palladium(II) complex or Pd(OAc)2 as the catalyst, which showed that the former could keep catalytic activity for a longer time. Abstract:A catalyst-free electrochemical cross-coupling reaction of aryl halides with trialkyl phosphite has been developed. This reaction proceeds in an undivided cell with a low-cost Ni anode and a graphite cathode under mild and neutral conditions. A wide range of functional groups are well-tolerated and the phosphorylated product can be obtained on the gram scale, showing that this transformation has the potential to be a valuable method for the construction of aromatic carbon-phosphorus bonds.

Abstract:Cross‐coupling reactions for constructing C‐N bonds represent a pivotal advancement in chemical science. Traditional methodologies, including nucleophilic aromatic substitution (SNAr) and transition metal‐catalyzed cross‐couplings, have limitations concerning aryl scope, reliance on toxic and costly transition‐metal catalysts, and issues related to atom economy and waste generation from ligands and additives. In this work, we introduce a novel method for aminating neutral, electron‐rich, and electron‐deficient aryl halides, eliminating the need for transition metals. Our approach involves the activation of aryl halides using solvated electrons generated from granulated lithium and sonication. This serves as a sustainable source of reducing power, facilitating the efficient formation of C‐N bonds under near ambient conditions. Competitive selectivity studies between halide and ester functionalities were explored. Reaction scope and conducted mechanistic studies support the proposed radical‐nucleophilic substitution (SRN1) mechanism for the reaction. Notably, the developed reaction has a highly competitive reductive dehalogenation pathway during the C‐N coupling reaction, and this mechanistic divergency was thoroughly explored. This work not only broadens the scope of C‐N coupling reactions which typically employs aryl bromides and iodides and rarely aryl fluorides which is also equally abundant, but also introduces a new way to do C‐N coupling reactions using solvated electrons.

Abstract:The defluoroborylation of fluoroarenes by chromium-catalyzed cleavage of unactivated C–F bonds is described. The reaction uses HBpin as the boron source, low-cost and commercially available chromium salt as the precatalyst, and terpyridine as crucial ligand, providing a protocol with atom-efficient benefits and a wide range of applicable substrates for the functionalization of aryl C–F bonds. Preliminary mechanistic studies indicate an unprecedant Cr-catalyzed magnesation of unactivated C–F bond occurred. The generated arylmagnesium intermediates then participated in the subsequent borylation reaction. The application of the stratey in the preparation of valuable derivatives is demonstrated by the late-stage functionalization of boronate ester groups.

Abstract:Four types of perfluoroarene-substituted and the corresponding non-fluorinated Pt(II) complexes, [PtCl2L] (L = 1 and 2), were prepared with 4,4′-bis(pentafluorophenyl)-2,2′-bipyridine (1a), 4,4′-diphenyl-2,2′-bipyridine (1b), 4,4′-bis(2-pentafluorophenylethynyl)-2,2′-bipyridine (2a), and 4,4′-bis(2-phenylethynyl)-2,2′-bipyridine (2b), respectively, to understand the role of perfluoroaromatic substitution and acetylene linkers on molecular structures and their induced supramolecular associations. The pentafluorophenyl groups lead to significant changes in electron distribution within the Pt(II) complexes, notably causing absorption bands to red-shift due to a metal-to-ligand charge transfer from nucleophilic platinum ions and demonstrating stabilization effects on the bands by fluorination in experimental and theoretical studies. The results of altering electron density and reducing the metal's nucleophilic tendencies through fluorination and the use of an acetylene linker are discussed, accompanied by crystal structures, the corresponding Hirshfeld surface analysis, and DFT calculations.

Abstract:Carbon-fluorine bond activation of the trifluoromethyl group represents an important approach to fluorine-containing molecules. While selective defluorinative functionalization reactions of CF3 -containing substrates have been achieved by invoking difluorocarbocation, difluorocarboradical, or difluoroorganometallic species as the key intermediates, the transformations via fluorocarbanion mechanism only achieved limited success. Furthermore, the enantioselective defluorinative transformation of the CF3 group remained a formidable challenge. Here we report a defluorinative functionalization reaction of 4-trifluoromethylpyridines involving difluoro(pyrid-4-yl)methyl anion as the key intermediate, which was developed based upon our previous studies on the N-boryl pyridyl anion chemistry. In particular, asymmetric defluoroallylation of 4-trifluoromethylpyridines and -pyrimidines could be achieved by using Ir-catalysis to forge a difluoroalkyl-substituted chiral center.

Abstract:Pd-catalyzed decarboxylative cross-couplings of 2-(2-azaaryl)acetates with aryl halides and triflates have been discovered. This reaction is potentially useful for the synthesis of some functionalized pyridines, quinolines, pyrazines, benzoxazoles, and benzothiazoles. Theoretical analysis shows that the nitrogen atom at the 2-position of the heteroaromatics directly coordinates to Pd(II) in the decarboxylation transition state.
