Abstract:A series of AZT 5'-O-hydrogen phospholipids 5a-e were synthesized by a tandem transesterification of diphenyl phosphite (DPP) with AZT and a long-chain alcohol. This method has the merits of easy operation and high yields. The corresponding phosphonates, phosphorothionates and phosphoroselenoates of 5d and 5e and the phosphoramidates conjugated with L-amino acid methyl ester 9a and 9b were also synthesized.

Abstract:Four 3'-azido-3'-deoxythymidine (AZT)-alpha-hydroxyphosphonate derivatives were prepared by the addition of diverse aryl aldehyde to the corresponding AZT-5'-yl H-phosphonate diester in the presence of base catalyst. The structures of all products were confirmed by P-31 NMR, H-1 NMR, ESI-MS and elemental analyses.

Abstract:5'-hydrogenphosphonothioate derivatives of AZT, d4T, and ddI as anti-HIV prodrug candidates were synthesized in 61-76% yields under mild conditions through sequential one-pot reactions, i.e., couple of triethylammonium phosphinate with different alcohols in the presence of pivaloyl chloride (PV-Cl), following oxidation with elemental sulfur and further condensation with AZT, d4T, or ddI in the presence of PV-Cl.

Abstract:Modified nucleosides such as 3'-azido-2',3'-dideoxythymide (AZT) and 2',3'-dideoxy-2',3'-didehydrothymide (d4T) are of importance as antiviral agents and in other medicinal applications. Consequently, there is interest in new analogs of these compounds and in new methods for preparing them. In this article, a simple but efficient, one-pot synthesis of 5'-diaryl esters and diamidates of phosphate, phosphorothioate, and phosphoroselenoate derivatives of AZT and d4T is described. The reaction of AZT/d4T with phosphorus trichloride, followed with phenols, aniline, or amino acid methyl esters in the presence of a base, led to the corresponding tricoordinated phosphite compounds of AZT/d4T, which were then oxidized with 3-chloroperoxybenzoic acid (m-CPBA), sulfur, or selenium powder to finally afford the corresponding target compounds in good yield.

Abstract:3'-Azido-3-deoxythymidine (AZT) was the first clinically approved drug against HIV infectiong despite its undesirable side reactions, such as bone marrow suppression. In our aim to develop new chemical entity of anti-tumor and anti-HIV, the H- phosphonate 6 of AZT conjugate with diosgenin was synthesized by a tandem transesterification reaction for the first time. There are the merits of easy operas tion and high yield in the reported method. It could be extended to synthesize other diosgenin phosphonate conjugates such as carbohydrate and peptide.

Abstract:An expeditious method to access 2-aminobenzoxazoles and 2-aminobenzothiazoles from various substituted 2-aminophenols and 2-aminothiophenols, respectively, with isoselenocyanates in a one pot procedure is reported. Elemental Se precipitates nearly quantitatively in the reaction without any deselenizing agent and it can be reused. A possible mechanism for the formation of the target products is proposed.

Abstract:The α-diazophosphonate was added to aryl (N-tosyl)imine to give β-aryl β-(N-tosyl)amino α-diazophosphonates, which were further subjected to TsOH-catalyzed diazo decomposition to yield α-aryl β-(N-tosyl)enaminophosphonates through 1,2 aryl migration. The α-aryl β-(N-tosyl)enamino phosphonates were hydrogenated to give α-aryl β-(N-tosyl)amino phosphonates.

Abstract:A practical and efficient method for the synthesis of 1,2,3-triazole-amino-bisphosphonates derivatives was developed through two steps: The Michael addition reaction of propargyl amine with vinylidene bisphosphonate, and the 1,3-dipolar click cycloaddition assisted by ultrasound irradiation. This approach has significant advantages in terms of experimental simplicity, mild reaction condition, and can be easily scaled up. The target compounds were characterized by H-1-, P-31-, C-13-NMR spectroscopy and high-resolution mass spectral analyses.

Abstract:Phosphorus(V) chloride was sequentially displaced by catechol N,O-bis(trimethylsilyl)amino acids and phenol (Method A) or catechol, phenol and N,O-bis(trimethylsilyl)amino acids (Method B) leading to a high yield synthesis of 2-phenoxy-2,7(5)-spiro[1,3,2-benzodioxaphosphole-2,2'-1,3,2-oxazophospholan]-5'-ones under mild conditions. The products were characterized by H-1, C-13, H-1-H-1 COSY, H-1-C-13 COSY and held desorption mass spectrometry.

Abstract:epsilon-Benzosultam derivatives are potential drug candidates with diverse biological activities. A series of chiral epsilon-benzosultams bearing phosphorus functionalities was synthesized by catalytic asymmetric hydrophosphonylation in the presence of a bifunctional phosphonium salt catalyst. The desired hydrophosphonylation products were obtained in good yields with high enantioselectivities, and scale-up reactions and further derivations were successfully accomplished. Some control experiments were also conducted to elucidate the plausible reaction mechanism of this chemical transformation.

Abstract:A study on the synthesis of the novel cyclic alpha-aminophosphonates and 2-alkoxy-2oxo-1,4,2-oxazaphosphinanes 4a-r has been carried out. The title compounds were obtained in good yields by one-pot procedure using o-aminophenol, alkyl dichlorophosphinite, and ketones or benzaldehyde. One of their geometric stereoisomers was isolated and characterized. Configurations of 4k and one isomer of 4r have been established by X-ray diffraction analysis. The synthetic methods provide an easy access to the organophosphorus heterocycles with the ring system mentioned above. The abnormal chemical shifts of alkyl-substitute protons in H-1 NMR spectra were given reasonable explanation. (c) 2007 Wiley Periodicals, Inc.
