Abstract:In the present study, we have synthesized a series of novel glucosyl triazoles for the first time. The glucosyl triazoles 4a-e were synthesized by reaction of some azidoglycosides with various terminal alkynes via a copper-catalyzed [3+2] cycloaddition (oclick chemistryo) and were deprotected to afford the corresponding glucosyl triazoles 5a-e in good yields. The structures of the new compounds were determined by IR, NMR spectroscopy, and mass spectrometry. The antitumor (human cervical cancer cell) activity was evaluated for the target compounds.

Abstract:Four 3'-azido-3'-deoxythymidine (AZT)-alpha-hydroxyphosphonate derivatives were prepared by the addition of diverse aryl aldehyde to the corresponding AZT-5'-yl H-phosphonate diester in the presence of base catalyst. The structures of all products were confirmed by P-31 NMR, H-1 NMR, ESI-MS and elemental analyses.

Abstract:A series of bile acid-polyamine amides conjugated with 3 '-azido-3 '-deoxythymidine (AZT) as potential antitumor prodrugs in the form of phosphoramidates were synthesized in good yields and their antitumor activities were assayed against two human cancer cells in vitro: cervix cancer HeLa cells and renal cancer 7860 cells. The improved antitumor activity probably derived from the enhanced delivery efficiency of AZT due to bile acid-polyamine conjugates. (C) 2007 Elsevier Ltd. All rights reserved.

Abstract:5'-hydrogenphosphonothioate derivatives of AZT, d4T, and ddI as anti-HIV prodrug candidates were synthesized in 61-76% yields under mild conditions through sequential one-pot reactions, i.e., couple of triethylammonium phosphinate with different alcohols in the presence of pivaloyl chloride (PV-Cl), following oxidation with elemental sulfur and further condensation with AZT, d4T, or ddI in the presence of PV-Cl.

Abstract:Six long-chain alkyl H-phosphonate derivatives of 3'-azido-2',3'-dideoxythymidine (AZT) and 2',3'-didehydro-2',3'-dideoxythymidine (d4T) were synthesized via one-pot reaction of AZT and d4T with phosphorus trichloride and different alcoholysis reagents in sequence. The products were characterized by P-31 NMR, H-1 NMR, C-13 NMR and ESI-MS.

Abstract:Modified nucleosides such as 3'-azido-2',3'-dideoxythymide (AZT) and 2',3'-dideoxy-2',3'-didehydrothymide (d4T) are of importance as antiviral agents and in other medicinal applications. Consequently, there is interest in new analogs of these compounds and in new methods for preparing them. In this article, a simple but efficient, one-pot synthesis of 5'-diaryl esters and diamidates of phosphate, phosphorothioate, and phosphoroselenoate derivatives of AZT and d4T is described. The reaction of AZT/d4T with phosphorus trichloride, followed with phenols, aniline, or amino acid methyl esters in the presence of a base, led to the corresponding tricoordinated phosphite compounds of AZT/d4T, which were then oxidized with 3-chloroperoxybenzoic acid (m-CPBA), sulfur, or selenium powder to finally afford the corresponding target compounds in good yield.

Abstract:3'-Azido-3-deoxythymidine (AZT) was the first clinically approved drug against HIV infectiong despite its undesirable side reactions, such as bone marrow suppression. In our aim to develop new chemical entity of anti-tumor and anti-HIV, the H- phosphonate 6 of AZT conjugate with diosgenin was synthesized by a tandem transesterification reaction for the first time. There are the merits of easy operas tion and high yield in the reported method. It could be extended to synthesize other diosgenin phosphonate conjugates such as carbohydrate and peptide.

Abstract:Nucleoside reverse transcriptase inhibitors are the only drugs so far approved for the treatment of AIDS. Nucleoside 5′-phosphorothioates is relatively resistant to enzymatic transformations. Novel 2′,3′- didehydro-2′, 3′-dideoxythymidine (d4T) 5′- thiophosphoramidates have been prepared by thiophosphorochloridate chemistry. These materials were designed to act as membrane-soluble prodrugs of the bioactive free nucleotides. In vitro evaluation reveals that the compounds have a pronounced, selective anti-HIV activity in MT-4 cells. The new compounds are characterized by NMR, IR and ESI-MS. The results from NMR experiments indicate that there are anti-orientation for glycosyl grouping in compounds 5a-5e.

Abstract:A series of AZT 5'-O-hydrogen phospholipids 5a-e were synthesized by a tandem transesterification of diphenyl phosphite (DPP) with AZT and a long-chain alcohol. This method has the merits of easy operation and high yields. The corresponding phosphonates, phosphorothionates and phosphoroselenoates of 5d and 5e and the phosphoramidates conjugated with L-amino acid methyl ester 9a and 9b were also synthesized.

Abstract:L‐2'‐deoxythreose nucleoside phosphonates PMDTA and PMDTT possess potent anti‐HIV activity. Herein, a novel class of 3'‐C‐branched‐l ‐threose nucleoside phosphonate analogs, 5'‐deoxy‐3'‐C‐hydroxymethyl‐3'‐O‐phosphonomethyl‐d ‐xylose nucleosides, were synthesized and biologically evaluated. The key sugar intermediate 3‐C‐benzyloxymethyl‐3‐O‐diethylphosphonomethyl‐1,2‐O‐isopropylidene‐α‐d ‐5‐deoxyxylose (8 ) was firstly synthesized, which may be an interesting scaffold for access to diverse 3'‐C‐branched l ‐threosyl nucleoside phosphonate derivatives. And the key synthesis involved Wittig olefination of 1,2‐O‐isopropylidene‐3‐oxo‐α‐d ‐5‐deoxyxylose, stereoselective dihydroxylation of alkenes by aqueous KMnO4, selective benzylation of hydroxymethyl group under activation of dibutyltin oxide, and introduction of phosphonate group by nucleophilic substitution. Eventually, glycosylation under Vorbrüggen conditions provided 3'‐C‐hydroxymethyl‐3'‐O‐phosphonomethyl‐β‐d ‐5'‐deoxyxylose nucleoside analogs in satisfying yield.
