Abstract:Arbuzov reaction of aryl phosphorodichloridite with mixture of one equivalent of AZT or d4T and one equivalent of tertbutyl alcohol led to the corresponding AZT/d4T aryl H-phosphonate diesters. and the following reaction of the H-phosphonate diesters with amino acid methyl esters in the presence of N-chloro-succinimide (NCS) produced membrane-soluble anti-HIV prodrugs AZT/d4T aryl phosphoramidate derivatives in good yields.

Abstract:5'-hydrogenphosphonothioate derivatives of AZT, d4T, and ddI as anti-HIV prodrug candidates were synthesized in 61-76% yields under mild conditions through sequential one-pot reactions, i.e., couple of triethylammonium phosphinate with different alcohols in the presence of pivaloyl chloride (PV-Cl), following oxidation with elemental sulfur and further condensation with AZT, d4T, or ddI in the presence of PV-Cl.

Abstract:3'-Azido-3-deoxythymidine (AZT) was the first clinically approved drug against HIV infectiong despite its undesirable side reactions, such as bone marrow suppression. In our aim to develop new chemical entity of anti-tumor and anti-HIV, the H- phosphonate 6 of AZT conjugate with diosgenin was synthesized by a tandem transesterification reaction for the first time. There are the merits of easy operas tion and high yield in the reported method. It could be extended to synthesize other diosgenin phosphonate conjugates such as carbohydrate and peptide.

Abstract:In the present study, we have synthesized a series of novel glucosyl triazoles for the first time. The glucosyl triazoles 4a-e were synthesized by reaction of some azidoglycosides with various terminal alkynes via a copper-catalyzed [3+2] cycloaddition (oclick chemistryo) and were deprotected to afford the corresponding glucosyl triazoles 5a-e in good yields. The structures of the new compounds were determined by IR, NMR spectroscopy, and mass spectrometry. The antitumor (human cervical cancer cell) activity was evaluated for the target compounds.

Abstract:Nucleoside reverse transcriptase inhibitors are the only drugs so far approved for the treatment of AIDS. Nucleoside 5′-phosphorothioates is relatively resistant to enzymatic transformations. Novel 2′,3′- didehydro-2′, 3′-dideoxythymidine (d4T) 5′- thiophosphoramidates have been prepared by thiophosphorochloridate chemistry. These materials were designed to act as membrane-soluble prodrugs of the bioactive free nucleotides. In vitro evaluation reveals that the compounds have a pronounced, selective anti-HIV activity in MT-4 cells. The new compounds are characterized by NMR, IR and ESI-MS. The results from NMR experiments indicate that there are anti-orientation for glycosyl grouping in compounds 5a-5e.

Abstract:A series of AZT 5'-O-hydrogen phospholipids 5a-e were synthesized by a tandem transesterification of diphenyl phosphite (DPP) with AZT and a long-chain alcohol. This method has the merits of easy operation and high yields. The corresponding phosphonates, phosphorothionates and phosphoroselenoates of 5d and 5e and the phosphoramidates conjugated with L-amino acid methyl ester 9a and 9b were also synthesized.

In this paper, 2',3'-O-alkoxymethylidene adenosine 5'-thiophosphoramidates were synthesized through a highly efficient approach. The new compounds were characterized by NMR, IR and ESI-MS.

Abstract:Several 1-(1-aryl-1,4-dihydro-3-carboxy-6-methylpyridazine4-one)-4-aryl thio-semicarbazides and their corresponding oxadiazole, thiadiazole and triazole derivatives were prepared and characterized by their spectral data. Abstract:Triazolo-fused 3′,4′-cyclic nucleoside 4′-spiro nucleoside analogues were synthesized by an intramolecular 1,3-dipolar cycloaddition of 4′-azido nucleoside derived azido-alkynes in a regio- and stereo-specific manner. The thymine nucleoside base in these target compounds was transformed into the corresponding 5-methyl cytosine component. The synthesized compounds were examined in an MAGI(multinuclear-activation galactosidase indicator) assay for exploring the anti-HIV activity and in a H9 T(human T lymphocytes H9) assay for measuring the cell toxicity.
