Significance of translation initiation factor C2 expression in hepatocellular carcinoma tissues

Abstract:AIM: To study the expression and its significance of C2 gene in hepatocellular carcinoma tissues. METHODS: In situ hybridization method was used to detect the expression of C2 mRNA in 21 samples of hepatocarcinoma tissues and pericancerous tissues, and ABC immunohistochemical method was used to detect the expression of C2 protein in 60 samples of hepatocarcinoma and 42 samples of pericancerous tissues, and Western blot method was used to detect the expression of C2 protein in hepatocarcinoma tissues and pericancerous tissues. RESULTS: In 21 samples of hepatocarcinoma and pericancerous tissue, the positive rates of C2 mRNA were 23.8% and 85.7%, respectively. In 60 samples of hepatocarcinomas and 42 samples of pericancerous tissues, the positive rates of C2 protein were 27.3% and 83.3%. In 27 samples of liver cirrhosis, the positive rates of C2 protein were 77.8%. Statistical analysis suggested that the positive rate of C2 mRNA and its protein were all significantly higher in pericancerous tissue than that in hepatocarcinoma (P<0.001). C2 protein was significantly correlated with patients age, HBsAg and AFP (P<0.05), but C2 protein was not correlated with pathological grading, tumor size, and lymph node metastases. Result of Western blot method was consistent with that of immunohistochemical method. CONCLUSION: Low expression of C2 gene might play an important role in the carcinogenesis, development and diagnosis of hepatocellular carcinoma in the early stage.
