Abstract:The preparation of a series of 2-pyridinamines and their alkyl derivatives is described.The target compounds are synthesized starting from corresponding 2-cyanopyridines, via incomplete hydrolysis in the presence of hydrogen peroxide in the dilute alkaline mixture of acetone-2% sodium hydroxide solution, and Hoffmann degradation reaction with freshly made NaBrO.This route is of industrial value because of cheap and readily available materials, moderate reaction conditions and convenient operations.

Abstract:AbstractIncorporating enzymatic reactions into natural product synthesis can significantly improve synthetic efficiency and selectivity. In contrast to the increasing applications of biocatalytic functional‐group interconversions, the use of enzymatic C−C bond formation reactions in natural product synthesis is underexplored. Herein, we report a concise and efficient approach for the synthesis of [7.7]paracyclophane natural products, a family of polyketides with diverse biological activities. By using enzymatic Friedel–Crafts alkylation, cylindrocyclophanes A and F and merocyclophanes A and D were synthesized in six to eight steps in the longest linear sequence. This study demonstrates the power of combining enzymatic reactions with contemporary synthetic methodologies and provides opportunities for the structure–activity relationship studies of [7.7]paracyclophane natural products.

Abstract:A novel heterogeneous strong acid catalyst was synthesized through the copolymerization of p -toluenesulfonic acid and paraformaldehyde and utilized for the synthesis of fructone. The results showed that the catalyst was very efficient for the reaction with the yield over 95%. The advantages of extremely high density of acidity, high thermal and chemical stability, low cost for the simple synthetic procedure, and reusability made the catalyst one of the best choices for the reaction.

Abstract:A substituted polyacetylene bearing a furan ring, poly(2-propynyl 2-furoate) (PPF), was synthesized for the first time with palladium acetylide complex catalysts in CHCl3 at 60degreesC to give a soluble and stable polymer with a yield of ca. 75%. Some features for polymerization of 2-propynyl 2-furoate are described and discussed. The resulting polymer was characterized by IR, GPC and H-1-NMR analysis.

Abstract:Cyclohexanone diethyl acetal is widely used as pharmaceutical intermediates, and it may be pyrolyzed to give 1-ethoxy cyclohexene which could be used in synthesizing both polymer material and pharmaceutical intermediates. Cyclohexanone diethyl acetal was prepared from cyclohexanone with anhydrous ethanol as reactant, ammonium chloride as catalyst, triethyl orthoformate as water scavenger. The proposed method, which is preferable for green process, has many advantages such as the catalyst used is cheap and easily obtained, the post treatment of the process is convenient, the process has few industrial waste and high yield, etc. The suitable technical conditions were investigated in detail, which include the effects of catalyst, reaction time and reactants mole ratio on yield. The results indicate that 91.8% yield could be obtained with the following optimal reaction conditions: cyclohexanone weight 29.4 g (0.3 mol), the mole ratio of cyclohexanone and triethyl orthoformate 1:1.2, the mole ratio of cyclohexanone and anhydrous ethanol 1: 3, the reaction time 90 minutes, the amount of catalyst 0.006 mole, and reaction with reflux. The purity and structure of the product was examined by GC GC-MS and NMR.

Abstract:Chiral [2.2]paracyclophane (PCP) is an intriguing prevalent scaffold widely utilized in pi-stacked polymers, luminescent materials, and asymmetric catalysis. However, current synthetic methods to access chiral PCP derivatives are still limited to physical separation and chemical or kinetic resolution. The direct desymmetric functionalization of nonsubstituted PCP has been a challenge since its discovery 70 years ago. Herein, we present a catalytic desymmetric dearomatization cyclopropanation reaction of PCP, providing rapid access to enantioenriched PCP cyclopropyl carboxylates with continuous tertiary/quaternary/quaternary chiral centers. The application of the chiral PCPs obtained has also been demonstrated by transformation to structurally novel C 2 symmetric dirhodium catalysts. Of equal importance, detailed density functional theory calculations have been carried out to reveal the mechanism of this reaction. The key to this success is a structurally symmetric bulky C 4 dirhodium catalyst: noncovalent interactions exist between substrates and catalysts, which form a cavity to host substrates, and mainly affect the regio- and enantioselectivity.

Abstract:A synthetic method for 2-formylcyclopent-2-enecarboxylic acid is described. This procedure comprises two steps. The first step is a [2 + 2] cyclo-addition reaction, and the second step is a hydrolysis and ring-opening reaction. A plausible mechanism of the hydrolysis and ring-opening reaction was supposed. Then, the effects of different solvents, base mol ratio, concentration of base, and reaction temperature on the yield of the second step were studied. Finally, under the obtained optimized conditions, the product was achieved and isolated in 59.4 % yield, which was more than double the ones reported previously. An improved and efficient procedure for the synthesis of 2-formylcyclopent-2-enecarboxylic acid has been developed.

Abstract:The condensation of ethyl cyanoacetate and salicylaldehyde would afford either coumarin-3-carboxylate ester or 3-cyancoumarin as the final product. Herein, comparative experiments and density functional theory (DFT) calculations were investigated to pursue a deeper understanding of the critical factor in this reaction. Experimental results indicated that condensation is accomplished through a cascade process, including the Knoevenagel procedure followed by selective cyclization of the phenolic hydroxyl group to the cyano or ester carbonyl group within the intermediate. Product distribution correlates well with the acid–base properties of the catalyst and the ability of the catalyst for forming complex by hydrogen bonds. Also, an efficient and facile approach was developed for the preparation of coumarin-3-carboxylate ester as a major product from the reaction of ethyl cyanoacetate with salicylaldehyde. Low-transition-temperature mixture (LTTM) formed from L-proline and oxalic acid was proved as an inexpensive, easily available, and efficient promoter which not only affords the products in high yields but also avoids the use of hazardous solvent and tedious isolation procedures.

Abstract:A novel 1,1,10,10,19,19-hexamethyl-5,14,23-trimethoxy[3.3.3]metacyclophane (2) was prepared in 25% yield by Friedel-Crafts cyclization of 4-(2-methoxyphenyl)-2-methylbutan-2-ol (1) at -78 degrees C using TiCl(4) as Lewis acid catalyst in anhydrous dichloromethane. The structure of cyclophane 2 was determined by single-crystal X-ray diffraction, and the impact of substrate concentration on the yield of macrocycle 2 was also examined. The study on the effect of substituents at the phenyl ring showed that the methoxy group in 1 is crucial for its trimerization to give the hexamethyl[3.3.3]metacyclophane derivative. Demethylation of 2 with BBr(3) gave 1,1,10,10,19,19-hexamethyl-5,14,23-trihydroxy[3.3.3]metacyclophane (4) in 96% yield, and its three hydroxyl groups provide the possible modification sites for further construction of supramolecular assemblies.

Abstract:Planar chiral [2.2]paracyclophanes consist of two functionalized benzene rings connected by two ethylene bridges. These organic compounds have a wide range of applications in asymmetric synthesis, as both ligands and catalysts, and in materials science, as polymers, energy materials and dyes. However, these molecules can only be accessed by enantiomer separation via a) time-consuming chiral separations and b) kinetic resolution approaches, often with a limited substrate scope, yielding both enantiomers. Here, we report a simple, efficient, metal-free protocol for organocatalytic desymmetrization of centrosymmetric diformyl[2.2]paracyclophanes. Our detailed experimental mechanistic study highlighted differences in the origin of enantiocontrol of pseudo-para and pseudo-ipso diformyl derivatives in NHC catalysed desymmetrizations based on whether a key Breslow intermediate is irreversibly or reversibly formed in this process. This gram-scale reaction enables a wide range of follow-up derivatizations of carbonyl groups, producing various enantiomerically pure planar chiral [2.2]paracyclophane derivatives, thereby underscoring the potential of this method.

Abstract:Over the past decades, the research on optically active polymers (OAPs) has significantly grown, and extensive studies have been carried out on their syntheses, conformations, and applications. The most commonly used OAPs are based on natural products such as sugars or amino acids, which limits their scope. A broader range of applications can be achieved by synthesizing lab-tailored monomers, which allow precise control over structure and properties. This research developed a four-step synthetic route to a previously unreported chiral [2.2]paracyclophane-based epoxide monomer. An aluminum catalyst and an alkylammonium initiating system were applied and optimized for its polymerization to provide access to a novel class of chiral polyethers. Furthermore, we demonstrated the copolymerization viability of the (4-[2.2]paracyclophanyl)oxirane monomer using phthalic anhydride.

Abstract:Polyfurans (PFus) are a type of conjugated polymer but have usually been considered to be inherently unstable and poorly processable. In this work, the multicomponent cyclopolymerization (MCC) of diisocyanide, dialkylacetylene dicarboxylates, and dialdehyde was developed to prepare modified PFus under mild conditions with catalyst-free one-pot reactions. The approach afforded high molecular weight poly(amine–furan–arylene)s (PAFAs) (M w up to 76400), which contained 3,4-substituted furan that in situ formed during the polymerization, with good thermal stability and film-processing properties. All the experimental conditions such as molar ratio of monomers, polymerization solvents and temperature, monomer structures, and reactivity were investigated in detail. PAFAs’ structures were well characterized by GPC, FTIR, and NMR. One of the PAFAs carrying dimethylbenzene moieties exhibited the characteristics of a black material. Thus, this MCC provides a new method to modify PFus with a variety of functional groups of main and/or side chains.

Abstract:For search the effective method of synthesis of citral intermediate,prenal and prenol was taken as starting material,using cetalization reaction 3-methyl-2-butenal diprenyl acetal was synthesized by senecioic acid catalysis and elimination reaction cis/trans-prenyl-3-methylbutadienyl ether with phosphoric acid catalysis.The conditions of acetalization reaction and elimination reaction were investigated in detail.It was found that with 0.3% senecioic acid as catalysis,acetalization reacting at 70~75℃ for 8 hrs by azeotropism,one-way conversion for prenal was 63~64%;after treatment 3-methyl-2-butenal diprenyl acetal yield was 96.8% with 97.5% purity.Using 100×10-6 phosphoric acid as catalyst,elimination reacting at 90~120℃ and 2.66 kPa and vaporizing out of the reaction products in a timely manner,after treatment cis/trans-prenyl-3-methylbutadienyl ether yield was 97% with 95.7% purity.
