Abstract:A practical Buchwald-Hartwig amination of [2.2]paracyclophanyl bromides with benzhydrylideneamine is developed. The method provides a Facile route to a variety of imino and amino [2.2]paracyclophanes that are otherwise not readily synthesized.

Abstract:A method for the amination of compounds with carbonyl groups was described.The process was carried out by using ketones as substrates,ammonium formate as the source of hydrogen and nitrogen,Pd / C as catalyst and the mixture of methanol and water as solvent under normal temperature and pressure.Ammonium formate as hydrogen supplier has the advantages of low cost,easy availability as well as excellent reducibility.while Pd / C as catalyst enables the reaction to take place under mild conditions.The process reacts quickly with convenient workup and high selectivity.The optimal reaction conditions were as follows : CH 3 OH∶H 2 O(V∶V) =9∶1,HCOONH 4 ∶ketone∶ Pd / C(M∶M∶M) =100∶10∶1.Here the amination of 2-adamantanone,5-hydroxy-2-adam antanone and 3-quinineone were studied with satisfactory results.All products were confirmed by 1 H-NMR,GC-MS or MS.

Abstract:An easily available N-heterocyclic carbene–palladium(II) complex was found to be an efficient catalyst for the Buchwald–Hartwig amination of aryl chlorides. Both secondary and primary amines were tolerated under the same reaction conditions. Under the optimal conditions, all reactions proceeded successfully to give the desired products in good to high yields within hours.Graphical Abstract

Abstract:A family of 2-(9H-carbazol-9-yl)phenyl-based phosphine ligands were synthesized and their efficacy in promoting the steric hindered Buchwald–Hartwig amination was evaluated. In the presence of Pd(OAc)2 (0.03–1.0 mol%) associated with the newly developed a carbazolyl-derived phosphine ligand, the synthesis of tetra-ortho-substituted diarylamines proceeded smoothly with excellent product yields (up to 99%). A remarkable result was obtained even for the coupling of highly sterically congested 2,6-diisopropylaniline and hindered 2-chloro-1,3,5-triisopropylbenzene (96% isolated yield). A possible decomposition pathway for the anthracenyl C–N coupling product is also reported.

Abstract:The scope limitation of substrates especially for five-member heteroaryl substrates often presenting in medical and bioactive molecules, along with high palladium loadings and sometimes needing strong bases, perplexes the application of the palladium-catalyzed C-N cross-coupling reactions. The terphenyl phosphine TXPhos supporting palladium catalyst [(TXPhos)(allyl)PdCl] dramatically broadened the existing scope of five-membered heteroaryl substrates and made the weak bases KHCO 3 and KOAc become general and optimal choices. Therefore, the barrier of bases has been broken and an attractive protocol for the preparation of secondary five-membered heteroaryl amines via the palladium-catalyzed C-N cross-coupling reaction has been established.

Abstract:An efficient and straightforward procedure toward the synthesis of amides-substituted 2H-benzopyrans H-benzopyrans has been explored through palladium-catalyzed one-pot aminocarbonylation reaction. By using nitro compounds as attractive nitrogen sources, with Mo(CO)6 6 as both CO source and reductant, a variety of amides-containing 2H- H- benzopyrans were obtained in moderate to high yields with good functional group tolerance. This protocol has advantages including atom-economy, no manipulation of toxic CO gas, and broad substrate scope.

Abstract:We report a diverse demonstration of synthetically useful chemoselectivity in the synthesis of di-, tri-, and tetraamines (62 examples) by use of Buchwald-Hartwig amination employing a single catalyst system ([Pd(cinnamyl)Cl](2)/L1; L1 = N-(2-(di(1-adamantyl)phosphino)phenyl)morpholine, Mor-DalPhos). Competition reactions established the following relative preference of this catalyst system for amine coupling partners: linear primary alkylamines and imines > unhindered electron-rich primary anilines, primary hydrazones, N,N-dialkylhydrazines, and cyclic primary alkylamines > unhindered electron-deficient primary anilines, α-branched acyclic primary alkylamines, hindered electron-rich primary anilines ≫ cyclic and acyclic secondary dialkylamines, secondary alkyl/aryl and diarylamines, α,α-branched primary alkylamines, and primary amides. The new isomeric ligand N-(4-(di(1-adamantyl)phosphino)phenyl)morpholine (p-Mor-DalPhos, L2) was prepared in 63% yield and was crystallographically characterized; the [Pd(cinnamyl)Cl](2)/L2 catalyst system exhibited divergent reactivity. Application of the reactivity trends established for [Pd(cinnamyl)Cl](2)/L1 toward the chemoselective synthesis of di-, tri-, and tetraamines was achieved. Preferential arylation was observed at the primary alkylamine position within 2-(4-aminophenyl)ethylamine with [Pd(cinnamyl)Cl](2)/L1 and 4-chlorotoluene (affording 5a); the alternative regioisomer (5a') was obtained when using [Pd(cinnamyl)Cl](2)/L2. These observations are in keeping with coordination chemistry studies, whereby binding of 2-(4-aminophenyl)ethylamine to the in situ generated [(L1)Pd(p-tolyl)](+) fragment occurred via the primary amine moiety, affording the crystallographically characterized adduct [(L1)Pd(p-tolyl)(NH(2)CH(2)CH(2)(4-C(6)H(4)NH(2))](+)OTf(-) (7) in 72% yield.

Abstract:De novo catalytic syntheses of diarylamines from a palladium‐catalyzed reductive Buchwald‐Hartwig amination of nitroarenes with aryl (pseudo)halides is described. The exquisite use of upstream nitroarenes as arylamine surrogates, the judicious selection of bis(pinacolato)diboron (B2pin2) as a stoichiometric reducing agent, and wide substrate scope including (hetero)aryl halides (Cl, Br and I) and aryl triflates, constitute the striking features of the current protocol. Moreover, application of this technique to the syntheses of advanced intermediates and active pharmaceutical ingredients also proved successful, thus providing an alternative step‐economical approach to the syntheses of diarylamine‐incorporated molecules. Preliminary mechanistic investigation demonstrates that an amine and a nitrosoarene intermediates might be involved in this reductive event.

Abstract:A novel PdCl2/bis(2-pyridylmethyl)amine-based ligand (1) catalytic system, which is water-soluble and air-stable, has been successfully synthesized and applied for Suzuki-Miyaura cross-coupling reaction. In the presence of catalytic amount of PdCl2/1 system, arylboronic acids can couple with a wide range of aryl halides, including aryl bromides and aryl chlorides. The reactions proceed under mild conditions to give excellent yields, and a wide range of functionalities is tolerated.

Abstract:An efficient palladium-catalyzed amination of aryl halides with amines, using rac-P-Phos as the ancillary ligand and Pd(OAc)(2) as the palladium source is developed. The ligand and all of the synthetic intermediates are stable to air and moisture, allowing the easy handling. The catalyst system performed well for a large number of different substrate combinations in 80-110 degrees C to furnish aromatic amines in high yields.

Abstract:The first aryl iodide catalyzed intramolecular C-H amination of phenylurea has been disclosed for high-efficiency synthesis of benzimidazolone derivatives in excellent yields (up to 97%) by an operationally simple one-step organocatalytic oxidative process. Fluorinated protic alcohols can efficiently accelerate the conversion of this transformation. The straightforward method has good functional group tolerance and can be performed with an inexpensive and readily accessible catalyst with high proficiency.

Abstract:With the readily available Gorlos-Phos as a ligand, halogenated diarylamines were prepared efficiently from Pd-catalyzed monoamination of bromophenyl chlorides and primary arylamines, tolerating functional groups such as aldehyde, ketone, nitrile, fluorine, sulphonyl and trifluoromethyl groups. The formation of the amination of the C-CI bond and diamination products were not observed.

Abstract:The intermingle of novel imidazolium salt (HL1 -Br) and Cu(OAc)2 marked down as a highly efficient catalytic system for Buchwald-Hartwig type C-N Coupling of a variety of amines with (hetero) aryl bromides/chlorides. In contrast to the whole realm of C−N Coupling, the developed protocol facilitates under base, solvent, and inert gas-free conditions. The established NHC-based Imidazolium salt 3-(2-(2-methoxyphenyl)-2-oxoethyl)-1-(pyridin-2- yl)-1H-imidazo-3-ium bromide (HL1-Br) along with Cu lend a hand to undergo various diversification. It broadens the range of C-N Coupling in numerous applications and gives rise to innovative solutions for the challenges in catalytic, synthetic chemistry, and drug discovery. The developed reaction proceeds at 110 °C and produces a broad spectrum of substrates in good to excellent yields.

Abstract:A series of novel pyridine stabilized Pd–NHC complexes were developed, which revealed high activities and broad substrates tolerance in the amination of various (hetero)-aryl chlorides. Besides various secondary amines, a wide range of primary anilines and aliphatic amines were also well tolerated. The results highlight us a new strategy to increase catalyst activity in the future catalyst design by alternating the σ-donor property and flexibility of NHC ligands.

Abstract:A bulky and electron-rich N-heterocyclic carbene–palladium complex (SIPr)Ph2Pd(cin)Cl was synthesized and characterized. It was found to be highly efficient and versatile for the coupling of different (hetero)aryl chlorides with various (hetero)aryl amines at room temperature, especially for the challenging amination of five- or six-membered ring heteroaryl chlorides with five- or six-membered ring heteroaryl amines. It was also successfully applied with high yields to the synthesis of various commercial pharmaceuticals and candidate drugs or compounds with potential pharmacological activities. All of these demonstrate its excellent catalytic efficacy in Buchwald–Hartwig amination and broad application prospects in relevant pharmaceutical preparations. DFT calculations suggest that the steric-induced electronic interaction makes the ligand more electron-donating, and the steric effect effectively regulates the rotation of the iPr-Ph-iPr group in the catalyzed system due to the introduction of the diphenyl skeleton. Considering the electronic effect and the steric effect together, the oxidative addition activation barriers of the (SIPr)Ph2 and (SIPr) ligands are close to each other. Reductive elimination is the rate-determining step of the (SIPr)Ph2Pd(cin)Cl-catalyzed system in the catalytic cycle, and the appropriate steric hindrance of the (SIPr)Ph2 ligand greatly reduces the energy barrier of this step. Abstract:A practical, efficient Pd(OAc)(2)/Cu(II)/PPh3 cocatalytic system has been developed. With only 1 mol% Pd(OAc)(2), 20 mol% copper(II) salt, and 0.5 equiv of inexpensive PPh3 as ligand, the direct arylation of benzoazoles with aryl bromides could be performed smoothly in mild condition, affording the desired arylated benzoazoles in good yields (75-93%).

Abstract:The first example of the Pd(II)-catalyzed enantioselective amination of aryl C–H bonds is reported. The key to the successful realization of this asymmetric catalytic transformation was the identification of mono-N-protected α-amino-O-methylhydroxamic acid (MPAHA) ligands, which promote reactivity under mild conditions and control enantioselectivity. Abstract:A coordinating activation strategy for selective para amination of arylamine with pyrazole is developed. Various substrates are compatible, giving the corresponding products in moderate to good yields. This strategy provides a practical solution for the efficient synthesis of arylamine‐containing pharmacophores from simple starting materials. A single electron transfer mechanism is suggested for this reaction.

Abstract:Amination reactions play a pivotal role in synthetic organic chemistry, facilitating the generation of nitrogen-containing scaffolds with broad applications in drug synthesis, material production, polymer formation, and the generation of amino acids and peptides. Amination offers the potential to fine tune the properties of natural products and produce functional materials for various applications. Palladium N -heterocyclic carbene (Pd–NHC) emerges as an innovative and highly effective catalyst in this context. Under favorable reaction conditions, this robust and simple catalyst efficiently facilitates the synthesis of a diverse range of compounds with varying complexity and utility. Pd–NHC complexes exhibit significant σ-electron donating potential, enhancing the ease of the oxidative addition process in their mechanistic pathway. Their steric topography further contributes to a rapid reductive elimination. These complexes demonstrate remarkable stability, a result of the strong Pd–ligand bond. The wide variety of Pd–NHC complexes has proven highly efficient in catalyzing reactions across a spectrum of complexities, from simple to intricate. The domain of aminations catalyzed by Pd–NHC has undergone significant diversification, presenting new opportunities, particularly in the realms of material chemistry and natural product synthesis. This review outlines the advancements in Pd–NHC-catalyzed amination reactions, covering literature up to date.

Abstract:A novel type of highly efficient chiral C2-symmetric bipyridine-N,N’-dioxides catalysts to catalyze Michael addition/Cyclization of 5-aminopyrazoles with α,β-unsaturated 2-acyl imidazoles has been developed, affording the corresponding adducts in 85-97% yield with up to 99% enantioselectivity. Remarkably, this protocol exhibits extraordinary advantages in terms of reactivity and enantioselectivity, given the fact that as low as 2.2 mol % of L1 and 2.0 mol % of Ni(OTf)2 can promote the title reaction on gram scale to afford desired product with excellent enantioselectivity.
