Abstract:A Streptomyces strain which could produce a novel kind of antifungal-antibiotics was screened out.The fermentation conditions were optimized: initial pH 6.5,inoculum percentage 10%,at 30℃,fermentation time (72 h) using conical beaker.And with the same K_(L)a,the reversed jet loop reactor(JLR) was more fit for antifungal-antibiotics fermentation than continuous stirred tank reactor(CSTR): the potency of fermentation liquor and the morphology of mycelium with JLR was much higher than those with CSTR.

Abstract:The removal efficiency of antibiotic resistance genes (ARGs) is the biggest challenge for the treatment of erythromycin fermentation residue (EFR). In the current research, 0% (control), 10% (T1), and 30% (T2) spray-dried EFR were composted with bulking materials, consisting of cattle manure and maize straw, for 30 days. Environmental factors and bacterial community on the behaviors of ARGs were further investigated. Apart from the high levels of erythromycin, the electrical conductivities were also increased by 66.7% and 291.7% in the samples of T1 and T2, respectively. After 30 days of composting, total ARGs in the samples of control were decreased by 78.1%-91.2%, but those of T1 and T2 were increased 14.5-16.7- and 38.5-68.7-fold. ARGs related to ribosomal protection (erm) dominated the samples of T1 and T2 at D 13 and 30, especially that ermF accounted for more than 80% of the total ARGs. Furthermore, the results of bacterial community revealed that EFR promoted the growth of Proteobacteria and Bacteroidetes, but inhibited that of Actinobacteria, Verrucomicrobia and Chloroflexi. Network analysis revealed that the enriched ARGs had strong correlation with seven bacterial genera, including Halomonas, Oceanobacillus, and Alcaligenes, most of which are halotolerant. Above all, erythromycin combined with high salinity can have synergistic effect on the enrichment of ARGs and their hosts.

Abstract:The kinetics of erythromycin fermentation in an air-lift bioreactor was studies.According to the kinetic equations,feeding strategies,the optimum process control parameters,the ratio of C/N(carbon/nitrogen)and oxygen uptake rates were obtained.It resulted in a significant improvement on the erythromycin fermentation.The total metabolic phase was reduced by 13% and the production was 10% higher than that by the traditional process.
Fumaric acid production in airlift loop reactor with porous sparger

Abstract:Airlift loop reactors with porous spargers were investigated and used in the process of fumaric acid production by Rhizopus oryzae ATCC 20344. In order to enhance oxygen mass transfer, which is very important for organic acid production, two kinds of porous spargers (stainless steel membrane tube and porcelain tube) were examined. Gas holdup, liquid circulation velocity, mixing time, bubble size, and bubble rise velocities were measured in a 50 L rectangular airlift loop reactor with different ratios of the cross-sectional area of the riser and downcomer. The local volumetric mass transfer coefficient (K(L)a) was also measured in the gas sparger zone. The results indicated that high K(L)a and excellent hydrodynamics can be obtained in the airlift loop reactor with a porous sparger. A 10 L laboratory airlift loop reactor was employed for the fumaric acid fermentation. Results showed that the turbulence of two-phase flow in the airlift loop reactor not only produced favorable conditions for mass transfer, but was also useful for forming and suspending small, well-distributed mycelial pellets (1-2 mm). A production rate of up to 0.814 g/L/h and efficiency yield of 50.1% (w/w) was obtained in the airlift loop reactor. The performance was compared with the typical stirred tank fermentor fermentation results.

Abstract:The effects of structure characteristics of external airlift loop reactor (EALR) on mass transfer behavior are studied. The parameters such as the ratio of height to diameter, the structure of air sparger, the location of air sparger and the distance between two pipes have effects on gas holdup and mass transfer of EALR. The optimum structure of reactor has been obtained and been used for fermentation of Bacillus Thuringensis. Compared with the traditional mechanical agitated fermentor, the cycle of fermentation in EALR is shortened by 9 hours and the cell concentration of broth is improved by 35%.

Abstract:The rational design of a bioreactor plays an important role in establishing an efficient biological wastewater treatment process. In this study, a multi-tube parallel airlift reactor (MP-ALR) composed of multiple draft tubes was designed and developed by computational fluid dynamics (CFD), and was applied to oily wastewater treatment. The CFD results showed that the gas holdup and liquid velocity of MP-ALR were higher than those in a traditional airlift reactor (T-ALR), and the flow field distribution of MP-ALR was more uniform. The optimum number of parallel draft tubes was determined to be 3 in the design of a 280 L reactor with a high aspect ratio. The experimental results of using Trichosporon fermentans to treat refined soybean oil wastewater (RSOW) in a 32 L MP-ALR showed that the maximum biomass concentration increased by 2.39 g/L and the chemical oxygen demand (COD) removal was increased by 17.73 % compared with those in T-ALR. The MP-ALR developed in this study not only improves the efficiency of the biological treatment of oily wastewater, but also provides a new idea for the design of bioreactors.

Abstract:The establishment of erythromycin production within the heterologous host E. coli marked an accomplishment in genetic transfer capacity. Namely, over 20 genes and 50 kb of DNA was introduced to E. coli for successful heterologous biosynthetic reconstitution. However, the prospect for production levels that approach those of the native host requires the application of engineering tools associated with E. coli. In this report, metabolic and genomic engineering were implemented to improve the E. coli cellular background and the plasmid platform supporting heterologous erythromycin formation. Results include improved plasmid stability and metabolic support for biosynthetic product formation. Specifically, the new plasmid design for erythromycin formation allowed for ≥89% stability relative to current standards (20% stability). In addition, the new strain (termed LF01) designed to improve carbon flow to the erythromycin biosynthetic pathway provided a 400% improvement in titer level. © 2017 American Institute of Chemical Engineers Biotechnol. Abstract:Erythromycin is a macrolide antibiotic produced by Saccharopolyspora erythraea. Its yield is greatly affected by the fermentation conditions and the bioreactor configurations. In this study, a novel scale-up method for erythromycin fermentation was developed based on computational fluid dynamics (CFD) and time constant analysis. Firstly, the dissolved oxygen (DO) was determined as a key parameter according to the physiological properties of S. erythraea cultivated in a 50 L bioreactor. It was found that the time constant of oxygen supply (tmt) in a 500 m3 bioreactor should be less than 6.25 s in order to satisfy the organism's oxygen uptake rate (OUR). Subsequently, a 500 m3 bioreactor was designed using the time constant method combined with empirical correlations. The impeller combination with one BDT8 impeller at bottom and two MSX4 impellers at upper part was determined, and then validated by numerical simulation. The results indicated that the tmt of the bioreactor (< 6.25 s) and the fluid properties, including gas hold-up, shear stress and fluid vector, met the requirements of erythromycin fermentation. Finally, the industrial production of erythromycin in the 500 m3 showed the design method was applicable in large scale fermentation.

Abstract:A process for efficient production of erythromycin by Saccharopolyspora erythraea using statistical designs and feeding strategy was developed. The critical nutrient components were selected in accordance with fractional factorial design and were further optimized via response surface methodology. Three significant components (ZnSO 4 , citric acid threonine) were identified for the optimization study. The optimum levels of these significant variables were determined with Box–Behnken design, which were ZnSO 4 0.039 g/l, citric acid 0.24 g/l and threonine 0.42 g/l, respectively. A novel feeding strategy based on oxygen uptake rate (OUR) measurement was developed successfully to increase the flux of erythromycin biosynthesis, in which the optimized nutrient components was fed in the 50 l stirred bioreactor when OUR began to decline at 46 h. The maximum erythromycin production reached 10,622 U/ml, which was 11.7% higher than the control in the same cultivation conditions. It was the first report to integrate physiological parameter OUR and statistical methods to optimize erythromycin production.

Abstract:The fermentation experiments of recombinant Ecc13B-1 for the production of PHB were carried out to achieve high cell density in 15L Air-lift Loop Reactor. The experimental results showed that the cell density can reach 57. 9g/cm3 in 53 hours in ALR with holeless draft tube. Furthermore, the production ability of cell and PHB significantly increased. Maintaining the stable specific growth rate in fermentation was helpful to the increase of cell density. The results also proved that ALR was superior to stirred tank for high cell density cultivation.

Abstract:Airlift loop reactors (ALRs) are important bioreactors for wastewater treatment. However, few studies have investigated the application of an ALR for simultaneous carbon and nutrient removal, especially for activated sludge systems. This study evaluated the performance of integrated nitrogen, phosphorus and COD removal in an ALR with a low height-to-diameter ratio in a limited filamentous bulking (LFB) state (SVI of 180-220mL/g). The average removal efficiencies for COD, NH(4)(+)-N, TN and TP were 91%, 92%, 86% and 94%, respectively. Additional research showed that only under the LFB state, the appropriate distribution of dissolved oxygen inside the ALR was established to promote a well-balanced aerobic and anoxic/anaerobic state. In addition, the macro-gradient of the substrate concentration at the inlet and the heavier bio-P sludge density compensated for the proliferation of filaments. Hence, the stable LFB state was achieved by balancing the floc-forming bacteria and the filamentous bacteria in the ALR.

Abstract:The physiological response of erythromycin fermentation scale-up from 50 L to 132 m 3 scale was investigated. A relatively high oxygen uptake rate (OUR) in early phase of fermentation was beneficial for erythromycin biosynthesis. Correspondingly, the maximal consistency coefficient ( K ) reflecting non-Newtonian fluid characteristics in 50 L and 132 m 3 fermenter also appeared in same phase. Fluid dynamics in different scale bioreactor was further investigated by real-time computational fluid dynamics modeling. The results of simulation showed that the impeller combination in 50 L fermenter could provide more modest flow field environment compared with that in 132 m 3 fermenter. The decrease of oxygen transfer rate (OTR) in 132 m 3 fermenter was the main cause for impairing cell physiological metabolism and erythromycin biosynthesis. These results were helpful for understanding the relationship between hydrodynamic environment and physiological response of cells in bioreactor during the scale-up of fermentation process.

Abstract:Abstract Airlift reactor (ALR) is a promising multiphase reactor for industrial applications. Abundant reports about modifications of the conventional ALR and optimization of their operation conditions for the purpose of performance enhancement have been accumulated in literatures, demanding a review paper to summarize the reactor design modifications and operation condition optimization of the ALR. In this review, the published research findings and results have been summarized. The basic concepts including the ALRs’ conventional design, classification, principles of operation, and characteristic parameters have been analyzed and systematically organized. The updated advances in the ALR design modifications have been reported. In particular, the concepts of the “groveled ALR” solving the scaling up problem in wastewater treatment, large-scale application, and the ALR with the cross-shaped geometry modifier stabilizing and strengthening the reactor were considered. Also, new operation modes and optimal conditions for enhancing the performance of the ALR have also been summed up. Except for conventional gas-driven methods, new driven methods for the ALR, such as mixture emission of the gas and the liquid and gas-inducing impeller, have been introduced. Optimization of operation conditions for the ALR includes varying position of the gas spargers, utilizing elevated pressure reactor, and exploring the impact of operation parameters, such as superficial gas velocity, static liquid level, and fluid properties. Comparisons between conventional ALRs and the modified systems are carried out paying attention to analogies, similarities, and differences. Most of the documented research results are obtained for various reactor designs at a laboratory scale; studies at pilot and full scale are still insufficient, which indicates that universal scale up design rules permitting the ALR design with a high confidence are required.

Abstract:BACKGROUND: The classical airlift loop reactor (ALR) has been widely used in petrochemical, biochemical, energy and environmental processes due to such advantages as simple structure, without motional mechanism, easy sealing and low energy consumption. A novel ALR has been designed using forced periodic reversible ventilation, termed an airlift reversible loop reactor (ARLR). RESULTS: Using computational fluid dynamics (CFD) simulation and experimental validation, the mass transfer characteristics of the ARLR were studied. The simulation results predicted the experimental data well, especially at low ventilation capacity. The oscillation period had significant effects on gas holdup and the mass transfer coefficient of the ARLR. When ventilation capacity was increased from 0.22 to 3.49 vvm, compared with central airlift and annular airlift reactors, ARLR increased the mass transfer coefficient by 931% and 1058%, respectively, according to simulation results, and by 1125% and 1458%, respectively, according to experimental data. CONCLUSION: The results showed that the ARLR could significantly enhance gas holdup and mass transfer coefficient compared with traditional central airlift and annular airlift reactors. Results indicated that the optimum oscillation period decreased with increase of ventilation capacity. ARLR has the potential for application in aerobic fermentation. Abstract:A novel source of biological nitrogen produced by multi-strain fermentation was developed for erythromycin production. The nitrogen composition of corn steep liquor, biological nitrogen and soybean flour was optimized by using Response surface methodology (RSM) firstly. A Box-Behnken design was used to estimate the optimum nitrogen composition for the production of erythromycin as follows: corn steep liquor: gamma = 5.1 g L-1, biological nitrogen gamma = 5.96 g L-1 and soybean flour gamma = 24.17 g Based on the optimum nitrogen source, the constant glucose fed-batch and pH control fed-batch strategies were used to further optimize the erythromycin production in 50 L-1 stirred bioreactor, respectively. It was found that pH control could serve as an effective control strategy for erythromycin production, and the maximum erythromycin production was 8528 U mL(-1) at 190 h. This work demonstrates that the new medium formulation based on a cheap nitrogen source, biological nitrogen, is a potential alternative for economic erythromycin production on a large scale.

Abstract:One important challenge in chemical industry, especially for processes of synthesis fuels, is the development of effective three-phase reactors for high pressure and temperature. In the present study, a novel airlift internal-loop reactor (ALR) was proposed and its hydrodynamic behaviors were experimentally studied. The local bubble rise velocity and liquid velocity are measured under different operating conditions. The influence of operating conditions including the superficial gas velocity and solid holdup on the bubble rise velocity and liquid velocity in the riser was studied. A mathematical model was proposed based on the energy balance to describe the multiphase flow characteristics in the internal-loop airlift reactor. The comparison between the calculated and experimental values shows that the proposed model can predict the flow behavior reasonably.

Abstract:<p>A pilot external airlift loop reactor (EALR) integrating mixing and separation was first proposed by combining directional flow in the EALR and liquid-solid separation in two parallel hydrocyclones. In this developed reactor, mixing, mass transfer, and liquid-solid separation can be simultaneously realized without extra energy input. Technical feasibility was firstly verified, and then hydrodynamics and mass transfer were measured. It was found that the minimum diameter of solid particles retained for circulation was 6.82 μm, and the treatment capacity of clean liquid was up to 3.0 m<sup>3</sup>/h. Additionally, global gas holdup, liquid circulating velocity, and volumetric mass transfer coefficient increased monotonously with increasing the superficial gas velocity while declined with the increase of solid concentration. Axial inhomogeneity of solid particles was decayed with increasing the superficial gas velocity and solid concentration. This new type of slurry EALR is promising to be applied in the gas-liquid-solid catalytic industries.</p>

Abstract:The concentric-tube internal loop airlift reactor is a typical reactor configuration which has been adopted for a myriad of chemical and biological processes. The reactor hydrodynamics (including mixing) and the mass transfer between the gas and liquid phases remarkably affect the operational conditions and thus are crucial to the overall performance. Hence, this study aims at providing a thorough description of the basic concepts and a comprehensive review of the relevant reported studies on the hydrodynamics and mass transfer of the concentric-tube internal loop airlift reactors for microalgae cultivation as an exemplary application. In particular, the reactor characteristics, geometry, CFD modeling, experimental characterization, and scale up considerations are elucidated. The research gaps for future research and development are also identified.

Abstract:In order to improve glycerol yield without extra i nv estment and complicated operation, an airlift reversible loop reactor (ARLR) with periodic oscillating airlift operation, in which the airflow could be alternated between one nozzle in the central region and eight nozzles in the annular region at the bottom of the reactor, was investigated during glycerol pr oduction by osmotolerant yeast Candida krusei.
