Abstract:In order to improve the conversion efficiency of lactic acid from cellulose,the spores containing cellobiase with high activity from Aspergillus niger ZU-07 and the cells of Lactobacillus delbrium were embedded together into calcium alginate gel beads,and the formed coimmobilized cell system was coupled with the enzymatic hydrolysis of cellulosic material to set up a novel coupling bioreactor.The research results showed that the cellobiose in the hydrolysate was converted to glucose rapidly by the cellobiase in the coimmobilized cells,and the glucose was utilized rapidly by L.delbrium to produce lactic acid.Therefore,the feedback inhibition on cellulase caused by cellobiose and glucose was eliminated effectively.When the hydrolysis temperature and fermentation temperature were controlled at 50℃ and 48℃, respectively,and the volume ratio of the coimmobilized cells to the column reactor was 40%,the produced lactic acid concentration and the yield of lactic acid from cellulose were 55.7 g·L-1 and 91.5%,respectively.Under the fed-batch process,the final concentration of lactic acid and productivity increased to 106.7 g·L-1 and 1.270 g·L-1·h-1,respectively,while the dosage of cellulase per gram substrate reduced by 25%.
Simultaneously Saccharification and Lactic Acid Fermentation for Using Cellulosic Material As Raw Material in a Coupling Bioreactor

Abstract:The enzymatic saccharification of cellulose and the fermentation of lactic acid were carried out simultaneously in a coupling bioreactor. The glucose from cellulose hydrolysis could be converted to lactic acid effectively by the immobilized Lactobacillus delbrueckii cells, and the yield and productivity of lactic acid were 70.3% and 0.713 g?(L?h)?1, respectively. The hydrolysis process of cellulose could be enhanced by adding cellobiase to the cellulase system of Trichoderma reesei. When an immobilized cellobiase column was joined into the coupling bioreactor, the accumulation of cellobiose disappeares and its feed back inhibition to the cellulase reaction is completely eliminated. The yield and productivity of lactic acid may be improved to 90.6% and 0.986 g?(L?h)?1, respectively. The bioreactor is quite stable, and the average lactic acid yield of repeated 10 batches is 89.6%. Under fed-batch process the final concentration of cellulosic substrate increases to 200 g?L?1, and the lactic acid concentration and productivity reach 105.2 g?L?1 and 1.315 g?(L?h)?1, respectively. The research results are helpful to advance the bioconversion of renewable resource.
Preparation of a Novel Soluble Inducer by Cellobiase-Release Microcapsules and Its Application in Cellulase Production

Abstract:Effective and low-cost inducer is a prerequisite for efficiency improvement in cellulase production by Trichoderma reesei. In this study, spores of Aspergillus niger enriched with cellobiase were embedded in Ca-alginate microcapsules, attaining a sustained cellobiase-release system (named CRMA) which was then used to catalyze the conversion of glucose into glucose-sophorose sugar mixture via transglycosylation. Further, the process and optimal parameters of transglycosylation reaction were studied in a 5 L reactor and the sophorose concentration reached 70.55 g/L at 50 degrees C, pH 4.0 with CRMA loading of 200 mL after 72 h of reaction. The obtained sugar mixture was subsequently used as both carbon source and inducer in cellulase production by Trichoderma reesei under fed-batch process. The filter paper activity (FPA) reached 90.28 IU/mL at 144 h fermentation time, 1.85-fold higher than that induced by lactose. These results indicate that the newly established strategy for soluble inducer synthesis has the potential for efficiency improvement and cost reduction in cellulase production.
Production and immobilization of cellobiase from Aspergillus niger ZU-07

Abstract:A high yield strain Aspergillus niger ZU-07 was used to produce cellobiase. It was found that the spores of A. niger ZU-07 were rich in cellobiase. By entrapping the spores into calcium alginate gels, the cellobiase was immobilized efficiently. The immobilized spores were quite stable, with a half-life 38 days. In repeated batch hydrolysis processes on 10gl−1 cellobiose by the immobilized spores, the yield of enzymic hydrolysis remained higher than 97% during 10 batches. In a continuous hydrolysis process, the yield reached 98% at 0.4h−1 dilution rate. The cellulosic residue of corncob pretreated with dilute acid was hydrolyzed with cellulase from Trichoderma reesei, the hydrolysis yield was only 69.5% and much cellobiose was accumulated in the hydrolysate. When the hydrolysate was further treated with immobilized spores of A. niger ZU-07, the hydrolysis yield of cellulose was improved to 84.2% because the feedback inhibition caused by cellobiose disappeared, the concentration of total reducing sugar increased from 48.50 to 58.78gl−1, and the ratio of glucose in the reducing sugar was increased from 53.6 to 89.5%. The research results are useful in the conversion and utilization of renewable biomass.

Abstract:Phenylacetic acid can be produced from 2-phenylethanol by selective oxidation. This process can be performed using Gluconobacter oxydans immobilized in LentiKats®, polyvinyl alcohol particles with lenticular shape. The biotransformation took place at non-growth conditions in an airlift bioreactor. The goal of this work was to examine the impact of the particles on the hydrodynamics and volumetric mass transfer coefficient for oxygen in the airlift reactor and to describe the activity of the immobilized biomass in such systems. For this purpose, the influence of aeration and 2-phenylethanol concentration on the activity of biomass was examined. Also, the effects of long-term storage of immobilized biomass, long duration of fermentation and repeated use of biomass on the rate of phenylacetic acid production were tested. The results were compared with the phenylacetic acid production by free cells of Gluconobacter oxydans. The results show that aeration of the airlift reactor has only a minor effect on the phenylacetic acid production for immobilized biomass but a significant one for free cells; however, the impact of 2-phenylethanol concentration on the production was notable for both forms of biocatalyst depending on the biomass concentration. Production ability of free cells was ten times higher than that of immobilized cells, but the production time was only a quarter of that of immobilized cells.

Production of Butyric Acid by Immobilized Cells in a Fibrous-Bed Bioreactor of Pilot-Scale

Abstract:In this paper,efficient production of butyric acid from glucose by Clostridum tyrobutyricum was performed with batch and fed-batch modes. Based on the results of 5-L fermentor's experiments,the fermentation process was successfully scaled up in a 5000-L fermentor. In 5-L fermentor batch modes,butyrate yield and productivity was reached to 0. 47 g / g and 0. 82 g / L·h respectively,which was improved by 31% and 134% compared with fermentations of free cells. In the fed-batch fermentation,the final concentration of butyric acid was 52. 8 g / L,increased by 46% compared to free-cell fermentation. From up-scaled fed-batch production of butyrate in the pilot plant,butyrate yield,productivity and butyrate concentration with immobilized-cells reached 0. 47 g / g,0. 63 g /( L·h) and 51. 63 g / L,which were improved by 37%,67% and 40% respectively. Results showed that compared with the laboratory scale,the fibrous bed reactor could maintain a higher level of butyric acid fermentation and the production efficiency when zoom up to 1000 times. The present work provides a powerful alternative to produce butyric acid as a bulk chemical in pilot scale.

Abstract:BACKGROUNDButyric acid is an important chemical with wide industrial applications, but large-scale bio-production still requires an efficient bioprocess. For this purpose, one novel moving fibrous-bed bioreactor (MFBB) was developed in this work for economical production of butyric acid.RESULTSThe cells of Clostridium tyrobutyricum were immobilized on a fibrous matrix packed in porous levitated sphere carriers. Each carrier was packed with a fiber quantity 2 cm x 6 cm (0.42 g) and was rotated at 100 rpm in a stirred-tank fermentor having pH and temperature controls. Batch and repeated-batch fermentations were carried out to produce butyric acid with a high yield (0.48 0.015 g g(-1)). Subsequent fed-batch fermentation using the MFBB resulted in a high concentration of butyric acid (65.4 +/- 1.4 g L-1). This novel MFBB could be employed to produce high concentration butyric acid (64.5 +/- 1.2 g L-1) by a simple repeated-batch addition of a high concentration of glucose (>100 g L-1).CONCLUSIONSThe MFBB system developed could practically and economically produce butyric acid from glucose for long-term operation. Abstract:In this research a bench scale rotating biological contactor (RBC) was designed and constructed to produce BC. The effects of variables including rotation speed of the disk, distance between disks, disk type and external aeration on BC productivity were investigated. Results showed that the highest weight of BC produced on the surface of integrated polyethylene discs which rotated at 13 rpm. It was also found that the highest amount of BC was obtained when the space between two adjacent discs was adjusted to 1 cm and the disk number was 16. An aquarium pump was used to investigate the impact of aeration on RBC made of 12 integrated polyethylene discs and operated at optimal rotation speed of 13 rpm. Disk spacing distance was adjusted to 1.5 cm to consider the possible increasing of the thickness of BC film by aeration. Wet weight and dry weight of BC resulted from aerated fermentation increased more than 64 and 47%, respectively as compared to non-aerated RBC. In comparison with static culture, wet weight and dry weight of BC produced in aerated RBC fermentation increased more than 90.7 and 71%, respectively. Nanoscale structure of produced bacterial cellulose was confirmed by SEM analysis.

Abstract:Bacterial cellulose (BC) is a remarkable biomacromolecule with potential applications in food, biomedical, and other industries. However, the low economic feasibility of BC production processes hinders its industrialization. In our previous work, we obtained candidate strains with improved BC production through random mutations in Gluconacetobacter . In this study, the molecular identification of LYP25 strain with significantly improved productivity, the development of chestnut pericarp (CP) hydrolysate medium, and its application in BC fermentation were performed for cost-effective BC production process. As a result, the mutant strain was identified as Gluconacetobacter xylinus . The CP hydrolysate (CPH) medium contained 30 g/L glucose with 0.4 g/L acetic acid, whereas other candidates known to inhibit fermentation were not detected. Although acetic acid is generally known as a fermentation inhibitor, it improves the BC production by G. xylinus when present within about 5 g/L in the medium. Fermentation of G. xylinus LYP25 in CPH medium resulted in 17.3 g/L BC, a 33 % improvement in production compared to the control medium, and BC from the experimental and control groups had similar physicochemical properties. Finally, the overall process of BC production from biomass was evaluated and our proposed platform showed the highest yield (17.9 g BC/100 g biomass).

Mitochondrial Ca(2+) buffering regulates synaptic transmission between retinal amacrine cells.

Abstract:The diverse functions of retinal amacrine cells are reliant on the physiological properties of their synapses. Here we examine the role of mitochondria as Ca(2+) buffering organelles in synaptic transmission between GABAergic amacrine cells. We used the protonophore p-trifluoromethoxy-phenylhydrazone (FCCP) to dissipate the membrane potential across the inner mitochondrial membrane that normally sustains the activity of the mitochondrial Ca(2+) uniporter. Measurements of cytosolic Ca(2+) levels reveal that prolonged depolarization-induced Ca(2+) elevations measured at the cell body are altered by inhibition of mitochondrial Ca(2+) uptake. Furthermore, an analysis of the ratio of Ca(2+) efflux on the plasma membrane Na-Ca exchanger to influx through Ca(2+) channels during voltage steps indicates that mitochondria can also buffer Ca(2+) loads induced by relatively brief stimuli. Importantly, we also demonstrate that mitochondrial Ca(2+) uptake operates at rest to help maintain low cytosolic Ca(2+) levels. This aspect of mitochondrial Ca(2+) buffering suggests that in amacrine cells, the normal function of Ca(2+)-dependent mechanisms would be contingent upon ongoing mitochondrial Ca(2+) uptake. To test the role of mitochondrial Ca(2+) buffering at amacrine cell synapses, we record from amacrine cells receiving GABAergic synaptic input. The Ca(2+) elevations produced by inhibition of mitochondrial Ca(2+) uptake are localized and sufficient in magnitude to stimulate exocytosis, indicating that mitochondria help to maintain low levels of exocytosis at rest. However, we found that inhibition of mitochondrial Ca(2+) uptake during evoked synaptic transmission results in a reduction in the charge transferred at the synapse. Recordings from isolated amacrine cells reveal that this is most likely due to the increase in the inactivation of presynaptic Ca(2+) channels observed in the absence of mitochondrial Ca(2+) buffering. These results demonstrate that mitochondrial Ca(2+) buffering plays a critical role in the function of amacrine cell synapses.
Enhancement of acetate productivity in a thermophilic (55 °C) hollow-fiber membrane biofilm reactor with mixed culture syngas (H2/CO2) fermentation

Abstract:Conversion of organic wastes to syngas is an attractive way to utilize wastes. The produced syngas can be further used to produce a variety of chemicals. In this study, a hollow-fiber membrane biofilm reactor with mix cultures was operated at 55 °C to convert syngas (H2/CO2) into acetate. A high concentration of acetate (42.4 g/L) was reached in batch experiment while a maximum acetate production rate of 10.5 g/L/day was achieved in the continuous-flow mode at hydraulic retention time (HRT) of 1 day. Acetate was the main product in both batch and continuous-flow experiments. n-Butyrate was the other byproduct in the reactor. Acetate accounted for more than 98.5 and 99.1% of total volatile fatty acids in batch and continuous modes, respectively. Illumina Miseq high-throughput sequencing results showed that microorganisms were highly purified and enriched in the reactor. The main genus was Thermoanaerobacterium (66% of relative abundance), which was usually considered as H2 producer in the literature, however, likely played a role as a H2 consumer in this study. This study provides a new method to generate the high producing rate and purity of acetate from syngas.

Abstract:In this study, a two-compartment biofilm reactor was designed for iturin A production. The biofilm reactor consists of a stirred-tank fermentor containing exclusively suspended cells and a packing column where the biofilm is attached. Polyester fiber with sphere shape and rough surfaces was chosen as the carrier of biofilm in packing column. Batch, fed-batch, and repeated-batch fermentation using Bacillus velezensis ND in the biofilm reactor were studied. Compared to conventional suspended cell fermentations, the productivity of iturin A in batch and fed-batch biofilm fermentation were increased by 66.7% and 63.3%, respectively. Maximum itutin A concentration of 6.8 ± 0.1 g/L and productivity of 46.9 ± 0.2 mg/L/h were obtained in fed-batch biofilm fermentation. Repeated-batch fermentation showed high stability, with almost same profile as batch fermentation. After a step-wise temperature control strategy was introduced in the biofilm reactor, productivity of iturin A was increased by 131.9% compared to suspended cell reactor. This superior performance of biofilm reactor confirms that it has great potential in industrial production of iturin A.

Fumaric acid production in airlift loop reactor with porous sparger

Abstract:Airlift loop reactors with porous spargers were investigated and used in the process of fumaric acid production by Rhizopus oryzae ATCC 20344. In order to enhance oxygen mass transfer, which is very important for organic acid production, two kinds of porous spargers (stainless steel membrane tube and porcelain tube) were examined. Gas holdup, liquid circulation velocity, mixing time, bubble size, and bubble rise velocities were measured in a 50 L rectangular airlift loop reactor with different ratios of the cross-sectional area of the riser and downcomer. The local volumetric mass transfer coefficient (K(L)a) was also measured in the gas sparger zone. The results indicated that high K(L)a and excellent hydrodynamics can be obtained in the airlift loop reactor with a porous sparger. A 10 L laboratory airlift loop reactor was employed for the fumaric acid fermentation. Results showed that the turbulence of two-phase flow in the airlift loop reactor not only produced favorable conditions for mass transfer, but was also useful for forming and suspending small, well-distributed mycelial pellets (1-2 mm). A production rate of up to 0.814 g/L/h and efficiency yield of 50.1% (w/w) was obtained in the airlift loop reactor. The performance was compared with the typical stirred tank fermentor fermentation results.

Abstract:Submerged cultivation of edible filamentous fungi ( Agaricomycetes ) in bioreactors enables maximum mass transfer of nutrients and has the potential to increase the volumetric productivity of fungal biomass compared to solid state cultivation. These aspects are paramount if one wants to increase the range of bioactives (e.g. glucans) in convenient time frames. In this study, Trametes versicolor (M9911) outperformed four other Agaricomycetes tested strains (during batch cultivations in an airlift bioreactor). This strain was therefore further tested in semi-continuous cultivation. Continuous and semi-continuous cultivations (driven by the dilution rate, D ) are the preferred bioprocess strategies for biomass production. We examined the semi-continuous cultivation of T. versicolor at dilution rates between 0.02 and 0.1 h −1 . A maximum volumetric productivity of 0.87 g/L/h was obtained with a D of 0.1 h −1 but with a lower total biomass production (cell dry weight, CDW 8.7 g/L) than the one obtained at lower dilution rates (12.3 g/L at D of 0.04 and vs 13.4 g/L, at a D of 0.02 h −1 ). However, growth at a D of 0.1 h −1 resulted in a very short fermentation (18 h) which terminated due to washout (the specific D exceeded the maximum growth rate of the fungal biomass). At a D of 0.04 h −1 , a CDW of 12.3 g/L was achieved without compromising the total residence time (184 h) of the fermentation. While the D of 0.04 h −1 and 0.07 h −1 achieved comparable volumetric productivities (0.5 g/L/h), the total duration of the fermentation at D of 0.07 h −1 was only 85 h. The highest glucan content of cells (27.8 as percentage of CDW) was obtained at a D of 0.07 h −1 , while the lowest glucan content was observed in T. versicolor cells grown at a D of 0.02 h −1 .

A novel electrolytic gas lift reactor for efficient microbial electrosynthesis of acetate from carbon dioxide

Abstract:The low current density impedes the practical application of microbial electrosynthesis for CO 2 fixation. Engineering the reactor design is an effective way to increase the current density, especially for H 2 -mediated microbial electrosynthesis reactors. The electrolytic bubble column microbial electrosynthesis reactor has shown great potential for scaling up, but the mixing and gas mass transfer still need to be enhanced to further increase the current density. Here, we introduced an inner draft tube to the bubble column to tackle the problem. The addition of draft tube resulted in a 76.6% increase in the volumetric mass transfer coefficient ( k La ) of H 2 , a 40% increase in the maximum current density (337 A/m 2 ) and a 72% increase in average acetate production rate (3.1 g/L/d). The computational fluid dynamics simulations showed that the addition of draft tube enhanced mixing efficiency by enabling a more ordered cyclic flow pattern and a more uniform gas/liquid distribution. These results indicate that the electro-bubble column reactor with draft tube holds great potential for industrial implementation.

Use of Pressurized and Airlift Bioreactors for Citric Acid Production by Yarrowia lipolytica from Crude Glycerol

Abstract:Citric acid production is generally carried out in an aqueous medium in stirred tank reactors (STR), where the solubility of oxygen is low and the oxygen demand of microbial cultures is high. Thus, for this bioprocess, providing adequate oxygen mass transfer rate (OTR) from the gas phase into the aqueous culture medium is the main challenge of bioreactor selection and operation. In this study, citric acid production by Yarrowia lipolytica W29 from crude glycerol, in batch cultures, was performed in two non-conventional bioreactors normally associated with high mass transfer efficiency: a pressurized STR and an airlift bioreactor. Increased OTR was obtained by raising the total air pressure in the pressurized STR and by increasing the aeration rate in the airlift bioreactor. An improvement of 40% in maximum citric acid titer was obtained by raising the air pressure from 1 bar to 2 bar, whereas, in the airlift bioreactor, a 30% improvement was attained by increasing the aeration rate from 1 vvm to 1.5 vvm. Both bioreactor types can be successfully applied for the citric acid production process using alternative ways of improving OTR than increasing mechanical stirring power input, thus leading to important operating saving costs.

Abstract:In order to improve glycerol yield without extra i nv estment and complicated operation, an airlift reversible loop reactor (ARLR) with periodic oscillating airlift operation, in which the airflow could be alternated between one nozzle in the central region and eight nozzles in the annular region at the bottom of the reactor, was investigated during glycerol pr oduction by osmotolerant yeast Candida krusei. It was found that glycerol yi eld could be greatly improved with a periodic oscillating airlift operation, com p ared with that with a steady-state central airlift operation, but the airlift m ode had no significant effect on glucose consumption rate. Abstract:Haematococcus pluvialis is traditionally cultivated in a suspension for astaxanthin production. This study presents the novel cultivation approach by immobilized H. pluvialis in bacterial cellulose produced from the symbiosis of Gluconacetobacter xylinus and H. pluvialis . It was observed that the immobilization itself was a regulator to increase the astaxanthin content. The key genes associated to astaxanthin synthesis, such as psy, lcy, bkt, chy, were significantly up-regulated after immobilization. BC immobilized gel can be utilized concurrently with different technologies to improve astaxanthin accumulation (e.g., amount of induction medium, area of biogel, et al). A small-scale screen panel photobioreactor was design to explore the application of the cultivation approach. Compared to suspended culture, the induction time was shortened from 7 days to 3 days. Astaxanthin productivity of red stage reached to 343.2 mg m -2 d -1 . This was greater than that of many other cultivation systems.
Insights into Agitated Bacterial Cellulose Production with Microbial Consortia and Agro-Industrial Wastes

Abstract:Bacterial cellulose (BC) is emerging as an attractive large-scale polymer due to its superior properties. The dominant static culture for BC fermentation by bacteria or microbial consortium results in low productivity. Agitated culture, as an industrially projected technique, has been widely investigated but exclusively for cellulose-producing bacterial strains. Addressing this concern and evaluating the potential of residues as feedstock, this study highlights the utilization of microbial consortium BA2 and seven agro-industrial wastes including cocoa husks, sugarcane bagasse and others. Remarkably, rice bran (RB) appears as a promising substrate, achieving 2.14 g/L (dry basis) and outperforming the traditional HS medium, evident from a 15-day fermentation. A complex interplay between oxygen availability, glucose consumption and BC yield was revealed; while orbital and magnetic stirring with forced air ventilation (AFV) showed low BC yields and early biomass saturation, 4.07 g/L (dry basis) was targeted by magnetic stirring at 100 rpm from the start using only headspace air. However, beyond controlled operating conditions, mechanical agitation and favorable cellulose adhesion to metal in the stirred tank bioreactor negatively affect BC yield. This pattern uncovers the need for a further approach to the design of bioreactors when the microbial consortium is considered.

Abstract:In this study, a cost‐effective complex culture media containing molasses and corn steep liquor (CSL) was developed for the high production of bacterial cellulose (BC) by investigating the effect of four effective factors on BC production at three levels using Taguchi and combined methods. The predicted and actual values of BC production in optimal conditions by Taguchi and combined methods were 8.41 and 14.52 g/L, respectively. These results showed that the combined method was more suitable for predicting the optimal conditions in the optimization of BC production, the cost of developed culture medium was around 94% cost of HS medium preparation, molasses was the most effective factor in both experimental design methods, and initial pH adjustment had little impact on BC production. Then, the effect of inoculation conditions containing three factors of inoculation age, ethanol addition time, and agitation rate on the increase of BC production at three levels was investigated using the response surface methodology with the Box–Behnken design algorithm. Under the optimal conditions including inoculum age of 3 days, ethanol addition time of 10 days, and stirring speed of 100 rpm, the predicted and experimental results of BC production were 21.61 and 20.21 g/L, respectively. This is among the highest ever reported for BC production, which was achieved with a more cost‐effective culture medium containing molasses and CSL.
