Abstract:Functionalized Nα-Fmoc-Ne-(γ-oxobutanoic acid)-L-lysine benzyl ester(4) was synthesized by benzyl esterification of carboxyl group,Boc deprotection of e-amino group and modification of e-amino group with succinic anhydride from Fmoc-Lys(Boc)-OH(1).Finally site-directed mPEGylation of the main chain and side chain of lysine were realized successfully by the coupling of 1 and 4 with monomethoxy-polyethylene glycol(mPEG) aided by DIC/HOBt,respectively.Ne-Boc-Nα-Fmoc-L-lysine polyethylene glycol ester(6) and Nα-Fmoc-Ne-(polyethylene glycol γ-oxobutyrate)-L-lysine benzyl ester(5) were firstly synthesized in the total yield of 93.7% and 52.6%,respectively.The structures of 5 and 6 were characterized by 1H NMR,IR and MS.

Abstract:Novel oligodeoxynucleotide analogues consisting of deoxyisonucleosides were obtained by the phosphoramidite approach on automated DNA-synthesizer. The phosphoramidite building blocks were synthesized by phosphorylation of corresponding isonucleosides. The oligodeoxynucleotide analogues A, B and C were evaluated with respect to hybridization properties and enzymatic stabilities. The oligomers are all stable towards snake venom phosphodiesterase, but only oligomer B and C displayed acceptable hybridization properties to complementary dA(14).

Abstract:A concise preparation of N(alpha)-Fmoc-N(epsilon)-(Boc, methyl)-lysine and its application in the synthesis of site-specifically lysine monomethylated peptide is described. N (alpha)-Fmoc-N(epsilon)-(Boc, methyl)-lysine is obtained, via consecutive reductive benzylation and reductive methylation in a one-pot reaction, followed by debenzylation through catalytic hydrogenolysis and Boc protection in another one-pot reaction. A peptide containing monomethylated lysine is successfully synthesized by incorporating N(alpha)-Fmoc-N(epsilon)-(Boc, methyl)-lysine as a building block via solid-phase peptide synthesis.

Abstract:In this article, O-diisoproppyloxyphosphoryl-L-serine -threonine and -tyrosine were designed and synthesized. Among them, O-diisopropyloxyphosphoryl-L-serine or -threonine was obtained by the migration of phosphoryl group from nitrogen to oxygen of the corresponding N-diisopropyloxyphosphoryl-L-serine or -threonine under ultrasonic irradiation. N-Diisoproppyloxyphosphoryl-L-serine or -threonine was successfully prepared from diisopropyl phosphite and serine or threonine in sodium hypochlorite aqueous media in good yield. O-Diisopropyloxyphosphoryl-L-tyrosine was synthesized via reaction of L-tyrosine cupric complex with diisopropyl phosphorochloridate, then unprotection by sulfide hydrogen.

Abstract:A direct method for the preparation of phosphonamidate- and phosphonate-linked phosphonopeptides has been developed. Using this method, both phosphonopeptides were prepared in acceptable yields directly from simple and commercially available chemicals in one-pot reactions of benzyl carbamate, aldehydes, and methyl dichlorophosphite, followed by aminolysis with amino acid esters or alcoholysis with hydroxy esters.

Abstract:With the development of synthetic biology, the demand for high-quality long oligonucleotides is increasing. The use of dinucleotide phosphoramidites (dimer amidites) will reduce half of the steps required in the stepwise monomer condensation, which is expected to lead to the high-quality synthesis of oligonucleotides. Herein, we report a convenient, unified procedure for the sustainable preparation of fully protected dimer amidites bearing a methyl phosphate and a β-cyanoethyl phosphoramidite. Key precursors of those amidites—dimers containing a 3′-hydroxyl group—have been prepared in high yield using a one-pot, scalable approach. The structures of all 16 dimer amidites and corresponding 3ʹ-hydroxyl dimers were confirmed by HPLC-MS and 31P NMR spectra. With these dimer amidites, modification of the standard method for automated solid-phase oligonucleotide synthesis enables high-yield block couplings. Oligonucleotides synthesized following this method are of sufficient quality to assemble gene-length DNA fragments. As a consequence, we develop a practical dinucleotide phosphoramidite chemistry for de novo DNA synthesis.

Abstract:Two novel monophosphoramidites were synthesized through a five-step transformation from commercially available L-proline. In the Rh-catalyzed asymmetric hydrogenation of alpha-dehydroamino acid derivatives, ligand (S-C,R-a)-lb showed good enantioselectivity and up to 91% e.e. was obtained.

Abstract:Novel oligonucleotide analogues built from isonucleosides were synthesized by the phosphoramidite approach on an automated DNA synthesizer. The phosphoramidite building blocks were synthesized by phosphitylation of the corresponding protected isonucleosides. The oligonucleotide analogues C - G containing the isonucleoside 1-3 were studied with respect to their hybridization properties and enzymatic stability. The oligomers bearing an isonucleoside at the end of the strands all proved stable towards snake-venom phosphodiesterase, but only the oligomers D-G exhibit acceptable duplex stability when hybridized with complementary d(A(14)).

Abstract:We synthesized a lysine-linked auxiliary-derived building block, epsilon-N-2-mercapto-1-(2,4-dimethoxyphenyl)ethyl lysine, in 20 % isolated yield over a six-step route. The new building block is well compatible with Fmoc solid-phase peptide synthesis (SPPS) and hydrazide-based native chemical ligation (NCL). Furthermore, we successfully incorporated the new lysine-linked auxiliary handle into a protein segment and performed the chemical synthesis of K48-linked diubiquitin with stepwise NCL.

Abstract:The group-assisted purification asymmetric synthesis of alpha,beta-diamino acid derivatives was achieved by treating chiral N-phosphonyl imines with a Ni-II-complexed glycine-derived enolate; traditional purification techniques such as chromatography and recrystallization are not required. Successful control of the syn stereochemistry of the vicinal diamino products complements our previous methods that afforded anti stereoisomers; thus, all four individual isomers can be synthesized simply by changing the geometry of the enolate. In contrast to our previous synthesis in which at least 5 equiv. of the glycine Schiff base enolate was required for complete conversion, the new synthesis only needs 1.1 equiv. of the glycine Schiff base enolate to give complete diastereoselectivity (>30:1dr) and high yields (91-97%). The absolute stereochemistry was unambiguously determined by X-ray structure analysis.

Abstract:A simple and efficient method for the synthesis of gamma,delta-alkynyl-beta-amino acid derivatives by a copper-catalyzed three-component amine-alkyne-alkyne addition reaction was developed. Various gamma,delta-alkynyl-beta-amino acid derivatives were synthesized in moderate to good yields in one step. With chiral prolinol derivatives employed as the amine component, excellent diastereoselectivities (up to >99:1 diastereomeric ratio (dr)) were obtained. The scope of the reaction and further transformations of the resulting amino acid derivatives, such as deprotection and cyclization are also described.

Abstract:It was found that nucleoside H-phosphonothioate, H-phosphonodithioate, and H-phosphonoselenoate monoesters in the presence of iodine underwent rapid oxidative esterification with various hydroxylic components to produce in "one pot" reactions the corresponding nucleoside phosphorothioate, nucleoside phosphorodithioate and nucleoside phosphoroselenoate diesters in good yields. The transformation represents a novel alternative method for the synthesis of internucleotide phosphate bridge, bearing sulfur or selenium atoms. Mechanistic studies have revealed metaphosphate analogues as the probable intermediates, involved in the reactions.

Abstract:Unsymmetrical N-benzyloxycarbonyl-protected 1-amino-1-arylalkylphosphonate mixed diesters were synthesized using a one-pot reaction involving benzyl carbamate, aromatic aldehydes and alkoxydichlorophosphine, followed by treatment with alcohols in the presence of triethylamine. The reactions were followed by 31P NMR and a mechanism is proposed.

Abstract:Reaction of nucleoside triphosphates (NTPs) with amines in pyridine mediated by trimethylsilyl chloride produced nucleoside 5'-phosphoramidates in moderate yields without any preprotection of nucleosides and amino acid methyl esters. The reaction pathway is very similar to the mechanism of the RNA capping reaction, DNA or RNA ligation reaction, and catalysis of hydrolases and nucleases involving the formation of covalent enzyme-NMP (nucleoside 5'-monophosphate) intermediates in biological systems, which could provide a valuable clue for the enzymatic reactions.

Abstract:Coupling of N-phosphoamino acids 2 and amino acid methyl esters by dichlorotriphenylphosphorane 3 produced N-phosphopeptide methyl esters 5, which were sequentially hydrolyzed in triethylamine or dilute NaOH aqueous solution. The target products N-phosphodipeptides 6 were obtained in reasonable yields and high purity after isolation using extraction method. The convenient and efficient approach could be generally used for synthesis of polypeptides.

Abstract:Facile, mild and efficient one-pot preparation of N-alpha-Fmoc-N-epsilon-(benzyl, methyl)-lysine, a building block for monomethylated peptide synthesis, was described. This building block was proved to be efficient for the synthesis of site-specifically monomethylated peptide.
