Abstract:Molecular recognition capabilities of a novel nucleolipid amphiphile, octadecanoyl ester of 1-(2 carboxyethyl) adenine, to the complementary nucleobases at the air/water interface were investigated by surface pressure-area (pi-A) isotherms and UV spectra measurements. All of the observations show that molecular recognition through complementary base pairing takes place at the air/water interface between the adenine moiety in the headgroup of the nucleolipid amphiphile and the dissolved complementary nucleobase substrates in the subphase. On the surface of pure water, the monolayer gave a limiting molecular area of 29.3 Angstrom(2) and collapse pressure at about 62 mN/m while on the subphase of aqueous 5 mM thymidine and uridine solution, the limiting molecular area and collapse pressure are 42.0 Angstrom(2) and 77 mN/m, 39.7 Angstrom(2) and 79 mN/m, respectively. In the Langmuir-Blodgett (LB) matrix, the constituent molecules are arranged regularly, which facilitates the photodimerization of the thymine moieties and uracil moieties that transferred into solid substrates along with the surface monolayer of the nucleolipid amphiphile as a result of the formation of the base-base complex, the photodimerization accomplished in 6 and 4 h for the thymine and uracil rings in the LB films under irradiation of a 254-nm ultraviolet light at room temperature, respectively. Novel Nucleolipids of Pyrimidine β-D-Ribonucleosides: Combinatorial Synthesis, Spectroscopic Characterization, and Cytostatic/Cytotoxic Activities

Abstract:Four series of nucleolipids with either uridine, 5-methyluridine, 5-fluorouridine, and 6-azauridine as β-D-ribonucleoside component have been prepared in a combinatorial (not parallel!) manner (see Formulae). All compounds have been characterized by elemental analyses, ESI mass spectrometry as well as by (1) H-, and (13) C-NMR, and UV spectroscopy. A selection of eight nucleolipids with different lipophilizing moieties, based on earlier findings, as well as of 5-fluorouridine as control were first tested on their cytotoxic effect towards PMA-differentiated human THP-1 macrophages. Those compounds which did not exhibit a significant inhibitory effect on the survival of the macrophages were next tested on their cytostatic/cytotoxic effect towards the human astrocytoma/oligodendroglioma GOS-3 cells as well as against the rat malignant neuroectodermal BT4Ca cell line. Additionally, induction of apoptosis of the cell lines was evaluated. It turned out that particularly a combined lipophilization of the nucleosides by an 2',3'-O-ethyl levulinate residue plus a farnesyl moiety at N(3) of the pyrimidine moiety of the corresponding nucleolipids leads to an active compound with the highest probability.

Abstract:Liposomes and other nanoparticles have been widely studied as innovative nanomaterials because of their unique properties. Pyridinium salts, on the basis of 1,4-dihydropyridine (1,4-DHP) core, have gained significant attention due to their self-assembling properties and DNA delivery activity. This study aimed to synthesize and characterize original N-benzyl substituted 1,4-dihydropyridines and evaluate the influence on structure modifications on compound physicochemical and self-assembling properties. Studies of monolayers composed of 1,4-DHP amphiphiles revealed that the mean molecular areas values were dependent on the compound structure. Therefore, the introduction of N-benzyl substituent to the 1,4-DHP ring enlarged the mean molecular area by almost half. All nanoparticle samples obtained by ethanol injection method possessed positive surface charge and average diameter of 395–2570 nm. The structure of the cationic head-group affects the size of the formed nanoparticles. The diameter of lipoplexes formed by 1,4-DHP amphiphiles and mRNA at nitrogen/phosphate (N/P) charge ratios of 1, 2, and 5 were in the range of 139–2959 nm and were related to the structure of compound and N/P charge ratio. The preliminary results indicated that more prospective combination are the lipoplexes formed by pyridinium moieties containing N-unsubstituted 1,4-DHP amphiphile 1 and pyridinium or substituted pyridinium moieties containing N-benzyl 1,4-DHP amphiphiles 5a–c at N/P charge ratio of 5, which would be good candidates for potential application in gene therapy.

Abstract:Oligonucleotides that contain up to three aminopropyl nucleoside analogues have been synthesized. Dimers of aminopropyl adenine and thymidine were prepared and used as building blocks by applying phosphoramidite chemistry. Both R and S isomers of the aminopropyl nucleosides were used. This incorporation led to a reduction of thermal stability of double-stranded DNA. Furthermore, the (R)-adenine analogue, which yielded (S)APNA, can be considered as a candidate for universal base pairing.

Abstract:We report a general solid-phase synthesis strategy for a novel class of DNA block copolymer amphiphiles. The assembled spherical micelle structures exhibit recognition properties defined by their DNA sequences and can be used to build higher-ordered structures through hybridization with materials that possess complementary DNA. This work is an important step toward creating DNA modified structures that can act as phase transfer agents and carriers of nanovesicles.

Abstract:Fourier transform infrared (FTIR) transmission, polarized transmission and attenuated total reflection (ATR) spectroscopy were employed to investigate the molecular recognition capabilities of a novel nucleolipid amphiphile, octadecanoyl ester of 1-(2-carboxyethyl) adenine, to the complementary nucleobases, thymidine and uridine, in the Langmuir-Blodgett film matrix. A broad band observed in the 3500-3000 cm(-1) region indicated the formation of the multiple hydrogen bonding between the adenine moiety in the headgroup of the nucleolipid amphiphile and the corresponding complementary nucleobase in the subphase was observed, which verified the molecular recognition mechanism suggested in the previous paper. In the thymidine-containing LB films of the nucleolipid amphiphile, the molecules are biaxially oriented, while in the uridine-containing LB films, the molecules are uniaxially oriented; similar to the LB films transferred from pure water, the corresponding anisotropy and isotropy were detected via FTIR polarized transmission spectroscopy. This is due to the steric effect caused by the methyl of the thymine ring. The order-disorder transitions of the LB films deposited from various subphases were investigated by using FTIR transmission spectroscopy. The results show that the existence of the complementary nucleobases in the LB films has little influence on the arrangement of the hydrocarbon chains of the nucleolipid amphiphile. The no-complementary base-containing and complementary base-containing LB films of the nucleolipid amphiphile all display high thermal stability. (C) 1998 Elsevier Science S.A. All rights reserved.

Abstract:Despite of great advances of phospholipids and liposomes in clinical therapy, very limited success has been achieved in the preparation of smart phospholipids and controlled-release liposomes for in vivo drug delivery and clinical trials. Here we report a supramolecular approach to synthesize novel supramolecularly engineered phospholipids based on complementary hydrogen bonding of nucleosides, which greatly reduces the need of tedious chemical synthesis, including reducing the strict requirements for multistep chemical reactions, and the purification of the intermediates and the amount of waste generated relative more traditional approaches. These upgraded phospholipids self-assemble into liposome-like bilayer structures in aqueous solution, exhibiting fast stimuli-responsive ability due to the hydrogen bonding connection. In vitro and in vivo evaluations show the resulted supramolecular liposomes from nucleoside phospholipids could effectively transport drug into tumor tissue, rapidly enter tumor cells, and controllably release their payload in response to an intracellular acidic environment, thus resulting in a much higher antitumor activity than conventional liposomes. The present supramolecularly engineered phospholipids represent an important evolution in comparison to conventional covalent-bonded phospholipid systems.

Abstract:Lipid derivatives of nucleoside analogs have been highlighted for their potential for effective gene delivery. A novel class of nucleobase-lipids are rationally designed and readily synthesized, comprising thymine/cytosine, an ester/amide linker and an oleyl lipid. The diversity of four nucleobase-lipids termed DXBAs (DOTA, DNTA, DOCA and DNCA) is investigated. Besides, DNCA is demonstrated to be an effective neutral transfection material for nucleic acid delivery, which enbles to bind to oligonucleotides via H-bonding and π-π stacking with reduced toxicity in vitro and in vivo. Several kinds of nucleic acid drugs including aptamer, ssRNA, antisense oligonucleotide, and plasmid DNAs can be delivered by DXBAs, especially DNCA. In particular, G4-aptamer AS1411 encapsulated by DNCA exhibits cellular uptake enhancement, lysosome degradation reduction, cell apoptosis promotion, cell cycle phase alteration in vitro and duration prolongation in vivo, resulting in significant anti-proliferative activity. Our results demonstrate that DNCA is a promising transfection agent for G4-aptamers and exhibites bright application prospects in the permeation improvement of single-stranded oligonucleotides or plasmid DNAs.

Abstract:All living organisms synthesize phospholipids as the primary constituent of their cell membranes. Enzymatic synthesis of diacylphospholipids requires preexisting membrane-embedded enzymes. This limitation has led to models of early life in which the first cells used simpler types of membrane building blocks and has hampered integration of phospholipid synthesis into artificial cells. Here we demonstrate an enzyme-free synthesis of natural diacylphospholipids by transacylation in water, which is enabled by a combination of ion pairing and self-assembly between lysophospholipids and acyl donors. A variety of membrane-forming cellular phospholipids have been obtained in high yields. Membrane formation takes place in water from natural alkaline sources such as soda lakes and hydrothermal oceanic vents. When formed vesicles are transferred to more acidic solutions, electrochemical proton gradients are spontaneously established and maintained. This high-yielding non-enzymatic synthesis of natural phospholipids in water opens up new routes for lipid synthesis in artificial cells and sheds light on the origin and evolution of cellular membranes.

Abstract:Monolayer behavior of a nucleolipid amphiphile, (3',5'-distearoyl)-2'-deoxythymidine, on pure water, aqueous 1 mM and 5 mM adenosine solutions was investigated by means of surface pressure-molecular area (pi-A) isotherms, which indicate that both hydrogen-bonding and stacking interactions are present between the (3',5'-distearoyl)-2'-deoxythymidine monolayer and adenosine in subphase, and the recognition of this amphiphile to adenosine is more complete in higher subphase concentration. The ultraviolet-visible (UV-vis), Fourier transform infrared (FTIR)transmission FTIR-attenuated total reflection (ATR), and Fourier transform surface-enhanced Raman scattering (FT-SERS) spectroscopic results indicate that the adenosine molecules in the subphase can be transferred onto solid substrates by Langmuir-Blodgett (LB) technique as a result of the formation of Watson-Crick base-pairing at the air/water interface. The regular and closed packing of the constituent molecules facilitates the photodimerization of the thymine moieties in the headgroup under ultraviolet irradiation even at room temperature. FT-SERS technique, for the first time, was introduced into the research area of molecular recognition occurring at an interface system. High-quality FT-SERS spectra of single LB monolayers obtained in the 1750-500 cm(-1) region, together with the FTIR-ATR results in the NH-stretching high-frequency region, provide direct evidences for the formation of multiple complementary hydrogen bonds between the base pairs.

Abstract:A new synthesis of the tetrasubstituted tetrahydrofuran fragment of the marine secondary metabolites amphidinolides X and Y is described. The oxygenated chiral quaternary carbon was assembled by asymmetric dihydroxylation in high enantioselectivity and the tetrahydrofuran ring was constructed by an acid-catalyzed 5-endo ring-opening cyclization of the epoxide possessing a vinyl moiety.

Abstract:A new single-chain nuclei acid base-containing amphiphile, octadecanyl ester of 1-(2-carboxyethyl)thymine, was synthesized, and its surface behaviors on pure water and on aqueous solutions of adenine were investigated by the method of surface pressure-area (π-A) isotherms. Molecular recognition through complementary base pairing takes place at the air/water interface between the thymine moiety in the headgroup of the amphiphile and adenine, which results in the pick-up of the base-base complex onto quartz plate by the Langmuir-Blodgett (LB) technique. Photodimerization of thymine rings was also investigated in the multilayer LB film of the amphiphile deposited from pure water surface, and the results show that the orderly and regular packing of the molecules in the LB matrix provides favorable conditions for the formation of thymine dimers.

Abstract:Dipeptide-conjugated nucleosides were efficiently synthesized from the intermediates of 3'-amino-3'-deoxy-nucleosides by using the solid-phase synthetic strategy with HOBt/HBTU (1-hydroxy-1H-benzotriazole/2-(1H-benzotriazol-1-yl)-1,1,3,3-tetramethyluronium hexafluoroborate) as the coupling reagents (Schemes 1-3). CD Spectra and thermal melting studies showed that the synthesized hydrophobic dipeptide-thymidine and -uridine derivatives 8a-8d. 13a-d, and 18 had a mild affinity with the polyA (.) polyU duplex and could induce the change of RNA conformation. The results also implied that the interaction of conjugates with RNA might be related to the sugar pucker conformation of the nucleoside.
