Abstract:A new ent-kaurane diterpenoid, 6β,7β,13α-trihydroxy-1α-acetoxy-7α,20-epoxy- ent-kaur-16-en-15-one (1) and five known compounds, epinodosinol (2), isodocarpin (3), nodosin (4), enmedol (5), nervosanin B (6), were isolated from the aerial part of Isodon nervosus. The structure of the new compound was determined by spectral methods (1D-,2D-NMR and MS). Four of the compounds were tested for their cytotoxicity towards HL60, SMMC-7721 and Hela cells.

Abstract:Two new C(20)-oxygenated ent-kaurene diterpenoids, taibaihenryiins A (1) and B (2), along with five known compounds, shikokianin (3), longikaurin D, 2 alpha -hydroxyursolic acid, longikaurin F, and lasiodin, were isolated from the EtOH extract of the leaves and tender branches of Isodon henryi (HEMSL.) KUDO. The structures of the new compounds were determined as 11 alpha -acetoxy-7,20-epoxy-1 alpha ,6 beta ,7 beta -trihydroxy-ent-kaur-16-en-15-one (1) and 7,20-epoxy-3 beta ,6 beta ,7 beta ,11 alpha -tetrahydroxy-ent-kaur-16-en-15-one (2) on the basis of detailed, spectroscopic analyses.

Abstract:Further investigation on the aerial parts of Isodon enanderianus afforded a novel asymmetric ent-kauranoid dimer, enanderinanin J (1). The structure of the dimer was elucidated by mews of spectroscopic methods (Including 2D NMR techniques). Enanderinanin J was a dimer of xerophilusin A and probably formed by [4 + 2] cycloaddition.

Abstract:Two new ent-kaurane diterpenoids, 16( S)-hydroxymethyl-1alpha, 6beta, 7beta, 11beta-tetrahydnoxy-7a, 20-epoxyent-kauran-15-one (1) and 16 ( R)-hydroxymethyl-1alpha, 6beta, 7beta, 14beta-tetrahydroxy-7alpha, 20-epoxy-ent-kauran-15-one (2), named rubescensins F (1) and G (2), respectively, were isolated from the EtOAc extract of the dried leaves of Isodon rubescens ( Hemsl.) Ham collected in Jiyuan Prefecture, Henan Province, together with fourteen known compounds lasiodonin (3), oridonin (4), ponicidin, rubescensins C and D, rabdoternin C, wikstroemioidin B, enmenol-1alpha-O-beta-D-glucoside, enmenol, pedalitin, salicylic acid, ursolic acid, beta-sitosterol and daucosterol. Their structures were determined on the basis of spectral data.

Abstract:Two novel ent-kauranoids with new sub-skeleton types, gesneroidins G and H were isolated as white powder from the ether extract of the leaves of Isodon gesneroides through normal phase column chromatography. Their structures were elucidated as 3beta,6alpha,7beta-triacetoxyl-15-hydroxyl-14-oxo-ent-15,16-seco-kaur-11,17-olide and 1alpha-hydroxyl-3beta,6alpha,7beta,11beta-tetraacetoxylent-nor-15,17-kaur-8,16-olide on the basis of the spectral evidences including ID and 2D NMR spectra.

Abstract:From the plants of the genus Isodon (Labiatae), many deterpenoids having ent-kaurene, 7,20-epoxy-ent-kaurene, 6,7-seco-ent-kaurene, 8,9-seco-ent-kaurene, 6-epi-ent-gibberelene, ent-isopimanthrene and abietene skeletons have been isolated and characterized.1These substances have shown various biological activities such as antibacterial, anitumor and cytotoxic effects2-3 etc.In a previous study of the chemical constituents of Isodon, we reported on the isolation of two new ent-kaurene diterpenoids, taibairubescensins A and B.4 During the course of our systematic search for biologically active substances from plants of the genus Isodon, recently, we examined diterpenoid constituents of Isodon Japonica (Burm.f.) Hara (Labiatae) collected from Taibai Mountain, China, and isolated a new diterpenoid, named taibaijaponicain E (1), along with four known compounds, norhendosin A (2), nodosin (3), rabdophyllin G (4), lasiodin (5).We report here the isolation and structure elucidation of new compound by a combination of NMR techniques, including DEPT, HMBC and NOESY.

Abstract:Nine new ent-kaurane diterpenoids, isoscoparins D-L (1-9), and an artificial product, the acetonide of rabdoloxin A (10), along with 16 known analogues (11-26), were isolated from the leaves of Isodon scoparius. The new structures were determined by ID and 2D NMR spectroscopic analysis. Selected compounds were evaluated for their cytotoxicity against NB4, A549, PC-3, MCF-7, and SH-SY5Y cell lines.

Abstract:Two undescribed ent-kaurene diterpenes, named guidongnins I (1) and J (2), were isolated from the medicinal plant Isodon rubescens. Compound 1 was determined to contain an unprecedented 23 carbons in the skeleton by bearing an extra isopropyl group at C-17 out of the diterpenoid parent structure, and compound 2 was the first example of 6,7-seco-7,20-olide-ent-kaurenes with two fused-tetrahydrofuran rings formed between C-6 and C-19/C-20 through oxygen bridges. Their structures, including their absolute configurations, were determined using the analyses of the spectroscopic and X-ray diffraction data. Guidongnins I (1) and J (2) were assessed for their anti-cancer activities against the growth of various cancer cell lines, and 2 displayed cytotoxic potency against HepG2 at IC50 27.14 ± 3.43 μM.

Abstract:A new ent -kaurane type diterpenoid glycoside, 17-O-β-D-glucopyranosyl-16α- ent -kauran-19-oic acid ( 1 ), together with 17-hydroxy-16α- ent -kauran-19-oic acid ( 2 ), 16α,17-dihydroxyl- ent -kauran-19-oic acid ( 3 ), and 16α-hydroxy-17-acetoxy- ent -kauran-19-oic acid ( 4 ) were isolated from the aerial parts of Inula japonica Thunb. The structure of 1 was determined mainly by use of 1D and 2D NMR spectroscopic techniques including HSQC, 1 H- 1 H COSY, HMBC, and NOESY. In addition, 4 exhibited significant inhibitory activity on NO production in LPS-stimulated RAW264.7 cells with IC 50 value of 14.3 µg/mL.
