Abstract:A new kaurenoid, taibairubescensin C (1), was isolated from the ethanol extract of the leaves and tender branches of Isodon rubescens (Hemsl.) Hara. Its structure was designated as 2beta,6alpha-diacetoxy-3beta, 11beta-dihydroxy-ent-kaur-16-en-15-one (1) on the basis of detailed spectroscopic analysis.

Abstract:From the plants of the genus Isodon (Labiatae), many deterpenoids having ent-kaurene, 7,20-epoxy-ent-kaurene, 6,7-seco-ent-kaurene, 8,9-seco-ent-kaurene, 6-epi-ent-gibberelene, ent-isopimanthrene and abietene skeletons have been isolated and characterized.1These substances have shown various biological activities such as antibacterial, anitumor and cytotoxic effects2-3 etc.In a previous study of the chemical constituents of Isodon, we reported on the isolation of two new ent-kaurene diterpenoids, taibairubescensins A and B.4 During the course of our systematic search for biologically active substances from plants of the genus Isodon, recently, we examined diterpenoid constituents of Isodon Japonica (Burm.f.) Hara (Labiatae) collected from Taibai Mountain, China, and isolated a new diterpenoid, named taibaijaponicain E (1), along with four known compounds, norhendosin A (2), nodosin (3), rabdophyllin G (4), lasiodin (5).We report here the isolation and structure elucidation of new compound by a combination of NMR techniques, including DEPT, HMBC and NOESY.

Abstract:Two new C(20)-oxygenated ent-kaurene diterpenoids, taibaihenryiins A (1) and B (2), along with five known compounds, shikokianin (3), longikaurin D, 2 alpha -hydroxyursolic acid, longikaurin F, and lasiodin, were isolated from the EtOH extract of the leaves and tender branches of Isodon henryi (HEMSL.) KUDO. The structures of the new compounds were determined as 11 alpha -acetoxy-7,20-epoxy-1 alpha ,6 beta ,7 beta -trihydroxy-ent-kaur-16-en-15-one (1) and 7,20-epoxy-3 beta ,6 beta ,7 beta ,11 alpha -tetrahydroxy-ent-kaur-16-en-15-one (2) on the basis of detailed, spectroscopic analyses.

Abstract:Two undescribed ent-kaurene diterpenes, named guidongnins I (1) and J (2), were isolated from the medicinal plant Isodon rubescens. Compound 1 was determined to contain an unprecedented 23 carbons in the skeleton by bearing an extra isopropyl group at C-17 out of the diterpenoid parent structure, and compound 2 was the first example of 6,7-seco-7,20-olide-ent-kaurenes with two fused-tetrahydrofuran rings formed between C-6 and C-19/C-20 through oxygen bridges. Their structures, including their absolute configurations, were determined using the analyses of the spectroscopic and X-ray diffraction data. Guidongnins I (1) and J (2) were assessed for their anti-cancer activities against the growth of various cancer cell lines, and 2 displayed cytotoxic potency against HepG2 at IC50 27.14 ± 3.43 μM.

Abstract:Six new ent-kaurane diterpenoids, lungshengenins B-G (1-6), together with three known diterpenoids, lungshengenin A (7), inflexin (8), and lushanrubescinsin C (9), were isolated from the leaves and tender branches of Isodon lungshengensis. Their structures were elucidated by means of spectroscopy, mainly 1D and 2D NMR techniques. Lungshengenins A (7), C (2), and G (6) were cytotoxic toward K562 cells, having IC(50) values equal to or less than 10 microg/mL.

Abstract:Ternifolide A (1), a new diterpenoid featuring a unique 10-membered lactone ring formed between C-6 and C-15, along with ternifolide B (2), a nor-diterpenoid, and ternifolide C (3) were isolated from the leaves of Isodon ternifolius. Both H-8 and H-9 being alpha-orientations in compound 1 were found for the first time. The absolute configurations of 1 and 3 were confirmed by X-ray diffraction study. Compounds 1 and 3 were evaluated for their cytotoxicity.

Compounds 3, 4, 6, 8, and 10 showed significant inhibitory effect toward K562 with IC(50) values ranging from 3.2 microg/ml to 8.2 microg/ml, while 3 and 6 exhibited potent antitumor activity against T24, but none exhibited cytotoxicity toward the cells of A549.

Abstract:A new ent-kauranoid named laxiflorin D (1) was isolated from the leaves of Isodon eriocalyx var. laxiflora, whose structure was determined by the spectroscopic methods. Six known ent-kaurane diterpenoids, laxiflorin A (2), maoecrystals A-C (3～5), Q (6) and eriocalyxin B (7), together with cirsimaritin (8) and 2α-hydroxyursolic acid (9), were also isolated.

Abstract:Two new ent-kaurane-derived diterpene derivatives, phyllostacins A (1) and B (2), were isolated from the aerial parts of Isodon phyllostachys, together with two known compounds, irroratin A (3) and serrin B (4). Both 1 and 2 were found to be present as diastereoisomers. In the case of 1, the corresponding diastereoisomeric diacetates 5 and 6 were prepared and separated. The structures of the new compounds were elucidated by extensive 1D- and 2D-NMR spectroscopic methods, in combination with MS experiments. In (D-5)pyridine solution, the two epimers of 1 are present in equal amounts, but in CDCl3 or CD3OD, the (S)-epimer predominates in the mixture of hemiacetals.

Abstract:Twelve new diterpenoids, isoresbins A-L (1-12), together with twenty-eight known ones, were isolated from the aerial parts of Isodon oresbius. Their diverse structures included 6,7-seco-ent-kaurane, 7,20-epoxy-ent-kaurane, 6,7:8,15-diseco-ent-kaurane, and abietanes skeletons, which were elucidated by spectroscopic data interpretation, single-crystal X-ray diffraction, and quantum chemical calculation. Isoresbins A (1) and B (2) possessed a new rearranged 15(8 → 11)-abeo-6,7-seco-ent-kaurane skeleton. 1 and 5 promoted lysosomal function, which was evaluated by LysoTracker Red staining and DQ-ovalbumin dequenching assay. 1 showed cytotoxicity against six human tumor cell lines with IC<sub>50</sub> values in 2.07-4.04 μM range. Moreover, 1 induced damage of mitochondrial membrane potential, G2/M cell cycle arrest and apoptosis in SW480 cells.
