Abstract:OBJECTIVE:To study the chemical constituents of aerial part of Rabdosia serra.METHOD:The compounds were isolated by extraction, coloum chromatography over silica gel and ODS, and preparative HPLC. Their structures were identified by various spectroscopic methods including MS, IR, 1D and 2D NMR data.RESULT:Six compounds were isolated from R. serra and were characterized as ent-1alpha, 7alpha, 14beta, 20-tetrahydroxy-11, 16-kauradien-15-one (1), kamebakaurin (2), dihydrokamebakaurin (3), rabdoinflexin A (4), daucosterol( 5), and beta-sitosterol (6).CONCLUSION:Compound 1 is a new compound, and coumpound 3 was obtained from this plant for the first time.

Abstract:Five new ent-pimarane diterpenoids ent-16-nor-2-oxopimar-8(14)-ene-15,19-dial (1), ent-16-nor-2α,19-dihydroxypimar-8-en-15-al (2), 3-O-acetyldarutigenol (3), 19-O-acetylkirenol (4), ent-16-nor-3β,15-dihydroxypimar-8(14)-ene (5) were isolated and characterized from the ethanol extract of Sigesbeckia pubescens. Their structures were elucidated on the basis of spectroscopic analysis. The absolute configuration of C-15 in compounds 3 and 4 was assigned using Snatzke's method. All these compounds were assessed for their anti-inflammatory potential by measuring the inhibitory effects on NO production in LPS-induced RAW 264.7 macrophage cells and compound 4 showed significantly inhibitory activity with IC50 value of 5.9 μM.

Abstract:Two undescribed ent-kaurene diterpenes, named guidongnins I (1) and J (2), were isolated from the medicinal plant Isodon rubescens. Compound 1 was determined to contain an unprecedented 23 carbons in the skeleton by bearing an extra isopropyl group at C-17 out of the diterpenoid parent structure, and compound 2 was the first example of 6,7-seco-7,20-olide-ent-kaurenes with two fused-tetrahydrofuran rings formed between C-6 and C-19/C-20 through oxygen bridges. Their structures, including their absolute configurations, were determined using the analyses of the spectroscopic and X-ray diffraction data. Guidongnins I (1) and J (2) were assessed for their anti-cancer activities against the growth of various cancer cell lines, and 2 displayed cytotoxic potency against HepG2 at IC50 27.14 ± 3.43 μM.

Abstract:Six new ent-kaurane diterpenoids, lungshengenins B-G (1-6), together with three known diterpenoids, lungshengenin A (7), inflexin (8), and lushanrubescinsin C (9), were isolated from the leaves and tender branches of Isodon lungshengensis. Their structures were elucidated by means of spectroscopy, mainly 1D and 2D NMR techniques. Lungshengenins A (7), C (2), and G (6) were cytotoxic toward K562 cells, having IC(50) values equal to or less than 10 microg/mL.

Abstract:From the plants of the genus Isodon (Labiatae), many deterpenoids having ent-kaurene, 7,20-epoxy-ent-kaurene, 6,7-seco-ent-kaurene, 8,9-seco-ent-kaurene, 6-epi-ent-gibberelene, ent-isopimanthrene and abietene skeletons have been isolated and characterized.1These substances have shown various biological activities such as antibacterial, anitumor and cytotoxic effects2-3 etc.In a previous study of the chemical constituents of Isodon, we reported on the isolation of two new ent-kaurene diterpenoids, taibairubescensins A and B.4 During the course of our systematic search for biologically active substances from plants of the genus Isodon, recently, we examined diterpenoid constituents of Isodon Japonica (Burm.f.) Hara (Labiatae) collected from Taibai Mountain, China, and isolated a new diterpenoid, named taibaijaponicain E (1), along with four known compounds, norhendosin A (2), nodosin (3), rabdophyllin G (4), lasiodin (5).We report here the isolation and structure elucidation of new compound by a combination of NMR techniques, including DEPT, HMBC and NOESY.

Abstract:Eight new diterpenoids (1-8) with varied structures were isolated from the aerial parts of Isodon xerophilus. Among them, xerophilsin A (1) was found to be an unusual meroditerpenoid representing a hybrid of an ent-kauranoid and a long-chain aliphatic ester, xerophilsins B-D (2-4) are dimeric ent-kauranoids, while xerophilsins E-H (5-8) are new ent-kauranoids. The structures of 1-8 were elucidated mainly through the analyses of their spectroscopic data. The absolute configurations of 2, 6, and 8 were confirmed by single-crystal X-ray diffraction, and the configuration of C-16 in 7 was established through quantum chemical calculation of NMR chemical shifts, as well as modeling of key interproton distances. Bioactivity evaluation of all isolated compounds revealed that 2, 3, and 5 inhibited NO production in LPS-stimulated RAW264.7 cells.

Abstract:The unprecedented oxidative dearomatization-induced [5+2] cycloaddition/pinacol-type 1,2-acyl migration cascade efficiently generates a quaternary carbon center and assembles the highly oxygenated bicyclo[3.2.1]octane framework of ent-kaurene diterpenoids. By incorporation of the subsequent retro-aldol/aldol process and singlet oxygen ene reaction, this concise and convergent approach has enabled the first asymmetric total syntheses of pharicin A, pharicinin B, 7-O-acetylpseurata C, and pseurata C.
