Methods and Applications in Fluorescence

Abstract:Fluorescent samples typically emit isotropically in all directions. Large lenses and other optical components are needed to capture a significant fraction of the emission, and complex confocal microscopes are required for high resolution focal-plane imaging. It is known that Bessel beams have remarkable properties of being able to travel over long distances, over 1000 times the wavelength, without diverging, and hence are called non-diffracting beams. In previous reports the Bessel beams were formed by an incident light source, typically with plane-wave illumination on a circular aperture. It was not known if Bessel beams could form from fluorescent light sources. We demonstrate transformation of the emission from fluorescent polystyrene spheres (FPS) into non-diverging beams which propagate up to 130 mm (13 cm) along the optical axis with a constant diameter. This is accomplished using a planar metal film, with no nanoscale features in the X-Y plane, using surface plasmon-coupled emission. Using samples which contain many FPS in the field-of-view, we demonstrate that an independent Bessel beam can be generated from any location on the metal film. The extremely long non-diffracted propagation distances, and self-healing properties of Bessel beams, offer new opportunities in fluorescence sensing and imaging.

Abstract:Dark hollow beams (DHBs) have great potential in the applications of optical manipulation, and the generation of DHBs is still a challenging and rewarding issue. In this paper, we present a beam shaping system for generating DHBs. The proposed system is composed of a freeform lens array and a non-classical room system which has a constant focal length but various image locations. The DHBs with a well-controlled intensity profile generated by the proposed system is not sensitive to the change of the intensity distribution of the incident beam, which allows flexible choices of light sources. Moreover, the annular pattern produced by the DHB remains unchanged when the image plane is moved a long distance of 17mm, and the energy efficiency of the beam shaping system is greater than 90% when Fresnel loss is considered. The proposed beam shaping system endows the generated DHBs with new properties and may have great potential in the field of optical tweezers and atom guides. Abstract:White-light non-diffraction beams such as Airy beam and Bessel beam have potential applications in multispectral imaging and micromanipulation. Generation of white-light Airy beam and Bessel beam with high quality and high efficiency still remains challenging for conventional diffractive or refractive optics which suffers from significant chromatic dispersion. In this paper, both high-quality white-light Airy beam and Bessel beam are generated using a deformable mirror by modulating the incident LED beam with tunable cubic and conical wavefronts. The main lobe of the generated white-light non-diffraction beams does not suffer from chromatic dispersion along the propagation. The results also show that the generation of the white-light Bessel beam has higher requirements for spatial coherence than white-light Airy beams. Our work expands the understanding of the white-light non-diffraction beams and paves the way for the applications.

Abstract:A new mathematical model named hollow Gaussian beams (HGBs) is introduced to describe the dark hollow beam. The area of the dark region across the HGBs can be easily controlled by properly choosing the beam parameters. An analytical propagation formula for HGBs through paraxial optical system is derived. The evolution properties of the intensity distribution of HGBs in free space propagation are illustrated graphically using the derived propagation formula. By using matrix eigenvalues method, the laser cavity with diffraction optical elements, which can generate the dark hollow beams, was constructed. Dark hollow beams were realized in experiment and its intensity distribution properties in near field and far field were studied. The results derived from the experiment is coincident with those predicted by the theory. The dark hollow beams we have obtained can be used conveniently to analyze atoms manipulation and may provide a new method for atom cooling.

Abstract:We propose a novel Schell model source for generating twisted partially coherent beams with an initial radius of curvature, which is called a twisted flat-topped cosine Gaussian Schell-model (TFCGSM) source. The TFCGSM beam comprises a wavefront phase and a flat-top structure, with the source degree of coherence determined by two cosine functions. Based on the Huygens–Fresnel principle, the general analytical expression of the cross-spectral density function of the TFCGSM beam propagating through the paraxial ABCD optical system is derived, and then its propagation properties are studied. The results show that the conversion of the array of the beam and the non-uniform structure can be realized by adjusting the parameters in the source plane. As the propagation distance of the TFCGSM beam increases, it rotates around the axis and increases the intensity of the array distribution. Surprisingly, the initial radius of curvature can cause the beam to rotate. The unique shape and properties of the TFCGSM beam create new possibilities for optical communication and enhanced optical functions.

Abstract:In the ABCD optical system, the propagation properties and the radiation forces are obtained by studying the cross spectral density of partially coherent self-shifting cosine-Gaussian beams. A self-shifting phenomenon occurs when the beams propagate in the strongly nonlocal nonlinear medium. The shifting parameters could influence the bend characteristics of the propagation trajectory and the beam center, while the power ratio affects the periods of the parabolic trajectory. Furthermore, the radiation forces on a Rayleigh particle in the focusing optical system are studied, and the obtained force distributions depend on the refractive index, the shifting parameters, and the coherence widths. What we report here has potential applications in optical communication and optical tweezing.

Abstract:A high-order Bessel-Gaussian mode is introduced to describe hollow beams. The results for high-order Bessel-Gaussian beams propagating through lens focusing system and free space are derived in terms of Collins integral formula. The diffraction patterns and profile for high-order Bessel-Gaussian beams propagating through the above-mentioned optical systems are illustrated.

Abstract:Limited-diffraction beams are a class of waves that may be localized in space and time. Theoretically, these beams are propagation invariant and can propagate to an infinite distance without spreading. In practice, when these beams are produced with wave sources of a finite aperture and energy, they have a very large depth of field, meaning that they can keep a small beam width over a large distance. Because of this property, limited-diffraction beams may have applications in various areas such as medical imaging and tissue characterization. In this paper, fundamentals of limited-diffraction beams are reviewed and the studies of these beams are put into a unified theoretical framework. Theory of limited-diffraction beams is further developed. New limited-diffraction solutions to Klein-Gordon Equation and Schrodinger Equation, as well as limited-diffraction solutions to these equations in confined spaces are obtained. The relationship between the transformation that converts any solutions to an (-1)-dimensional wave equation to limited-diffraction solutions of an -dimensional equation and the Lorentz transformation is clarified and extended. The transformation is also applied to the Klein-Gordon Equation. In addition, applications of limited-diffraction beams are summarized.

Abstract:The expression of spectral density of cosine-Gaussian-correlated Schell-model (CGSM) beams diffracted by an aperture is derived, and used to study the changes in the spectral density distribution of CGSM beams upon propagation, where the effect of aperture diffraction is emphasized. It is shown that, comparing with that of GSM beams, the spectral density distribution of CGSM beams diffracted by an aperture has dip and shows dark hollow intensity distribution when the order-parameter n is big enough. The central intensity increases with increasing truncation parameter of aperture. The comparative study of spectral density distributions of CGSM beams with aperture and that of without aperture is performed. Furthermore, the effect of order-parameter n and spatial coherence of CGSM beams on the spectral density distribution is discussed in detail. The results obtained may be useful in optical particulate manipulation.

Abstract:On-chip channelless diffraction-free beam guiding enables dense integration of optical circuits in a reconfigurable manner, where total internal reflection, which is considered the cornerstone of guided-wave optics, is utilized to confine light in the out-of-plane direction. Here, we theoretically propose a physical mechanism to achieve on-chip channelless diffraction-free beam guiding beyond the light cone, utilizing the physics of bound states in the continuum. A bound state in the continuum with a tailored spatial dispersion plays an important role in cancelling both the in-plane diffraction and the out-of-plane scattering when the condition for total internal reflection is not satisfied. As a proof-of-concept verification, we experimentally demonstrate such an effect based on an all-dielectric platform using microwaves. The on-chip channelless diffraction-free beam guiding beyond the light cone also allows direct free-space coupling to such self-collimation modes. Our results may open up an avenue for exploring the physics and applications of guided-wave optics.

Abstract:The central spot of COS beam (CS-COS) can be obtained from COS beam by using the zero field truncation technique. The transversal intensity distributions of CS-COS through three different kinds of aperture functions are simulated numerically from the near field to the far field. It is shown that the CS-COS has the property of eliminating the diffraction fringes in the whole propagation process if a suitable aperture is introduced.

Abstract:We introduce a new class of twisted sources with twisted cosine-Gaussian Schell-model correlation structure. The spectral intensity and the degree of coherence of the field upon propagation are discussed. Such novel twisted field is characterized by unfamiliar twist pattern and controllable far-zone lattice profile. It exhibits a Gaussian or a lattice-like intensity distribution in the source plane, while always turns into a 2 × 2 lattice profile in the far zone. Notably, the array profile twists around the propagation axis instead of each element rotating about its own lobe center, which is different from most of the twisted array models. Moreover, the splitting tendency in the intensity distribution could be flexibly modulated by the twisted factor, the source coherence and the beam width. The coherence distribution could rotate in the same direction as the intensity with appropriate choice of parameters. Finally, the cross-spectral density's phase distribution exhibits a spiral windmill structure and coherent singularities could be observed upon propagation.

Abstract:Diffraction-free optical beams propagate freely without change in shape and scale. Monochromatic beams that avoid diffractive spreading require two-dimensional transverse profiles, and there are no corresponding solutions for profiles restricted to one transverse dimension. Here, we demonstrate that the temporal degree of freedom can be exploited to efficiently synthesize one-dimensional pulsed optical sheets that propagate self-similarly in free space. By introducing programmable conical (hyperbolic, parabolic, or elliptical) spectral correlations between the beam's spatio-temporal degrees of freedom, a continuum of families of axially invariant pulsed localized beams is generated. The spectral loci of such beams are the reduced-dimensionality trajectories at the intersection of the light-cone with spatio-temporal spectral planes. Far from being exceptional, self-similar axial propagation is a generic feature of fields whose spatial and temporal degrees of freedom are tightly correlated. These one-dimensional `space-time' beams can be useful in optical sheet microscopy, nonlinear spectroscopy, and non-contact measurements.

Abstract:We theoretically investigate the propagation of a paraxial beam in two-dimensional media with spatial dispersion. Based on the spatial dispersion theory and the (1 + 1)-dimensional paraxial wave equation, we get an expression which determines the diffraction of the beam. By fitting the dispersion surface of a typical spatial dispersion medium (a photonic crystal) calculated by the plane-wave-expansion method, the value of the diffraction term is determined, with which one can predict the diffraction of the paraxial beam that propagates in such media. Numerical simulations based on the finite-difference time-domain method confirm the theoretical results.

Abstract:Nondiffracting light beams receive substantial attention in various areas due to their properties of diffractionless propagation and achievable subwavelength beam size. Extending the propagation distance and compressing the beam size of nondiffracting beams are crucial to their practical applications. However, it remains challenging to achieve nondiffracting light beams with a subwavelength subdiffracting transverse size and a long propagation distance with controllable intensity profiles in both transverse and longitudinal directions. Here, we propose an optimization method to generate an ultralong-superoscillation nondiffracting light beam with full control of the beam intensity profile. A 45 λ -long-superoscillation nondiffracting light beam, the transverse sizes of which are within the range of 0.37 λ –0.4 λ along the propagation axis, is experimentally demonstrated by a metalens based on continuous phase modulation using geometrical phase metasurfaces. Our method provides an attractive way to controllably generate ultralong-superoscillation nondiffracting beams on a subwavelength scale and might find fascinating applications in optical manipulation, materials processing, data storage, microscopy, and spectroscopy. https://doi.org/10.1103/PhysRevApplied.19.014041 © 2023 American Physical Society
