Abstract:Improving the sensitivity of the fluorescence method is crucial in biosensor fields. In this study, an ultrasensitive and excellent selectivity method was developed for the detection of trace Cu2+ ions using 3-mercaptopropionic acid capped CdSSe/CdS/ZnS quantum dots (MPA-CdSSe/CdS/ZnS QDs) as a fluorescent probe. Through energy band engineering to optimize the interior inorganic structure and passivate the surface defects, the photoluminescence quantum yield of the MPA-CdSSe/CdS/ZnS QDs in aqueous solution had as high as 70 %. The X-ray diffraction (XRD) and Transmission Electron Microscope (TEM) analysis revealed the prepared CdSSe/CdS/ZnS QDs were cubic zinc blende structure with average diameter 7.2 nm. Further, the fluorescence of the MPA-CdSSe/CdS/ZnS QDs was significantly quenched by the Cu2+ with fast response time through an efficient photo-induced electron transfer (PET) progress. By optimizing the thickness of CdS shell, the MPA-CdSSe/CdS/ZnS QDs probe with appropriate CdS shell thickness exhibited excellent linear linearity and sensitivity for the detection of Cu2+ ions with limit of detection as low as 0.05 nM. Additionally, ions interference experiments demonstrated the good selectivity of the probe towards Cu2+. Furthermore, this sensing system could also be applied to the measurement of Cu2+ ions in real water samples with satisfactory recoveries. This work provided new insights to design and synthesize high-performance QDs-based fluorescence probes with controllable exciton emission behavior for their applications in biosensor and biological fields.

Abstract:In this work, a hydrophilic naphthalimide-based fluorescence chemosensor (sensor 1) was synthesized for Cu2+ recognition, in which 2-(2-aminoethoxy)ethanol was introduced to improve the hydrophily and Schiff base acted as the multidentate ligand for Cu2+. The effect factors, sensing mechanism and regenerability of sensor 1 for Cu2+ sensing were systematically investigated. It was found that sensor 1 displayed a long emission wavelength of 532 nm upon excited in visible light region (436 nm), and the good water solubility made it utilized in aqueous media. It could selectively react with Cu2+ over other common metal ions to form a 2:1 complex within 1 min and result in significant fluorescence quench. The fluorescence change was linear to 0.5-10.0 μmol L-1 of Cu2+ with a low detection limit of 3.74 × 10-8 mol L-1. Sensor 1 has been successfully utilized for analyzing Cu2+ in water samples as well as imaging cellular Cu2+. Moreover, in view of fluorescence "on-off-on" switches of sensor 1 induced by Cu2+ and EDTA, an IMPLICATION logic gate was constructed based on fluorescence mode with Cu2+ and EDTA as inputs.

Abstract:In this paper, we propose a fluorescent biosensor for the sequential detection of Pb2+ ions and the cancer drug epirubicin (Epn) using the interactions between label-free guanine-rich ssDNA (LFGr-ssDNA), acridine orange (AO), and a metal-phenolic nanomaterial (i.e., nano-monoclinic copper-tannic acid (NMc-CuTA)). An exploration of the sensing mechanism shows that LFGr-ssDNA and AO strongly adsorb on NMc-CuTA through π-π stacking and electrostatic interactions, and this results in the fluorescence quenching of AO. In order to sense the target Pb2+, initially, LFGr-ssDNA specifically binds with Pb2+ ions to form a G4 complex (G-Pb2+-G base pair), which was released from the surface of NMc-CuTA with strong AO fluorescence enhancement (Turn-ON). The subsequent addition of a biothiol, like cysteine (Cys), to the G4 complex decreases the fluorescence, as the Pb2+ ions released from the G4 complex have a higher interaction affinity with the sulfur atoms of Cys; this further induces the unwinding of the G4 complex to form LFGr-ssDNA. Finally, Epn was added to this, which intercalates with LFGr-ssDNA to form a G4 complex via G-Epn-G, resulting in fluorescence recovery (Turn-ON). Accordingly, the Turn-ON fluorescent probe had subsequent limits of detection of 1.5 and 5.6 nM for Pb2+ and Epn, respectively. Hence, the reported NMc-CuTA-based sensing platform has potential applications for the detection of Pb2+ and Epn in real samples with good sensitivity and selectivity.

Abstract:Fluorescent sensor has demonstrated to be a facile and effective method to detect heavy metal ions owing to its unique characteristics such as simplicity, ease of operation and cost-effectiveness. In this work, a novel fluorescent probe with aggregation-induced emission (AIE) feature that contains an amide ligand base on the tetraphenylethylene (TPE) dye was designed and successfully synthesized. The utilization of the AIE-active fluorescent probe (named as FDPA) for detection of Cu2+ in aqueous solution has also been examined. We demonstrated that the resultant AIE-active molecule displays amphiphilic property and can self-assemble in aqueous solution with remarkable enhancement of fluorescent intensity owing to its AIE feature. The detection limit of probe for Cu2+ determination is 6.11 × 10-9 M and shows excellent selectivity in the presence of competitive ions. This work provides a useful route to overcome the fluorescence quenching of conventional fluorescent probes in aqueous solution and an elegant way to prepare fluorescent probes with better fluorescence properties.

Abstract:A new ratiometric fluorescent sensor for Cu(2+), WLN, has been developed via integrating a 1,8-naphthalimide fluorophore with 8-aminoquinoline. WLN exhibits a highly selective ratiometric response to Cu(2+) over other transition metal ions in aqueous media. Moreover, its practical ratiometric imaging ability for intracellular Cu(2+) has been confirmed in human breast adenocarcinoma cells (MCF-7 cells) using a confocal microscope.

Abstract:A simple turn-off fluorescent probe, 5-(diethylamino)-2-(hydrazonomethyl)phenol (DHP), is designed and synthesized for the sensitive and selective detection of Cu2+. The bright green fluorescence of DHP is quenched after the addition of Cu2+. The probe DHP exhibits good anti-interference performance against Cu2+ in the presence of multiple metal ions. The fluorescence intensity of DHP (10 μM) at 522 nm is well linearized with the Cu2+ concentration at 0-5 μM, and it has a detection limit of 0.29 nM (R2 = 0.9949). The complexation ratio of the probe DHP to Cu2+ is 2 : 1 and the complexation constant is 3.44 × 104 M-1 (R2 = 0.9974). In addition, the probe DHP can be recovered using EDTA and Cu2+ can be effectively monitored at pH 5-11, with good results in dipstick experiments and actual water samples. HepG-2 cells remained viable in excess of 90% after being exposed to DHP (50 μM) for 24 h, which demonstrates the extremely low toxicity of DHP, and it can be used for in vivo cell imaging.

Abstract:A novel DNA-templated click chemistry strategy for homogenous fluorescent detection of Cu(2+) has been developed based on click ligation-dependent DNA structure switch and the selective quenching ability of graphene oxide (GO) nanosheet. The clickable duplex probe consists of two DNA strands with alkyne and azide group, respectively, and Cu(+)-catalyzed alkyne-azide cycloaddition (CuAAC) reaction can chemically ligate these two strands. Toehold sequence displacement was consequently exploited to achieve DNA structure transformation bearing fluorescent tag FAM. Cu(2+)-induced chemical ligation caused the probe transfer to hybrid structure with single stranded DNA (ssDNA) tail, while only duplex structure was obtained without Cu(2+). This structural difference can be probed by GO-based fluorescence detection due to the preferential binding of GO to ssDNA. Under the optimum conditions, this sensor can sensitively and specifically detect Cu(2+) with a low detection limit of 58 nM and a linear range of 0.1-10 μM. This new strategy is highly sensitive and selective for Cu(2+) detection because of the great specificity of click chemistry and super-quenching ability of GO. Moreover, with the aid of high efficient DNA templated synthesis, the detection process requires only about half an hour which is much quicker than previous click-chemistry-based Cu(2+) sensors.

Abstract:A novel ionophore-based fluorescent nanosensor has been successfully fabricated for the sensitive and selective detection of Cu2+ ions. The nanosensor was constructed through self-assembly of amphiphilic block copolymers, incorporating elesclomol as a Cu2+ ionophore and long-chain dialkylcarbocyanines (DiD) as a fluorescent dye. This design exhibits an "ON/OFF" fluorescence response, where Cu2⁺ ions are selectively sequestered within the nanosensors, resulting in fluorescence quenching of DiD. This strategy enables rapid and highly selective Cu2⁺ sensing with remarkable fluorescence quenching efficiency (up to 93.5 %) and an exceptionally low detection limit of 28.6 nM. The linear detection range extends over two orders of magnitude (0.05-10 μM). Furthermore, the feasibility of this nanosensor for practical applications was confirmed through successful determination of Cu2+ in real water and beer samples, with excellent recovery rates. This nanosensor offers advantages of simplicity, rapidity, and cost-effectiveness, holding significant potential for sensitive and selective Cu2+ detection in various biological and environmental samples.

Abstract:A homogenous fluorescence method was constructed for Cu2+ detection by employing DNA-templated click chemistry and exonuclease reaction. In this strategy, a dumbbell shaped DNA probe, which contained an alkyne group and an azide group at its ends, was designed as the template for the click chemistry reaction, and also the signal probe. In the absence of Cu2+, the DNA probe was digested into small oligonucleotide fragments by exonuclease, resulting in a low fluorescence background. However, this DNA probe can be sealed at its two ends by Cu2+-induced click chemistry ligation in the presence of Cu2+. This closed structure of DNA would remain stable after addition of exonuclease, and could then be stained by SYBR Green I. A strong fluorescence signal was observed, which was related to the concentration of Cu2+. This assay showed high selectivity and reached the detection limit of 39 nM. Moreover, the proposed strategy exhibited satisfactory detection results in real complex sample analysis, and has promising application in environmental monitoring and food safety.

Abstract:A novel fluorescent biosensor for detecting uranyl ion (UO22+) in aqueous environment has been developed based on the specific recognition of DNAzyme and the fluorescence quenching ability of molybdenum disulfide (MoS2) nanosheets. The DNAzyme contains a DNA enzyme strand and a 6-carboxylfluorescein (FAM)-labeled DNA substrate strand. We demonstrated that MoS2 nanosheets have low affinity to the substrate-enzyme complex DNAzyme. Whereas, in the presence of UO22+, UO22+ can specifically cleave DNAzyme to release FAM-labeled single-strand DNA and the released FAM-labeled single-strand DNA can be firmly adsorbed on the surface of MoS2 nanosheets, which resulted in an obvious decrease of fluorescence intensity. This provided a sensing platform for the rapid, simple and sensitive fluorescent detection of UO22+. By using the sensing platform, a sensitive and selective fluorescent method for the rapid detection of UO22+ has been developed. In comparison with previous biosensor, the proposed method has obvious analytical advantage such as relatively high sensitivity and good stability, short analytical time and low cost. It can be used to detect as low as 2.14 nM of UO22+ in aqueous environment with a recovery of 96-102% and a RSD < 5% (n = 6). Abstract:Copper ion (Cu2+) plays important role in life system and industry, therefore, efficient detection of Cu2+ is very important. To improve the selectivity and sensitivity of Cu2+, herein, quinoline and coumarin groups were rationally employed into a novel fluorescent sensor CQ to efficiently sensing Cu2+ through the collaboration of these two groups. Interestingly, the CQ possesses aggregation‐induced emission (AIE) properties and shows nice selective and sensitive fluorescent response to Cu2+. The lowest limit of detection (LOD) for Cu2+ is 2.81×10‐8 M. Moreover, the Cu2+ sensing process has good anti‐interference properties. The possible recognition mechanism was investigated by experiments and theoretical calculations, which supplied a deep insight not only on the collaboration of the quinoline and coumarin groups in the Cu2+ recognition process, but also on the charge transfer induced fluorescent response mechanism. The results shown that the CQ could specifically detected Cu2+ via the collaboration of the quinoline and coumarin groups through multiple coordination interactions. Importantly, test strips based on CQ were fabricated, which could act as test kits for convenient and efficient detection of Cu2+.

Abstract:A BODIPY-based turn-on fluorescent probe 1 has been designed and synthesized for fluorescence detection of Cu2+ in aqueous media and imaging in living cells. The CN bonds are selectively hydrolyzed by the reaction with Cu2+ to generate formyl-BODIPY. Abstract:Through molecular design of a new phenanthro[9,10-d]imidazole-based compound, the probe 4-VB-PI-DMBA (4-VB-PI-DMBA = N,N-dimethyl-4-(1-(4- vinylbenzyl)-1H-phenanthro[9,10-d]imidazole-2-yl)aniline) strongly emissive at λ em = 462 nm, is capable of the uniquely fluorescent “turn-off” for Cu2+ ion with a detection limit of 30.2 nM. This result shows that the phenanthro[9,10-d]imidazole-derivative with a twisting structure and without proton-donating groups, renders a selective fluorescence and visual sensor platform to Cu2+ ion.

Ultrasensitive and Colorimetric Detection of Copper-Based on Dual-DNAzymes

Abstract:Copper is an essential element for life and constitutes copper proteins, and attends life activities in human body. However, an excess of copper is poisonous, and can cause hepatitis, gastroenteritis and pneumonia. copper enters into biosphere from ores by weathering and erosion and exists in water in the form of a divalent copper ion. Therefore, it is necessary to detect copper ions in an ultrasensitive and quantitative way. Herein, a dual-functional DNAzymatic visual biosensor was developed, optimized and validated to detect divalent copper ions sensitively and selectively in tap water. In the method, dual DNAzymes with different functions were designed within one biosensor. One DNAzyme was Cu2+-dependent cleaving DNAzyme, which could be cleaved by Cu2+ for signal recognition. The other one was G-quadruplex DNAzyme, which was induced by terminal deoxynucleotidyl transferase (TdT) for "one-to-multiplex" signal amplification and output. In the middle of two steps, the exponential amplification reaction (EXPAR) was applied to amplify the recognitive signal exponentially. Under optimized conditions, Cu(2+ c)ould be selectively observed with the naked eyes with the detection limit (LOD) of 10 pmol/L and with the microplate reader with LOD of 7.6 pmol/L in about 2 h. Furthermore, Cu2+ in the concentration range of 10 pmol/L-200 pmol/L in tap water samples could be discriminated by naked eyes. The platform provides an ultrasensitive and colorimetric detective tool for the analysis of trace Cu2+ in tap water with high specificity, and will have broad application prospect in many fields.

Abstract:Excessive copper ions (Cu2+) cause serious environmental pollution and even endanger the health of organisms. Fluorescence chemosensing materials are widely used in the detection of metal ions due to their simple operation and high sensitivity. In this study, SiO2-encapsulated single perovskite quantum dot (PQD@SiO2) core-shell nanostructures which show strong, stable, and green fluorescence are synthesized and composited with gold nanoclusters (AuNCs) which show Cu2+-sensitive and red light-emitting fluorescence to obtain a visualized ratiometric fluorescence sensor (AuNCs/PQD@SiO2) for the detection of Cu2+. In the visualized detection of Cu2+, the green fluorescence emitted from the ion-insensitive PQD@SiO2 component is used as a reference signal and the red fluorescence emitted by ion-sensitive AuNC component is adopted as a sensing signal. In the presence of Cu2+, the red fluorescence is quenched whereas the green fluorescence remains stable, which results in a visualized fluorescence color change from orange-red to yellow and finally to green with increasing Cu2+ concentration. The significant change in the fluorescence color of AuNCs/PQD@SiO2 in response to Cu2+ enables a rapid, sensitive, and visualized detection of Cu2+. Further accurate and sensitive ratiometric fluorescence analysis of Cu2+ can be accomplished by measuring the ratio of fluorescence intensities at 643 and 520 nm (I643/I520) at a certain Cu2+ level. The developed AuNCs/PQD@SiO2-based sensor has been validated by its satisfactory application in the detection of Cu2+ in human serum and environmental water samples.

Abstract:Based on the typical Suzuki coupling reaction and Schiff base reaction, a novel fluorescent molecular PCBW is synthesized and applied as a fluorescence and colorimetric sensor to detect Cu2+ in aqueous solution. The PCBW sensor presents the aggregation-caused quenching (ACQ) effect and at 1 × 10-5 mol L-1 it emits the strongest turquoise fluorescence in the DMSO-H2O system (fw = 40%). The sensor exhibits a 'turn-off' fluorescent characteristic by adding Cu2+, and its fluorescent intensity shows a reliable linear relationship with the Cu2+ concentration in the range of 0-6 × 10-6 mol L-1, with a detection limit of 1.19 × 10-8 mol L-1. Meanwhile, the PCBW sensor also exhibits the colorimetric sensing from colorless to light yellow. The sensor has good selectivity and anti-interference and its pH application range can be extended from 5 to 10. The intramolecular charge transfer (ICT) is speculated as the main fluorescence mechanism of PCBW. In addition, the sensor presents good reusability and is practicable to detect Cu2+ in diverse aqueous samples.
