Abstract:A mild method for the preparation of tryptanthrin and its derivatives by copper(I) catalysis with two identical isatins in the presence of KHCO 3 as a base is described. Furthermore, more complicated tryptanthrin derivatives are obtained in two steps with two different isatins in one pot. This work features readily available starting materials, mild reaction conditions, simple operation, good functional group tolerance, high yields and selectivity. This transformation not only provided a new access to tryptanthrin and its derivatives, but also enriched the chemistry of isatins and copper catalysis.

Abstract:The first example of cyclization cross-coupling of tryptophols and tryptamines has been realized by copper catalysis with air or oxone as the terminal oxidant, resulting in the direct construction of a new class of heterocyclic 3a,3a′-bisindolines in moderate to good yields with high chemoselectivities. A series of mechanistic control experiments were also conducted, indicating that the copper catalyst selectively coordinates with the nitrogen moiety of the tryptamine to initiate the oxidation, and a nucleophilic-alkylation process is proposed for the carbon–carbon bond-forming in the reaction. Abstract:A highly effective and straightforward method to construct a wide range of functionalized 2,3-disubstituted indoles has been developed. The method involves the tandem reaction of rhodium(II)-catalyzed denitrogenative annulation of triazole-based benzyl anilines and oxygen-mediated oxidative aromatization. The developed method can also be used to synthesize 2,3-disubstituted benzofurans by replacing the benzyl anilines with benzyl phenols.

Abstract:Oxidative rearrangement of 3-substituted indoles is a fundamental organic transformation for the synthesis of 2-oxindole scaffolds. However, this time-consuming and complex task produces a single type of product via two-electron oxidation using stoichiometric chemical oxidants. Herein, we report a unified and efficient copper-catalyzed selective single-electron transfer strategy for three oxidation reactions of 3-substituted indoles using air (O2) as the terminal oxidant, providing a streamlined and practical synthetic approach to access 3-hydroxyl-, 3-alkoxyl-, and 3-hygrogenous-2-oxindoles via a 3-radical-2-hydroxyl indoline intermediate. This copper catalysis protocol demonstrates high chemoselectivity, good functional group tolerance, and broad application potential in the modular synthesis of biologically 3-functionalized oxindole derivatives.

Abstract:2,3'‐Bisindoles with C–C linkages have attracted interest in medicinal chemistry, yet their synthesis is intricate with many steps. Notably, direct C–H/C–H cross‐coupling of non‐directed heteroaromatics remains challenging, often requiring precious metals and oxidants to enhance coupling efficiency. Herein, we present a copper‐catalyzed C–H/C–H cross‐coupling method for N‐substituted indoles without directing groups, facilitated by molecular oxygen under gentle conditions. It showed reasonable functional group compatibility and provided one‐pot access to a variety of 2,3'‐bisindoles derivatives in moderate to good yields.

Abstract:Transition-metal-catalyzed C-N bond formation reactions have been widely studied in recent decades, but site-selective intermolecular C-N cross-coupling between the carbon and nitrogen sites of two identical molecules is less explored. Herein, we report the highly selective copper-catalyzed oxidative cyclization/C-N cross-coupling of two tryptamines with O2 as the terminal oxidant, which provides practical and direct access to a new class of indole-linked-pyrroloindoline compounds in moderate to good yields. Mechanistic studies suggested that the coordination between the copper catalyst and the tryptamine substrate is key for chemoselectivity control.

Abstract:An electrooxidative reconstruction of indole core under ambient conditions is reported, by constructing tryptanthrin architecture successfully. This electrochemical strategy simplifies molecular skeleton‐hopping by employing commercially available NH‐indole feedstock as the starting materials. Furthermore, the synthetic value of this skeletal reconstruction was demonstrated by the readily accessible and divergent further transformations, which provided a diverse array of biologically active molecules. A preliminary mechanism investigation involving control experiments and cyclovoltammetric studies have evidenced the indispensability of electricity, as well as the importance of oxygen and TEMPO, thereby revealing a possible reaction pathway.

Abstract:3-Sulfonyl[1,2,3]triazolo[4,5-b]indoles were efficiently prepared via a tandem catalysis process involving intramolecular ligand stabilized CuAAC and Cu-catalyzed C–N coupling. The obtained 3-sulfonyl[1,2,3]triazolo[4,5-b]indoles could be utilized as α-imino rhodium carbene precursors for the construction of a range of valuable indole molecules including pyrrolo[2,3-b]indoles, spirocyclopropyl iminoindoles, 2,3-dihydropyrrolo[2,3-b]indoles, 3,3′-biindoles, and 2,3′-biindoles.

Abstract:The Pictet-Spengler type condensation of tryptamine derivatives and aldehydes or ketones is a classic reaction, and has been previously applied to assemble indole-annulated 5-, 6- and 8-membered heterocyclic rings. In this work, we further expand the synthetic scope of this reaction to the 7-membered azepino[4,5-b]indole skeleton through the direct C-H functionalization of 2-alkyl tryptamines, in which the non-activated methylene group participates in a 7-membered ring formation with aldehydes. By combining this unprecedented ring-forming process with a second C-H olefination at the same carbon, the concise total synthesis of natural products ngouniensines is achieved, demonstrating the synthetic potential of the developed chemistry in simplifying retrosynthetic disconnections.

Abstract:[GRAPHICS]One-pot synthesis of indoles by a palladium-catalyzed annulation of ortho-haloanilines and aldehydes has been developed. Coupling of ortho-iodoaniline with aldehyde is realized under mild ligandless conditions [Pd(OAc)(2), DABCO, DMF, 85 degrees C], whereas X-Phos is found to be the ligand of choice for coupling reactions involving ortho-chloroanilines/ortho-bromoanilines and aldehydes. A variety of ortho-haloanilines with different electronic properties are suitable substrates, and aldehydes including chiral ones participated in this reaction without racemization. Coupling of (S)-2-N,N-di-tert-butoxycarbonyl-5-oxopentanoate, derived from L-glutamic acid, with ortho-haloanilines provides a rapid access to the ring-A-substituted tryptophans in good to excellent yields.

Abstract:Tryptophan and its non-canonical variants play critical roles in pharmaceutical molecules and enzymes. Facile access to this privileged class of amino acids from readily available building blocks remains a long-standing challenge. Here, we report a regioselective synthesis of non-canonical tryptophans bearing C4-C7 substituents via Rh-catalyzed annulation between structurally diverse tert-butyloxycarbonyl (Boc)-protected anilines and alkynyl chlorides readily prepared from amino acid building blocks. This transformation harnesses Boc-directed C–H metalation and demetalation to afford a wide range of C2-unsubstituted indole products in a redox-neutral fashion. This umpolung approach compared to the classic Larock indole synthesis offers a novel mechanism for heteroarene annulation and will be useful for the synthesis of natural products and drug molecules containing non-canonical tryptophan residues in a highly regioselective manner.

Abstract:The first successful copper-catalyzed decarboxylative cyclization reaction of ethynylbenzoxazinones and thiols has been developed. A few-studied α-addition process of the copper-allenylidene intermediate promoted this reaction. With this protocol, a range of 2-thiomethylene indole compounds have been obtained. This methodology displayed significant advantages including its mild reaction conditions, cheap catalysts, good yields and wide scope of substrate compatibility.
