Abstract:An efficient [bis-(trifluoroacetoxy)iodo]benzene (PIFA)-mediated oxidative C-N bond formation is developed for the synthesis of 1H-indazoles from readily available arylhydrazones. The reaction tolerates a wide range of functional groups and has broad scope of substrates. Moreover, this method is a relative green and reliable method for rapid preparation of substituted 1H-indazoles under mild conditions.

Abstract:An efficient and environmentally friendly methodology for 3,3‐dichlorination and 2‐oxidation of indole derivatives is described. Using KI as promotor, MCl (M=K, Na) as chlorine source, and oxone as oxidant, the reaction proceeds smoothly affording various functionalized 3,3‐dichloro‐2‐oxindoles in moderate to excellent yields. This strategy can also be extended to 3‐substituted indoles. With a broad substrate scope, it is a practical approch to 3,3‐disubstituted‐2‐oxindoles.

Abstract:The copper-catalyzed sulfenylation of indoles with aryl iodide and sulfur powder has been investigated both experimentally and theoretically. This protocol provides a direct and facile approach to prepare 3-sulfenylindoles with moderate to excellent yields and good functional-group tolerance. The in-situ IR analysis provided evidence for that NaOAc could promote the synthesis of diphenyl disulfide by the coupling of aryl iodide and sulfur powder. According to DFT calculations, the coupling pathway involving the intermediate N-methyl-3-iodoindole is more favored than the direct coupling by the CH activation of indole. The N-methyl-3-iodoindole was identified as a crucial intermediate in the catalytic cycle.

Abstract:2,3'‐Bisindoles with C–C linkages have attracted interest in medicinal chemistry, yet their synthesis is intricate with many steps. Notably, direct C–H/C–H cross‐coupling of non‐directed heteroaromatics remains challenging, often requiring precious metals and oxidants to enhance coupling efficiency. Herein, we present a copper‐catalyzed C–H/C–H cross‐coupling method for N‐substituted indoles without directing groups, facilitated by molecular oxygen under gentle conditions. It showed reasonable functional group compatibility and provided one‐pot access to a variety of 2,3'‐bisindoles derivatives in moderate to good yields.

Abstract:We report the development of a rapid approach for directly converting indoles into 2-oxindoles promoted by HOCl formed in situ from the combination of PIFA and n-Bu4NCl·H2O. The procedure is widely functional group tolerant and provides 2-oxindoles in up to 94% yield within 5 min. The potential applications of the developed methodology are demonstrated by the gram-scale preparation of 3-methyl-2-oxindole (11a), the one-pot two-step syntheses of spiro-oxindoles 26a and 26b, and the formal synthesis of (-)-folicanthine (2).

Abstract:A novel and efficient transition metal-free C-H bond halogenation of indole derivatives has been developed. 3-Halogenated (3-Br, 3-I) indoles are highly regioselectively produced by this protocol. Simple and readily available halide salts (TBAB, KI) are employed as the halogen source. The transition metal-free and the mild conditions make this protocol very easy to handle and practical. Abstract:Described herein is the discovery of a palladium‐catalyzed chlorocylization strategy for the synthesis of 3‐chloroindoles tolerated with a wide range of functional groups in moderate to excellent yields. In this transformation, the solvent effect is so important that only THF presents the positive effect. Furthermore, the present method provides an essay access to the N‐unprotected 3‐chloroindoles. A mechanism study showed that 2‐alkynlanilines were catalyzed by palladium to from corresponding indoles, followed by copper‐mediated oxidation chlorination to deliver the 3‐chloroindoles products.

Abstract:Oxidative rearrangement of 3-substituted indoles is a fundamental organic transformation for the synthesis of 2-oxindole scaffolds. However, this time-consuming and complex task produces a single type of product via two-electron oxidation using stoichiometric chemical oxidants. Herein, we report a unified and efficient copper-catalyzed selective single-electron transfer strategy for three oxidation reactions of 3-substituted indoles using air (O2) as the terminal oxidant, providing a streamlined and practical synthetic approach to access 3-hydroxyl-, 3-alkoxyl-, and 3-hygrogenous-2-oxindoles via a 3-radical-2-hydroxyl indoline intermediate. This copper catalysis protocol demonstrates high chemoselectivity, good functional group tolerance, and broad application potential in the modular synthesis of biologically 3-functionalized oxindole derivatives.

Abstract:A highly effective and straightforward method to construct a wide range of functionalized 2,3-disubstituted indoles has been developed. The method involves the tandem reaction of rhodium(II)-catalyzed denitrogenative annulation of triazole-based benzyl anilines and oxygen-mediated oxidative aromatization. The developed method can also be used to synthesize 2,3-disubstituted benzofurans by replacing the benzyl anilines with benzyl phenols.

Abstract:Oxidative rearrangement of indoles is an essential transformation that has been widely used in the synthesis of many pharmaceutical agents and bioactive natural products. Herein, we report an efficient, practical and metal-free method for the oxidative rearrangement of indoles to spirooxindoles, 2-oxindoles, 2,2-disubstituted indolin-3-ones, and oxa-spirooxindoles using 10 mol% HBr and hydrogen peroxide as the terminal oxidant. The protocol displays a broad substrate scope (35 examples) and a wide range of functional group tolerance and has been applied in the synthesis of (+/-)-coerulescine and the formal synthesis of (+/-)-horsfiline. Notably, the distinct advantages of this protocol involve metal-free catalysis, waste prevention, and simple operation.(c) 2022 Elsevier Ltd. All rights reserved.

Abstract:C2-formyl indoles are important building blocks for the organic synthesis of alkaloids and small molecules with broad biological activities. Generally, its preparation involves both direct formylation of indoles at the activated C2-position with as unstable carbonyl sources and oxidative cleavage of tedious prepared tetrahydro- β -carbolines (TH β Cs) in the harsh conditions. In continuation of our research on the oxidative functionalization of indoles, in this letter, we report that straightforward effectively oxidation of easily available 2-methylindoles bearing C3-heteroatom functional group proceeds regioselectively C2-formyl indoles in good and excellent yields employing an oxidative combination of NCS/DMSO. This methodology presents a reasonably broad substrate scope and excellent functional group tolerance, thus enabling the preparation of highly versatile building blocks susceptible to further functionalization. An additional feature of this approach includes high efficiency, ease of operation, facile purification, and short reaction time, making it a promising strategy for organic synthesis and drug discovery.

Abstract:In continuation of our research interest in the green oxidation of indoles, we further explore the direct oxidation of 2-methylindoles to 2-formyl indoles promoted by NCS and associated with H2O as the oxygen source. This methodology was demonstrated to be a robust protocol consisting of chlorination, SN2', and oxidation processes, and presents a reasonably broad substrate scope and excellent functional group tolerance, thus enabling the preparation of high added-value versatile building blocks susceptible to further functionalization.

Abstract:A visible-light-promoted aerobic hydroxyazidation of indole derivatives with TMSN3 is described. The reaction proceeded under photocatalyst-free conditions to furnish trans-2-azidoindolin-3-ols with high regioselectivity and stereoselectivity. The protocol is operationally simple, and the starting materials (e.g., 1-(pyrimidin-2-yl) indoles, azidotrimethylsilane, and air) are readily available. The proposed mechanism in which substrates act as photocatalysts upon excitation using blue light-emitting diodes (LEDs) was supported by experimental studies.
