Abstract:Herein, we report the first intramolecular C-N bond formation of phenylpropanamide derivatives via organocatalytic oxidative reactions, affording 3,3-disubstituted oxindole derivatives with up to 99% yield. The high efficiency of this reaction is exemplified by the transition metal-free mild conditions and the ability to perform the reaction on a gram scale. Meanwhile, the DFT calculation of the catalytic oxidative transformation pathway has also been studied.

Abstract:An efficient [bis-(trifluoroacetoxy)iodo]benzene (PIFA)-mediated oxidative C-N bond formation is developed for the synthesis of 1H-indazoles from readily available arylhydrazones. The reaction tolerates a wide range of functional groups and has broad scope of substrates. Moreover, this method is a relative green and reliable method for rapid preparation of substituted 1H-indazoles under mild conditions.

Abstract:The oxidative C2-C3 π bond strategy has been the most efficient tool to synthesize indoline with oxygen-containing, which was frequently appeared in natural products with various biological activities as a structural units. Recently, the oxidation-induced cascade strategy through oxygenation-activating indolic C2-C3 π bond of indoles received much more attention in swiftly establishing the complex indoline with oxygen-containing molecular architectures and held tremendous potential in total synthesis of indole alkaloids, and could be carried out between the quitely probable and activated indole radical cation or imine cation intermediates produced via oxidative C2-C3 π bond of indole with various nucleophiles or ring-forming reagents by employing simple and non-decorated indoles as starting substrates. Herein, we have reviewed recent advances in the oxidation-induced cascade strategies connecting intra-cyclization or inter-annulation reactions, nucleophilic or radical additions and rearrangement via oxidative C2-C3 π bond of indoles in about the past two decades, providing diverse oxygen-contained indolines such as indoxyls, indoline oxygen-heterocycles and indolones, respectively. The features and mechanisms of different types of oxidation-induced cascade reactions have been summarized and represented, and their asymmetric reactions and applications in the total synthesis of indole alkaloids have been exampled.

Abstract:C2-formyl indoles are important building blocks for the organic synthesis of alkaloids and small molecules with broad biological activities. Generally, its preparation involves both direct formylation of indoles at the activated C2-position with as unstable carbonyl sources and oxidative cleavage of tedious prepared tetrahydro- β -carbolines (TH β Cs) in the harsh conditions. In continuation of our research on the oxidative functionalization of indoles, in this letter, we report that straightforward effectively oxidation of easily available 2-methylindoles bearing C3-heteroatom functional group proceeds regioselectively C2-formyl indoles in good and excellent yields employing an oxidative combination of NCS/DMSO. This methodology presents a reasonably broad substrate scope and excellent functional group tolerance, thus enabling the preparation of highly versatile building blocks susceptible to further functionalization. An additional feature of this approach includes high efficiency, ease of operation, facile purification, and short reaction time, making it a promising strategy for organic synthesis and drug discovery.

Abstract:An efficient organoiodine‐catalysed intramolecular direct C–N bond oxidative coupling reaction is presented. Structurally diverse 3‐(mono)substituted oxindoles were rapidly obtained in a complex oxidative system in up to 87% yield. Various N‐alkoxy‐2‐phenylpropanamides were well‐tolerated. This study showed that the electronic effects on the aromatic ring of reactants have a crucial effect on the selectivity (C−N/C−O) of cyclisation products. In addition, gram‐scale synthesis and late‐stage modification of 3‐(mono)substituted oxindole derivatives revealed the practical application of this transformation.

Abstract:A one‐pot approach for the asymmetric synthesis of C2‐tetrasubstituted indolin‐3‐ones from 2‐substituted indoles was developed via merging transition metal catalysis with organocatalysis. This strategy involves two processes, including CuI catalyzed oxidative dearomatization of 2‐substituted indoles using O2 as green oxidant, and followed by an proline‐promoted asymmetric Mannich reaction with ketones or aldehydes. A series of C2‐tetrasubstituted indolin‐3‐ones were obtained in 35–86% yields, 2:1‐>20:1 dr and 48–99% ee. Moreover, the synthetic 2‐tetrasubstituted indolin‐3‐ones could be easily transformed into 1H‐[1,3] oxazino [3,4‐a]indol‐5(3H)‐ones via a [4+1] cyclization process. In addition, the synthetic compound 3 s show certain antibacterial activity against S. aureus ATCC25923 and multi‐drug resistance bacterial strain of S. aureus (20151027077) and its MIC values up to 8 μg/mL and 16 μg/mL, respectively.

Abstract:3‐Idolenone is a key intermediate in the synthesis of many drugs and plays an important role in synthetic chemistry and biochemistry. A new method for the synthesis of trifluoromethylated 3‐indolenones by carbonylation catalyzed by Pd(0) is described. In the absence of an additive, 1‐chloro‐3,3,3‐trifluoro‐propylene (an inexpensive ozone‐friendly CF3 structural unit) reacts with indole and carbon monoxide to synthesize trifluoromethylatedindolenone in good yield, regioselectivity and chemoselectivity and with strong resistance to basal functional groups such as alkynes, aldehydes and esters. Notably, in addition to indole compounds being well transformed into the corresponding products, pyrrole, heteroindole can also is also suitable for the corresponding chemical transformation. This study provides a scientific method for the late construction of trifluoromethylated indole/pyrrole 3‐ketene.

Abstract:A scalable and practical synthesis of functionalized indoles via Pd- tBuONO cocatalyzed aerobic cycloisomerization of o-allylanilines is reported. Using molecular oxygen as a terminal oxidant, a series of substituted indoles were prepared in moderate to good yields. The avoidance of hazardous oxidants, heavy-metal cocatalysts, and high boiling point solvents such as DMF and DMSO enables this method to be applied in pharmaceutical synthesis. A practical gram-scale synthesis of indomethacin demonstrates its application potential.

Abstract:Oxidative rearrangement of 3-substituted indoles is a fundamental organic transformation for the synthesis of 2-oxindole scaffolds. However, this time-consuming and complex task produces a single type of product via two-electron oxidation using stoichiometric chemical oxidants. Herein, we report a unified and efficient copper-catalyzed selective single-electron transfer strategy for three oxidation reactions of 3-substituted indoles using air (O2) as the terminal oxidant, providing a streamlined and practical synthetic approach to access 3-hydroxyl-, 3-alkoxyl-, and 3-hygrogenous-2-oxindoles via a 3-radical-2-hydroxyl indoline intermediate. This copper catalysis protocol demonstrates high chemoselectivity, good functional group tolerance, and broad application potential in the modular synthesis of biologically 3-functionalized oxindole derivatives.
