Abstract:A new cascade radical addition/cyclization of unactivated alkenes and phosphine oxides has been developed. A wide variety of bioactive phosphine-containing quinazolinones were obtained in up to 91% yield for 34 examples under transition-metal-free conditions. This cascade protocol represents an environmentally friendly and efficient way to build phosphine-containing quinazolinones in excellent yields under mild conditions with a broad substrate scope and high atomic economy.

Abstract:The first example of cyclization cross-coupling of tryptophols and tryptamines has been realized by copper catalysis with air or oxone as the terminal oxidant, resulting in the direct construction of a new class of heterocyclic 3a,3a′-bisindolines in moderate to good yields with high chemoselectivities. A series of mechanistic control experiments were also conducted, indicating that the copper catalyst selectively coordinates with the nitrogen moiety of the tryptamine to initiate the oxidation, and a nucleophilic-alkylation process is proposed for the carbon–carbon bond-forming in the reaction. Abstract:Transition-metal-catalyzed C-N bond formation reactions have been widely studied in recent decades, but site-selective intermolecular C-N cross-coupling between the carbon and nitrogen sites of two identical molecules is less explored. Herein, we report the highly selective copper-catalyzed oxidative cyclization/C-N cross-coupling of two tryptamines with O2 as the terminal oxidant, which provides practical and direct access to a new class of indole-linked-pyrroloindoline compounds in moderate to good yields. Mechanistic studies suggested that the coordination between the copper catalyst and the tryptamine substrate is key for chemoselectivity control.

Abstract:A photoinduced copper-catalyzed alkoxyl triggered C−C bond cleavage/aminocarbonylation cascade is presented. Through adjusting the structure of alkoxyl radical precursors, functionalized lactones and keto-amides were synthesized with good yields and excellent functional group tolerance under redox-neutral conditions. Notably, this protocol enables the integration of lactone fragments with many amine drugs and drug fragments.

Abstract:We report a safe and convenient open-flask copper-catalyzed selective oxidation/functionalization methodology for tetrahydrocarbolines and tetrahydro-β-carbolines that employs atmospheric O<sub>2</sub> as the terminal oxidant. The system is applicable to oxidative rearrangement of tetrahydro-β-carbolines, tetrahydrocarboline oxidation to α-alkoxy carbazoles and spirooxindoles, and Witkop oxidation. Mechanistic experiments indicated that a single-electron oxidation process is responsible for the tunable selectivity control. This copper-catalysis protocol represents a significant advance in the field of indole oxidation.

Abstract:The copper-catalyzed cyclization of activated alkenes with cyclobutanone O-acyl oximes under redox-neutral conditions has been reported. This facile protocol provided an efficient approach to a variety of cyanoalkylated oxindoles and dihydroquinolin-2(1H)-ones with a broad substrate scope and excellent functional group tolerance. In this reaction, sequential C-C bond cleavage, radical addition, and cyclization processes were involved, wherein multiple bonds were constructed in a one-pot reaction. Mechanistic studies suggest that the reaction probably proceeded via a radical pathway.

Abstract:A visible-light-induced radical cascade reaction for the direct alkynylation of tryptamines with alkynyl sulfones was developed, affording a series of C3-alkynyl pyrroloindolines in excellent yields. This method features readily available substrates, good functional group tolerability, and scalability. C3-alkynyl pyrroloindolines show great potential as useful building blocks in the preparation of complex and valuable pyrroloindoline alkaloids.

Abstract:Inspired by the heme iron-catalyzed radical insertion of dioxygen to the tryptophan indole ring, herein we utilize alkylperoxy radical species as a coupling partner to trigger a peroxycyclization of readily accessible tryptophan derivatives and enable the first synthesis of peroxypyrroloindolenines. A preliminary biological evaluation revealed promising anti-cancer activities (IC50 = 22.00 μM for compound 2a), and revealed that both the indolenine core and the peroxy functionality are responsible for the antiproliferation effect against Hela cell lines.

Abstract:Herein, we introduce copper-catalyzed cascade radical addition/cyclization reactions for the synthesis of oxindole derivatives. The in situ generated copper complex can solely server as a standalone photocatalyst. The generation of alkyl radical from unactivated alkyl iodides can be achieved by copper catalysis under visible-light irradiation at room temperature in absence of any additives or oxidants. Various functional groups are well compatible. Primary, secondary, and tertiary alkyl iodides all exhibit good reactivity, leading to products with good yields.

Abstract:Oxidative rearrangement of 3-substituted indoles is a fundamental organic transformation for the synthesis of 2-oxindole scaffolds. However, this time-consuming and complex task produces a single type of product via two-electron oxidation using stoichiometric chemical oxidants. Herein, we report a unified and efficient copper-catalyzed selective single-electron transfer strategy for three oxidation reactions of 3-substituted indoles using air (O2) as the terminal oxidant, providing a streamlined and practical synthetic approach to access 3-hydroxyl-, 3-alkoxyl-, and 3-hygrogenous-2-oxindoles via a 3-radical-2-hydroxyl indoline intermediate. This copper catalysis protocol demonstrates high chemoselectivity, good functional group tolerance, and broad application potential in the modular synthesis of biologically 3-functionalized oxindole derivatives.

Abstract:Oxazoles are among the most important heterocyclic scaffolds in the fields of natural products and medicinal chemistry. Herein is developed a tandem reaction for the synthesis of a diverse array of 4,5-difunctionalized oxazoles utilizing easily-accessible ethyl 2-isocyanoacetate and aldehydes (26 examples, 31-83% yields). This cascade reaction is facilitated by catalytic CuBr and molecular oxygen as the oxidant. The process involves a catalytic cycloaddition oxidative dehydroaromatization mechanism. The broad aldehyde substrate scope, mild reaction conditions, and atom economy make this protocol an attractive alternative to access functionalized oxazoles.

Abstract:A copper-catalyzed aerobic oxidative intramolecular alkene C-H amination has been developed using readily available substituted 3-benzylidene-2-pyridin-2-ylmethyl-2,3-dihydro-isoindol-1-ones as the starting materials, and the corresponding N-heterocycles were obtained in good to excellent yields. Abstract:A new copper-catalyzed aerobic synthesis of 4-oxobutanenitriles by nitrogen-migration cyanation and oxygenation of γ,δ-unsaturated keto oximes at room temperature is developed. By means of the copper(0) powder catalyst, a nitrogen-migration cyanation and oxygenation of unsaturated keto oximes with O2 via a radical intramolecular 5-exo-trig cyclization, C=C double bond cleavage and ring-opening process to assemble 4-oxobutanenitriles. Mechanistic studies indicate that these processes include radical initiating the reaction from the oxime side and the superoxide radical formation.

Abstract:The first synthetically useful interrupted Witkop oxidation has been disclosed through a radical triggered oxidative peroxycyclization of tryptamine derivatives, using a TBAI/TBHP catalytic system. This differs from known tryptophan oxidations, which typically cleave the 2,3-double bond. Upon one-pot reduction, this system renders an easy access to structurally distinct peroxypyrroloindoles, which are demonstrated to be useful precursors in various downstream synthetic elaborations.

Abstract:A mild method for the preparation of tryptanthrin and its derivatives by copper(I) catalysis with two identical isatins in the presence of KHCO 3 as a base is described. Furthermore, more complicated tryptanthrin derivatives are obtained in two steps with two different isatins in one pot. This work features readily available starting materials, mild reaction conditions, simple operation, good functional group tolerance, high yields and selectivity. This transformation not only provided a new access to tryptanthrin and its derivatives, but also enriched the chemistry of isatins and copper catalysis.

Abstract:Herein, we report a general copper‐catalyzed method for the tunable oxygenative rearrangement of tetrahydrocarbazoles to cyclopentyl‐bearing spiroindolin‐2‐ones and spiroindolin‐3‐ones. The method demonstrates excellent chemoselectivity, regioselectivity, and product control simply by using the H2O and O2 as oxygen source, respectively. This open‐flask method is safe and simple to operate, and no other chemical oxidants are required. Besides, inspired from the unique pathway of 1, 2‐migration rearrangement, a highly controllable hydroxylation of indoles for the construction of C3a‐hydroxyl iminium indolines was also developed. Mechanistic experiments suggest that a single‐electron transfer‐induced oxidation process is responsible for the tunable selectivity control.

Abstract:AbstractA novel radical [1,3]‐nitrogen shift catalyzed by copper diacetate under an oxygen atmosphere (1 atm) has been developed for the construction of a diverse range of indole derivatives from α,α‐disubstituted benzylamine. In this reaction, oxygen was used as a clean terminal oxidant, and water was produced as the only by‐product. Five inert bonds were cleaved, and two C−N bonds and one C−C double bond were constructed in one pot during this transformation. This unique method demonstrated broad application protential for the late‐stage modification of biologically active natural products and drugs. Mechanistic investigations indicate that a unique 4‐exo‐trig cyclization of an aminyl radical onto a phenyl ring is involved in the catalytic cycle.
