Abstract:Here we report the examination of two convenient strategies, the use of a d-amino acid residue or a glycoside segment, for increasing the proteolytic resistance of supramolecular hydrogelators based on small peptides. Our results show that the introduction of d-amino acid or glycoside to the peptides significantly increases the resistance of the hydrogelators against proteinase K, a powerful endopeptidase. The insertion of d-amino acid in the peptide backbone, however, results relatively low storage moduli of the hydrogels, likely due to the disruption of the superstructures of the molecular assembly. In contrast, the introduction of a glycoside to the C-terminal of peptide enhances the biostability of the hydrogelators without the significant decrease of the storage moduli of the hydrogels. This work suggests that the inclusion of a simple glycogen in hydrogelators is a useful approach to increase their biostability, and the gained understanding from the work may ultimately lead to development of hydrogels of functional peptides for biomedical applications that require long-term biostability.

Abstract:Peptide-based supramolecular hydrogels have been extensively explored in biomaterials owing to their unique bioactive, stimulus-responsive, and biocompatible features. However, peptide-based hydrogels often have low mechanical stability with storage moduli of 10-1000 Pa. They are susceptible to mechanical destruction and solvent erosion, greatly hindering their practical application. Here, we present a photo-cross-linking strategy to enhance the mechanical stability of a peptide-based hydrogel by 10(4)-fold with a storage modulus of ~100 kPa, which is one of the highest reported so far for hydrogels made of small peptide molecules. This method is based on the ruthenium-complex-catalyzed conversion of tyrosine to dityrosine upon light irradiation. The reinforcement of the hydrogel through photo-cross-linking can be achieved within 2 min thanks to the fast reaction kinetics. The enhancement of the mechanical stability was due to the formation of a densely entangled fibrous network of peptide dimers through a dityrosine linkage. We showed that in order to implement this method successfully, the peptide sequence should be rationally designed to avoid the cross talk between self-assembly and cross-linking. This method is convenient and versatile for the enhancement of the mechanical stability of tyrosine-containing peptide-based hydrogels. We anticipate that the photo-cross-linked supramolecular hydrogels with much improved mechanical stability will find broad applications in tissue engineering and drug controlled release.

Abstract:The application of a peptide-based supramolecular hydrogel in the field of bioactive materials is limited by its poor stability in extreme environments. Increasing the number of hydrophobic amino acids in the peptides may enhance the stability of the hydrogel, but an organic solvent is always required to dissolve peptides because of its strong hydrophobic properties. In this study, we linked hydrophilic amino acids to super hydrophobic peptides using a self-hydrolyzed ester bond as a linker to enhance its solubility in water, which resulted in two highly potent peptide hydrogels. The hydrogels showed excellent stability in highly acidic, highly basic and saturated salt solutions for 15 days. This system may be particularly useful in designing a robust drug release system that can maintain a constant release rate for days and against abrupt changes in the environment.

Abstract:This article reports the synthesis of a new class of conjugates containing a nucleobase, a peptidic epitope, and a saccharide and the evalution of their gelation, biostability, and cell compatibility. We demonstrate a facile synthetic process, based on solid-phase peptide synthesis of nucleopeptides, to connect a saccharide with the nucleopeptides for producing the target conjugates. All the conjugates themselves (1–8) display excellent solubility in water without forming hydrogels. However, a mixture of 5 and 8 self-assembles to form nanofibers and results in a supramolecular hydrogel. The proteolytic stabilities of the conjugates depend on the functional peptidic epitopes. We found that TTPV is proteolytic resistant and LGFNI is susceptible to proteolysis. In addition, all the conjugates are compatible to the mammalian cells tested.

Abstract:The development of small molecules that can efficiently gel water is of great interest for researchers in the field of self-assembly. We recently found that a short peptide-based molecule (Nap-GFFpY-OMe) could form hydrogels at a minimum gelation concentration of 0.01 wt% after enzymatic conversion, which was the most efficient small molecular hydrogelator reported up to now. In order to study the relationship between the chemical structure and the gelation ability for these short peptide-based gelators, we designed and synthesized a small library of compounds with similar chemical structures to Nap-GFFpY-OMe. They have different capping groups for the amine on glycine (G), different numbers of glycines, different numbers of phenylalanines (F), and different groups at the C-terminus of the peptides. Based on the results, we concluded that the compounds were 'super gelators' when their amine capping group was naphthalene, the number of G residues was odd (one and three), the number of F residues was two, and the C-terminus of the peptide was a methyl ester. We found that 10 compounds within the library could form hydrogels and the hydrogels were characterized by rheology, transmission electron microscopy (TEM), and fluorescence spectroscopy. The cytotoxicity of all the precursors of the gelators were also evaluated by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay and we found that three precursors possessed obviously bigger IC50 values than their corresponding minimum gelation concentrations (MGCs). This study not only suggests the potential applications of our gel system in biomedical fields, but also provides useful information for the molecular design of short peptide-based hydrogelators with excellent gelation abilities.

Abstract:In addition to the widely used polymeric hydrogels, molecular hydrogels are also emerging as promising biomaterials. However, molecular hydrogels typically suffer from poor mechanical properties and relatively low in vivo stability. Here we report on the preparation of two hybrid hydrogels (Hgel I and Hgel II) by the combination of two molecular hydrogelators and alginate, respectively. First, the molecular hydrogelator and sodium alginate were dissolved in water, and then a hydrogel was formed upon triggering by enzymes or reductants. Afterward the resulting hydrogel was soaked in calcium ion solution to form the hybrid hydrogel. The preparation process was easy and mild which allowed the resulting hybrid hydrogels to act as carriers for biomacromolecules such as enzymes. Both Hgel I and Hgel II had better stabilities and mechanical properties than the calcium alginate gel (CAgel) alone. Hgel I had exceptional stability compared with Hgel II and CAgel. The reason we propose is that molecular hydrogelators in Hgel I interacted with alginate more strongly than molecular hydrogelators in Hgel II according to the fluorescence results of these gels. Unlike the porous microstructure of molecular hydrogels, the microstructure of two hybrid hydrogels was almost the same as that of CAgel (film-like morphology) except with some nanofibers embedded. We found that Hgel I was an ideal carrier for enzyme immobilization with high recyclable properties and excellent preservation of enzyme activities due to its good mechanical strength. However, Hgel II was not suitable as a carrier for immobilized lactase probably due to poor affinity between Hgel II and the substrate. Combining the advantages of molecular hydrogels and polymeric hydrogels would broaden the applications of both kinds of hydrogels in the future.

Abstract:Dipeptide self-assembled hydrogels have potential biomedical applications because of their great biocompatibility, bioactivity, and tunable physicochemical properties, which can be modulated in the molecular level by design of amino acid sequences. Herein, a series of dipeptides (Fmoc-FL, -YL, -LL, and -YA) are designed to form shear-thinning hydrogels with self-healing and tunable mechanical properties by adjusting the synergetic effect of hydrophobic interactions (π-π stacking and hydrophobic effect) and hydrogen bonds of peptides through substitution of amino acid residues. The enhancement of hydrophobic interactions is a primary factor to promote mechanical rigidity of hydrogels, and strong hydrogen-bonding interactions between molecules contribute to the instantaneous self-healing property, which is supported by experimental studies (FTIR, CD, SEM, AFM, and rheology) and molecular dynamics simulations. The injectable dipeptide hydrogels were certified as an ideal endoscopic submucosal dissection filler to make operation convenient and secure in mice and living mini-pig's experiments with a longer duration time, higher stiffness, and lower inflammatory response than commercial clinical fillers.

Abstract:Here we report a new class of molecular hydrogelators based on the conjugates of dipeptide and (naphthalen-2-yloxy)acetic acid. They form hydrogels efficiently the lowest concentration for them to gel water is 0.07 wt%. Two hydrogelators show the ability to form helical nanofibers within the hydrogels, and the chirality of the hydrogelators apparently dictates the handedness of the nanofibers-D-peptide and L-peptide afford left-handed and right-handed self-assembled nanofibers in the hydrogels, respectively. Moreover, MTT assay of the viability of cells indicates that these molecular hydrogelators are biocompatible.

Abstract:Three new hydrogelators based on the conjugates of three naturally occurring biological building blocks: nucleobase, saccharide, and amino acids, were explored. Being synthesized via a facile solid phase peptide synthesis route, the hydrogelators self-assemble in water to afford supramolecular nanofibers and hydrogels. Transmission electron microscopy, oscillation rheometry, and circular dichroism reveal that the hydrogels consist of largely helix-based nanofibers of the widths of 5-12 nm and exhibit storage moduli up to 1 kPa. These hydrogelators also exhibit excellent cell-compatibility. This work illustrates a new platform for constructing molecular soft materials for nanobiotechnology.

Abstract:Molecular hydrogels hold big potential for tissue engineering and controlled drug delivery. Our lab focuses on short-peptide-based molecular hydrogels formed by biocompatible methods and their applications in tissue engineering (especially, 3D cell culture) and controlled drug delivery. This feature article firstly describes our recent progresses of the development of novel methods to form hydrogels, including the strategy of disulfide bond reduction and assistance with specific protein-peptide interactions. We then introduce the applications of our hydrogels in fields of controlled stem cell differentiation, cell culture, surface modifications of polyester materials by molecular self-assembly, and anti-degradation of recombinant complex proteins. A novel molecular hydrogel system of hydrophobic compounds that are only formed by hydrolysis processes was also included in this article. The hydrogels of hydrophobic compounds, especially those of hydrophobic therapeutic agents, may be developed into a carrier-free delivery system for long term delivery of therapeutic agents. With the efforts in this field, we believe that molecular hydrogels formed by short peptides and hydrophobic therapeutic agents can be practically applied for 3D cell culture and long term drug delivery in near future, respectively.

Abstract:In this work, a novel starch-based zwitterionic copolymer, starch-graft-poly(sulfobetaine methacrylate) (ST-g-PSBMA), was synthesized via Atom Transfer Radical Polymerization. Starch, which formed the main chain, can be degraded completely in vivo, and the pendent segments of PSBMA endowed the copolymer with excellent protein resistance properties. This ST-g-PSBMA copolymer could self-assemble into a physical hydrogel in normal saline, and studies of the formation mechanism indicated that the generation of the physical hydrogel was driven by electrostatic interactions between PSBMA segments. The obtained hydrogels were subjected to detailed analysis by scanning electron microscopy, swelling ratio, protein resistance, and rheology tests. Toxicity and hemolysis analysis demonstrated that the ST-g-PSBMA hydrogels possess excellent biocompatibility and hemocompatibility. Moreover, the cytokine secretion assays (IL-6, TNF-α, and NO) confirmed that ST-g-PSBMA hydrogels had low potential to trigger the activation of macrophages and were suitable for in vivo biomedical applications. On the basis of these in vitro results, the ST-g-PSBMA hydrogels were implanted in SD rats. The tissue responses to hydrogel implantation and the hydrogel degradation in vivo were determined by histological analysis (Hematoxylin and eosin, Van Gieson, and Masson's Trichrome stains). The results presented in this study demonstrate that the physical cross-linking, starch-based zwitterionic hydrogels possess excellent protein resistance, low macrophage-activation properties, and good biocompatibility, and they are a promising candidate for an in vivo biomedical application platform.

Abstract:As a new class of biomaterials, most supramolecular hydrogels formed by small peptides require the attachment of long alkyl chains, multiple aromatic groups, or strong electrostatic interactions. Based on the fact that the most abundant protein assemblies in nature are dimeric, we select short peptide sequences from the interface of a heterodimer of proteins with known crystal structure to conjugate with nucleobases to form nucleopeptides. Being driven mainly by hydrogen bonds, the nucleopeptides self-assemble to form nanofibers, which results in supramolecular hydrogels upon simple mixing of two distinct nucleopeptides in water. Moreover, besides being biocompatible to mammalian cells, the heterodimer of the nucleopeptides exhibit excellent proteolytic resistance against proteinase K. This work illustrates a new and rational approach to create soft biomaterials by a supramolecular hydrogelation triggered by mixing heterodimeric nucleopeptides.

Abstract:Peptide-based hydrogels have gained considerable attention as a compelling platform for various biomedical applications in recent years. Their attractiveness stems from their ability to seamlessly integrate diverse properties, such as biocompatibility, biodegradability, easily adjustable hydrophilicity/hydrophobicity, and other functionalities. However, a significant drawback is that most of the functional self-assembling peptides cannot form robust hydrogels suitable for biological applications. In this study, we present the synthesis of novel peptide-PEG conjugates and explore their comprehensive hydrogel properties. The hydrogel comprises double networks, with the first network formed through the self-assembly of peptides to create a β-sheet secondary structure. The second network is established through covalent bond formation via N-hydroxysuccinimide chemistry between peptides and a 4-arm PEG to form a covalently linked network. Importantly, our findings reveal that this hydrogel formation method can be applied to other peptides containing lysine-rich sequences. Upon encapsulation of the hydrogel with antimicrobial peptides, the hydrogel retained high bacterial killing efficiency while showing minimum cytotoxicity toward mammalian cells. We hope that this method opens new avenues for the development of a novel class of peptide-polymer hydrogel materials with enhanced performance in biomedical contexts, particularly in reducing the potential for infection in applications of tissue regeneration and drug delivery.

Abstract:Here we report the generation of a novel class of supramolecular hydrogelators based on the integration of nucleobase, Arg-Gly-Asp (RGD) peptides, and glucosamine in a single molecule. These novel small molecule hydrogelators self-assemble in water to form stable supramolecular nanofibers/hydrogels and exhibit useful biostability. This approach provides a new opportunity for systematic exploration of the self-assembly of small biomolecules by varying any individual segment to generate a large array of supramolecular hydrogels for biological functions and for biomedical applications.
