Abstract:A polypeptide-DNA hydrogel is prepared by Z. Li, D. Liu, and co-workers, employing the “X”-shaped DNA assembling structure as crosslinker. The hydrogel can be modified with multifunctional components (here fluorescent molecules as a model), and possess excellent self-healing and thixotropic properties, enabling the direct-writing of arbitrary 3D structures. The study on page 1138 provides a simple, universal strategy for the assembly of functionalized hydrogels with biomolecules, especially enzymes.

Abstract:Dipeptide self-assembled hydrogels have potential biomedical applications because of their great biocompatibility, bioactivity, and tunable physicochemical properties, which can be modulated in the molecular level by design of amino acid sequences. Herein, a series of dipeptides (Fmoc-FL, -YL, -LL, and -YA) are designed to form shear-thinning hydrogels with self-healing and tunable mechanical properties by adjusting the synergetic effect of hydrophobic interactions (π-π stacking and hydrophobic effect) and hydrogen bonds of peptides through substitution of amino acid residues. The enhancement of hydrophobic interactions is a primary factor to promote mechanical rigidity of hydrogels, and strong hydrogen-bonding interactions between molecules contribute to the instantaneous self-healing property, which is supported by experimental studies (FTIR, CD, SEM, AFM, and rheology) and molecular dynamics simulations. The injectable dipeptide hydrogels were certified as an ideal endoscopic submucosal dissection filler to make operation convenient and secure in mice and living mini-pig's experiments with a longer duration time, higher stiffness, and lower inflammatory response than commercial clinical fillers.

Abstract:A precursor of a β-peptide derivative transforms into a hydrogelator and forms supramolecular hydrogels upon phosphatase-catalyzed dephosphorylation. This new strategy works in vitro and in vivo (see image). The enzymatic hydrogelation in vivo indicates that the β-peptide-based hydrogel exhibits a longer lifetime than that of α-peptide hydrogel in a subcutaneous environment. Such an excellent in vivo stability renders the β-peptide-based hydrogel as a potential candidate for biomedical applications.

Abstract:Here we report the examination of two convenient strategies, the use of a d-amino acid residue or a glycoside segment, for increasing the proteolytic resistance of supramolecular hydrogelators based on small peptides. Our results show that the introduction of d-amino acid or glycoside to the peptides significantly increases the resistance of the hydrogelators against proteinase K, a powerful endopeptidase. The insertion of d-amino acid in the peptide backbone, however, results relatively low storage moduli of the hydrogels, likely due to the disruption of the superstructures of the molecular assembly. In contrast, the introduction of a glycoside to the C-terminal of peptide enhances the biostability of the hydrogelators without the significant decrease of the storage moduli of the hydrogels. This work suggests that the inclusion of a simple glycogen in hydrogelators is a useful approach to increase their biostability, and the gained understanding from the work may ultimately lead to development of hydrogels of functional peptides for biomedical applications that require long-term biostability.

Abstract:Peptide-based supramolecular hydrogels have attracted great attention due to their good biocompatibility and biodegradability and have become promising candidates for biomedical applications. The bottom-up self-assembly endows the peptides with a highly ordered secondary structure, which has proven to be an effective strategy to improve the mechanical properties of hydrogels through strong physical interactions and energy dissipation. Inspired by the excellent mechanical properties of spider-silk, which can be attributed to the rich β-sheet crystal formation by the hydrophobic peptide fragment, a hydrophobic peptide (HP) that can form a β-sheet assembly was designed and introduced into a poly(vinyl alcohol) (PVA) scaffold to improve mechanical properties of hydrogels by the cooperative intermolecular physical interactions. Compared with hydrogels without peptide grafting (P-HP0), the strong β-sheet self-assembly domain endows the hybrid hydrogels (P-HP20, P-HP29, and P-HP37) with high strength and toughness. The fracture tensile strength increased from 0.3 to 2.1 MPa (7 times), the toughness increased from 0.4 to 21.6 MJ m–3 (54 times), and the compressive strength increased from 0.33 to 10.43 MPa (31 times) at 75% strain. Moreover, the hybrid hydrogels are enzymatically degradable due to the dominant contribution of the β-sheet assembly for network cross-linking. Combining the good biocompatibility and sustained drug release of the constructed hydrogels, this hydrophobic β-sheet peptide represents a promising candidate for the rational design of hydrogels for biomedical applications.

Abstract:The application of a peptide-based supramolecular hydrogel in the field of bioactive materials is limited by its poor stability in extreme environments. Increasing the number of hydrophobic amino acids in the peptides may enhance the stability of the hydrogel, but an organic solvent is always required to dissolve peptides because of its strong hydrophobic properties. In this study, we linked hydrophilic amino acids to super hydrophobic peptides using a self-hydrolyzed ester bond as a linker to enhance its solubility in water, which resulted in two highly potent peptide hydrogels. The hydrogels showed excellent stability in highly acidic, highly basic and saturated salt solutions for 15 days. This system may be particularly useful in designing a robust drug release system that can maintain a constant release rate for days and against abrupt changes in the environment.

Abstract:Supramolecular hydrogels hold great promise for controlled drug delivery. Herein we report a supramolecular hydrogel based on a peptide bearing a terminal aldehyde. The hydrogel was prepared via an enzyme instructed self-assembly (EISA) process, and the resulting hydrogels showed ultra-stable properties in highly acidic or basic aqueous solutions. The hydrogelator could form Schiff bases with amine drugs. Owing to the pH-responsive properties of Schiff bases, the hydrogels could be used for controlled release of encapsulated amine drugs. Our study provides a peptide-based hydrogel that may be applied for controlled drug delivery.

Abstract:A supramolecular hydrogel based on D-amino acids, which resists hydrolysis catalyzed by proteinase K and offers long-term biostability, exhibits controlled release in vivo, as proved by the pharmacokinetics of encapsulated 125I tracers and the SPECT imaging of the hydrogel-encapsulated 131I tracers. As the first in vivo imaging investigation of the drug release properties of the supramolecular hydrogel, isotope encapsulation serves as a valid, useful assay for characterizing the controlled release properties of supramolecular hydrogels in vivo. Our results indicate that supramolecular hydrogels promise new biomaterials for controlled drug release.

Abstract:Peptide-based hydrogels (PHGs) are biocompatible materials suitable for biological, biomedical, and biotechnological applications, such as drug delivery and diagnostic tools for imaging. Recently, a novel class of synthetic hydrogel-forming amphiphilic cationic peptides (referred to as series K), containing an aliphatic region and a Lys residue, was proposed as a scaffold for bioprinting applications. Here, we report the synthesis of six analogues of the series K, in which the acetyl group at the N-terminus is replaced by aromatic portions, such as the Fmoc protecting group or the Fmoc-FF hydrogelator. The tendency of all peptides to self-assemble and to gel in aqueous solution was investigated using a set of biophysical techniques. The structural characterization pointed out that only the Fmoc-derivatives of series K keep their capability to gel. Among them, Fmoc-K3 hydrogel, which is the more rigid one (G’ = 2526 Pa), acts as potential material for tissue engineering, fully supporting cell adhesion, survival, and duplication. These results describe a gelification process, allowed only by the correct balancing among aggregation forces within the peptide sequences (e.g., van der Waals, hydrogen bonding, and π–π stacking).

Abstract:Zwitterionic polymer materials have been extensively studied, but zwitterionic peptides supramolecular hydrogel materials are rarely studied. In this study, the preparation of two zwitterionic hydrogels using self-assembled peptides were reported. The hydrogels could be fabricated easily by changing the temperature or enzyme catalysis in a short time. And the differences in structure and function of the zwitterion peptide hydrogels caused by the two preparation methods were also be compared. We found that the hydrogel prepared by enzyme induced self-assembly has better solubility and lower cytotoxicity than that prepared by the heating-cooling process. The result showed the enzyme induced self-assembly way to form zwitterionic peptides supramolecular hydrogel materials could have further biomedical applications.
Photo-cross-linking Approach to Engineering Small Tyrosine-Containing Peptide Hydrogels with Enhanced Mechanical Stability.

Abstract:Peptide-based supramolecular hydrogels have been extensively explored in biomaterials owing to their unique bioactive, stimulus-responsive, and biocompatible features. However, peptide-based hydrogels often have low mechanical stability with storage moduli of 10-1000 Pa. They are susceptible to mechanical destruction and solvent erosion, greatly hindering their practical application. Here, we present a photo-cross-linking strategy to enhance the mechanical stability of a peptide-based hydrogel by 10(4)-fold with a storage modulus of ~100 kPa, which is one of the highest reported so far for hydrogels made of small peptide molecules. This method is based on the ruthenium-complex-catalyzed conversion of tyrosine to dityrosine upon light irradiation. The reinforcement of the hydrogel through photo-cross-linking can be achieved within 2 min thanks to the fast reaction kinetics. The enhancement of the mechanical stability was due to the formation of a densely entangled fibrous network of peptide dimers through a dityrosine linkage. We showed that in order to implement this method successfully, the peptide sequence should be rationally designed to avoid the cross talk between self-assembly and cross-linking. This method is convenient and versatile for the enhancement of the mechanical stability of tyrosine-containing peptide-based hydrogels. We anticipate that the photo-cross-linked supramolecular hydrogels with much improved mechanical stability will find broad applications in tissue engineering and drug controlled release.

Abstract:The combination of polymers and low molecular weight (LMW) compounds is a powerful approach to prepare new supramolecular materials. Here we prepare two-component hydrogels made by a well-known and biologically active polymer, hyaluronic acid (HA), and a dipeptide-based supramolecular gelator. We undertake a detailed study of materials with different compositions including macroscopic (hydrogel formation, rheology) and micro/nanoscopic characterization (electron microscopy, X-ray powder diffraction). We observe that the two components mutually benefit in the new materials: a minimum amount of HA helps to reduce the polymorphism of the LMW network leading to reproducible hydrogels with improved mechanical properties; the LMW component network holds HA without the need for an irreversible covalent crosslinking. These materials have a great potential for biomedical application as, for instance, extracellular matrix mimetics for cell growth. As a proof of concept, we have observed that this material is effective for cell growth in suspension and avoids cell sedimentation even in the presence of competing cell-adhesive surfaces. This may be of interest to advanced cell delivery techniques.
New Supramolecular Hydrogels Based on Diastereomeric Dehydrotripeptide Mixtures for Potential Drug Delivery Applications

Abstract:Self-assembly of peptide building blocks offers unique opportunities for bottom-up preparation of exquisite nanostructures, nanoarchitectures, and nanostructured bulk materials, namely hydrogels. In this work we describe the synthesis, characterization, gelation, and rheological properties of new dehydrotripeptides, Cbz-L-Lys(Cbz)-L,D-Asp-∆Phe-OH and (2-Naph)-L-Lys(2-Naph)-L,D-Asp-∆Phe-OH, containing a N-terminal lysine residue Nα,ε-bis-capped with carboxybenzyl (Cbz) and 2-Naphthylacetyl (2-Naph) aromatic moieties, an aspartic acid residue (Asp), and a C-terminal dehydrophenylalanine (∆Phe) residue. The dehydrotripeptides were obtained as diastereomeric mixtures (L,L,Z and L,D,Z), presumably via aspartimide chemistry. The dehydrotripeptides afforded hydrogels at exceedingly low concentrations (0.1 and 0.04 wt%). The hydrogels revealed exceptional elasticity (G' = 5.44 × 104 and 3.43 × 106 Pa) and self-healing properties. STEM studies showed that the diastereomers of the Cbz-capped peptide undergo co-assembly, generating a fibrillar 3D network, while the diastereomers of the 2-Naph-capped dehydropeptide seem to undergo self-sorting, originating a fibril network with embedded spheroidal nanostructures. The 2-Naph-capped hydrogel displayed full fast recovery following breakup by a mechanical stimulus. Spheroidal nanostructures are absent in the recovered hydrogel, as seen by STEM, suggesting that the mechanical stimulus triggers rearrangement of the spheroidal nanostructures into fibers. Overall, this study demonstrates that diastereomeric mixtures of peptides can be efficacious gelators. Importantly, these results suggest that the structure (size, aromaticity) of the capping group can have a directing effect on the self-assembly (co-assembly vs. self-sorting) of diastereomers. The cytotoxicity of the newly synthesized gelators was evaluated using human keratinocytes (HaCaT cell line). The results indicated that the two gelators exhibited some cytotoxicity, having a small impact on cell viability. In sustained release experiments, the influence of the charge on model drug compounds was assessed in relation to their release rate from the hydrogel matrix. The hydrogels demonstrated sustained release for methyl orange (anionic), while methylene blue (cationic) was retained within the network.

Abstract:In recent years, supramolecular hydrogels have made groundbreaking research progress in biomedical fields such as drug delivery, biosensing, imaging analysis, and tissue engineering. Peptides, with their unique characteristics of facile preparation, low immunogenicity and easy biodegradability, are commonly used as building blocks of supramolecular hydrogels. Peptide-based supramolecular hydrogels loaded with drugs, prepared via physical means or covalent crosslinking, exhibit unique three-dimensional network structures and strong water retention capacities. These properties enhance drug bioavailability and reduce side effects, enabling drug accumulation and responsive release at disease sites, significantly improving the therapeutic efficacy. Here, we review recent advancements in peptide-based supramolecular hydrogels and their biotherapeutic applications, including chemotherapy, photothermal therapy, photodynamic therapy, immunotherapy, gene therapy, antibacterial and anti-inflammatory treatments, and other biological applications. This review aims to provide new inspiration for the development of biomaterials in the therapeutic field and provide more personalized options for disease treatment. Additionally, challenges and limitations in this field are briefly discussed.

Abstract:Here we report the generation of a novel class of supramolecular hydrogelators based on the integration of nucleobase, Arg-Gly-Asp (RGD) peptides, and glucosamine in a single molecule. These novel small molecule hydrogelators self-assemble in water to form stable supramolecular nanofibers/hydrogels and exhibit useful biostability. This approach provides a new opportunity for systematic exploration of the self-assembly of small biomolecules by varying any individual segment to generate a large array of supramolecular hydrogels for biological functions and for biomedical applications.

Abstract:Self-assembly peptide-based hydrogels are well known and popular in biomedical applications due to the fact that they are readily controllable and have biocompatibility properties. A dipeptide is the shortest self-assembling motif of peptides. Due to its small size and simple synthesis method, dipeptide can provide a simple and easy-to-use method to study the mechanism of peptides’ self-assembly. This review describes the design and structures of self-assembly linear dipeptide hydrogels. The strategies for preparing the new generation of linear dipeptide hydrogels can be divided into three categories based on the modification site of dipeptide: 1) COOH-terminal and N-terminal modified dipeptide, 2) C-terminal modified dipeptide, and 3) uncapped dipeptide. With a deeper understanding of the relationship between the structures and properties of dipeptides, we believe that dipeptide hydrogels have great potential application in preparing minimal biocompatible materials.
